The cellular FLICE inhibitory protein (c-FLIP) is an endogenous inhibitor of the caspase-8 proapoptotic signaling pathway downstream of death receptors. Recent evidence indicates that the long form of c-FLIP (c-FLIP(L)) is required for proliferation and effector T-cell development. However, the role of c-FLIP(L) in triggering autoimmunity has not been carefully analyzed. We now report that c-FLIP(L) transgenic (Tg) mice develop splenomegaly, lymphadenopathy, multiorgan infiltration, high titers of auto-antibodies, and proliferative glomerulonephritis with immune complex deposition in a strain-dependent manner. The development of autoimmunity requires CD4(+) T cells and may result from impaired thymic selection. At the molecular level, c-FLIP(L) overexpression inhibits the zeta chain-associated protein tyrosine kinase of 70 kDa (ZAP-70) activation, thus impairing the signaling pathway derived from ZAP-70 required for thymic selection. Therefore, we have identified c-FLIP(L) as a susceptibility factor under the influence of epistatic modifiers for the development of autoimmunity.

O objetivo deste trabalho foi avaliar a tolerância de estirpes de Cupriavidus necator a zinco, cádmio, cobre e chumbo, além de determinar a eficiência simbiótica das estirpes mais tolerantes em associação a espécies leguminosas com potencial para revegetação. A tolerância foi testada em meio LB, suplementado com 2,5; 5,0; 7,5; 10; 12,5 e 15 mmol L-1 de ZnSO4.7H2O, CdSO4.8H2O, CuSO4.5H2O e PbCl2, respectivamente, em comparação ao controle sem adição de metal. Determinou-se a eficiência simbiótica das quatro estirpes de C. necator mais tolerantes aos metais avaliados (UFLA02-71, UFLA02-73, UFLA01-659 e UFLA01-663), as quais foram inoculadas nas espécies: Leucaena leucocephala, Enterolobium contortisiliquum, Acacia mangium, Mimosa caesalpiniifolia, M. pudica, M. pigra e M. acutistipula. Em vasos com solos, avaliaram-se L. leucocephala, M. pudica e M. caesalpiniifolia e as estirpes UFLA01-659 e UFLA02-71, selecionadas na avaliação de eficiência simbiótica. A estirpe UFLA02-71 proporcionou incrementos de matéria seca da parte aérea de 870% em M. caesalpiniifolia, enquanto que UFLA01-659 proporcionou 885% em M. pudica e 924% em L. leucocephala. As estirpes UFLA01-659 e UFLA02-71, além da alta tolerância a metais pesados, apresentaram eficiência em fixar nitrogênio, em simbiose com essas leguminosas, em solos com rizóbios nativos capazes de nodulá-las, e devem ser avaliadas quanto ao seu potencial de utilização em programas de recuperação de áreas degradadas.

O objetivo deste trabalho foi parametrizar e avaliar o modelo DSSAT/Canegro para cinco variedades brasileiras de cana-de-açúcar. A parametrização foi realizada a partir do uso de dados biométricos e de crescimento das variedades CTC 4, CTC 7, CTC 20, RB 86-7515 e RB 83-5486, obtidos em cinco localidades brasileiras. Foi realizada análise de sensibilidade local para os principais parâmetros. A parametrização do modelo foi feita por meio da técnica de estimativa da incerteza de probabilidade generalizada ("generalized likelihood uncertainty estimation", Glue). Para a avaliação das predições, foram utilizados, como indicadores estatísticos, o coeficiente de determinação (R²), o índice D de Willmott e a raiz quadrada do erro-médio (RMSE). As variedades CTC apresentaram índice D entre 0,870 e 0,944, para índice de área foliar, altura de colmo, perfilhamento e teor de sacarose. A variedade RB 83-5486 apresentou resultados similares para teor de sacarose e massa de matéria fresca do colmo, enquanto a variedade RB 86-7515 apresentou valores entre 0,665 e 0,873, para as variáveis avaliadas.

Background: Myotragus balearicus was an endemic bovid from the Balearic Islands (Western Mediterranean) that became extinct around 6,000-4,000 years ago. The Myotragus evolutionary lineage became isolated in the islands most probably at the end of the Messinian crisis, when the desiccation of the Mediterranean ended, in a geological date established at 5.35 Mya. Thus, the sequences of Myotragus could be very valuable for calibrating the mammalian mitochondrial DNA clock and, in particular, the tree of the Caprinae subfamily, to which Myotragus belongs. Results: We have retrieved the complete mitochondrial cytochrome b gene (1,143 base pairs), plus fragments of the mitochondrial 12S gene and the nuclear 28S rDNA multi-copy gene from a well preserved Myotragus subfossil bone. The best resolved phylogenetic trees, obtained with the cytochrome b gene, placed Myotragus in a position basal to the Ovis group. Using the calibration provided by the isolation of Balearic Islands, we calculated that the initial radiation of caprines can be dated at 6.2 ± 0.4 Mya. In addition, alpine and southern chamois, considered until recently the same species, split around 1.6 ± 0.3 Mya, indicating that the two chamois species have been separated much longer than previously thought. Conclusion: Since there are almost no extant endemic mammals in Mediterranean islands, the sequence of the extinct Balearic endemic Myotragus has been crucial for allowing us to use the Messinian crisis calibration point for dating the caprines phylogenetic tree.

INTRODUCTION: Dietary supplement (DS) use increased rapidly in recent years. However, evidence of benefits of many DSs for healthy users is scarce and may not equate with known risks of overdose, drug interaction and recently discovered negative long-term effects. This exploratory study aimed to investigate the perceptions and motivations of DS users in Lausanne, Switzerland. METHOD: A convenience sample (n = 147) was recruited at the entrances of local sales points. Data were collected in on-site semistructured interviews that assessed dietary supplementation habits. RESULTS: The majority of DSs were all-in-one products, containing a mixture of minerals and vitamins, or products containing only minerals. Among the 147 users, 72 (49%) used one all-in-one product and 3 (2%) used two all-in-one products. Thirty-one (21%) consumers did not know for at least one product what the purpose of their DS use was. Seventy-five percent of participants thought that DS use presents no risk or nearly no risk. Only 49% of participants stated that their physicians were informed about their consumption. Although men searched more often for potential risks (p <0.001), they turned less frequently to health professionals to get this information (p = 0.007). DISCUSSION: As in other surveys performed elsewhere, our study shows that, in Lausanne (Switzerland), DSs are commonly used as mixed products. Risk perception seems generally low among DS users. Physicians should be trained to evaluate patients' perceived needs and DS consumption in order to provide good evidence-based information or to propose alternatives to DS use.
