Schiff base vanadium(IV) complexes of phenyl esters of the two acidic amino acids, i.e., aspartic and glutamic acid, were synthesized. The phenyl esters of these amino acids were synthesized by conventional method whereas the Schiff base vanadium(IV) complexes were synthesized using microwave irradiation. The complexes were characterized by spectroscopic tools such as IR, 1H NMR, mass (ES), ESR, and UV visible spectroscopy. All the complexes were studied for antibacterial and antifungal activity and found to be moderately active.

The transition metal complexes of salicylhydrazone of anthranilhydrazide (H2L) were synthesised. The structures of metal complexes were characterized by various spectroscopic [IR, NMR, UV-Vis, EPR], thermal and other physicochemical methods. The single-crystal X-ray diffraction study of [Cu(HL)Cl]center dot H2O reveal its orthorhombic system with space group P2(1)2(1)2 and Z=4. The copper center has a distorted square planar geometry with ONO and Cl as the donor atoms. The ligand and its metal chelates have been screened for their antimicrobial and anti-tubercular activities using serial dilution method. Metal complexes in general have exhibited better antibacterial and antifungal activity than the free ligand and in few cases better than the standard used. Among the bacterial strains used, the complexes are highly potent against Gram-positive strains compared to Gram-negative. Anti-tubercular activity exhibited by the Co(II) complex is comparable with the standard used. (C) 2011 Elsevier B. V. All rights reserved.

A series of (2-aminothiazol-4-yl)methylester (5a-t) derivatives were synthesized in good yields and characterized by H-1 NMR, C-13 NMR, mass spectral and elemental analyses. The crystal structure of 5a was evidenced by X-ray diffraction study. The compounds were evaluated for their preliminary in vitro antibacterial, antifungal activity and were screened for antitubercular activity against Mycobacterium tuberculosis H37Rv strain. A new class of sulfone linked bis heterocycles viz., pyrrolyl/pyrazolyl arylaminosulfonylmethyl 1,3,4-oxadiazoles, 1,3,4-thiadiazoles, and 1,2,4-triazoles were prepared and tested for antimicrobial activity and cytotoxicity. The chloro-substituted compounds 5c, 8c and 14c showed comparable antibacterial activity to chloramphenicol against Pseudomonasaeruginosa and compound 5c exhibited comparable antifungal activity to ketoconazole against Penicilliumchrysogenum. One of the compounds, vinylsulfonyl oxadiazole showed appreciably cytotoxic activity on A549 lung carcinoma cells with an IC50 at a concentration of 31.7 mu M. (C) 2012 Elsevier Masson SAS. Mass spectrometric analysis of a banyan endophyte, Bacillus subtilis K1, extract showing broad spectrum antifungal activity revealed a complex mixture of lipopeptides, iturins, surfactins, and fengycins. Fractionation by reversed-phase high performance liquid chromatography (HPLC) facilitated a detailed analysis of fengycin microheterogeneity. Matrix assisted laser desorption ionization (MALDI) and electrospray ionization (ESI) mass spectrometric studies permitted the identification of several new fengycin variants. Four major sites of heterogeneity are identified: (1) N-terminus beta-hydroxy fatty acid moiety, where chain length variation and the presence of unsaturation occur, (2) position 6 (Ala/Val/Ile/Leu), (3) position 10 (Val/Ile) within the macrocyclic ring, and (4) Gln to Glu replacement at position 8, resulting in fengycin variants that differ in mass by 1 Da. Diagnostic fragment ions provide a quick method for localizing the sites of variation in the macrocycle or the linear segment. Subsequent establishment of the sequences is achieved by MS/MS analysis of linear fengycin species produced by hydrolysis of the macrocyclic lactone. Unsaturation in the fatty acid chain and the presence of linear precursors in the B. subtilis K1 extract are also established by mass spectrometry. The anomalous distribution of intensities within isotopic multiplets is a diagnostic for Gln/Glu replacements. High resolution mass spectrometry facilitates the identification of fengycin species differing by 1 Da by localizing the variable position (Gln(8)/Glu(8)) in the fengycin variants.

Benzoyl phenyl urea, a class of insect growth regulator's acts by inhibiting chitin synthesis. Carvacrol, a naturally occurring monoterpenoid is an effective antifungal agent. We have structurally modified carvacrol (2-methyl-5-1-methylethyl] phenol) by introducing benzoylphenyl urea linkage. Two series of benzoylcarvacryl thiourea (BCTU, 4a-f) and benzoylcarvacryl urea (BCU, 5a-f) derivatives were prepared and characterized by elemental analysis, IR, H-1 and C-13 NMR and Mass spectroscopy. Derivatives 4b, 4d, 4e, 4f and 5d, 5f showed comparable insecticidal activity with the standard BPU lufenuron against Dysdercus koenigii. BCTU derivatives 4c, 4e and BCU 5c showed good antifungal activity against phytopathogenic fungi viz. Magnaporthe grisae, Fusarium oxysporum, Dreschlera oryzae; food spoilage yeasts viz. Debaromyces hansenii, Pichia membranifaciens; and human pathogens viz. Candida albicans and Cryptococcus neoformans. Compounds 5d, 5e and 5f showed potent activity against human pathogens. Moderate and selective activity was observed for other compounds. All the synthesized compounds were non-haemolytic. These compounds have potential application in agriculture and medicine. A new class of sulfonamidomethane pyrrolyl-oxadiazoles/thiadiazoles and pyrazolyl-oxadiazoles/thiadiazoles was prepared from arylsulfonylaminoacetic acid hydrazides and E-cinnamic acid. The lead compounds were tested for antimicrobial and cytotoxic activities. The thiadiazole compounds having chloro substituent on the aromatic ring 4c, 8c and 10c exhibited comparable antibacterial activity against Pseudomonas aeruginosa and also antifungal activity against Penieillium ehrysogenunz. The styryl oxadiazole compound 3c showed appreciable cytotoxic activity on A549 lung carcinoma cells which can be used as a lead compound in the future studies.

In this Letter, we report the structure activity relationship (SAR) studies on series of positional isomers of 5(6)-bromo-1-(phenyl)sulfonyl]-2-(4-nitrophenoxy)methyl]-1H-benzim idazoles derivatives 7(a-j) and 8(a j) synthesized in good yields and characterized by H-1 NMR, C-13 NMR and mass spectral analyses. The crystal structure of 7a was evidenced by X-ray diffraction study. The newly synthesized compounds were evaluated for their in vitro antibacterial activity against Staphylococcus aureus, (Gram-positive), Escherichia coil and Klebsiella pneumoniae (Gram-negative), antifungal activity against Candida albicans, Aspergillus flavus and Rhizopus sp. and antitubercular activity against Mycobacterium tuberculosis H37Rv, Mycobacterium smegmatis, Mycobacterium fortuitum and MDR-TB strains. The synthesized compounds displayed interesting antimicrobial activity. The compounds 7b, 7e and 7h displayed significant activity against Mycobacterium tuberculosis H37Rv strain.

A simple and efficient protocol for the synthesis of novel 2,6-bis(4-methoxyphenyl)-1-methylpiperidin-4-one oxime esters 4(a-q) is described. Initially, p-anisaldehyde 1 was condensed (Mannich reaction) with acetone and ammonium acetate trihydrate afforded 2,6-bis(4-methoxyphenyl)piperidin-4-one 2. Then, methylation followed by oximation with hydroxylamine hydrochloride (NH(2)OHa (TM) HCl) furnished a key scaffold 4. Further, to explore the enhanced biological properties of the piperidin-4-one core i.e. the key scaffold 4 was conjugated with substituted benzoyl chlorides in the presence of anhydrous K2CO3 as base to obtain novel 2,6-bis(4-methoxyphenyl)-1-methylpiperidin-4-one oxime esters 4(a-q) in excellent yields. The newly synthesized compounds were characterized by elemental analysis, IR, H-1 NMR, C-13 NMR and mass spectroscopic techniques, and screened for their in vitro antioxidant and antimicrobial activities. Most of the compounds exerted positive efficacy towards the biological assays performed. Among the synthesized analogues, compounds 4l and 4m exhibited promising antioxidant activity and on the other hand compounds 4b and 4d manifested persuasive antibacterial activity, whereas compound 4b displayed stupendous antifungal activity against A. flavus strain.

Background: Candida-associated denture stomatitis is a frequent infectious disease. Treatment of this oral condition is difficult because failures and recurrences are common. The aim of this study was to test the in vitro antifungal activity of pure constituents of essentials oils. -- Methods: Eight terpenic derivatives (carvacrol, farnesol, geraniol, linalool, menthol, menthone, terpinen-4-ol, and aterpineol), a phenylpropanoid (eugenol), a phenethyl alcohol (tyrosol) and fluconazole were evaluated against 38 Candida isolated from denture-wearers and 10 collection Candida strains by the CLSI M27-A3 broth microdilution method. -- Results: Almost all the tested compounds showed antifungal activity with MIC ranges of 0.03-0.25% for eugenol and linalool, 0.03-0.12% for geraniol, 0.06-0.5% for menthol, a-terpineol and terpinen-4-ol, 0.03-0.5% for carvacrol, and 0.06-4% for menthone. These compounds, with the exception of farnesol, menthone and tyrosol, showed important in vitro activities against the fluconazole-resistant and susceptible-dose dependent Candida isolates. -- Conclusions: Carvacrol, eugenol, geraniol, linalool and terpinen-4-ol were very active in vitro against oral Candida isolates. Their fungistatic and fungicidal activities might convert them into promising alternatives for the topic treatment of oral candidiasis and denture stomatitis.

Neste trabalho, objetivou-se avaliar as propriedades físicas e microbiológicas de resinas acrílicas a base de polimetilmetacrilato após a incorporação de sal de diacetato de clorexidina (CDA) às mesmas. Para tal, foram confeccionados corpos de prova (CDPs) com as resinas VIPI COR e Duralay, sem e com incorporação de 0,5%; 1,0% e 2,0% de CDA, totalizando 8 grupos. A cromatografia líquida foi utilizada para mensurar a liberação de CDA pelas resinas acrílicas, e ainda, mensurar sua lixiviação de monômeros residuais.Para isso, os CDPs foram armazenados individualmente em placas para cultura celular de 24 poços contendo 1 ml de água destilada estéril em cada poço. Após tempo de armazenagem de 2 horas, 7 dias, 14 dias, 21 dias e 28 dias, a 37oC, a solução foi retirada e a liberação de clorexidina ou monômeros residuais foi avaliada utilizando-se HPLC associado a espectrometria ultravioleta. A atividade antifúngica para C. albicans foi avaliada utilizando teste de difusão em ágar, no qual os CDPs foram colocados em placas de BHI previamente inoculadas com C. albicans, com medição do halo de inibição após 48 horas de incubação a 37C. A análise do grau de conversão das resinas se deu através da técnica de espectroscopia de infravermelho transformada de Fourier FTIR utilizando-se uma amostra de resina não polimerizada de cada grupo e realizados 4 scans de absorbância. Para a mensuração da sorção de água por parte das resinas contendo CDA, foram confeccionados 10 corpos de prova para cada grupo, que foram posicionados em suporte dentro de dessecador a 37C para remoção de umidade intrínseca (m1) e depois imersos em 100 ml de água deionizada por 7 dias a 37C, tendo a água trocada diariamente. Após este intervalo, os corpos foram secos para obter a nova massa da resina (m2). As massas obtidas foram incluídas em fórmula matemática para obtenção do grau de sorção. Após obtenção dos resultados, quando comparou-se o halo inibição entre os grupos testados e de mesma marca, apenas as análises entre grupo CDA 2% x grupo CDA 1% e entre CDA 1% x CDA 0,5% não apresentaram diferenças significantes. Quanto a liberação de CDA, a análise de variância demonstrou que dois dos três fatores avaliados (concentração do fármaco e tempo de armazenagem) alteram de maneira significativa a taxa de liberação da clorexidina (p<0,0001), entretanto a marca do material pareceu não influenciar de maneira significativa na liberação do fármaco. Quanto ao grau de conversão, os valores obtidos não foram significantes e apresentou-se menor apenas nos grupos com CDA 2% . Para ambas a sorção de água aumentou conforme a incorporação do sal cresceu e houve aumento significativo nas concentrações de 1.0% e 2.0%. Podemos concluir que a incorporação da clorexidina às resinas a base de PMMA: é capaz de inibir o crescimento de C. albicans; não alterou o grau de conversão das resinas testadas; não altera a liberação de monômeros residuais; e, altera a sorção de água das resinas acrílicas a base de PMMA quando concentrações maiores de CDA são adicionadas.

Resinas macias para reembasamento de próteses são largamente utilizadas após cirurgias para estabilizarem a prótese e condicionarem o tecido, aguardando a completa cicatrização. É importante que o material não seja facilmente colonizado por biofilme oral e se possível, evite a contaminação do sítio cirúrgico. Objetivou-se avaliar o efeito da incorporação de clorexidina às resinas acrílicas macias para o reembasamento de próteses totais, através de análises de liberação, citotoxicidade e efeito inibitório de um biofilme de C. albicans. Foram confeccionados corpos de provas (CDPs) com as resinas Trusoft e Coe-soft, com incorporação de 0%, 0,5%, 1,0% e 2,0% de clorexidina, totalizando 8 grupos. A liberação de clorexidina foi avaliada através da mensuração da mudança na densidade óptica da solução de armazenamento, na qual ficaram imersos os CDPs, por espectrometria UV, a cada 48 horas, durante 40 dias. A citotoxicidade celular foi avaliada em fibroblastos (linhagem L929), que ficaram 24 horas em contato com meio de cultura no qual os CDPs ficaram previamente imersos, pela técnica de absorção de corante vermelho neutro após 24, 48 e 72 horas e semanalmente até o 28 dia. E, por fim, a atividade antifúngica contra a C. albicans (ATCC 10231) foi avaliada de duas maneiras: (1) teste de difusão em ágar, no qual os CDPs foram colocados em placas de BHI previamente inoculadas com C. albicans, com medição do halo de inibição após 48 horas de incubação a 37C; (2) a avaliação da inibição da formação de um biofilme de C. albicans sobre a superfície dos CDPs pela quantificação por metil tetrazólio (MTT) a cada 48 horas, durante 22 dias, com leitura feita em espectrofotômetro de UV. Os dados obtidos foram inseridos no programa SigmaStat (versão 3.1, USA) para realizar as análises estatísticas. As diferenças estatísticas foram determinadas por análises de variâncias do tipo ANOVA e todos os procedimentos para comparações múltiplas pareadas foram feitos utilizando-se o método Holm-Sidak, com nível de significância global igual a 0,05. A clorexidina adicionada às resinas testadas foi capaz de ser liberada para o meio de armazenagem, proporcionalmente à quantidade de clorexidina incorporada, porém com diferentes cinéticas de liberação entre as resinas, visto que a Trusoft libera até 71% do total de clorexidina liberada nas primeiras 48 horas e a Coe-soft, até 44%. Ambas as resinas com incorporação de clorexidina apresentaram efeito citotóxico adicional, se comparadas às resinas sem clorexidina, porém para a Coe-soft não houve diferença estatística dos valores, apenas para a Trusoft (p<0,001). Ocorreu formação de halo de inibição proporcionalmente às concentrações de resinas adicionadas, com maiores halos para a resina Trusoft (p<0,001), e sem formação de halo para as resinas sem clorexidina; a inibição da formação de biofilme, realizada somente com a resina Coe-soft, mostrou total inibição durante 8, 12 e 16 dias, para a incorporação de 0,5%, 1,0% e 2,0% respectivamente, sendo uma diminuição estatisticamente significativa (p<0,001) em relação à resina sem incorporação de clorexidina, que não apresentou inibição do biofilme.

枯草芽孢杆菌(Bacillus subtilis)是革兰氏阳性细菌研究的模式菌株,为重要的生防菌剂。本论文以枯草芽孢杆菌ATCC6051作对照,首次对新型菌株KB-1111和KB-1122进行了生物学特性、生防潜力以及与水稻稻瘟病致病菌互作的蛋白质组学研究。 形态学观察表明,菌株KB-1111和KB-1122在细胞形态、芽孢的大小、运动性、菌落褶皱和色素的生成等方面与菌株ATCC6051相似,具有枯草芽孢杆菌的典型特征。生理生化测定以及对多种碳源的利用结果显示,三个菌株大部分指标检测结果相同,只在几个方面等存在差异。 体外平板对峙抑菌试验说明,枯草芽孢杆菌ATCC6051、KB-1111和KB-1122对8种作物、果蔬代表性病害致病真菌具有明显的拮抗效果。其中,菌株KB-1122的广谱抗真菌活性优于KB-1111,而KB-1111又强于对照菌株ATCC6051,尤其是对稻瘟病(M. grisea P131)和蔬菜菌核病(S. sclerotiorum)显现出强烈的抑制作用,具备生防拮抗菌的优秀性能。 比较蛋白质组学分析结果表明,液体悬浮培养枯草芽孢杆菌KB-1111、KB-1122二维蛋白质组表达谱至少有11个胞内蛋白和10个胞外蛋白出现丰度差异。其中,菌株KB-1122中胁迫或逆境反应相关ATP酶、顺乌头酸水合酶和alpha-淀粉酶前体在细胞内蛋白质组,以及分泌型蛋白―内切葡聚糖酶在胞外蛋白质组中的高丰度表达可能与菌株KB-1122的优势拮抗能力相关。 将对数生长期的枯草芽孢杆菌KB-1122与菌丝丰富期的稻瘟病菌P131悬浮混合共培养发现,在24小时的共培养过程中,稻瘟病菌P131菌丝体及芽管经历了致变、破裂、细胞质溢出直至菌丝体崩溃等一系列变化,枯草芽孢杆菌KB-1122表现出强烈的拮抗效应。差异显示蛋白质组学研究表明,共培养菌体蛋白质组至少有39个蛋白点丰度发生显著变化,其中33个蛋白点得到成功鉴定,包括12个上调蛋白和21个下调蛋白。根据鉴定结果分析,这些上调的蛋白质全部来源于枯草芽孢杆菌,而下调的蛋白全部属于稻瘟病菌。共培养过程中的培养液蛋白质组至少有20个蛋白点丰度发生显著变化,其中18个蛋白点得到成功鉴定。根据以上分析结果初步认为,3-磷酸甘油醛脱氢酶、丝氨酸蛋白激酶和内切葡聚糖酶在枯草芽孢杆菌KB-1122与稻瘟病菌P131相互作用的过程中可能是B. subtilis KB-1122发挥抗真菌活性的关键性蛋白。

壳聚糖是一种天然的聚阳离子氨基多糖,由甲壳素经脱乙酰反应得到。作为天然可再生资源,壳聚糖以其特有的安全无毒、可生物降解、生物相容等特性,在农业、生物工程、制药、环保等领域引起广泛关注。随着对壳聚糖研究的不断深入,壳聚糖的抑菌活性及机理成为研究热点之一,但是,大多集中于壳聚糖分子量和脱乙酰度对其抑菌活性的影响,而对壳聚糖衍生物的抑菌性能及机理研究较少。本文合成了N–季铵盐,N,O–季铵盐,羧甲基壳聚糖希夫碱及其铜配合物,壳聚糖有机酸盐,并对它们的抑菌活性进行了研究,同时探讨了引入基团与抑菌活性之间的关系。 测定了壳聚糖及其季铵盐对黄瓜枯萎病菌、黄瓜炭疽病菌和桃褐腐病菌的抑制活性,结果表明壳聚糖季铵盐的抑制活性高于壳聚糖,其中,抑制活性最强的是N–异丁基–N,N–二甲基壳聚糖,当样品浓度为500μg/mL时,对三种病原菌的抑制率分别为:67.52%、70.42%和76.25%。壳聚糖分子带正电荷的氨基可以与菌体表面带负电的物质作用,导致菌体死亡,而壳聚糖季铵盐分子中的正电性可以加强这种作用,从而增强抑菌活性,而且随着正电性的增强,抑菌活性增强。 为了进一步研究壳聚糖季铵盐的正电性与其抑菌活性的关系,合成了含有氟、溴、氯吸电子基团苯环取代的壳聚糖季铵盐,通过计算分别得到取代基团的电负性以及氨基正电性。结果表明,随着取代基团电负性的增强,季铵盐氨基正电性增强,抑菌活性增强。其中,抑菌活性最强的是N–(2–氟)苄基–N,N–二甲基壳聚糖,当样品浓度为500μg/mL时,对三种病原菌的抑制率分别为:80.85%、100%和100%。 将壳聚糖的–NH2与有机酸的–H通过离子键结合制备得到了系列的固态壳聚糖有机酸盐:壳聚糖甲酸盐、壳聚糖乙酸盐、壳聚糖丙酸盐、壳聚糖丁酸盐和壳聚糖戊酸盐,抑菌活性结果表明,随着衍生物链长的增加,抑菌活性增强,而且衍生物抑菌活性与取代基团的电负性成正相关。 以羧甲基壳聚糖为原料,合成了羧甲基壳聚糖希夫碱及其铜配合物,各类衍生物的抑菌活性强弱顺序如下:羧甲基壳聚糖希夫碱铜配合物>羧甲基壳聚糖希夫碱>羧甲基壳聚糖,从而说明了基团的相互增效作用能增强衍生物的抑菌活性。 以N–季铵盐为原料,以环氧丙基三甲基氯化铵为改性剂合成了具有–N+(CH3)2R和–N+(CH3)3两种基团的N,O–季铵盐,抑菌活性结果表明,N,O–季铵盐的抑菌活性

Amphibian skin secretions are rich in antimicrobial peptides acting as important components of innate defense system against invading microorganisms. A novel type of peptide, designated as maximin S, was deduced by random sequencing of 793 clones from a constructed Bombina maxima skin cDNA library. The putative primary structures of maximin S peptides can be grouped into five species, in which maximin S I has 14 amino acid residues and the rest of maximin S peptides (S2-S5) all have 18 amino acid residues. Unlike most of the amphibian antimicrobial peptides so far identified, the newly characterized four maximin S precursors are composed of maximin S I and different combinations of tandem repeated maximin S2-S5 linked by internal peptides. Except maximin S I, the predicted secondary structures of maximin S2-S5 show a similar amphipathic alpha-helical structure. MALDI-TOF mass spectrometry analysis of partially isolated skin secretions of the toad indicates that most of the deduced maximin S peptides are expressed. Two deduced maximin S peptides (S1, S4) were synthesized and their antimicrobial activities were tested. Maximin S4 only had an antibiotic activity against mycoplasma and had no antibacterial or antifungal activity toward tested strains. Maximin S1 had no activity under the same conditions.
