The knowledge of electronic and local structures is a fundamental step towards understanding the properties of ferroelectric ceramics. X-ray absorption near-edge structure (XANES) of Pb1-xLaxZr0.40Ti0.60O3 ferroelectric samples was measured in order to know how the local order and electronic structure are related to their ferroelectric property, which was tailored by the substitution of lead by lanthanum atoms. The analysis of XANES spectra collected at Ti K- and L-edges XANES showed that the substitution of Pb by La leads to a decrement of local distortion around Ti atoms on the TiO6 octahedron. The analysis of O K-edge XANES spectra showed that the hybridization between O 2p and Pb 6sp states is related to the displacement of Ti atoms in the TiO6 octahedra. Based on these results, it is possible to determine that the degree of ferroelectricity in these samples and the manifestation of relaxor behavior are directly related to the weakening of O 2p and Pb 6sp hybridization. Abstract Background Overflow metabolism is an undesirable characteristic of aerobic cultures of Saccharomyces cerevisiae during biomass-directed processes. It results from elevated sugar consumption rates that cause a high substrate conversion to ethanol and other bi-products, severely affecting cell physiology, bioprocess performance, and biomass yields. Fed-batch culture, where sucrose consumption rates are controlled by the external addition of sugar aiming at its low concentrations in the fermentor, is the classical bioprocessing alternative to prevent sugar fermentation by yeasts. However, fed-batch fermentations present drawbacks that could be overcome by simpler batch cultures at relatively high (e.g. 20 g/L) initial sugar concentrations. In this study, a S. cerevisiae strain lacking invertase activity was engineered to transport sucrose into the cells through a low-affinity and low-capacity sucrose-H+ symport activity, and the growth kinetics and biomass yields on sucrose analyzed using simple batch cultures. Results We have deleted from the genome of a S. cerevisiae strain lacking invertase the high-affinity sucrose-H+ symporter encoded by the AGT1 gene. This strain could still grow efficiently on sucrose due to a low-affinity and low-capacity sucrose-H+ symport activity mediated by the MALx1 maltose permeases, and its further intracellular hydrolysis by cytoplasmic maltases. Although sucrose consumption by this engineered yeast strain was slower than with the parental yeast strain, the cells grew efficiently on sucrose due to an increased respiration of the carbon source. Consequently, this engineered yeast strain produced less ethanol and 1.5 to 2 times more biomass when cultivated in simple batch mode using 20 g/L sucrose as the carbon source. Conclusion Higher cell densities during batch cultures on 20 g/L sucrose were achieved by using a S. cerevisiae strain engineered in the sucrose uptake system. Such result was accomplished by effectively reducing sucrose uptake by the yeast cells, avoiding overflow metabolism, with the concomitant reduction in ethanol production. The use of this modified yeast strain in simpler batch culture mode can be a viable option to more complicated traditional sucrose-limited fed-batch cultures for biomass-directed processes of S. cerevisiae.

The need for high bandwidth, due to the explosion of new multi\-media-oriented IP-based services, as well as increasing broadband access requirements is leading to the need of flexible and highly reconfigurable optical networks. While transmission bandwidth does not represent a limit due to the huge bandwidth provided by optical fibers and Dense Wavelength Division Multiplexing (DWDM) technology, the electronic switching nodes in the core of the network represent the bottleneck in terms of speed and capacity for the overall network. For this reason DWDM technology must be exploited not only for data transport but also for switching operations. In this Ph.D. thesis solutions for photonic packet switches, a flexible alternative with respect to circuit-switched optical networks are proposed. In particular solutions based on devices and components that are expected to mature in the near future are proposed, with the aim to limit the employment of complex components. The work presented here is the result of part of the research activities performed by the Networks Research Group at the Department of Electronics, Computer Science and Systems (DEIS) of the University of Bologna, Italy. In particular, the work on optical packet switching has been carried on within three relevant research projects: the e-Photon/ONe and e-Photon/ONe+ projects, funded by the European Union in the Sixth Framework Programme, and the national project OSATE funded by the Italian Ministry of Education, University and Scientific Research. The rest of the work is organized as follows. Chapter 1 gives a brief introduction to network context and contention resolution in photonic packet switches. Chapter 2 presents different strategies for contention resolution in wavelength domain. Chapter 3 illustrates a possible implementation of one of the schemes proposed in chapter 2. Then, chapter 4 presents multi-fiber switches, which employ jointly wavelength and space domains to solve contention. Chapter 5 shows buffered switches, to solve contention in time domain besides wavelength domain. Finally chapter 6 presents a cost model to compare different switch architectures in terms of cost.

This thesis deals with a novel control approach based on the extension of the well-known Internal Model Principle to the case of periodic switched linear exosystems. This extension, inspired by power electronics applications, aims to provide an effective design method to robustly achieve the asymptotic tracking of periodic references with an infinite number of harmonics. In the first part of the thesis the basic components of the novel control scheme are described and preliminary results on stabilization are provided. In the second part, advanced control methods for two applications coming from the world high energy physics are presented.

Deutsch:Diese Arbeit beschäftigt sich zum einen mit der Synthese neuer, vernetzbarer, ferroelektrischer Verbindungen, welche eine höhere spontane Polarisation und damit ein besseres Schaltverhalten nach Vernetzung aufweisen sollten. Dazu wurde in bekannte Systemen die Halogene Fluor, Chlor und Brom erfolgreich eingebaut. Desweiteren konnten neue Untersuchungmethoden für ferroelektrische, flüssigkristalline Netzwerke erfolgreich angewendet und weiterentwickelt werden. Damit gelang es z. B. neue Erkenntnisse über die elastischen Eigenschaften von LC-Elastomeren zu gewinnen, wobei es erstmalig gelang, Seifenblasen aus LC-Polymeren herzustellen und durch UV-Bestrahlung zu vernetzen. Durch die Messung des Radius in Abhängigkeit des Druckes war es möglich festzustellen, daß sich das Verhalten des Polymers, welches zunächst oberflächenspannungskontrolliert war, nach UV-Bestrahlung, in ein elastisches Verhalten änderte. Aus der Radius vs. Druckbeziehung war es möglich, Daten über die elastischen Eigenschaften zu erhalten. Die Ballone zeigten dabei typische, gummielastische Eigenschaften. Ein Einfluß der Mesophase (d.h. SA oder SC-Phase) auf die Eigenschaften der Ballone konnte dabei nicht festgestellt werden. Für die beiden hier untersuchten Systeme des inter- und intralyer vernetzbaren System konnte festgestellt werden, daß ihr elastisches Verhalten sehr ähnlich ist, ganz im Gegensatz zu den früheren elektrooptischen Untersuchungen. D. h. beide Systeme zeigten nach der Vernetzung bis auf einen Faktor 2 das gleiche elastische Verhalten. Im Gegensatz zu nematischen Elastomeren, welche am Phasenübergang zum Teil große thermoelastische Änderungen zeigen, zeigten die hier untersuchten Elastomere keine Änderung der elastischen Eigenschaften beim Phasenübergang, was sich u.a. auf die relativ hohen Vernetzungsdichten zurückführen läßt. Weiterhin wurde die Elektrostriktion in ferroelektrischen flüssigkristallinen Elastomerenfilmen untersucht, welche zu einem neuen Weltrekord des elektrostriktiven Effektes führte. Es wurden Schichtdickenänderungen von 4% bei einem angelegten Feld von 1,5 kV gemessen. Röntgenstreuexperimente an gespincoateten, vernetzten Polymerfilmen haben überdies gezeigt, daß der gemessene Effekt voll und ganz auf den elektroklinen Effekt zurück zu führen ist. Zum Schluß wurde ein neuer Weg ausgearbeitet, um flüssigkristalline Netzwerke unter Einsatz von weniger präparativer Chemie zu erhalten. Dazu wurde die Möglichkeit der Netzwerkbildung mit organischen Gelbildnern untersucht. In diesem Zusammenhang ist es erstmalig gelungen, ferroelektrische Flüssigkristalle reversibel in dem einen oder anderen Zustand orientiert zu stabilisieren, wobei beliebig oft zwischen den stabilisierten Zuständen gewechselt werden konnte.

Es werden zwei komplementäre "bottom-up" Methoden präsentiert, die den kontrollierten Einbau von "intelligenten" planaren Defekten in selbstorganisierte kolloidale photonische Kristalle (KPKs) ermöglichen. Die Defektschicht basiert auf einem funktionellen, nanometer-skalierten dünnen Film, der entweder durch schichtweise ("layer-by-layer") Selbstorganisation und Mikrokontakttransferübertragung oder durch Aufschleudern und einer KPK-Opferfüllung hergestellt wird. Die entwickelten Techniken gestatten die Integration von maßgeschneiderten dünnen Defektfilmen bestehend aus einer enorm großen Vielfalt an Materialien; sie sind kostengünstig und können im größeren Maßstab angewendet werden. Optische Untersuchungen zeigen einen engen, durch den Defekt hervorgerufenen Transmissionszustand in der photonischen Bandlücke. Die Defektwellenlänge hängt von der optischen Dicke der Defektschicht ab. Aktives Schalten der Defektwellenlänge wird erreicht, indem Defektschichten aus Makromolekülen hergestellt werden, die über externe Erreger wie Licht, Temperatur, Redoxzyklen und mechanischen Druck adressiert werden können. Die Ergebnisse der Untersuchungen sind im Einklang mit separat durchgeführten Ellipsometrie-Messungen und theoretischen "scalar wave approximation"-Berechnungen. Darüber hinaus werden KPKs mit funktionellen biomolekularen Defekten vorgestellt. Über Verschiebungen der Defektmode können DNA-Konformationsänderungen, die enantioselektive Einlagerung eines chiralen Antitumormedikaments sowie Enzymaktivitäten optisch beobachtet werden. Die Einlagerung von fluoreszierenden Farbstoffen und Quantenpunkten in Defekt-KPKs führt zu einer eindeutigen, durch die photonische Bandlücke und den Defektzustand hervorgerufenen Modifizierung der Photolumineszenz (PL)-Spektren. Schaltbare PL-Modifizierungen werden detektiert, wenn adressierbare Defekt-KPKs verwendet werden.

Nowadays the rise of non-recurring engineering (NRE) costs associated with complexity is becoming a major factor in SoC design, limiting both scaling opportunities and the flexibility advantages offered by the integration of complex computational units. The introduction of embedded programmable elements can represent an appealing solution, able both to guarantee the desired flexibility and upgradabilty and to widen the SoC market. In particular embedded FPGA (eFPGA) cores can provide bit-level optimization for those applications which benefits from synthesis, paying on the other side in terms of performance penalties and area overhead with respect to standard cell ASIC implementations. In this scenario this thesis proposes a design methodology for a synthesizable programmable device designed to be embedded in a SoC. A soft-core embedded FPGA (eFPGA) is hence presented and analyzed in terms of the opportunities given by a fully synthesizable approach, following an implementation flow based on Standard-Cell methodology. A key point of the proposed eFPGA template is that it adopts a Multi-Stage Switching Network (MSSN) as the foundation of the programmable interconnects, since it can be efficiently synthesized and optimized through a standard cell based implementation flow, ensuring at the same time an intrinsic congestion-free network topology. The evaluation of the flexibility potentialities of the eFPGA has been performed using different technology libraries (STMicroelectronics CMOS 65nm and BCD9s 0.11μm) through a design space exploration in terms of area-speed-leakage tradeoffs, enabled by the full synthesizability of the template. Since the most relevant disadvantage of the adopted soft approach, compared to a hardcore, is represented by a performance overhead increase, the eFPGA analysis has been made targeting small area budgets. The generation of the configuration bitstream has been obtained thanks to the implementation of a custom CAD flow environment, and has allowed functional verification and performance evaluation through an application-aware analysis.

Capire come modellare l'attività del cervello a riposo, resting state, è il primo passo necessario per avvicinarsi a una reale comprensione della dinamica cerebrale. Sperimentalmente si osserva che, quando il cervello non è soggetto a stimoli esterni, particolari reti di regioni cerebrali presentano un'attività neuronale superiore alla media. Nonostante gli sforzi dei ricercatori, non è ancora chiara la relazione che sussiste tra le connessioni strutturali e le connessioni funzionali del sistema cerebrale a riposo, organizzate nella matrice di connettività funzionale. Recenti studi sperimentali mostrano la natura non stazionaria della connettività funzionale in disaccordo con i modelli in letteratura. Il modello implementato nella presente tesi per simulare l'evoluzione temporale del network permette di riprodurre il comportamento dinamico della connettività funzionale. Per la prima volta in questa tesi, secondo i lavori a noi noti, un modello di resting state è implementato nel cervello di un topo. Poco è noto, infatti, riguardo all'architettura funzionale su larga scala del cervello dei topi, nonostante il largo utilizzo di tale sistema nella modellizzazione dei disturbi neurologici. Le connessioni strutturali utilizzate per definire la topologia della rete neurale sono quelle ottenute dall'Allen Institute for Brain Science. Tale strumento fornisce una straordinaria opportunità per riprodurre simulazioni realistiche, poiché, come affermato nell'articolo che presenta tale lavoro, questo connettoma è il più esauriente disponibile, ad oggi, in ogni specie vertebrata. I parametri liberi del modello sono stati scelti in modo da inizializzare il sistema nel range dinamico ottimale per riprodurre il comportamento dinamico della connettività funzionale. Diverse considerazioni e misure sono state effettuate sul segnale BOLD simulato per meglio comprenderne la natura. L'accordo soddisfacente fra i centri funzionali calcolati nel network cerebrale simulato e quelli ottenuti tramite l'indagine sperimentale di Mechling et al., 2014 comprovano la bontà del modello e dei metodi utilizzati per analizzare il segnale simulato.

The concept of platform switching has been introduced to implant dentistry based on observations of reduced peri-implant bone loss. However, randomized clinical trials are still lacking. This study aimed to test the hypothesis that platform switching has a positive impact on crestal bone-level changes.
