Migration, invasion and protease activity are essential for tumor progression and metastasis. Metastatic cells rely on invadopodia to degrade and invade extracellular matrix (ECM). Invadopodia are membrane protrusions with enzymes required for ECM degradation. These protrusions contain cortactin and membrane type I matrix metalloproteinase (MT1-MMP) superimposed to areas of digested matrix. Here we characterized invadopodia in a cell line (CAC2) derived from human adenoid cystic carcinoma. We carried out fluorescent-substrate degradation assay to assess in situ protease activity of CAC2 cells. Digestion spots in fluorescent substrate appear as black areas in green background. Cells were cultured on Matrigel-gelatin-FITC and fixed after 1 h and 3 h. CAC2 cells were double labeled to actin and cortactin. Cells were also double stained to actin and MT1-MMR Samples were studied by laser scanning confocal microscopy. In all time points CAC2 cells showed actin, cortactin, and MT1-MMP colocalized with digestion spots in fluorescent substrate. We searched for other proteases involved in invadopodia activity. We have previously demonstrated that MMP9 influences adenoid cystic carcinoma behavior. This prompted us to investigate role played by MMP9 on invadopodia formation. CAC2 cells had MMP9 silenced by siRNA. After I h in fluorescent substrate, cells with silenced MMP9 showed clear decrease in matrix digestion compared with controls. No differences were found in cells with silenced MMP9 grown for 3 h on fluorescent substrate. Our results showed that CAC2 cells exhibit functional invadopodia containing cortactin and MT1-MMR Furthermore, MMP9 would be required in the initial steps of invadopodia formation. Microsc. Res. Thermoluminescence, electron paramagnetic resonance and optical absorption properties of rhodonite, a natural silicate mineral, have been investigated and compared to those of synthetic crystal, pure and doped. The TL peaks grow linearly for radiation dose up to 4 kGy, and then saturate. In all the synthetic samples, 140 and 340 degrees C TL peaks are observed; the difference occurs in their relative intensities, but only 340 degrees C peak grows strongly for high doses. Al(2)O(3) and Al(2)O(3) + CaO-doped synthetic samples presented several decades intenser TL compared to that of synthetic samples doped with other impurities. A heating rate of 4 degrees C/s has been used in all the TL readings. The EPR spectrum of natural rhodonite mineral has only one huge signal around g = 2.0 with width extending from 1,000 to 6,000 G. This is due to Mn dipolar interaction, a fact proved by numerical calculation based on Van Vleck dipolar broadening expression. The optical absorption spectrum is rich in absorption bands in near-UV, visible and near-IR intervals. Several bands in the region from 540 to 340 nm are interpreted as being due to Mn(3+) in distorted octahedral environment. A broad and intense band around 1,040 nm is due to Fe(2+). It decays under heating up to 900 degrees C. At this temperature it is reduced by 80% of its original intensity. The pink, natural rhodonite, heated in air starts becoming black at approximately 600 degrees C.

In this work, TG/DTG and DSC techniques were used to the determination of thermal behavior of prednicarbate alone and associated with glyceryl stearate excipient ( 1: 1 physical mixture). TG/DTG curves obtained for the binary mixture showed a reduction of approximately 37 degrees C to the thermal stability of drug (T(dm/dt-0) (Max)(DTG)). The disappearance of stretching band at 1280 cm(-1) (nu(as) C-O, carbonate group) and the presence of streching band with less intensity at 1750 cm(-1) (nu(s) C-O, ester group) in IR spectrum obtained to the binary mixture submitted at 220 degrees C, when compared with IR spectrum of drug submitted to the same temperature, confirmed the chemical interaction between these substances due to heating. Kinetics parameters of decomposition reaction of prednicarbate were obtained using isothermal (Arrhenius equation) and non-isothermal (Ozawa) methods. The reduction of approximately 45% of activation energy value (E(a)) to the first step of thermal decomposition reaction of drug in the 1:1 (mass/mass) physical mixture was observed by both kinetics methods.

Nanocomposite electrolytes of a fully amorphous trifunctional polyether (3PEG) and poly- (methylene ethylene oxide) (PMEO) have been complexed with two lithium salts and nanoparticulate (~20 nm) fillers of TiO2 and Al2O3. Addition of the fillers to the polymer salt complexes shows a significant change in the conformational modes of both polymers, especially the D-LAM region between 200 and 400 cm-1, indicating a reduced segmental flexibility of the chain. These changes are more pronounced with the use of TiO2 than Al2O3. Incorporation of the nanoparticulate fillers to the electrolytes fails to influence the degree of ion association, suggesting that the number of charge carriers available for conduction in both polymers using both LiClO4 and LiCF3SO3 is not the source of any conductivity increase. Addition of the fillers, which was seen to increase the conductivity in PEO-based systems, generally lowers the conductivity in the present PMEO systems, while the addition of TiO2 has little or no effect except in the cases of 3PEG 1.5 and 1.25 mol/kg LiClO4. In this case, 10 wt % TiO2 provides a conductivity increase of half an order of magnitude at approximately 60 °C. We also report for the first time a Raman spectroscopy investigation into the PEO-based nanocomposite electrolytes. The present results are discussed in terms of the electrostatic interactions involving dielectric properties of the fillers, of special interest being the interactions between the polymer and the fillers and between the ionic species and the fillers, when the effect of crystallization can be ignored.

Food security is a significant issue in the remote, high altitude areas of Nepal. Malnutrition is common because the harsh climate restricts production and access to fresh food. Humla is a province in north-west Nepal where the severe weather conditions and limited arable land restrict the growing season of vegetables to only 4-5 months a year. Passive solar greenhouses are a technology that can be used to extend the growing period of vegetables. For the greenhouse to be effective in Humla, the design must be appropriate to the climatic conditions, available resources and the social requirements of the community. This paper describes the design and evaluation of a family-sized solar greenhouse proposed for Humla. Three solar greenhouses built in other high altitude areas have initially been reviewed to see what techniques might be replicated in Humla. The thermal performance of the proposed family-sized greenhouse has been predicted using the solar simulation software, TRNSYS, and compared against an existing design in the area. The proposed design was found to be superior and average night-time temperatures in the coldest month were predicted to be approximately 7°C higher. Overheating was controlled in the hottest month by shading and natural ventilation.

Single pass warm rolling and compression experiments were carried out from ambient to 800°C for ultra-low carbon (ULC) steel with ∼100 ppm carbon and interstitial free (IF) steels, both with two levels of silicon. Subsequently, annealing was done in order to recrystallize the deformed specimens. The main purpose of this study was to understand the effects of rolling temperature and silicon on stress responses and textures. This study comprises two main themes: flow stress and strain rate sensitivity during compression and shear banding and textures in warm rolled specimens. The effects of deformation temperature on in-grain shear bands were different between ULC-Si and IF-Si steels. As in previous work with more conventional steels, in-grain shear bands in the IF grade had low sensitivity to rolling temperature, while those in the ULC grade depended significantly on the deformation temperature. However, the temperature profile of shear banding in the ULC grade was approximately 150°C higher than in previous work. Deformation and recrystallisation textures for both IF and ULC grades depended on their rolling temperatures. The variation of both grain size and texture after annealing can be explained by the rise and fall of in-grain shear banding activity which is related to the strain rate sensitivity.

Oxide type spinel AB2O4 presents structure adjusted for application in the automobile industry. The spinel of cobalt has many practical applications had its excellent physical and chemical properties such as catalyst in hydrocarbon oxidation reaction. The CeO2 has been used in many of these processes because it assigns to a material with excellent thermal resistance and mechanics, high capacity of oxygen stockage (OSC) among others properties. This work deals with the synthesis, characterization and catalytic application of spinel of cobalt and CeO2 with fluorita structure, obtained for method of Pechini and method of Gel-Combustion. The process of Pechini, the puff was obtained at 300 ºC for 2 h in air. In the process of Gel-Combustion the approximately at 350 ºC material was prepared and burnt for Pyrolysis, both had been calcined at 500 ºC, 700 ºC, 900 ºC and 1050 ºC for 2 h in air. The materials of the calcinations had been characterized by TG/DTA, electronic microscopy of sweepings (MEV), spectroscopy of absorption in the infra-red ray (FTIR) and diffraction of X-rays (DRX). The obtained material reaches the phase oxide at 450 oC for Pechini method and 500 °C for combustion method. The samples were submitted catalytic reaction of n-hexane on superficies of materials. The reactor function in molar ration of 0, 85 mol.h-1.g-1 and temperature of system was 450 °C. The sample obtained for Pechini and support in alumine of superficial area of 178,63 m2.g-1 calcined at 700 ºC, give results of catalytic conversions of 39 % and the sample obtained for method of gel-combustion and support in alumina of 150 mesh calcined at 500 ºC result 13 % of conversion. A semi solid thin strip continuous casting process was used to obtain 50%wt Pb/50%wtSn strip by single and twin roll processing at speed of 15 m/min. A 50%wt Pb/50%wtSn plate ingot was also cast for rolling conventionally into strips of 1.4 mm thickness and 45 mm width for comparison with those achieved non-conventionally. This hypoeutectic alloy has a solidification interval and fusion temperature of approximately 31 degrees C and 215 degrees C respectively. The casting alloy temperature was around 280 degrees C as measured by a type K immersion thermocouple prior to pouring into a tundish designed to maintain a constant melt flow on the cooling slope during semi solid material production. A nozzle with a weir ensures that the semi solid material is dragged smoothly by the lower roll, producing strip with minimum contamination of slag/oxide. The temperatures of the cooling slope and the lower roll were also monitored using K type thermocouples. The coiled semi solid strip, which has a thickness of 1.5 mm and 45 mm width, was rolled conventionally in order to obtain 1.2 mm thick strip. The coiled thixorolled strip had a thickness of 1.2 mm and achieved practically the same width as the conventional strips. Blanks of 40 mm diameter were cut from the strips in a mechanical press, ready for deep drawing and ironing for mechanical characterization. All the strips achieved from non-conventional processing had the same mechanical performance as those achieved conventionally. The limiting drawing ratio (LDR) achieved was approximately 2.0 for all strips. Microscopy examination was made in order to observe phase segregation during processing.

The thermal properties of plums (Prunus domestica) and prunes were investigated in the moisture content of 14.2-80.4% (wet basis) near room temperature (approximately 28 degrees C). The apparent density of the fruits increased from 1042.9 to 1460.0 kg/m(3), and the bulk density increased from 706.6 to 897.5 kg/m(3) as the plums were dried, following classical empirical models as a function of moisture content. It was found that specific heat, effective thermal diffusivity, and effective thermal conductivity of the prunes increased with the moisture content of the samples, which can be represented by using different empirical models.

The objective of this study was to evaluate the effect of immersion for varying periods in different concentrations of gibberellic acid and separate methods of scarification on the germination of seeds tucum. In the first trial, testing different soaking periods (24 and 48 hours), different forms of soaking (fast and slow) and different concentrations of gibberellic acid (0, 100 and 200 mg L-(1)) compared to the control (water) under completely randomized design in a factorial 2x2x3. The second experiment evaluated the different scarification treatments being: physical (seed coat removal in the hilar region with the aid of a scalpel), chemical (sulfuric acid 98 PA for 2:04 minutes) and thermal (hot water at approximately 98 degrees C and cold water at about 2 degrees C for 4 minutes). In the first trial were evaluated % of contaminated seeds (seeds infected by microorganisms) and hard seeds (who did not start the germination process, but not soiled), and in the second we assessed germination percentage (%) every two days for three months; germination Speed Index (GSI), time to occurrence of 50 % germination (T50) and Emergency Speed Index (ESI). The use of gibberellic acid was ineffective in promoting germination of Tucum (Astrocaryum Huaimi Mart.). Scarification treatments were effective in promoting germination and emergence of seedlings in the nursery and the most efficient physical removal scarification of the seed coat in the hilar region with the highest percentage of germination.
