Arachis villosulicarpa is a perennial species cultivated for its soft and tasty seeds by indigenous inhabitants of the Mate Grosso State, Brazil. Besides A. hypogaea, this species is considered as the only species of Arachis which represents a valuable food source for human consumption. Due to the lack of knowledge concerning the genetic diversity of A. villosulicarpa, this study was conducted to evaluate the genetic variability of the accessions from the Germplasm Collection of CENARGEN/EMBRAPA (Brasilia, DF, Brazil) and Institute Agronomico (IAC, Campinas, SP, Brazil). In addition, the genetic similarity between A. villosulicarpa, the related wild species A. pietrarellii, and the cultivated peanut A. hypogaea cv. Tatu was evaluated. From the entire sample analyzed of A. villosulicarpa, the accession from Institute Agronomico showed the highest indices of diversity for both enzymatic systems analyzed, pointing this accession as a promising source of genetic variability that must be preserved in the Germplasm Bank. A high level of genetic similarity was observed between A. pietrarellii and A. villosulicarpa, supporting previous suggestions that A. pietrarellii could be the ancestral progenitor species of A. villosulicarpa or that both species originated from a common ancestor.

Three populations of Scythrophrys from Brazil were studied cytogenetically. The karyotype found in Piraquara (Parana State) and Sao Bento do Sul (Santa Catarina State) (karyotype 1) differed from that of Rancho Queimado (Santa Catarina State) (karyotype 11) in NOR location and in the occurrence of some C-bands. In karyotype I, pair 5 contained NOR-bearing chromosomes, while in karyotype 11 the NOR occurred in pair 10. This difference probably accounts for the inverted classification of pairs 5 and 6 in these karyotypes, which appear to be homologous, as suggested by the presence of heterochromatic markers. These karyotypes also differed in the size of the non-centromeric C-bands in pairs 1 and 8. Comparison of the Scythrophrys karyotypes with those of the related genus Paratelmatobius revealed several similarities, especially between karyotype 11 of Scythrophrys and that of P. poecilogaster. Both karyotypes had a NOR in pair 10, so it was inferred to be present also in the common ancestor for these genera. The non-centromeric C-bands in pairs 7, 10, and in pair 5 of karyotype I and pair 6 of karyotype II of Scythrophrys are characters exclusively found in this genus.

The occurrence, number of insertion sites and antisense RNA expression of micropia transposable element were studied in 26 species that belong to three subgroups (mercatorum, mulleri and hydei) of repleta group of Drosophila. Under high specific PCR, micropia sequences were detected in 11 species, but under less stringent condition, this retrotransposon was detected in all species. The widespread distribution of micropia suggests that this element was already present at the common ancestor of the repleta group of Drosophila. Southern blot analysis showed a variation from 0 to 17 different insertion sites and the occurrence of male-specific sequences. We found that the expression of the 1.0 kb micropia antisense RNA is variable among the species and tissues (soma and testis), which suggests that more than one mechanism regulates transposition in these species. Variation of amplification by PCR and of antisense RNA expression, as well as divergence of nucleotide sequences among the species allow us to suggest that at least two subfamilies of micropia transposable element are harbored by the genome of this species group.

Eutherian mammals share a common ancestor that evolved into two main placental types, i.e., hemotrophic (e.g., human and mouse) and histiotrophic (e.g., farm animals), which differ in invasiveness. Pregnancies initiated with assisted reproductive techniques (ART) in farm animals are at increased risk of failure; these losses were associated with placental defects, perhaps due to altered gene expression. Developmentally regulated genes in the placenta seem highly phylogenetically conserved, whereas those expressed later in pregnancy are more species-specific. To elucidate differences between hemotrophic and epitheliochorial placentae, gene expression data were compiled from microarray studies of bovine placental tissues at various stages of pregnancy. Moreover, an in silico subtractive library was constructed based on homology of bovine genes to the database of zebrafish - a nonplacental vertebrate. In addition, the list of placental preferentially expressed genes for the human and mouse were collected using bioinformatics tools (Tissue-specific Gene Expression and Regulation [TiGER] - for humans, and tissue-specific genes database (TiSGeD) - for mice and humans). Humans, mice, and cattle shared 93 genes expressed in their placentae. Most of these were related to immune function (based on analysis of gene ontology). Cattle and women shared expression of 23 genes, mostly related to hormonal activity, whereas mice and women shared 16 genes (primarily sexual differentiation and glycoprotein biology). Because the number of genes expressed by the placentae of both cattle and mice were similar (based on cluster analysis), we concluded that both cattle and mice were suitable models to study the biology of the human placenta. The cocoon, produced by most holometabolous insects, is built with silk that is usually produced by the larval salivary gland. Although this silk has been widely studied in the Lepidoptera, its composition and macromolecular arrangement remains unknown in the Hymenoptera. The macromolecular array patterns of the silk in the larval salivary gland of some meliponids, wasps, and ants were analyzed with polarized-light microscopy, and they were compared with those of Bombyx mori (Lepidoptera). There is a birefringent secretion in the glandular lumen of all larvae, due to filamentous structural proteins that display anisotropy. The silk in the distal, middle and proximal regions of the secretory portion of Formicidae and Vespidae glands presented a lattice optical pattern. We found a different pattern in the middle secretory portion of the Meliponini, with a zigzag rather than a lattice pattern. This indicates that the biopolymer fibers begin their macromolecular reorganization at this glandular region, different from the Formicidae and the Vespidae, in which the zigzag optical pattern was only found at the lateral duct. Probably, the mechanism of silk production in the Hymenoptera is a characteristic inherited from a common ancestor of Vespoidea and Sphecoidea; the alterations in the pattern observed in the Meliponini could be a derived characteristic in the Hymenoptera. We found no similarity in the macromolecular reorganization patterns of the silk between the Hymenoptera species and the silkworm.

The known diversity of dart-poison frog species has grown from 70 in the 1960s to 247 at present, with no sign that the discovery of new species will wane in the foreseeable future. Although this growth in knowledge of the diversity of this group has been accompanied by detailed investigations of many aspects of the biology of dendrobatids, their phylogenetic relationships remain poorly understood. This study was designed to test hypotheses of dendrobatid diversification by combining new and prior genotypic and phenotypic evidence in a total evidence analysis. DNA sequences were sampled for five mitochondrial and six nuclear loci (approximately 6,100 base pairs [bp]; x=3,740 bp per terminal; total dataset composed of approximately 1.55 million bp), and 174 phenotypic characters were scored from adult and larval morphology, alkaloid profiles, and behavior. These data were combined with relevant published DNA sequences. Ingroup sampling targeted several previously unsampled species, including Aromobates nocturnus, which was hypothesized previously to be the sister of all other dendrobatids. Undescribed and problematic species were sampled from multiple localities when possible. The final dataset consisted of 414 terminals: 367 ingroup terminals of 156 species and 47 outgroup terminals of 46 species. Direct optimization parsimony analysis of the equally weighted evidence resulted in 25,872 optimal trees. Forty nodes collapse in the strict consensus, with all conflict restricted to conspecific terminals. Dendrobatids were recovered as monophyletic, and their sister group consisted of Crossodactylus, Hylodes, and Megaelosia, recognized herein as Hylodidae. Among outgroup taxa, Centrolenidae was found to be the sister group of all athesphatanurans except Hylidae, Leptodactyidae was polyphyletic, Thoropa was nested within Cycloramphidae, and Ceratophryinae was paraphyletic with respect to Telmatobiinae. Among dendrobatids, the monophyly and content of Mannophryne and Phyllobates were corroborated. Aromobates nocturnus and Colostethus saltuensis were found to be nested within Nephelobates, and Minyobates was paraphyletic and nested within Dendrobates. Colostethus was shown to be rampantly nonmonophyletic, with most species falling into two unrelated cis- and trans-Andean clades. A morphologically and behaviorally diverse clade of median lingual process-possessing species was discovered. In light of these findings and the growth in knowledge of the diversity of this large clade over the past 40 years, we propose a new, monophyletic taxonomy for dendrobatids, recognizing the inclusive clade as a superfamily (Dendrobatoidea) composed of two families (one of which is new), six subfamilies (three new), and 16 genera (four new). Although poisonous frogs did not form a monophyletic group, the three poisonous lineages are all confined to the revised family Dendrobatidae, in keeping with the traditional application of this name. We also propose changes to achieve a monophyletic higher-level taxonomy for the athesphatanuran outgroup taxa. Analysis of character evolution revealed multiple origins of phytotelm-breeding, parental provisioning of nutritive oocytes for larval consumption (larval oophagy), and endotrophy. Available evidence indicates that transport of tadpoles on the dorsum of parent nurse frogs-a dendrobatid synapomorphy-is carried out primitively by male nurse frogs, with three independent origins of female transport and five independent origins of biparental transport. Reproductive amplexus is optimally explained as having been lost in the most recent common ancestor of Dendrobatoidea, with cephalic amplexus arising independently three times. © American Museum of Natural History 2006.

We present a historical perspective about the ideas concerning the origin of life. After displaying the main topics necessary for understanding life's origin, the main characteristics of the present life forms and their relationships are shown, suggesting a common ancestor. The conditions for prebiotic chemistry in terrestrial or interplanetary environments are reviewed. We put in context the arguments about the early origin of replicators versus metabolism. The very narrow window for life settlement in the early Earth is stressed, pointing to the likelihood of life arising in other places in the Universe. Finally, we present the cornerstones of current search for life outside our planet.

A febre do dengue é uma das mais importantes arboviroses distribuída por todas as áreas tropicais do mundo. O vírus dengue (VDEN) é transmitido principalmente pela picada do mosquito Aedes aegypti infectado. A dispersão do vetor e o aumento do fluxo migratório entre países possibilitaram a ocorrência de grandes epidemias e manifestações clínicas severas, como febre hemorrágica do dengue (FHD) e Síndrome do choque do dengue (SCD). O objetivo deste trabalho foi realizar a caracterização molecular de isolados do VDEN sorotipo 1 (VDEN-1) no Brasil ao nível dos genes estruturais C/prM/M/E de 29 cepas isoladas durante epidemias ocorridas no Brasil no período de 1994 a 2008. A identidade nucleotídica entre as cepas de VDEN-1 do estudo em relação às outras isoladas no Brasil variou de 96,1% a 100%, enquanto o percentual de identidade de aminoácidos foi determinado entre 98,4% a 100%. As diferenças de aminoácidos entre as cepas do estudo, quando comparadas com a cepa FGA/89 (Guiana Francesa), mostraram a presença de importantes substituições não-sinônimas com mudança de caráter bioquímico, tais como os resíduos E297 (Met Tre) e E338 (Ser Leu), sendo necessário estudos para verificar se essas alterações podem ou não estar relacionadas à virulência. A análise filogenética para a proteína E, realizada por meio do método de máxima verossimilhança para as cepas do estudo e outras cepas selecionadas do banco de dados do Genbank, mostraram que as cepas de VDEN-1 isoladas no Brasil desde 1982 pertencem ao genótipo V, corroborando com os resultados publicados anteriormente. O tempo de divergência do VDEN-1, estimado através da hipótese de relógio molecular, mostrou que este vírus teve sua origem a partir de uma linhagem ancestral, há aproximadamente, 113 anos e observou-se ainda que as cepas de VDEN-1 circulantes no Brasil e as provenientes da África possuem um ancestral comum, sendo necessário estudos de filogeografia que mostrem as possíveis rotas de entrada do VDEN-1 no Brasil.

As análises citogenéticas de diversos Falconiformes mostraram que os acipitrídeos têm uma organização cromossômica atípica na classe Aves, com um número diplóide relativamente baixo (média de 2n= 66) e poucos pares de microcromossomos (4 a 6 pares). Propostas baseadas em citogenética clássica sugeriram que esse fato devia-se à fusão de microcromossomos presentes no cariótipo ancestral das Aves. No intuito de contribuir para o esclarecimento das questões referentes à evolução cromossômica e filogenética dessa família, três espécies da subfamília Buteoninae (Rupornis magnirostris, Buteogallus meridionales e Asturina nitida) e duas espécies da subfamília Harpiinae (Harpia harpyja e Morphnus guianensis) foram analisados citogeneticamente através da aplicação de técnicas de citogenética clássica e molecular. As espécies de Buteoninae apresentaram cariótipos muito semelhantes, com número diplóide igual a 68; o número de cromossomos de dois braços entre 17 e 21, o cromossomo Z submetacêntrico e o W metacêntrico em R. magnirostris e submetacêntrico em Asturina nitida. O uso de sondas de 18/28S rDNA mostrou a localização de regiões organizadoras de nucléolo em um par submetacêntrico médio nas três espécies, correspondendo ao braço curto do par 7. Sequências teloméricas foram mapeadas não só na região terminal dos braços, mas também em algumas posições intersticiais. Sondas de cromossomo inteiro derivadas dos pares 1 a 10 de Gallus gallus (GGA) produziram o mesmo número de sinais nessas três espécies. A disponibilidade das sondas de cromossomos totais derivadas de Leucopternis albicollis confirmou a existência de uma assinatura citogenética comum para as espécies de Buteoninae analisadas por FISH, que se trata da associação entre GGA1p e GGA6, inclusive com um sítio de sequência telomérica intersticial reforçando esse fato. As espécies de Harpiinae analisadas mostraram que o número diplóide das espécies de H. harpyja e M. guianensis foi igual a 58 e 54, respectivamente, e que ambas as espécies apresentam vinte e dois pares de cromossomos de dois braços, mesmo Harpia apresentando dois pares a mais. 18/28S rDNA produziram sinais no braço curto do par 1 em M. guianensis e em dois pares em H. harpyja (pares 6 e 25). Sequências teloméricas intersticiais também foram observadas em alguns pares. Apesar da similaridade na morfologia cromossômica, não foram observadas associações compartilhadas por essas duas espécies. As diferentes associações observadas em Morphnus e Harpia mostram que essas espécies sofreram uma reorganização genômica expressiva após sua separação em linhagens independentes. Além disso, ausência de associações semelhantes sugere que houve fissões nos macrocromossomos do ancestral em comum desse grupo, e as fusões foram subsequentes ao seu isolamento como linhagens diferentes. Os resultados aqui apresentados, somados àqueles publicados anteriormente com outras espécies de Accipitridae indicam que os processos de fissões envolvendo os macrocromossomos de GGA e fusões entre esses segmentos e entre esses e microcromossomos são rearranjos recorrentes nesse grupo. Apesar dos Falconidae também apresentarem cariótipos atípicos, e números diploides baixos, os dados globais da citogenética de Accipitridae indicam que, assim como postulado para as semelhanças morfológicas entre esses dois grupos, os cariótipos rearranjados corresponderiam a homoplasias, do ponto de vista evolutivo, apoiando que essas duas famílias não formam um grupo monofilético.
