Lectins have been classified into a structurally diverse group of proteins that bind carbohydrates and glycoconjugates with high specificity. They are extremely useful molecules in the characterization of saccharides, as drug delivery mediators, and even as cellular surface makers. In this study, we present camptosemin, a new lectin from Camptosema ellipticum. It was characterized as an N-acetyl-d-galactosamine-binding homo-tetrameric lectin, with a molecular weight around 26 kDa/monomers. The monomers were stable over a wide range of pH values and exhibited pH-dependent oligomerization. Camptosemin promoted adhesion of breast cancer cells and hemagglutination, and both activities were inhibited by its binding of sugar. The stability and unfolding/folding behavior of this lectin was characterized using fluorescence and far-UV circular dichroism spectroscopies. The results indicate that chemical unfolding of camptosemin proceeds as a two-state monomer-tetramer process. In addition, small-angle X-ray scattering shows that camptosemin behaves as a soluble and stable homo-tetramer molecule in solution.

Coal mining and incineration of solid residues of health services (SRHS) generate several contaminants that are delivered into the environment, such as heavy metals and dioxins. These xenobiotics can lead to oxidative stress overgeneration in organisms and cause different kinds of pathologies, including cancer. In the present study the concentrations of heavy metals such as lead, copper, iron, manganese and zinc in the urine, as well as several enzymatic and non-enzymatic biomarkers of oxidative stress in the blood (contents of lipoperoxidation = TBARS, protein carbonyls = PC, protein thiols = PT, alpha-tocopherol = AT, reduced glutathione = GSH, and the activities of glutathione S-transferase = GST, glutathione reductase = GR, glutathione peroxidase = GPx, catalase = CAT and superoxide dismutase = SOD), in the blood of six different groups (n = 20 each) of subjects exposed to airborne contamination related to coal mining as well as incineration of solid residues of health services (SRHS) after vitamin E (800 mg/day) and vitamin C (500 mg/day) supplementation during 6 months, which were compared to the situation before the antioxidant intervention (Avila et al., Ecotoxicology 18:1150-1157, 2009; Possamai et al., Ecotoxicology 18:1158-1164, 2009). Except for the decreased manganese contents, heavy metal concentrations were elevated in all groups exposed to both sources of airborne contamination when compared to controls. TBARS and PC concentrations, which were elevated before the antioxidant intervention decreased after the antioxidant supplementation. Similarly, the contents of PC, AT and GSH, which were decreased before the antioxidant intervention, reached values near those found in controls, GPx activity was reestablished in underground miners, and SOD, CAT and GST activities were reestablished in all groups. The results showed that the oxidative stress condition detected previously to the antioxidant supplementation in both directly and indirectly subjects exposed to the airborne contamination from coal dusts and SRHS incineration, was attenuated after the antioxidant intervention.

The fractionation through bioguided antileishmanial activity of the dichloromethane extract of Cassia fistula fruits (Leguminosae) led to the isolation of the active isoflavone biochanin A, identified by spectroscopic methods. This compound showed 50% effective concentration (EC(50)) value of 18.96 mu g/mL against promastigotes of Leishmania (L.) chagasi. The cytotoxicity of this substance against peritoneal macrophages resulted in an EC(50) value of 42.58 mu g/mL. Additionally, biochanin A presented an anti-Trypanosoma-cruzi activity, resulting in an EC(50) value of 18.32 mu g/mL and a 2.4-fold more effectiveness than benznidazole. These results contribute with novel antiprotozoal compounds for future drug design studies.

O presente trabalho teve por objetivo estudar o comportamento de 7 cultivares de milho (Zea mays L.) quanto à interferência da comunidade de plantas daninhas. Para tanto, os 7cultivares de milho (ESALQ VF-7, ESALQ VD-8, COMPOSTOS ARQUITETURA, FLINT e DENTADO, PIRANÃO e o híbrido CARGILL 501) foram submetidos a duas condições de interferência: com e sem, totalizando 14 tratamentos arranjados em esquema fatorial (7x2), instalados, no campo, em área experimental da FCAV/UNESP, Campus de Jaboticabal, seguindo o delineamento experimental de blocos ao acaso com 4 repetições. A comunidade infestante cujas espécies mais importantes foram SIDCO, BRAPL e ELEIN, estabeleceu-se a partir dos 14 dias após a semeadura, passando a interferir negativamente sobre a cultura a partir dos 35 dias. Os cultivares CD e CF apresentaram maior crescimento que os demais. A interferência da comunidade infestante reduziu a altura de inserção da primeira espiga; o comprimento e a circunferência das espigas, os pesos da espigas e dos grãos e a produção estimada da cultura, independente do cultivar. Os cultivares PIR e ARQ, independente da interferência, mostraram-se os menos produtivos. O cultivar Carg mostrou-se o mais produtivo mesmo quando sob interferência. O cultivar ARQ foi o mais sensível à interferência, enquanto PIR e VF-7 foram os menos sensíveis, podendo ser utilizados em programas de melhoramento genético visando tolerância a interferência de plantas daninhas.

Avaliou-se o efeito da adição de enzimas fibrolíticas (celulase e xilanase) sobre as taxas de passagem de partículas e de diluição e os parâmetros ruminais (pH e nitrogênio amoniacal) em animais consumindo dietas contendo silagem de milho e feno de tifton 85 (Cynodon spp.) como volumoso. Oito bovinos com fístulas no rúmen e no duodeno foram distribuídos em dois quadrados latinos 4 x 4, com os tratamentos distribuídos em esquema fatorial 2 x 2 (duas fontes de volumoso e adição ou não de enzimas). O complexo enzimático foi proveniente de fonte comercial e extraído dos fungos Aspergillus niger e Trichoderma longibrachiatum, sendo fornecido na proporção de 12 g/animal/dia, misturado à ração total. Para as taxas de passagem, foram utilizados como marcadores das fases sólida e líquida o cromo-mordente e o cobalto-EDTA, respectivamente. Não houve efeito das enzimas sobre o pH e as concentrações de N-NH3, que apresentaram médias de 6,5 e 22,10 mg/100 mL, respectivamente. A taxa de passagem de partículas no rúmen não foi influenciada pela suplementação enzimática, com médias de 3,4 e 3,0%/h para as dietas contendo silagem de milho e feno de tifton, respectivamente. A taxa de diluição aumentou de 10,07 para 11,84%/h com a adição de enzimas. A suplementação com enzimas fibrolíticas em dietas formuladas com silagem de milho e feno de tifton como volumoso praticamente não alterou os parâmetros avaliados.

The food industry is interested in natural products. Anthocyanins are phenolic antioxidants of great importance with health-relevant applications. Several studies have linked the intake of fruits and vegetables with reduced risk of chronic degenerative diseases because of its antioxidant properties. This study aimed to compare different strategies for obtaining natural pigments from red jambo (Syzygium malaccence) and analyze its functional potential. Two different strategies were studied: (1) solid-liquid extraction (SLE) in reactor with controlled parameters, (2) powder obtention. The investigation of the functional potential was conducted taking into account the total phenolic content (TPC), the antioxidant activity (AA), the total anthocyanins concentration (TA) and α-amylase and α-glucosidase inhibition. The best extracts obtained by SLE showed TPC of 174.15 mg GAE/100g, AA of 3.56 μmol Trolox eq/g and TA of 133.59 mg cyd-3-glu/100 g. The best results for the second strategy were TPC of 1024.22 mg GAE/100 g, AA of 29.03 μmol Trolox eq/g and TA of 1193.41 mg cyd-3- glu/100 g. It was observed moderate amylase inhibition (26.30%) and high glucosidase inhibitory activity (97.47%). Skin extracts showed, in general, superior results when compared to whole red jambo, with superior values for dehydrated products. Based on our result, red jambo can be considered as a rich source of phenolic antioxidants, as well on amylase and glucosidase inhibitors

The present work reports on the preparation of Al2O3-TiO2 ceramics by high-energy ball milling and sintering, varying the molar fraction in 1:1 and 3:1. The powder mixtures were processed in a planetary Fritsch P-5 ball mill using silicon nitride balls (10 mm diameter) and vials (225 mL), rotary speed of 250 rpm and a ball-to-powder weight ratio of 5:1. Samples were collected into the vial after different milling times. The milled powders were uniaxially compacted and sintered at 1300 and 1500 degrees C for 4h. The milled and sintered materials were characterized by X-ray diffraction and electron scanning microscopy (SEM). Results indicated that the intensity of Al2O3 and TiO2 peaks were reduced for longer milling times, suggesting that nanosized particles can be achieved. The densification of Al2O3-TiO2 ceramics was higher than 98% over the relative density in samples sintered at 1500 degrees C for 4h, which presented the formation of Al2TiO5.

The aim of this study was characterize the positivity of the bovine leukosis virus in the Microregion of the Serra de Botucatu. Sera from 1193 bovine from 65 properties of the Microregion of the Serra de Botucatu were evaluated throught ELISA test. All the evaluated animals were adult and 16 of them only were male; 85.5% were crossbred, 6.45% Nellore and 8% dutch. of the analyzed samples, 618 sera had resulted positive to the test. In only one flock it was not found seroreagents animals, the regional positivity was 52% (the seropositivity in the properties varied of 10% to 67%), the higher the percentage of positivity was in the animals of the dutch race (94.7%), followed for the crossbred (43.7%). The high percentage of positivity of the disease in our region is distinguished.

O objetivo do presente trabalho foi avaliar uma equação de predição das exigências de proteína bruta (PB) para reprodutoras pesadas na fase de produção. O experimento foi realizado com 600 aves reprodutoras pesadas, Hubbard HI-Y, durante o período de 31 a 46 semanas de idade, alojadas em boxes num delineamento experimental inteiramente casualizado com três tratamentos e cinco repetições de 40 aves. Os tratamentos consistiram de: T1- Fornecimento de PB de acordo com o manual da linhagem (controle), T2- Fornecimento de PB de acordo com a equação de predição determinada, utilizando os dados de desempenho médio das aves do tratamento controle para predizer as exigências e T3- Fornecimento de PB de acordo com a equação de predição determinada, utilizando os dados de desempenho de cada parcela experimental para predizer as exigências, onde a equação de predição avaliada foi: PB=2,282.P0,75+0,356.G+0,262.MO, sendo PB a exigência de proteína bruta (g/ave/dia), P o peso corporal (kg), G o ganho de peso (g) e MO a massa de ovos (g). As rações foram formuladas para atender as exigências nutricionais e quando necessário eram incluídos os aminoácidos sintéticos, metionina, lisina, triptofano, treonina e arginina. As aves alimentadas de acordo com a equação ingeriram menores quantidades de proteína (20,8g/dia) quando comparadas às alimentadas de acordo com as recomendações (23,80g), entretanto isto levou a menores pesos dos ovos refletindo no peso dos pintos. A equação de predição proporcionou melhores resultados quanto à eficiência protéica. Assim, concluiu-se que a equação de predição não forneceu a quantidade mínima de proteína bruta para atender as exigências dos aminoácidos não suplementados na dieta.
