Objective: Archipelago (AGO, also known as hCdc4, Fbw7, or Sel-10) is an F-box containing component of the SCF complex implicated in the ubiquitination and proteolysis of cyclin E and c-Myc, and found to be mutated in 16% of endometrial carcinomas. We have previously reported somatic mutations in AGO in 3/10 ovarian cancer cell lines, but the frequency of such mutations in primary ovarian cancer is unknown.

Methods: The coding sequence of AGO was analyzed in 95 primary sporadic ovarian tumors and 16 cases of familial ovarian cancer, and correlated with levels of cyclin E and c-Myc protein expression. Constructs encoding mutations in AGO were transfected into an AGO-null cell line to directly test their ability to regulate cyclin E and c-Myc levels.

Results: Mutations were present in only 2 of 95 sporadic cases: a premature stop within the WD domain (471 Ter) and a missense change near the F-box (S245T). Both primary tumor specimens containing these mutations showed high levels of cyclin E and c-Myc, but reconstitution of an AGO-null cell line with constructs encoding these mutations showed 471 Ter to be inactive in regulating endogenous cyclin E and c-Myc levels, while the S245T mutant was indistinguishable from wild-type. No germ-line mutations were found in familial cases of ovarian cancer.

Conclusion: Somatic AGO mutations are infrequent in primary ovarian cancers and are unlikely to contribute to familial ovarian cancer. Reconstitution experiments, rather than measuring tumor levels of cyclin E and c-Myc, provide an effective approach to determine the functional significance of AGO mutations identified in human cancers.

Geological, biological, morphological, and hydrochemical data are presented for the newly discovered Moytirra vent field at 45oN. This is the only high temperature hydrothermal vent known between the Azores and Iceland, in the North Atlantic and is located on a slow to ultraslow-spreading mid-ocean ridge uniquely situated on the 300 m high fault scarp of the eastern axial wall, 3.5 km from the axial volcanic ridge crest. Furthermore, the Moytirra vent field is, unusually for tectonically controlled hydrothermal vents systems, basalt hosted and perched midway up on the median valley wall and presumably heated by an off-axis magma chamber. The Moytirra vent field consists of an alignment of four sites of venting, three actively emitting "black smoke," producing a complex of chimneys and beehive diffusers. The largest chimney is 18 m tall and vigorously venting. The vent fauna described here are the only ones documented for the North Atlantic (Azores to Reykjanes Ridge) and significantly expands our knowledge of North Atlantic biodiversity. The surfaces of the vent chimneys are occupied by aggregations of gastropods (Peltospira sp.) and populations of alvinocaridid shrimp (Mirocaris sp. with Rimicaris sp. also present). Other fauna present include bythograeid crabs (Segonzacia sp.) and zoarcid fish (Pachycara sp.), but bathymodiolin mussels and actinostolid anemones were not observed in the vent field. The discovery of the Moytirra vent field therefore expands the known latitudinal distributions of several vent-endemic genera in the north Atlantic, and reveals faunal affinities with vents south of the Azores rather than north of Iceland. © 2013. Os nemátodes são um grupo de invertebrados, não segmentados que formam um Filo (Nematoda) bem definido e claramente distinto dos outros grupos de organismos. Este Filo constitui um dos grupos animais mais disseminados no planeta, e em termos de número de indivíduos os nemátodes são o grupo animal mais abundante na Terra: quatro em cada cinco animais da Biosfera são nemátodes. Apesar de microscópicos, os animais multicelulares que constituem este grupo são capazes de explorar uma enorme variedade de habitats, nos mares, nas águas doces, nos solos, como parasitas de animais ou de plantas, ou mesmo em condições extremas, como nos solos secos da Antárctida ou em fontes termais (Baldwin et al. 1999). A actual informação sobre a nematofauna do arquipélago dos Açores encontra-se fraccionada e espalhada em diversas publicações científicas, mas igualmente em dados ainda não publicados (Sturhan, comunicação pessoal). Entre as diversas publicações de nemátodes para os Açores, há que salientar os trabalhos realizados por Sturhan (1973, 1975, 1983) e Macara (1994), que muito contribuíram para o conhecimento e distribuição actual das espécies de vida livre e parasitas de plantas; bem como os trabalhos de Afonso-Roque (1995) e Casanova et al. (1996) relativamente a espécies parasitas de animais, reportadas para as diferentes ilhas do arquipélago. A lista de nemátodes apresentada neste capítulo, tem como base as publicações conhecidas para o arquipélago (ver listagem de publicações até 2010 em www. naturdata.com), bem como o registo de espécies assinaladas pela primeira vez para o arquipélago (Sturhan, comunicação pessoal). A classificação utilizada baseia-se na recente revisão da sistemática e filogenia proposta por De Ley & Blaxter (2002) para todo o Filo, até ao nível da Família. A classificação dos restantes taxa segue o critério utilizado para a fauna terrestre da Fauna Europaea (http://www. faunaeur.org). É apresentada a distribuição das espécies nas nove ilhas dos Açores, usando-se a seguinte simbologia: COR – Corvo; FLO – Flores; FAI – Faial; PIC – Pico; GRA – Graciosa; SJG – São Jorge; TER – Terceira; SMG – São Miguel e SMR – Santa Maria.

Poilão dam reservoir (Cape Verde Archipelago) is in critical conditions, owing the excessive silting up, the high concentration of nitrates and the pronounced anoxia all over the lake. Considering that the most suitable remediation strategy is the removal of the bottom sediments where nutrients are preferentially concentrated, we have done a geochemical study, in order of evaluating their suitability to agricultural use. Analyses indicate that sediments are rich in a few key nutrients, when compared with parent soils. Thus, adding suitable sediments to nearby degraded soils can improve food crops for smallholder farmers living in close proximity to this system.

Deep-sea resources have been increasingly exploited, and due to that, several ecosystems and species have been considerably affected. Deep-water sharks populations have been of the most disturbed by practices of unselected fisheries, bycatch and discard, mainly due to their low commercial value. Those practices make deep-water sharks very vulnerable to overfishing given their life-history traits, increasing their extinction risk. With the prohibition of the direct fishery, and implementation of quotas and TACs (Total Allowable Catches) regarding the deep-sea shark landings, the official landings have dramatically decreased after the 1990s. However, the IUU (Illegal, unreported and unregulated) catch has exponentially increased. With the analysis of catch per unit effort (CPUE), the depths, and the mean weight of the individuals over the years for each one of the nine most caught species in the Azores, we produced a descriptive analysis of the effect of fisheries in those species. The results show that some of these species have been suffering from a great fishing pressure, and their populations will be greatly affected in the near future if drastic measures are not taken when it comes to managing their long term sustainability.
