The creation of three-dimensional (3D) drawings for proposed designs for construction, re-construction and rehabilitation activities are becoming increasingly common for highway designers, whether by department of transportation (DOT) employees or consulting engineers. However, technical challenges exist that prevent the use of these 3D drawings/models from being used as the basis of interactive simulation. Use of driving simulation to service the needs of the transportation industry in the US lags behind Europe due to several factors, including lack of technical infrastructure at DOTs, cost of maintaining and supporting simulation infrastructure—traditionally done by simulation domain experts—and cost and effort to translate DOT domain data into the simulation domain.

A autora comenta evidências obtidas por pesquisa de avaliação da educação infantil no Brasil, relacionando-as ao debate sobre a qualidade dos serviços educacionais oferecidos às crianças pequenas em âmbito internacional. Refere-se a dados de inúmeros estudos realizados no exterior, os quais indicam que a boa qualidade dos programas de educação infantil impacta o desenvolvimento das crianças a longo prazo. Sugere que, para alcançar maiores níveis de qualidade, é necessário pensar a educação como um sistema constituído de duas partes principais: os programas de educação infantil e a infraestrutura para mantê-los

Bacterial reporters are live, genetically engineered cells with promising application in bioanalytics. They contain genetic circuitry to produce a cellular sensing element, which detects the target compound and relays the detection to specific synthesis of so-called reporter proteins (the presence or activity of which is easy to quantify). Bioassays with bacterial reporters are a useful complement to chemical analytics because they measure biological responses rather than total chemical concentrations. Simple bacterial reporter assays may also replace more costly chemical methods as a first line sample analysis technique. Recent promising developments integrate bacterial reporter cells with microsystems to produce bacterial biosensors. This lecture presents an in-depth treatment of the synthetic biological design principles of bacterial reporters, the engineering of which started as simple recombinant DNA puzzles, but has now become a more rational approach of choosing and combining sensing, controlling and reporting DNA 'parts'. Several examples of existing bacterial reporter designs and their genetic circuitry will be illustrated. Besides the design principles, the lecture also focuses on the application principles of bacterial reporter assays. A variety of assay formats will be illustrated, and principles of quantification will be dealt with. In addition to this discussion, substantial reference material is supplied in various Annexes.

O artigo analisa as diferentes modalidades de ações afirmativas raciais em vigor nas universidades públicas brasileiras até 2012. A partir de um levantamento das leis e resoluções que regulamentam tais políticas, cruzadas com microdados produzidos pelo Instituto Nacional de Estudos e Pesquisas Educacionais, traçamos um panorama da ação afirmativa no país. Nosso esforço tem por objetivo explicitar os diferentes diagnósticos acerca das desigualdades educacionais no Brasil em que as universidades se baseiam para elaborar suas políticas e investigar de que maneira adotam procedimentos para mitigar iniquidades na admissão aos seus cursos, lidando com o desafio de transformar categorias sociológicas como classe, raça e etnicidade, bem como demandas governamentais e de movimentos sociais em critérios norteadores de políticas públicas.

Este artigo discute os rumos da política pública de inclusão digital nas escolas com base nos resultados da pesquisa de avaliação de impacto e de processo do Projeto UCA-Total (Um Computador por Aluno) realizada em cinco municípios onde foi implementado no decorrer de 2010 e 2011. A avaliação de impacto, de caráter longitudinal, foi feita através de um survey domiciliar aplicado em duas ondas. A pesquisa de processo associou diversas metodologias qualitativas, como grupos focais, entrevistas e visitas a campo. Limita-se aqui aos resultados derivados da pesquisa de painel, que revelam o baixo nível de aproveitamento do Projeto e um padrão de funcionamento bastante divergente em razão de vantagens e debilidades locais. Seu enorme potencial não foi até hoje completamente aproveitado.

Indoleamine 2,3-dioxygenase 1 (IDO1) is an important therapeutic target for the treatment of diseases such as cancer that involve pathological immune escape. Starting from the scaffold of our previously discovered IDO1 inhibitor 4-phenyl-1,2,3-triazole, we used computational structure-based methods to design more potent ligands. This approach yielded highly efficient low molecular weight inhibitors, the most active being of nanomolar potency both in an enzymatic and in a cellular assay, while showing no cellular toxicity and a high selectivity for IDO1 over tryptophan 2,3-dioxygenase (TDO). A quantitative structure-activity relationship based on the electrostatic ligand-protein interactions in the docked binding modes and on the quantum chemically derived charges of the triazole ring demonstrated a good explanatory power for the observed activities.

Abstract: As a part of an innovation project funded by the Federal Highway Administration (FHWA) Highways for LIFE program, a full-depth precast, ultra-high-performance concrete (UHPC) waffle deck panel and appropriate connections suitable for field implementation of waffle decks were developed. Following a successful full-scale validation test on a unit consisting of two panels with various types of connections under laboratory conditions, the waffle deck was installed successfully on a replacement bridge in Wapello County, Iowa. The subsequent load testing confirmed the desirable performance of the UHPC waffle deck bridge. Using the lessons from the completed project and outcomes from a series of simple and detailed finite element analyses of waffle decks, this report was developed to serve as a guide for broadening the design and installation of the UHPC waffle deck panel in new and existing bridges. Following an introduction to UHPC and waffle deck panels and a summary of completed work, this document presents information on waffle deck design, design of connections, redecking using waffle deck panels, and guidance on precast fabrication, construction, and installation of UHPC waffle deck panels.

The purpose of this study was to investigate the effect of cement paste quality on the concrete performance, particularly fresh properties, by changing the water-to-cementitious materials ratio (w/cm), type and dosage of supplementary cementitious materials (SCM), and airvoid system in binary and ternary mixtures. In this experimental program, a total matrix of 54 mixtures with w/cm of 0.40 and 0.45; target air content of 2%, 4%, and 8%; a fixed cementitious content of 600 pounds per cubic yard (pcy), and the incorporation of three types of SCMs at different dosages was prepared. The fine aggregate-to- total aggregate ratio was fixed at 0.42. Workability, rheology, air-void system, setting time, strength, Wenner Probe surface resistivity, and shrinkage were determined. The effects of paste variables on workability are more marked at the higher w/cm. The compressive strength is strongly influenced by the paste quality, dominated by w/cm and air content. Surface resistivity is improved by inclusion of Class F fly ash and slag cement, especially at later ages. Ternary mixtures performed in accordance with their ingredients. The data collected will be used to develop models that will be part of an innovative mix proportioning procedure.

This guide specification and commentary for concrete pavements presents current state-of-the art thinking with respect to materials and mixture selection, proportioning, and acceptance. This document takes into account the different environments, practices, and materials in use across the United States and allows optional inputs for local application. The following concrete pavement types are considered: jointed plain concrete pavement, the most commonly used pavement type and may be doweled or non-doweled at transverse joints; and continuously reinforced concrete pavement, typically constructed without any transverse joints, typically used for locations with high truck traffic loads and/or poor support conditions.
