Foram feitas avaliações do zinco no solo através do teste microbiológico do Aspergillus nigêr (WALLACE, 1961) em amostras de solo colhidas dos horizontes de 8 perfis de solos de 8 séries de solos do município de Piracicaba, Estado de São Paulo, Brasil. De cada sub horizonte foram tomados 0,25 g de solo onde se adicionou 50 ml de solução nutritiva sem zinco deixando-se em incubação em estufa adequada, à temperatura de 28° C, por um período de 7 dias. Passado este período colheu-se o micélio produzido e pesou-se após ser sêco a 70 - 80° C. Estes testes foram comparados com provas em branco e soluções (adições) contendo doses diferentes de zinco. No ensaio microbiológico fez-se, também, em amostras correspondentes ao Ap dos solos, a aplicação de doses crescentes de zinco (0 a 16 microgramas de zinco), nas mesmas condições usadas do teste, para se verificar as reações de cada solo à adição do zinco. Nas mesmas amostras colhidas dos perfis determinou-se em extratos, zinco solúvel, respectivamente de HCl 0,1 N de EDTA-(NH4)2CO3 e de Ditizona-acetato de amônio. Foram estudadas as correlações entre os resultados do Zinco das soluções extratoras e do teste microbiológico. O trabalho permitiu as seguintes conclusões: - O teste microbiológico do Aspergillus niger revelou-se eficiente na avaliação do zinco do solo. - Das 3 solúveis extratoras apenas a ditizona mostrou correlação com o teste microbiológico. - O teste microbiológico permitiu separar os solos em 3 grupos; segundo a sua reação a aplicação de zinco: Bem suprida em Zinco "Luiz de Queiroz"; medianamente supridas: Quebra-Dente, Bairrinho e Lageadinho; mal supridas: Iracema, Monte Olimpo, Guamium e Paredão Vermelho.

O objetivo deste trabalho foi avaliar a eficiência de solubilização de FePO4 por Aspergillus niger, em meio de cultura com diferentes fontes de carbono (C) e nitrogênio (N) e com concentrações crescentes de fosfato. A quantidade de fosfato solúvel, acidez e pH final foram determinados após o crescimento do fungo, em cultura estacionária a 30ºC. A eficiência de solubilização aumentou conforme o crescimento do fungo, atingiu o máximo no 11º dia (68%) e depois regrediu. Das fontes de C e de N testadas, as maiores eficiências de solubilização foram obtidas com manitol (21%) e ácido glutâmico (17%). Com o aumento da concentração de fosfato (0 a 1.330 µg mL-1), a máxima eficiência de solubilização (70%) foi obtida com 330 µg mL-1 de PO4(3-) e decresceu até 47% com 1.330 µg mL-1. A produção de ácidos foi o principal mecanismo de solubilização do FePO4, com base na correlação positiva e significativa entre a produção de fosfato e a acidez.

Agricultural wastes from cactus Cereus peruvianus and Opuntia ficus indica were investigated for protein production by solid substrate fermentation. Firstly, the polyelectrolytes were extracted and used in water cleaning as auxiliary of flocculation and coagulation. The remaining fibrous material and peels were used as substrate for fermentation with Aspergillus niger. Glucoamylase and cellulase were the main enzymes produced. Amino acids were determined by HPLC and protein by Lowry's method. After 120 hours of fermentation the protein increased by 12.8%. Aspartic acid (1.27%), threonine (0.97%), glutamic acid (0.88%), valine (0.70%), serine (0.68%), arginine (0.82%), and phenylalanine (0.51%) were the principal amino acids produced.

Microbial lipases have a great potential for commercial applications due to their stability, selectivity and broad substrate specificity because many non-natural acids, alcohols or amines can be used as the substrate. Three microbial lipases isolated from Brazilian soil samples (Aspergillus niger; Geotrichum candidum; Penicillium solitum) were compared in terms of their stability and as biocatalysts in the enantioselective esterification using racemic substrates in organic medium. The lipase from Aspergillus niger showed the highest activity (18.2 U/mL) and was highly thermostable, retaining 90% and 60% activity at 50 ºC and 60 ºC after 1 hour, respectively. In organic medium, this lipase provided the best results in terms of enantiomeric excess of the (S)-active acid (ee = 6.1%) and conversion value (c = 20%) in the esterification of (R,S)-ibuprofen with 1-propanol in isooctane. The esterification reaction of the racemic mixture of (R,S)-2-octanol with decanoic acid proceeded with high enantioselectivity when lipase from Aspergillus niger (E = 13.2) and commercial lipase from Candida antarctica (E = 20) were employed.

We report herein the synthesis of aryl beta-N-acetylglucosaminides containing azido, amino and acetamido groups at C-6 as potential antimicrobial agents. It was expected that these compounds could interfere with the biosynthesis and/or biotransformation of N-acetylglucosamine in fungi and bacteria. None of the compounds showed antimicrobial activity against bacteria (Bacillus subtilis, Micrococcus luteus, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa), filamentous fungus (Aspergillus niger) and yeasts (Saccharomyces cerevisae, Candida albicans and Candida tropicallis), at the concentration of 1 mg/mL in agar diffusion assay.

The aim of this work was to verify the effects of initial medium moisture content (U), addition of ammonium sulphate (N) and of potassium phosphate (P) in the production of the polygalacturonase through the solid-state fermentation, using cashew apple husk as substrate and Aspergillus niger CCT0916 as transformation agent. We also studied the best extraction conditions of the produced enzyme. The best condition of production was with U of 40%, 1% of N and 0% of P being reached an activity of the poligalacturonase of 10.1 U/g. The best extraction condition is an agitation system with a time of 100 min and a solvent-fermented medium volume ratio of 5 mL/g.

The new iridoid glucoside 10-O-vanilloyl-geniposidic acid has been isolated from the aerial parts of Alibertia myrciifolia along with hydroxyhopanone, 3α,22-dihydroxyhopane, ursolic acid, luteolin-3´,4´-dimethyl ether, caffeic acid and geniposidic acid. The structures of the isolated compounds were determined by means of mass spectrometry and nuclear magnetic resonance spectral analyses. The antifungal activities of the iridoids 10-O-vanilloyl-geniposidic acid and geniposidic acid were evaluated against the phytopathogenic fungi strains Colletotrichum gloeosporioides, Fusarium solani and Aspergillus niger.

In the present study, various amides of 2-amino-5-(4-methylphenyl)-diazenyl-4-phenyl-1, 3-thiazole was synthesized and their biological activities were evaluated. All the synthesized compounds were characterized by the combination of elemental analysis and standard spectroscopic methods. They are screened for anti-bacterial activity against Escherichia coli and Staphylococcus aureus as well as screened for antifungal activity against Aspergillus niger and Apergillus oryzae by cup plate method at 1 µg/ mL concentration in DMF.

O trabalho teve como objetivos avaliar e correlacionar a qualidade sanitária e fisiológica de sementes de abóbora, variedade Menina Brasileira (Cucurbita moschata.). Foram avaliados dois lotes de sementes de abóbora produzidas no sistema agroecológico e quatro no sistema convencional, com e sem tratamento químico. Os lotes foram submetidos aos testes de sanidade, seguindo a metodologia do "Blotter test", com congelamento, germinação e vigor (primeira contagem, índice de velocidade de germinação, envelhecimento acelerado e emergência de plântulas). Os resultados indicaram a separação dos lotes de diferentes origens a partir da qualidade sanitária e fisiológica, onde as maiores incidências de fungos foram observadas nos lotes agroecológicos e o maior potencial fisiológico foi observado nos lotes de origem convencional não tratados. Foram encontrados os fungos Fusarium oxysporum, Alternaria alternata, Cladosporium cucumerinum, Aspergillus niger, Penicillium digitatum, Rhizopus stolonifer e Phoma terrestris. A qualidade sanitária não interferiu na qualidade fisiológica das sementes de abóbora, variedade Menina Brasileira.

Ricinus communis L. is of great economic importance due to the oil extracted from its seeds. Castor oil has been used for pharmaceutical and industrial applications, as a lubricant or coating agent, as a component of plastic products, as a fungicide or in the synthesis of biodiesel fuels. After oil extraction, a castor cake with a large amount of protein is obtained. However, this by-product cannot be used as animal feed due to the presence of toxic (ricin) and allergenic (2S albumin) proteins. Here, we propose two processes for detoxification and allergen inactivation of the castor cake. In addition, we establish a biological test to detect ricin and validate these detoxification processes. In this test, Vero cells were treated with ricin, and cell death was assessed by cell counting and measurement of lactate dehydrogenase activity. The limit of detection of the Vero cell assay was 10 ng/mL using a concentration of 1.6 x 10(5) cells/well. Solid-state fermentation (SSF) and treatment with calcium compounds were used as cake detoxification processes. For SSF, Aspergillus niger was grown using a castor cake as a substrate, and this cake was analyzed after 24, 48, 72, and 96 h of SSF. Ricin was eliminated after 24 h of SSF treatment. The cake was treated with 4 or 8% Ca(OH)2 or CaO, and both the toxicity and the allergenic properties were entirely abolished. A by-product free of toxicity and allergens was obtained.

O desenvolvimento de alimentos é uma constante nas indústrias, nas universidades e nos institutos de pesquisa visando otimizar processamentos, agregar diferentes ingredientes em produtos já existentes, entre outros fatores. Após o desenvolvimento de um produto, informações adicionais podem ser obtidas pela determinação do rendimento e do custo do produto final. O objetivo deste trabalho foi estimar o rendimento e o custo de queijos Minas frescal tradicionais e adicionados de extrato hidrossolúvel de soja desengordurado (EHS) processados com coalho bovino e com coagulante microbiano (Aspergillus niger var. awamori). A introdução de 8% do EHS (em relação ao teor de sólidos totais do leite) no queijo Minas frescal processado com coalho bovino proporcionou rendimento em gramas de sólidos totais por litro de leite (g ST/L) superior de 16,67% em relação ao queijo Minas frescal tradicional processado com o mesmo coalho. O queijo tipo Minas frescal processado com coagulante microbiano e com 8% de EHS apresentou rendimento (g ST/L) superior de 16,54%, quando comparado ao seu tradicional. O custo das formulações-base (kg de queijo/100L de leite) processadas com coalho bovino e com coagulante microbiano foi US$ 36,75 e US$ 37,50, respectivamente, ao passo que os queijos tradicionais processados com os mesmos agentes coagulantes apresentaram custo equivalente a US$ 35,12 e US$ 35,36.

The high demands for sugars and the development of enzymatic technology have increased the production of sweeteners, especially for glucose and fructose syrups. This work describe a technology for glucose and fructose syrups from Brazilian cassava starch using enzymes produced by soil microrganisms isolated from the Brazilian Cerrado soil. Firstly, Aspergillus niger and Streptomyces sp. were isolated from the soil and used as glucoamylase (GA) and glucose isomerase (GI) producer sources. After characterization, GA and GI exhibited optimum pH 4.5 and 8.0, respectively. GA showed maximum activity at 60 ºC and GI at 85 ºC. GA and GI retained 65 and 80%, respectively, of initial activity after 180 minutes of incubation at 60 ºC. The kinetic parameters Km and Vmáx were 0.476 (mg.mL-1) and 8.58 (µmol/minute) for GA and 0.082 (M) and 48.20 (µmol/minute) for GI. The maximum glucose syrups production occurred after 24 hours of reaction with a 98% yield. The production of fructose syrups with 42% (w/v) was reached after 96 hours of reaction.

In the present study, the effect of pectinases on the production of pulp and juice of araçá and the presence of bioactive compounds were evaluated. An enzyme extract (EE) produced by Aspergillus niger LB-02-SF in solid state fermentation and the commercial enzyme Ultrazym®AFP-L were used in this study. After enzyme treatment with the EE preparation, the extraction yield increased by 23.1% and viscosity decreased by 42.8%, during pulp maceration. During juice processing, there was an increase of 70.6% in clarification and a decrease of 72.87% in turbidity. Higher values of these parameters, 47.7, 69.0, 80.7, and 79.7%, respectively, were obtained using the Ultrazym®AFP-L, which also led to a significant increase in the polyphenol content, both in the pulp (24%) and in the juice (28%), with a less pronounced effect when the EE was applied (10 and 21%, respectively). The anthocyanins content in the araçá pulp increased after treatment with the commercial preparation (23%), and there was no significant increase with the use of EE. The use of Ultrazym®AFP-L increased the β-carotene content by 29.4% in the fruit pulp, while the treatment with EE did not result in significant changes compared with those of the juice and pulp controls.

The goat placental immunomodulatory peptides were produced by fermentation with Aspergillus Niger. The objective of the present study was to investigate the effects of fermentation parameters (carbon source content, pH, and time) on spleen lymphocyte proliferation for the highest immune activity of the fermentation broth using response surface methodology (RSM). According to the data analysis by the Design-Expert® software, the stimulation index value (23.51%), which is the maximum immune activity, was obtained under the following conditions: content of carbon source 1.97 g·L-1, initial pH 5.0, and 74.43 h of fermentation time. Under the optimized fermentation conditions, at a certain concentration range, the fermentation broth produced a significant effect on the proliferation of mouse spleen lymphocytes. Ultrafiltration technique was performed to separate the fermentation broth with different MW (molecular weight). It was found that peptides in the range of <10 KDa were the main bioactivity fractions for the immunomodulatory and antioxidant activities.
