La fenomenología como posibilidad

Heidegger, en el parágrafo séptimo de Ser y Tienzpo, reconoció que el valor de la fenomenología reside en ser una posibilidad. Pues bien, se trata de indagar qué sentido pueda tener tal afirmación. Para ello, se analiza el sentido de los conceptos clave del planteamiento husserliano, teniendo encuenta toda su evolución y viendo cómo, de alguna manera, desde ellos quedan posibilitados la consideración y el estudio de ámbitos que, en cierta medida, significaban la ruptura del idealismo egológico y conciencialista, abriéndose a paradigmas filosóficos en los que se incluyen nuevas caracterizaciones de la subjetividad.

L'enorme afirmació sens límits. Nietzsche en Maragall

En este artículo se examina una de las primeras y principales adaptaciones del pensamiento de Nietzsche en España, donde no tardaron en publicarse muchas de sus obras. Se admite generalmenteque el poeta y escritor catalán Joan Maragall, perteneciente al Modernismo de fines del siglo XIX, se inspiró en algunas ideasentresacadas del filósofo de la "voluntad de poder". Mediante el cotejo de conceptos y términos, se concluye ahora que la deudade este autor con Así habló Zaratustra es prácticamente literal. El texto que sigue muestra, pues, en qué forma y grado el nietzscheanismotoma cuerpo en la literatura de Maragall y de qué modo influye en su ideario personal.

Del historicismo a la hermeneutica:La recepción de Dilthey

El análisis de la recepción de la obra de Dilthey en la hermenéutica gadamerianapermite no sólo el acercamiento a la obra de estos dos autores, sino también esbozar elmapa de los problemas teóricos que caracterizan la transición del siglo XIX al XX, mapa enel que destaca la cuestión relativa a la relación de la filosofía con su historia. El objetivode este ensayo es clarificar qué tipo de presencia tiene Dilthey en la obra de Gadamer, paraquien no es un interlocutor más: la propuesta hermenéutica es una respuesta a los problemasque Dilthey plantea; en esta propuesta, el reconocimiento de Heidegger como referentefundamental se lleva a cabo en un campo de intereses compartido con Dilthey queproporciona elementos necesarios para hacer de la conciencia histórica "conciencia de lahistoria efectual".

Exploring the rate-limiting steps in visual phototransduction recovery by bottom-up kinetic modeling

Phototransduction in vertebrate photoreceptor cells represents a paradigm of signaling pathways mediated by G-protein-coupled receptors (GPCRs), which share common modules linking the initiation of the cascade to the final response of the cell. In this work, we focused on the recovery phase of the visual photoresponse, which is comprised of several interacting mechanisms. We employed current biochemical knowledge to investigate the response mechanisms of a comprehensive model of the visual phototransduction pathway. In particular, we have improved the model by implementing a more detailed representation of the recoverin (Rec)-mediated calcium feedback on rhodopsin kinase and including a dynamic arrestin (Arr) oligomerization mechanism. The model was successfully employed to investigate the rate limiting steps in the recovery of the rod photoreceptor cell after illumination. Simulation of experimental conditions in which the expression levels of rhodospin kinase (RK), of the regulator of the G-protein signaling (RGS), of Arr and of Rec were altered individually or in combination revealed severe kinetic constraints to the dynamics of the overall network. Our simulations confirm that RGS-mediated effector shutdown is the rate-limiting step in the recovery of the photoreceptor and show that the dynamic formation and dissociation of Arr homodimers and homotetramers at different light intensities significantly affect the timing of rhodopsin shutdown. The transition of Arr from its oligomeric storage forms to its monomeric form serves to temper its availability in the functional state. Our results may explain the puzzling evidence that overexpressing RK does not influence the saturation time of rod cells at bright light stimuli. The approach presented here could be extended to the study of other GPCR signaling pathways.

Population pharmacokinetic approach to evaluate the effect of CYP2D6, CYP3A, ABCB1, POR and NR1I2 genotypes on donepezil clearance.

AIMS: A large interindividual variability in plasma concentrations has been reported in patients treated with donepezil, the most frequently prescribed antidementia drug. We aimed to evaluate clinical and genetic factors influencing donepezil disposition in a patient population recruited from a naturalistic setting. METHODS: A population pharmacokinetic study was performed including data from 129 older patients treated with donepezil. The patients were genotyped for common polymorphisms in the metabolic enzymes CYP2D6 and CYP3A, in the electron transferring protein POR and the nuclear factor NR1I2 involved in CYP activity and expression, and in the drug transporter ABCB1. RESULTS: The average donepezil clearance was 7.3 l h(-1) with a 30% interindividual variability. Gender markedly influenced donepezil clearance (P < 0.01). Functional alleles of CYP2D6 were identified as unique significant genetic covariate for donepezil clearance (P < 0.01), with poor metabolizers and ultrarapid metabolizers demonstrating, respectively, a 32% slower and a 67% faster donepezil elimination compared with extensive metabolizers. CONCLUSION: The pharmacokinetic parameters of donepezil were well described by the developed population model. Functional alleles of CYP2D6 significantly contributed to the variability in donepezil disposition in the patient population and should be further investigated in the context of individual dose optimization to improve clinical outcome and tolerability of the treatment.
