Background: Candidal species, particularly Candida albicans are common pathogens in the oral cavity and perioral region. Many of the manifestations of candidiasis are associated with the formation of Candida biofilms on host surfaces and/or implanted biomaterials. Biofilms are clinically important due to their increased resistance to therapeutic intervention and the ability of cells within the biofilm to withstand host immune defences.
Objectives: The present study was designed to investigate the antifungal activity of two peptides found in skin secretions of the African volcano frog (Xenopus amieti) against the type strain of C. albicans NCTC 3179.
Methods: The antifungal activity of magainin-AM1 and peptide glycine-leucine-amide (PGLa-AM1) against C. albicans NCTC 3179 was studied in both planktonic and biofilm forms. Radial diffusion assays were used to obtain the minimum inhibitory concentration (MIC) of magainin-AM1 and PGLa-AM1 against planktonic C. albicans. Time kill assays were used to determine the time dependent fungicidal action of the peptides at both 4oC and 37oC. A 96 well microtitre plate model for candidal biofilm formation was employed to study the ability of the peptides to disrupt the early biofilm development (up to 24 hours) compared with the antifungal drug fluconazole. Biofilm formation was determined quantitatively using the crystal violet assay.
Results: Both magainin-AM1 and PGLa-AM1 demonstrated inhibitory activity against Candida albicans, with MIC values of 24.3 uM and 7.5uM respectively. Time-kill assays revealed bactericidal activity of both peptides at 37oC and 4oC. Magainin-AM1 and PGLa-AM1 inhibited biofilm formation in microtitre plate assays. The peptides were particularly effective during early biofilm establishment when compared with fluconazole treatment.
Conclusions: Magainin-AM1 and PGLa-AM1 are active against C albicans in both planktonic and biofilm forms. Further testing of this peptide family against candidal biofilms is recommended.

Une série de dimères composés de thiophène-aniline encombrée stériquement a été synthétisée. Les différents processus de désactivation de l’état singulet excité ont été étudiés par UV-visible, fluorescence, phosphorescence, photolyse par impulsion laser et calculs théoriques. Les graphiques de Stern-Volmer obtenus à partir des expériences de désactivation des états singulet et triplet ont démontré l’efficacité de l’azométhine à désactiver les fluorophores. Les calculs semi-empiriques AM1 examinant l’effet des substituants encombrés ont démontrés que les groupements tert-butyls sur l’aniline ont moins d’influence sur la barrière de rotation N-aryl que les substitutions alkyles en ont sur la rotation de thiophène-C. Les calculs Rehm-Weller basés sur les potentiels d’oxydation et de réduction ont montré que l’autodésactivation de l’état excité des azométhines se fait par transfert d’électron photoinduit menant à une éradication complète de la fluorescence. Des complexes métalliques contenant des ligands azométhines ont aussi été préparés. Le ligand est composé d’une unité hydroxyquinoline lié à un cycle thiophène. Les données photophysiques de ces complexes indiquent un déplacement bathochromique aussi bien en absorbance qu’en fluorescence. Des dispositifs de détection d’ion métallique ont été préparés et un exemple à partir d’une solution de cuivre a montré un déplacement bathochromique.

L’opossum nait dans un état très immature, mais rampe avec ses membres antérieurs (MA) de l’orifice urogénital de la mère à une tétine, où il s’attache pour poursuivre son développement. Des informations sensorielles sont nécessaires pour guider le nouveau-né vers une tétine et déclencher son attachement. Des expériences précédentes ont montré que le système du trijumeau, dont dépend l’innervation somesthésique du museau, influence les mouvements précoces des MA. Le présent projet vise à déterminer si les mécanorécepteurs faciaux sont fonctionnels et exercent une influence sur les MA. On s’intéresse particulièrement aux cellules de Merkel, un mécanorécepteur épidermique innervé par des fibres à adaptation lente de type I (SA I). Ces cellules ont été localisées sur le pourtour du museau de l’opossum nouveau-né en utilisant un traceur cellulaire, l’AM1-43. Nous avons analysé les réponses musculaires des MA consécutives à l’application de forces calibrées au museau sur des préparations in vitro. Ces réponses sont bilatérales et simultanées, très variables, et leur intensité augmente avec la force de la stimulation. Lors de stimulations répétitives pendant 60 min, les réponses diminuent avec le temps. Le retrait de la peau faciale abolit presque ces réponses. De plus, l’application d’un antagoniste des récepteurs métabotropiques du glutamate, qui affecte l’activité des fibres SA I, ou d’un antagoniste des récepteurs purinergiques les diminue fortement, suggérant une participation des cellules de Merkel. Ces résultats soutiennent que le sens du toucher facial relayé par le système du trijumeau est fonctionnel chez l’opossum nouveau-né et qu’il pourrait influencer les mouvements des MA.

Seventeen fungal isolates were tested in vitro as potential antagonists of two isolates of the root rot pathogen, Armillaria mellea. Some of the isolates were also added on mushroom composts with living mycelia to the roots of Armillaria-inoculated potted strawberry plants in the glasshouse to find out if they had the same degree of efficacy against the disease. Dactylium dendroides isolate SP was the most effective in reducing mycelial growth of A. mellea isolate 1 (Am1), followed by Trichoderma harzianum isolate Th2 and T. viride isolate Tv4. Th2, Th22, Tv3 and SP grew extensively over Am1 colonies, disintegrating the rhizomorphs. Isolate Tham1 of T hamatum was the most effective in reducing mycelial growth of A. mellea isolate 2 (Am2), followed by Tv3. Th12, Th22, Tv1, Tv3 and SP inhibited the initiation and growth of rhizomorphs of Am2. Regeneration tests showed that both Am1 and Am2 attacked by Trichoderma isolates and SP were no longer viable. Th23 and SP were almost as effective in vivo as in vitro. But isolate Co of Chaetomium olivaceum, which was ineffective in vitro, was found effective in vivo. Conversely, Th2, which exhibited good antagonistic activity in vitro, performed poorly in vivo. These results show that the in vitro and in vivo efficacies of potential antagonists may not necessarily be closely correlated. Hence, there is a danger that potentially effective isolates may be discarded if decisions are made only on the basis of preliminary screening tests carried out under laboratory conditions.

PPAR gamma is a ligand regulated transcriptional factor that modulates the transcription of several genes involved in fat and sugar metabolism. Due to its easy bacterial expression and crystallization, several crystal structures of holo-PPAR gamma have been reported and deposited in the Protein Data Bank. Here, we investigated the three-dimensional electrostatic properties of 55 PPAR gamma ligands and used this information for clustering them through principal component analysis. We found out that, according to their electrostatic potential, these ligands can be separated in three groups, with different binding features. We also observed that non-selective and selective ligands show different 3D electrostatic properties and are separated in different clusters. The relevance of this analysis for the development of new binders is discussed. (C) 2010 Elsevier Masson SAS. All rights reserved.

This article describes the integration of the LSD (Logic for Structure Determination) and SISTEMAT expert systems that were both designed for the computer-assisted structure elucidation of small organic molecules. A first step has been achieved towards the linking of the SISTEMAT database with the LSD structure generator. The skeletal descriptions found by the SISTEMAT programs are now easily transferred to LSD as substructural constraints. Examples of the synergy between these expert systems are given for recently reported natural products.

New lanthanide complexes with benzeneseleninic (ABSe) and 4-chloro-benzeneseleninic (ABSeCl) acids have been synthesized and characterized by elemental analysis, infrared and UV-visible spectroscopies. The emission spectra of the trivalent europium complexes presented the typical electronic (5)D(0) -> (7)F(j) transitions of the ion (J = 0-4). The ground-state geometries of the europium complexes have been calculated by using the Sparkle/AM1 model. From these results, the 4f-4f intensity parameters and energies of the ligand singlet and triplet excited states have been obtained. The lower emission quantum yield for the [Eu(ABSe)(3)(H(2)O)(2)](H(2)O)(2) compound, as compared to the [Eu(Al(3)SeCl)(3)(H(2)O)(2)] one, can be associated to the higher numbers of water molecules, in the first and second coordination spheres, that contribute to the luminescence quenching. The [Eu(Al(3)Se)(3)(H(2)O)(2)](H(2)O)(2) complex presents an intermediate state whose energy difference with respect to the first excited singlet state is resonant with three phonons from the water molecules, favouring a multiphonon relaxation process from the singlet state followed by a fast internal conversion process; this effect is less pronounced in the complex with the ABSeCl ligand. The luminescence decay curves of the gadolinium complexes indicate that the level responsible for the intramolecular energy transfer process has a triplet character for both compounds. The nephelauxetic effect in these compounds was investigated under the light of a recently proposed covalency scale based on the concept of overlap polarizability of the chemical bond. In order to extend previous SAR and QSAR studies, 3D-QSAR analysis has been performed using CoMFA and CoMSIA approaches applied to a set of 39 alpha-(N)-heterocyclic carboxaldehydes thiosemicarbazones with their inhibitory activity values (IC(50)) evaluated against ribonucleotide reductase (RNR) of H.Ep.-2 cells (human epidermoid carcinoma), taken from selected literature. Both rigid and field alignment methods, taking the unsubstituted 2-formylpyridine thiosemicarbazone in its syn conformation as template, have been used to generate multiple predictive CoMFA and CoMSIA models derived from training sets and validated with the corresponding test sets. Acceptable predictive correlation coefficients (Q(cv)(2) from 0.360 to 0.609 for CoMFA and Q(cv)(2) from 0.394 to 0.580 for CoMSIA models) with high fitted correlation coefficients (r` from 0.881 to 0.981 for CoMFA and r(2) from 0.938 to 0.993 for CoMSIA models) and low standard errors (s from 0.135 to 0.383 for CoMFA and s from 0.098 to 0.240 for CoMSIA models) were obtained. More precise CoMFA and CoMSIA models have been derived considering the subset of thiosemicarbazones (TSC) substituted only at 5-position of the pyridine ring (n=22). Reasonable predictive correlation coefficients (Q(cv)(2) from 0.486 to 0.683 for CoMFA and Q(cv)(2) from 0.565 to 0.791 for CoMSIA models) with high fitted correlation coefficients (r(2) from 0.896 to 0.997 for CoMFA and r(2) from 0.991 to 0.998 for CoMSIA models) and very low standard errors (s from 0.040 to 0.179 for CoMFA and s from 0.029 to 0.068 for CoMSIA models) were obtained. The stability of each CoMFA and CoMSIA models was further assessed by performing bootstrapping analysis. For the two sets the generated CoMSIA models showed, in general, better statistics than the corresponding CoMFA models. The analysis of CoMFA and CoMSIA contour maps suggest that a hydrogen bond acceptor near the nitrogen of the pyridine ring can enhance inhibitory activity values. This observation agrees with literature data, which suggests that the nitrogen pyridine lone pairs can complex with the iron ion leading to species that inhibits RNR. The derived CoMFA and CoMSIA models contribute to understand the structural features of this class of TSC as antitumor agents in terms of steric, electrostatic, hydrophobic and hydrogen bond donor and hydrogen bond acceptor fields as well as to the rational design of this key enzyme inhibitors.

Este trabalho, consiste em um estudo para avaliar os níveis de vibrações do corpo humano, do segmento mão-braço, e a transmissibilidade a que estão sujeitos os motoristas de ônibus urbanos. As medições foram realizadas, no eixo vertical, de acordo com as recomendações das normas ISO 2631 (1974 e 1997) e ISO 5349 (1986), utilizando-se acelerômetros uni-axiais, um computador com placa conversora analógica para digital (A/D), e programas desenvolvidos na plataforma de programação gráfica HPVee para aquisição e avaliação dos níveis de vibrações. A vibração vertical do corpo humano (direção z – ISO 2631) foi simultaneamente medida no assento do motorista e no suporte do banco junto ao piso do veículo. A vibração no segmento mão-braço (direção x – ISO 5349) foi medida no terceiro metacarpal, no triquetrum carpal e no ombro dos motoristas. Diferentes modelos de ônibus foram utilizados em uma rota de 18km que interliga as zonas norte e sul da cidade. Quatro acelerômetros uni-axiais (Endevco Isotron 2250A/AM1-10) foram usados para medir os níveis de vibração na mão, pulso, ombro do motorista e no assento; enquanto que, no piso, foi utilizado um acelerômetro uni-axial da Brüel & Kjaer 4338. Os resultados indicaram que, para o segmento mão-braço, os níveis de vibrações equivalentes a 4 horas de exposição são menores do que 2,0 m/s2, na maioria dos modelos, o que determina que os motoristas expostos até 25 anos possivelmente não apresentarão problemas relacionados à vibração. Porém, na análise em freqüência, os níveis de vibrações, nas faixas de freqüência de 6,3 a 50Hz, no terceiro metacarpal e no triquetrum carpal são superiores a 2,0 m/s2 podendo ocasionar danos a partir de 15 anos de exposição. Os níveis de vibrações do corpo humano e a transmissibilidade mostraram que os motoristas estão expostos a níveis perigosos de vibrações principalmente na área da ressonância da coluna vertebral.

Este trabalho tem por objetivo determinar e avaliar a transmissibilidade da vibração no corpo humano além de desenvolver um modelo numérico de quatro graus de liberdade representativo do sistema banco e corpo humano. Para auxiliar no desenvolvimento deste trabalho foi realizado um resgate de pesquisas publicadas por pesquisadores anteriormente na área de modelamento numérico, além das medições e avaliações da transmissibilidade da vibração no corpo humano. Foram realizadas medições da transmissibilidade da vibração no corpo humano na posição sentado em um banco comumente utilizado para motoristas de ônibus urbanos. As medições foram realizadas ao longo do eixo z, de acordo com as recomendações das normas ISO 2631 (1974 e 1997), utilizando-se de acelerômetros uni-axiais, um computador com placa conversora analógica para digital (A/D), além de programas desenvolvidos na plataforma de programação gráfica HPVee para aquisição dos níveis de vibração e avaliação das transmissibilidades da vibração. A vibração vertical foi simultaneamente medida no corpo humano (direção z – ISO 2631). Para medir os níveis de vibração no assento, na pélvis, no ombro e na cabeça foram utilizados quatro micro-acelerômetros uni-axiais (Endveco Isotron 2250A/AM1-10) enquanto que no piso, foi utilizado um acelerômetro uni-axial da Brüel & Kjaer 4338. O grupo estudado nos experimentos era composto por cinco indivíduos, sendo três homens e duas mulheres. Avaliou-se a transmissibilidade entre o assento do banco e o piso, entre a pélvis o assento, entre o ombro e o assento e entre a cabeça e o assento. Os resultados indicaram que o sistema apresentou uma amplificação da transmissibilidade entre o assento e o piso em até 2,5 vezes, enquanto que, para as demais transmissibilidades (pélvis/assento, ombro/assento, cabeça/assento) houve uma atenuação gradual da transmissibilidade da vibração Um modelo linear de quatro graus de liberdade foi desenvolvido para representar o comportamento biodinâmico de indivíduos brasileiros submetidos à vibração forçada proveniente de sinais medidos em ônibus urbanos brasileiros. O modelo responde simultaneamente de acordo com os dados obtidos experimentalmente da transmissibilidade entre o assento e o piso e entre o ombro e o assento para uma faixa de freqüência de 4 até 40 Hz. Foi desenvolvida uma rotina no Maple 5.5 (anexo 10.4) a qual consiste em ajustar uma curva de transmissibilidade calculada com os limites estabelecidos dos parâmetros biomecânicos definidos nos ensaios biométricos e literatura às curvas de transmissibilidade obtidas experimentalmente. Os resultados indicaram que para a curva de transmissibilidade entre o assento e o ombro o erro foi de 37,78% enquanto que para as curvas de transmissibilidade entre o assento e o piso apresentaram um erro de 17,74 %. Apesar dos valores de erro percentual terem sido relativamente elevados, os valores de ambas as curvas de transmissibilidade numérica apresentaram resultados de ajuste muito próximos às curvas experimentais.

The Topliss method was used to guide a synthetic path in support of drug discovery efforts toward the identification of potent antimycobacterial agents. Salicylic acid and its derivatives, p-chloro, p-methoxy, and m-chlorosalicylic acid, exemplify a series of synthetic compounds whose minimum inhibitory concentrations for a strain of Mycobacterium were determined and compared to those of the reference drug, p-aminosalicylic acid. Several physicochemical descriptors (including Hammett's sigma constant, ionization constant, dipole moment, Hansch constant, calculated partition coefficient, Sterimol-L and -B-4 and molecular volume) were considered to elucidate structure-activity relationships. Molecular electrostatic potential and molecular dipole moment maps were also calculated using the AM1 semi-empirical method. Among the new derivatives, m-chlorosalicylic acid showed the lowest minimum inhibitory concentration. The overall results suggest that both physicochemical properties and electronic features may influence the biological activity of this series of antimycobacterial agents and thus should be considered in designing new p-aminosalicylic acid analogs.

The isotypes of RAR and RXR are retinoic acid and retinoid X acid receptors, respectively, whose ligand-binding domain contains the ligand-dependent activation function, with distinct pharmacological targets for retinoids, involved in the treatment of various cancers and skin diseases. Due to the major challenge which cancer treatment and cure still imposes after many decades to the international scientific community, there is actually considerable interest in new ligands with increased bioactivity. We have focused on the retinoid acid receptor, which is considered an interesting target for drug design. In this work, we carried out density functional geometry optimizations, and different docking procedures. We performed screening in a large database (hundreds of thousands of molecules which we optimized at the AM1 level) yielding a set of potential bioactive ligands. A new ligand was selected and optimized at the B3LYP/6-31G* level. A flexible docking program was used to investigate the interactions between the receptor and the new ligand. The result of this work is compared with several crystallographic ligands of RAR. Our theoretically more bioactive new-ligand indicates stronger and more hydrogen bonds as well as hydrophobic interactions with the receptor. (c) 2005 Wiley Periodicals, Inc.
