BACKGROUND: There is an increasing interest in obtaining natural products with bioactive properties, using fermentation technology. However, the downstream processing consisting of multiple steps can be complicated, leading to increase in the final cost of the product. Therefore there is a need for integrated, cost-effective and scalable separation processes. RESULTS: The present study investigates the use of colloidal gas aphrons (CGA), which are surfactant-stabilized microbubbles, as a novel method for downstream processing. More particularly, their application for the recovery of astaxanthin from the cells of Phaffia rhodozyma is explored. Research carried out with standard solutions of astaxanthin and CGA generated from the cationic surfactant hexadecyl. trimethyl ammonium bromide (CTAB) showed that up to 90% recovery can be achieved under optimum conditions, i.e., pH 11 with NaOH 0.2 mol L-1. In the case of the cells' suspension from the fermentation broth, three different approaches were investigated: (a) the conventional integrated approach where CGA were applied directly; (b) CGA were applied to the clarified suspension of cells; and finally (c) the in situ approach, where CGA are generated within the clarified suspension of cells. Interestingly, in the case of the whole suspension (approach a) highest recoveries (78%) were achieved under the same conditions found to be optimal for the standard solutions. In addition, up to 97% recovery of total carotenoids could be achieved from the clarified suspension after pretreatment with NaOH. This pretreatment led to maximum cell disruption as well as optimum conditioning for subsequent CGA separation. CONCLUSIONS: These results demonstrate the potential of CGA for the recovery of bioactive components from complex feedstock. (c) 2008 Society of Chemical Industry.

The model amyloid peptide AAKLVFF was expressed as a His-tagged fusion protein with the immunoglobulin-binding domain B1 of streptococcal protein G (GB1), a small (56 residues), stable, single-domain protein. It is shown that expression of this model amyloid peptide is possible and is not hindered by aggregation. Formylation side reactions during the CNBr cleavage are investigated via synthesis of selectively formylated peptides.

Irrigation is used frequently in potato cultivation to maximize yield, but water availability may also affect the composition of the crop, with implications for processing properties and food safety. Five varieties of potatoes, including drought-tolerant and -sensitive types, which had been grown with and without irrigation, were analyzed to show the effect of water supply on concentrations of free asparagine, other free amino acids, and sugars and on the acrylamide-forming potential of the tubers. Two varieties were also analyzed under more severe drought stress in a glasshouse. Water availability had profound effects on tuber free amino acid and sugar concentrations, and it was concluded that potato farmers should irrigate only if necessary to maintain the health and yield of the crop, because irrigation may increase the acrylamide-forming potential of potatoes. Even mild drought stress caused significant changes in composition, but these differed from those caused by more extreme drought stress. Free proline concentration, for example, increased in the field-grown potatoes of one variety from 7.02 mmol/kg with irrigation to 104.58 mmol/kg without irrigation, whereas free asparagine concentration was not affected significantly in the field but almost doubled from 132.03 to 242.26 mmol/kg in response to more severe drought stress in the glasshouse. Furthermore, the different genotypes were affected in dissimilar fashion by the same treatment, indicating that there is no single, unifying potato tuber drought stress response.

BACKGROUND: Chemical chitin extraction generates large amounts of wastes and increases partial deacetylation of the product. Therefore, the use of biological methods for chitin extraction is an interesting alternative. The effects of process conditions on enzyme assisted extraction of chitin from the shrimp shells in a systematic way were the focal points of this study. RESULTS: Demineralisation conditions of 25C, 20 min, shells-lactic acid ratio of 1:1.1 w/w; and shells-acetic acid ratio of 1:1.2 w/w, the maximum demineralisation values were 98.64 and 97.57% for lactic and acetic acids, respectively. A total protein removal efficiency of 91.10% by protease from Streptomyces griseus with enzyme-substrate ratio 55 U/g, pH 7.0 and incubation time 3 h is obtained when the particle size range is 50-25 μm, which was identified as the most critical factor. The X-ray diffraction and 13C NMR spectroscopy analysis showed that the lower percent crystallinity and higher degree of acetylation of chitin from enzyme assisted extraction may exhibit better solubility properties and less depolymerisation in comparison with chitin from the chemical extraction. CONCLUSION: The present work investigates the effects of individual factors on process yields, and it has shown that, if the particle size is properly controlled a reaction time of 3 h is more than enough for deproteination by protease. Physicochemical analysis indicated that the enzyme assisted production of chitin seems appropriate to extract chitin, possibly retaining its native structure.

In the seventies, Nolan was the first to address the need for a descriptive stage theory concerning the planning, organizing and controlling activities associated with managing the organizational computer resource. The arrival of newer technologies, such as those based on the Internet, calls for fresh approaches in terms of their implementation and management. Intranet technology, which is based on Internet technology, differs from other types of IT in terms of its characteristics, use and implementation. We propose a four-stage model for intranet implementation and management. Each stage is portrayed by seven general characteristics. We propose that, in order to ensure intranet institutionalization, three existential crises must be overcome. First, if a sponsor does not nurture the intranet, it cannot evolve beyond its experimental beginnings. Second, if a critical mass of both users and content cannot be reached simultaneously, the intranet will not progress. Finally, if the intranet remains uncontrolled, it will be perceived to be useless, and therefore users will abandon it.

Os debates e estudos sobre a importância do conhecimento organizacional têm sido cada vez mais freqüentes nos últimos anos. As empresas e universidades de negócios consideram a Gestão do Conhecimento uma vantagem competitiva para as organizações que conseguem estabelecer e aplicar modelos voltados para este fim. A bibliografia atual sobre este tema tem demonstrado duas tendências de propostas práticas para a Gestão do Conhecimento. A primeira enfatiza a importância do desenvolvimento de softwares responsáveis pela disseminação e estocagem dos conhecimentos organizacionais adquiridos de diversas fontes. A segunda, aplicada neste trabalho, é focada na capacidade e potencial dos indivíduos, e destaca a importância de criar e compartilhar conhecimentos através das pessoas. Este trabalho demonstra, a partir de um modelo de criação e gestão do conhecimento organizacional desenvolvido pelos professores Ikujiro Nonaka e Hirotaka Takeuchi, e denominado "Espiral do Conhecimento", uma aplicação prática de Gestão do Conhecimento numa equipe de representantes de vendas de uma indústria farmacêutica. Evidenciam-se, como conseqüência desta adaptação, novas rotinas do processo de "Espiral do Conhecimento", apresentando-se melhorias qualitativas e quantitativas nos resultados da equipe.

Esta pesquisa trata do processo de formação de estratégias de manufatura em empresas brasileiras pequenas e médias. Fazendo uso de extensa revisão bibliográfica, é desenvolvida uma ferramenta para o auxílio da tipificação e análise dos processos de formação de estratégias de manufatura, que utiliza duas principais dimensões: a direção predominante (top-down ou bottom-up) e a consideração ou não da existência de trade-offs entre critérios de desempenho no processo de formação das estratégias de manufatura. Esta ferramenta é então aplicada na análise em profundidade de quatro estudos de caso de empresas brasileiras de médio porte. Conclusões são tiradas e hipóteses formuladas sobre como as empresas brasileiras pequenas e médias formam suas estratégias de manufatura. Também com base nos achados dos estudos de caso e na revisão bibliográfica, são identificados 10 requisitos para um processo ideal de gestão da formação de estratégias de manufatura em empresas brasileiras pequenas e médias. Com base nestes requisitos, e como orientação à prática, é então desenvolvido e proposto um processo passo-a-passo para permitir que gestores das empresas brasileiras pequenas e médias possam de forma relativamente simples, gerenciar melhor seus processos de gestão da formação de suas estratégias de manufatura, beneficiando-se do estado da arte dos desenvolvimentos conceituais na área.

The aim of this work was to develop an efficient reactor for the production of low methoxyl pectin, using pectinmethylesterase (PME, EC 3.1.1.11) from acerola immobilized on silica. The immobilized enzyme was used in up to 50 successive bioconversion runs at 50 degrees C with an efficiency loss of less than 20%. The fixed-bed reactor (6.0 x 1.5 cm) was prepared using PME immobilized in glutaraldehyde-activated silica operated at 50 degrees C with an optimum flow rate of 10 mL h(-1). The bioconversion yield was shown to strongly depend on the nature of the enzymatic preparation. An efficiency of 44% was achieved when concentrated PME was used, compared with only 30% with purified PME, both after an 8-h run. The process described could provide the basis for the development of a commercial-scale process. (c) 2006 Society of Chemical Industry.

A partially purified extract of pectinmethylesterase (PME) from acerola fruit was immobilized on various supports: glass, celite, chrysotile, agarose, concanavalin A Sepharose 4B, egg shell, polyacrylamide and gelatin. In addition, reticulation with glutaraldehyde was assessed, as well as the use of gelatin in the presence of celite, glass and silica. The highest immobilization yields were obtained when the pectinmethylesterase was immobilized in concanavalin A Sepharose 4B (81.7%) and in gelatin-water (78.0%).
