Apple (Malus domestica Borkh.) trees, cultivars Gala and Fuji, were sprayed at full bloom with thidiazuron (TDZ) at doses of 0, 5, 10, or 20 g ha-1 of a.i. in order to evaluate its effects on plant growth and development, return bloom, fruit set, nutrition, and fruit yield. Fruit set increased with TDZ dose in 'Gala' but not in 'Fuji'. TDZ did not affect fruit yield in any cultivar. In 'Gala', the return bloom was reduced in about 50% at TDZ doses of 10 or 20 g ha-1. TDZ increased shoot growth in both cultivars. In leaves, TDZ decreased the concentration of Ca and Mg in 'Gala' and of Mg in 'Fuji', but did not affect the chlorophyll content, leaf area, length, width, and dry mass per cm² regardless of cultivar. In fruits, the effect of TDZ varied according to the portion evaluated. Highest doses of TDZ decreased the concentration of Ca and K in 'Gala' and of K in 'Fuji' in the entire fruits (flesh + skin); in the skin, highest doses of TDZ reduced the levels of N, Ca, and Mg in both cultivars, in addition to the level of K in 'Fuji'.

Background:Besides tobacco and alcohol, dietary habits may have a relevant role in oral cavity and pharyngeal (OCP) cancer.Methods:We analysed the role of selected food groups and nutrients on OCP cancer in a case-control study carried out between 1997 and 2009 in Italy and Switzerland. This included 768 incident, histologically confirmed squamous cell carcinoma cases and 2078 hospital controls. Odds ratios (ORs) were estimated using logistic regression models including terms for tobacco, alcohol and other relevant covariates.Results:Significant inverse trends in risk were observed for all vegetables (OR=0.19, for the highest vs the lowest consumption) and all fruits (OR=0.39), whereas significant direct associations were found for milk and dairy products (OR=1.50), eggs (OR=1.71), red meat (OR=1.55), potatoes (OR=1.85) and desserts (OR=1.68), although trends in risk were significant only for potatoes and desserts. With reference to nutrients, significant inverse relations were observed for vegetable protein (OR=0.45, for the highest vs the lowest quintile), vegetable fat (OR=0.54), polyunsaturated fatty acids (OR=0.53), α-carotene (OR=0.51), β-carotene (OR=0.28), β-cryptoxanthin (OR=0.37), lutein and zeazanthin (OR=0.34), vitamin E (OR=0.26), vitamin C (OR=0.40) and total folate (OR=0.34), whereas direct ones were observed for animal protein (OR=1.57), animal fat (OR=2.47), saturated fatty acids (OR=2.18), cholesterol (OR=2.29) and retinol (OR=1.88). Combinations of low consumption of fruits and vegetables, and high consumption of meat with high tobacco and alcohol, led to 10- to over 20-fold excess risk of OCP cancer.Conclusion:Our study confirms and further quantifies that a diet rich in fruits and vegetables and poor in meat and products of animal origin has a favourable role against OCP cancer.

The cross-recognition of peptides by cytotoxic T lymphocytes is a key element in immunology and in particular in peptide based immunotherapy. Here we develop three-dimensional (3D) quantitative structure-activity relationships (QSARs) to predict cross-recognition by Melan-A-specific cytotoxic T lymphocytes of peptides bound to HLA A*0201 (hereafter referred to as HLA A2). First, we predict the structure of a set of self- and pathogen-derived peptides bound to HLA A2 using a previously developed ab initio structure prediction approach [Fagerberg et al., J. Mol. Biol., 521-46 (2006)]. Second, shape and electrostatic energy calculations are performed on a 3D grid to produce similarity matrices which are combined with a genetic neural network method [So et al., J. Med. Chem., 4347-59 (1997)] to generate 3D-QSAR models. The models are extensively validated using several different approaches. During the model generation, the leave-one-out cross-validated correlation coefficient (q (2)) is used as the fitness criterion and all obtained models are evaluated based on their q (2) values. Moreover, the best model obtained for a partitioned data set is evaluated by its correlation coefficient (r = 0.92 for the external test set). The physical relevance of all models is tested using a functional dependence analysis and the robustness of the models obtained for the entire data set is confirmed using y-randomization. Finally, the validated models are tested for their utility in the setting of rational peptide design: their ability to discriminate between peptides that only contain side chain substitutions in a single secondary anchor position is evaluated. In addition, the predicted cross-recognition of the mono-substituted peptides is confirmed experimentally in chromium-release assays. These results underline the utility of 3D-QSARs in peptide mimetic design and suggest that the properties of the unbound epitope are sufficient to capture most of the information to determine the cross-recognition.

The objective of this work was to characterize mandarin (Citrus spp.) germplasm from Southern Brazil by morphological and molecular analyses. Thirty seven cultivars from 34 distinct mandarin varieties were evaluated by morphological and agronomic traits of leaves, flowers and fruits, and by microsatellite markers. The morphological and agronomic characteristics suggested that almost all varieties can be produced for commercial use, and some, as the Satsuma variety, are recommended for breeding programs. Pooled DNA samples from 1-5 plants belonging to each cultivar were tested. Eight of the nine primers detected polymorphisms. Specific markers were found for some accessions. The dendrogram constructed with the morphological results divided the 37 cultivars into four groups, while that obtained with the microsatellites clustered 35 of the 37 cultivars into three groups only. Generally, intervarietal differences are not high, and this lack of agreement in the two multifactorial analyses indicates that diverse evolutionary factors are acting at these two levels of investigation.

Els resultats de l'estudi arqueobotànic realitzat sobre diferents àmbits de l'assentament de Sant Jaume aporta noves dades al coneixement de la gestió dels recursos vegetals de la protohistòria del nordest peninsular. El registre arqueològic constata una certa homogeneïtzació en la funcionalitat dels àmbits, que s'interpreten com a magatzems en el pis superior i espais d'estabulació a l'inferior. L'àmbit millor representat és l'A4, que conserva com a fet singular un elevat nombre de restes de llavors i fruits amb molt bon estat de conservació. Els tàxons conreats més importants són la pisana (Triticum dicoccum) i les guixes i guixols (Lathyrus sativus i Lathyrus cicera), seguits d¿una menor proporció d'ordi vestit (Hordeum vulgare), blat nu (comú/dur) (Triticum aestivum/durum), pèsol (Pisum sativum), llentia (Lens culinaris) i erb (Vicia ervilia). D'altra banda, el registre arqueobotànic del pis inferior de l'àmbit A4 sembla posar en evidència la presència de l¿associació de pisana i guixa, destinada a l'alim.

The inhibition of ethylene action by 1-methylcyclopropene (1-MCP) extends shelf and storage life of many climacteric fruits. However, 1-MCP appears to have limited effects on stone fruit depending on specie and cultivar. The effects of 1-MCP on ripening and quality of 'Laetitia' plums were determined during ripening at 23ºC following harvest and cold storage. Japanese plums (Prunus salicina, cv. Laetitia) were harvested at mature pre-climacteric stage, cooled to 2ºC within 36 hours of harvest and then treated with 0, 0.05, 0.10, 0.50 or 1.00 muL L-1 of 1-MCP at 1°C for 24 hours. Following treatment, fruits were either held at 23ºC for 16 days or stored at 1ºC for 50 days. Fruits were removed from cold storage at 10-day intervals and allowed to ripe at 23°C for five days. A delay of climacteric respiration and ethylene production by 1-MCP treatment during ripening following harvest and cold storage was associated to a slow rate of fruit softening. 1-MCP treatment also delayed the loss of titratable acidity and changes of flesh and skin color, whereas it had little or no effect on soluble solids content. 1-MCP effects were concentration- and storage duration-dependent and, generally, a saturation fruit response to 1-MCP occurred between 0.5 and 1.0 muL L-1. During ripening, 1-MCP treated fruits attained quality similar to that of controls. Results indicated that 1-MCP treatment may extend shelf life (23ºC) and storage life (1ºC) of 'Laetitia' plums by approximately six and 20 days, respectively.

The search for techniques that extend shelf life of guava (Psidium guajava) fruits, and reduce its postharvest losses is desirable. The objective of this work was to evaluate the effects of concentrations of competitive ethylene antagonist 1-methylcyclopropene (1-MCP) on conservation of 'Pedro Sato' guava fruits. Treatments consisted of 0, 100, 300 or 900 nL L-1 of 1-MCP for 3 hours followed by storage at 25ºC, and 10ºC with 90±5% RH. The application of 900 nL L-1 1-MCP for 3 hours was efficient in delaying loss of skin color, and in keeping fruit firmness at both storage temperatures. The 300 nL L-1 of 1-MCP concentration was efficient in delaying skin color loss only when fruits were stored at 10ºC. The effect of 1-MCP was quite significant on the reduction of rot incidence at both storage temperatures. The respiratory rate was lower in fruits treated with 300 and 900 nL L-1 of 1-MCP during storage at 25ºC. The product was efficient in delaying the ripening of fruits, and the concentration of 900 nL L-1 showed the best effect.

The present study was carried out in Northeastern Region of Brazil, in order to investigate the use of stingless bee Melipona subnitida Ducke in the pollination of greenhouse sweet pepper (Capsicum annuum L.). Treatments of hand cross-pollination, hand self-pollination, pollination by bees and restricted pollination were performed. Results showed that despite sweet pepper flowers are considered autogamous, this crop benefits from pollination by M. subnitida, producing fruits significantly heavier and wider, containing a greater number of seeds and of better quality (lower percentage of malformed fruits) than self-pollinated sweet pepper. Thus, M. subnitida can be considered an efficient pollinator of greenhouse sweet pepper.

En la actualidad, las cooperativas recolectan, seleccionan, tratan y separan la fruta según su calibre (peso, diámetro máximo, medio y/o mínimo) para que esta llegue al consumidor final según la categoría (calibre). Para poder competir en un mercado cada vez más exigente en calidad y precios, se requieren sistemas de clasificación automáticos que nos permitan obtener óptimos resultados con altos niveles de producción y productividad. Para realizar estas tareas existen calibradoras industriales que pesan la fruta mediante células de carga y con el peso obtenido las clasifican asignando las piezas a su salida correspondiente (mesa de confección) a través de un sistema de electroimanes. Desafortunadamente el proceso de calibración de la fruta por peso no es en absoluto fiable ya que en este proceso no se considera el grosor de la piel, contenido de agua, de azúcar u otros factores altamente relevantes que influyen considerablemente en los resultados finales. El objeto de este proyecto es el de evolucionar las existentes calibradoras de fruta instalando un sistema industrial de visión artificial (rápido y robusto) que trabaje en un rango de espectro Infrarrojo (mayor fiabilidad) proporcionando óptimos resultados finales en la clasificación de las frutas, verduras y hortalizas. De este modo, el presente proyecto ofrece la oportunidad de mejorar el rendimiento de la línea de clasificación de frutas, aumentando la velocidad, disminuyendo pérdidas en tiempo y error humano y mejorando indiscutiblemente la calidad del producto final deseada por los consumidores.

Recent studies on coffee (Coffea arabica L.) cultivation in agroforestry systems in Southern Brazil have shown the potential of partial shading to improve management of this crop. The objective of this work was to evaluate microclimatic conditions and their effects on coffee production of plants shaded with pigeon pea (Cajanus cajan) in comparison to unshaded ones, from May 2001 to August 2002 in Londrina, State of Paraná, Brazil. The appraised microclimatic characteristics were: global radiation, photosynthetic and radiation balance; air, leaf and soil temperatures; and soil humidity. Shading caused significant reduction in incident global solar radiation, photosynthetically active radiation and net radiation, and attenuated maximum leaf, air and soil temperatures, during the day. Shade also reduced the rate of cooling of night air and leaf temperatures, especially during nights with radiative frost. Soil moisture at 0-10 cm depth was higher under shade. The shaded coffee plants produced larger cherries due to slower maturation, resulting in larger bean size. Nevertheless, plants under shade emitted less plagiotropic branches, with smaller number of nodes per branch, and fewer nodes with fruits, resulting in a large reduction in coffee production. These results show the need to find an optimal tree density and management that do not compromise coffee production and protect against extreme temperatures.

Rapport de synthèseCette recherche s'intéresse à investiguer d'une part si les résidents en Suisse entre 15 et 24 ans suivent les recommandations nutritionnelles actuelles et d'autre part, s'il y a des différences entre ceux en surpoids et ceux en poids normal. Les données sont tirées de l'Enquête Suisse sur la Santé ESS 2007. Pour notre étude, seules les 1813 personnes âgées entre 15 et 24 ans ont été sélectionnées.Dans une première analyse bivariée, l'adhérence aux recommandations alimentaires a été comparée entre hommes et femmes. Les quantités proposées par le questionnaire ESS sont: 1 à 2 portions de fruits, 3 portions de légumes et 3 portions de produits laitiers par jour, ainsi qu'un maximum de 5 portions de viande et au moins une portion de poisson par semaine. Ces quantités sont en accord avec les recommandations de la Société Suisse de Nutrition.L'adhérence aux recommandations alimentaires est plutôt basse quelque soit le genre, en particulier pour les légumes et les produits laitiers. Les femmes ont tendance à manger les quantités recommandées de fruits, légumes et viande, tandis que les hommes ont tendance à manger la quantité recommandée de produits laitiers. Pour le poisson aucune différence n'a été observée entre personnes du sexe opposé.Ensuite, les personnes en surpoids ont été comparées dans une analyse bivariée aux personnes en poids normal, en fonction du sexe. Plusieurs facteurs ont été considérés: participation à une activité physique avec essoufflement pendant au mois 20 minutes et au moins 4 jours par semaine, attitude envers leur poids corporel (satisfaction avec le poids, désir de perdre du poids et régime suivi pour perdre du poids), consommation d'alcool, de cigarettes et présence d'un état dépressif majeur. Finalement, des facteurs potentiellement confondants (âge, nationalité et lieu de domicile) ont été inclus dans une analyse multivariée.Concernant l'adhérence aux recommandations alimentaires, la seule différence significative est une consommation de légumes plus basse chez les femmes en surpoids. Seulement 4.2% des femmes en surpoids mangent 3 portions de légumes ou plus par jour comparé à 18.1% des femmes avec un poids normal. La consommation de produits laitiers est très basse dans tous les groupes, seulement environ 10% des répondants mangent les 3 portions recommandées de produits laitiers.Pour le niveau d'activité physique, aucune différence significative n'a été observée. L'analyse bivariée montre que les femmes en surpoids mangent moins de légumes, sont moins satisfaites de leur corps et ont plus souvent suivi un régime pour perdre du poids. Cependant elles ne souhaitent pas plus souvent perdre du poids en comparaison des femmes en poids normal. Les hommes en surpoids sont moins satisfaits de leur corps, désirent plus souvent perdre du poids, mais n'ont pas plus souvent suivi un régime que les hommes ayant un poids normal.Ces résultats montrent qu'il est difficile de suivre les recommandations alimentaires pour les 15-24 ans. Une recherche plus approfondie est nécessaire afin de déterminer comment ces recommandations pourraient être mieux adaptées à la vie quotidienne des jeunes. La faible consommation de produits laitiers est préoccupante, vu que la période entre l'adolescence et l'âge adulte est un moment crucial pour le développement de la densité osseuse. De nouvelles stratégies doivent être trouvées pour améliorer la consommation de produits laitiers chez les jeunes. Comme le comportement alimentaire et l'attitude envers le poids corporel diffèrent beaucoup selon le sexe, les programmes de prévention de surpoids devraient cibler les jeunes spécifiquement par sexe.

Myeloid cell leukemia-1 (MCL1) is an anti-apoptotic member of the BCL2 family that is deregulated in various solid and hematological malignancies. However, its role in the molecular pathogenesis of diffuse large B-cell lymphoma (DLBCL) is unclear. We analyzed gene expression profiling data from 350 DLBCL patient samples and detected that activated B-cell-like (ABC) DLBCLs express MCL1 at significantly higher levels compared with germinal center B-cell-like DLBCL patient samples (P=2.7 × 10(-10)). Immunohistochemistry confirmed high MCL1 protein expression predominantly in ABC DLBCL in an independent patient cohort (n=249; P=0.001). To elucidate molecular mechanisms leading to aberrant MCL1 expression, we analyzed array comparative genomic hybridization data of 203 DLBCL samples and identified recurrent chromosomal gains/amplifications of the MCL1 locus that occurred in 26% of ABC DLBCLs. In addition, aberrant STAT3 signaling contributed to high MCL1 expression in this subtype. Knockdown of MCL1 as well as treatment with the BH3-mimetic obatoclax induced apoptotic cell death in MCL1-positive DLBCL cell lines. In summary, MCL1 is deregulated in a significant fraction of ABC DLBCLs and contributes to therapy resistance. These data suggest that specific inhibition of MCL1 might be utilized therapeutically in a subset of DLBCLs.

Over three years the postharvest quality of 'Marli' peaches harvested from the integrated (IFP) and conventional production (CFP) systems was evaluated. The peaches were harvested from commercial orchards of Prunus persica at two locations close to the city of São Jerônimo, RS, Brazil, and stored at 0.5°C for 10, 20 or 30 days. The peaches were evaluated at harvest, at retrieval from storage and after ripening periods at 20°C. No differences in fruit weight losses were determined. Decay incidence was low, and no differences were detected amongst systems in both 2001 and 2002 seasons, but in the 2000 season CFP peaches were more decayed. Flesh firmness of peaches from the IFP system were greater than CFP fruits in the years 2000 and 2001. In 2002, firmness changed little during storage and ripening. Peaches from the IFP in 2000 had higher titratable acidity and lower soluble solids. In the 2000 season, flesh browning was observed in decayed fruits, always at ripening after 20 or 30 days of cold storage Chilling injuries such as browning, woolliness and leatheriness ocurred in 2002. There were no differences amongst systems related to peach quality.

Resveratrol is a polyphenol that is mainly found in grapes and red wine and has been reported to be a caloric restriction (CR) mimetic driven by Sirtuin 1 (SIRT1) activation. Resveratrol increases metabolic rate, insulin sensitivity, mitochondrial biogenesis and physical endurance, and reduces fat accumulation in mice. In addition, resveratrol may be a powerful agent to prevent age-associated neurodegeneration and to improve cognitive deficits in Alzheimer's disease (AD). Moreover, different findings support the view that longevity in mice could be promoted by CR. In this study, we examined the role of dietary resveratrol in SAMP8 mice, a model of age-related AD. We found that resveratrol supplements increased mean life expectancy and maximal life span in SAMP8 and in their control, the related strain SAMR1. In addition, we examined the resveratrol-mediated neuroprotective effects on several specific hallmarks of AD. We found that long-term dietary resveratrol activates AMPK pathways and pro-survival routes such as SIRT1 in vivo. It also reduces cognitive impairment and has a neuroprotective role, decreasing the amyloid burden and reducing tau hyperphosphorylation.
