Abstract BACKGROUND: There is an imperative necessity for alternative sources of energy able to reduce the world dependence of fossil oil. One of the most successful options is ethanol obtained mainly from sugarcane and corn fermentation. The foremost residue from sugarcane industry is the bagasse, a rich lignocellulosic raw material uses for the production of ethanol second generation (2G). New cellulolytic and hemicellulytic enzymes are needed, in order to optimize the degradation of bagasse and production of ethanol 2G. RESULTS: The ability to produce hemicellulases and related enzymes, suitable for lignocellulosic biomass deconstruction, was explored using 110 endophytic fungi and 9 fungi isolated from spoiled books in Brazil. Two initial selections were performed, one employing the esculin gel diffusion assay, and the other by culturing on agar plate media with beechwood xylan and liquor from the hydrothermal pretreatment of sugar cane bagasse. A total of 56 isolates were then grown at 29°C on steam-exploded delignified sugar cane bagasse (DEB) plus soybean bran (SB) (3:1), with measurement of the xylanase, pectinase, β-glucosidase, CMCase, and FPase activities. Twelve strains were selected, and their enzyme extracts were assessed using different substrates. Finally, the best six strains were grown under xylan and pectin, and several glycohydrolases activities were also assessed. These strains were identified morphologically and by sequencing the internal transcribed spacer (ITS) regions and the partial β-tubulin gene (BT2). The best six strains were identified as Aspergillus niger DR02, Trichoderma atroviride DR17 and DR19, Alternaria sp. DR45, Annulohypoxylon stigyum DR47 and Talaromyces wortmannii DR49. These strains produced glycohydrolases with different profiles, and production was highly influenced by the carbon sources in the media. CONCLUSIONS: The selected endophytic fungi Aspergillus niger DR02, Trichoderma atroviride DR17 and DR19, Alternaria sp. DR45, Annulohypoxylon stigyum DR47 and Talaromyces wortmannii DR49 are excellent producers of hydrolytic enzymes to be used as part of blends to decompose sugarcane biomass at industrial level.

The relation between the intercepted light and orchard productivity was considered linear, although this dependence seems to be more subordinate to planting system rather than light intensity. At whole plant level not always the increase of irradiance determines productivity improvement. One of the reasons can be the plant intrinsic un-efficiency in using energy. Generally in full light only the 5 – 10% of the total incoming energy is allocated to net photosynthesis. Therefore preserving or improving this efficiency becomes pivotal for scientist and fruit growers. Even tough a conspicuous energy amount is reflected or transmitted, plants can not avoid to absorb photons in excess. The chlorophyll over-excitation promotes the reactive species production increasing the photoinhibition risks. The dangerous consequences of photoinhibition forced plants to evolve a complex and multilevel machine able to dissipate the energy excess quenching heat (Non Photochemical Quenching), moving electrons (water-water cycle , cyclic transport around PSI, glutathione-ascorbate cycle and photorespiration) and scavenging the generated reactive species. The price plants must pay for this equipment is the use of CO2 and reducing power with a consequent decrease of the photosynthetic efficiency, both because some photons are not used for carboxylation and an effective CO2 and reducing power loss occurs. Net photosynthesis increases with light until the saturation point, additional PPFD doesn’t improve carboxylation but it rises the efficiency of the alternative pathways in energy dissipation but also ROS production and photoinhibition risks. The wide photo-protective apparatus, although is not able to cope with the excessive incoming energy, therefore photodamage occurs. Each event increasing the photon pressure and/or decreasing the efficiency of the described photo-protective mechanisms (i.e. thermal stress, water and nutritional deficiency) can emphasize the photoinhibition. Likely in nature a small amount of not damaged photosystems is found because of the effective, efficient and energy consuming recovery system. Since the damaged PSII is quickly repaired with energy expense, it would be interesting to investigate how much PSII recovery costs to plant productivity. This PhD. dissertation purposes to improve the knowledge about the several strategies accomplished for managing the incoming energy and the light excess implication on photo-damage in peach. The thesis is organized in three scientific units. In the first section a new rapid, non-intrusive, whole tissue and universal technique for functional PSII determination was implemented and validated on different kinds of plants as C3 and C4 species, woody and herbaceous plants, wild type and Chlorophyll b-less mutant and monocot and dicot plants. In the second unit, using a “singular” experimental orchard named “Asymmetric orchard”, the relation between light environment and photosynthetic performance, water use and photoinhibition was investigated in peach at whole plant level, furthermore the effect of photon pressure variation on energy management was considered on single leaf. In the third section the quenching analysis method suggested by Kornyeyev and Hendrickson (2007) was validate on peach. Afterwards it was applied in the field where the influence of moderate light and water reduction on peach photosynthetic performances, water requirements, energy management and photoinhibition was studied. Using solar energy as fuel for life plant is intrinsically suicidal since the high constant photodamage risk. This dissertation would try to highlight the complex relation existing between plant, in particular peach, and light analysing the principal strategies plants developed to manage the incoming light for deriving the maximal benefits as possible minimizing the risks. In the first instance the new method proposed for functional PSII determination based on P700 redox kinetics seems to be a valid, non intrusive, universal and field-applicable technique, even because it is able to measure in deep the whole leaf tissue rather than the first leaf layers as fluorescence. Fluorescence Fv/Fm parameter gives a good estimate of functional PSII but only when data obtained by ad-axial and ab-axial leaf surface are averaged. In addition to this method the energy quenching analysis proposed by Kornyeyev and Hendrickson (2007), combined with the photosynthesis model proposed by von Caemmerer (2000) is a forceful tool to analyse and study, even in the field, the relation between plant and environmental factors such as water, temperature but first of all light. “Asymmetric” training system is a good way to study light energy, photosynthetic performance and water use relations in the field. At whole plant level net carboxylation increases with PPFD reaching a saturating point. Light excess rather than improve photosynthesis may emphasize water and thermal stress leading to stomatal limitation. Furthermore too much light does not promote net carboxylation improvement but PSII damage, in fact in the most light exposed plants about 50-60% of the total PSII is inactivated. At single leaf level, net carboxylation increases till saturation point (1000 – 1200 μmolm-2s-1) and light excess is dissipated by non photochemical quenching and non net carboxylative transports. The latter follows a quite similar pattern of Pn/PPFD curve reaching the saturation point at almost the same photon flux density. At middle-low irradiance NPQ seems to be lumen pH limited because the incoming photon pressure is not enough to generate the optimum lumen pH for violaxanthin de-epoxidase (VDE) full activation. Peach leaves try to cope with the light excess increasing the non net carboxylative transports. While PPFD rises the xanthophyll cycle is more and more activated and the rate of non net carboxylative transports is reduced. Some of these alternative transports, such as the water-water cycle, the cyclic transport around the PSI and the glutathione-ascorbate cycle are able to generate additional H+ in lumen in order to support the VDE activation when light can be limiting. Moreover the alternative transports seems to be involved as an important dissipative way when high temperature and sub-optimal conductance emphasize the photoinhibition risks. In peach, a moderate water and light reduction does not determine net carboxylation decrease but, diminishing the incoming light and the environmental evapo-transpiration request, stomatal conductance decreases, improving water use efficiency. Therefore lowering light intensity till not limiting levels, water could be saved not compromising net photosynthesis. The quenching analysis is able to partition absorbed energy in the several utilization, photoprotection and photo-oxidation pathways. When recovery is permitted only few PSII remained un-repaired, although more net PSII damage is recorded in plants placed in full light. Even in this experiment, in over saturating light the main dissipation pathway is the non photochemical quenching; at middle-low irradiance it seems to be pH limited and other transports, such as photorespiration and alternative transports, are used to support photoprotection and to contribute for creating the optimal trans-thylakoidal ΔpH for violaxanthin de-epoxidase. These alternative pathways become the main quenching mechanisms at very low light environment. Another aspect pointed out by this study is the role of NPQ as dissipative pathway when conductance becomes severely limiting. The evidence that in nature a small amount of damaged PSII is seen indicates the presence of an effective and efficient recovery mechanism that masks the real photodamage occurring during the day. At single leaf level, when repair is not allowed leaves in full light are two fold more photoinhibited than the shaded ones. Therefore light in excess of the photosynthetic optima does not promote net carboxylation but increases water loss and PSII damage. The more is photoinhibition the more must be the photosystems to be repaired and consequently the energy and dry matter to allocate in this essential activity. Since above the saturation point net photosynthesis is constant while photoinhibition increases it would be interesting to investigate how photodamage costs in terms of tree productivity. An other aspect of pivotal importance to be further widened is the combined influence of light and other environmental parameters, like water status, temperature and nutrition on peach light, water and phtosyntate management.

[EN]In this paper we present a video streaming solution. It describes the functionality of streaming server at our university and the workflow to embed videos into the learning platform Moodle. By the server-side role management teachers can get access to upload their recorded lectures to the server. Videos will be converted into different streaming formats and embed into the learning platform with provided embed code. The number of teachers who use this proposal is continuously growing at our university. Beside the lecture scripts the videos watched often by the students what the numbers of video accesses in the Moodle course and the streaming server traffic significantly show.

The 3-UPU three degrees of freedom fully parallel manipulator, where U and P are for universal and prismatic pair respectively, is a very well known manipulator that can provide the platform with three degrees of freedom of pure translation, pure rotation or mixed translation and rotation with respect to the base, according to the relative directions of the revolute pair axes (each universal pair comprises two revolute pairs with intersecting and perpendicular axes). In particular, pure translational parallel 3-UPU manipulators (3-UPU TPMs) received great attention. Many studies have been reported in the literature on singularities, workspace, and joint clearance influence on the platform accuracy of this manipulator. However, much work has still to be done to reveal all the features this topology can offer to the designer when different architecture, i.e. different geometry are considered. Therefore, this dissertation will focus on this type of the 3-UPU manipulators. The first part of the dissertation presents six new architectures of the 3-UPU TPMs which offer interesting features to the designer. In the second part, a procedure is presented which is based on some indexes, in order to allows the designer to select the best architecture of the 3-UPU TPMs for a given task. Four indexes are proposed as stiffness, clearance, singularity and size of the manipulator in order to apply the procedure.

The focus of this study is the relationship among three different manuscripts (Modena, Bibl. Estense, MS α.R.4.4; Firenze, Bibl. Laurenziana MS Rediano 9; and London, BL, MS Harley, 2253) and the poetry they transmit. The aim of this research is to show the ways that the Bible was used in the transmission of the lyric poetry in the three literatures that they represent: Occitan (primarily through Marcabru’s songs), Italian (through the love poetry of Guittone d’Arezzo), and Middle English (through the Harley love lyrics and the MS.’s primary scribe), in a medieval European context.

Many of developing countries are facing crisis in water management due to increasing of population, water scarcity, water contaminations and effects of world economic crisis. Water distribution systems in developing countries are facing many challenges of efficient repair and rehabilitation since the information of water network is very limited, which makes the rehabilitation assessment plans very difficult. Sufficient information with high technology in developed countries makes the assessment for rehabilitation easy. Developing countries have many difficulties to assess the water network causing system failure, deterioration of mains and bad water quality in the network due to pipe corrosion and deterioration. The limited information brought into focus the urgent need to develop economical assessment for rehabilitation of water distribution systems adapted to water utilities. Gaza Strip is subject to a first case study, suffering from severe shortage in the water supply and environmental problems and contamination of underground water resources. This research focuses on improvement of water supply network to reduce the water losses in water network based on limited database using techniques of ArcGIS and commercial water network software (WaterCAD). A new approach for rehabilitation water pipes has been presented in Gaza city case study. Integrated rehabilitation assessment model has been developed for rehabilitation water pipes including three components; hydraulic assessment model, Physical assessment model and Structural assessment model. WaterCAD model has been developed with integrated in ArcGIS to produce the hydraulic assessment model for water network. The model have been designed based on pipe condition assessment with 100 score points as a maximum points for pipe condition. As results from this model, we can indicate that 40% of water pipeline have score points less than 50 points and about 10% of total pipes length have less than 30 score points. By using this model, the rehabilitation plans for each region in Gaza city can be achieved based on available budget and condition of pipes. The second case study is Kuala Lumpur Case from semi-developed countries, which has been used to develop an approach to improve the water network under crucial conditions using, advanced statistical and GIS techniques. Kuala Lumpur (KL) has water losses about 40% and high failure rate, which make severe problem. This case can represent cases in South Asia countries. Kuala Lumpur faced big challenges to reduce the water losses in water network during last 5 years. One of these challenges is high deterioration of asbestos cement (AC) pipes. They need to replace more than 6500 km of AC pipes, which need a huge budget to be achieved. Asbestos cement is subject to deterioration due to various chemical processes that either leach out the cement material or penetrate the concrete to form products that weaken the cement matrix. This case presents an approach for geo-statistical model for modelling pipe failures in a water distribution network. Database of Syabas Company (Kuala Lumpur water company) has been used in developing the model. The statistical models have been calibrated, verified and used to predict failures for both networks and individual pipes. The mathematical formulation developed for failure frequency in Kuala Lumpur was based on different pipeline characteristics, reflecting several factors such as pipe diameter, length, pressure and failure history. Generalized linear model have been applied to predict pipe failures based on District Meter Zone (DMZ) and individual pipe levels. Based on Kuala Lumpur case study, several outputs and implications have been achieved. Correlations between spatial and temporal intervals of pipe failures also have been done using ArcGIS software. Water Pipe Assessment Model (WPAM) has been developed using the analysis of historical pipe failure in Kuala Lumpur which prioritizing the pipe rehabilitation candidates based on ranking system. Frankfurt Water Network in Germany is the third main case study. This case makes an overview for Survival analysis and neural network methods used in water network. Rehabilitation strategies of water pipes have been developed for Frankfurt water network in cooperation with Mainova (Frankfurt Water Company). This thesis also presents a methodology of technical condition assessment of plastic pipes based on simple analysis. This thesis aims to make contribution to improve the prediction of pipe failures in water networks using Geographic Information System (GIS) and Decision Support System (DSS). The output from the technical condition assessment model can be used to estimate future budget needs for rehabilitation and to define pipes with high priority for replacement based on poor condition. rn

In questa tesi dottorale viene preso in analisi il tema della famiglia, uno degli elementi fondanti della riflessione pedagogica, crocevia di una molteplicità di nuclei interpretativi con diramazioni e contaminazioni, con mutamenti attraverso le epoche storiche, rappresentati in pagine contenute nei Classici della letteratura per l’infanzia e nei migliori libri di narrativa contemporanea. Si tratta di un tema di grande ampiezza che ha comportato una scelta mirata di Autori che, nei loro romanzi hanno trattato questioni riguardanti la famiglia nelle sue pluralità delle sue tante accezioni, dalla vita familiare agli abbandoni, dalle infanzie senza famiglia alle famiglie altre. Nelle diverse epoche storiche, le loro narrazioni hanno lasciato un segno per l'originalità interpretativa che ancora oggi ci raccontano storie di vie familiale Dai romanzi ottocenteschi alle saghe fantasy degli ultimi cinquant’anni, fino ai picturebook, destinati ai più piccoli, le families stories possono costituire un materiale pedagogico privilegiato, sia offrendo occasioni di scoperta e conoscenza di sé e del mondo, attraverso le quali i lettori bambini, enigmatici frontalieri, varcano soglie verso altrovi misteriosi, sia fornendo spunti agli studiosi per approfondire tematiche multiple e complesse. Le families stories riflettono spesso in maniera critica le divergenze che possono manifestarsi tra le prassi individuate e studiate dalle scienze umane e sociali e le metafore narrative proposte dai numerosi Autori della letteratura per l’infanzia. Proponendo una prospettiva spesso spiazzante, esse interpretano la realtà a fondo, cogliendo i più piccoli ed inosservati particolari che, invece, hanno la capacità di rompere gli schemi socio-educativi dell’epoca storica in cui le storie prendono vita.

Zusammenfassung: Primo Levis 1975 publiziertes Buch Il sistema periodico (Das periodische System, deutsch 1986) wurde 2006 bei einer Befragung des Imperial College in London zum „best science book ever“ gekürt. Zweifellos gehört Levis Buch zu den berühmtesten literarischen Werken mit chemischem Inhalt. Wohl jeder Chemiker kennt es oder hat schon davon gehört. Es gliedert sich in 21 Element-Erzählungen, die berühmteste von diesen ist die letzte, die vom Kohlenstoff handelt. Im Folgenden versuche ich, den Nachweis zu führen, dass diese Geschichte eine Vorlage hatte, nämlich die Erzählung ‚Lebensgeschichte eines Kohlenstoffatoms‘ von Hermann Römpp, die 1946 als Kosmos-Bändchen bei Franckh erschien. Hermann Römpp, der während des ‚Dritten Reiches‘ die antisemitischen und eugenischen Maßnahmen der Nationalsozialisten in einigen Schriften gepriesen und gerechtfertigt hatte, publizierte die Geschichte als einzige seines Oeuvres nicht unter eigenem Namen, sondern unter dem Pseudonym „Dr. Helmut Schmid“. rnDer Nachweis, dass Römpps Geschichte eine Vorlage für Levi war, soll eine vertiefte Lektüre von Levis Erzählung ermöglichen.

Die vorliegende kulturwissenschaftlich ausgerichtete Arbeit befasst sich mit der Konstruktion und Inszenierung von Realität, insbesondere im Verhältnis zu Körperdiskursen und Körperpraktiken in Makeover-Sendungen sowie mit deren komplexen politischen und kulturellen Implikationen. Die leitenden Fragen der vorliegenden Arbeit sind: Wie wird der Körper in den ausgewählten Formaten dargestellt? Wie wird er erzählt und mit welchen (Film-)Techniken in Szene gesetzt? Welche expliziten und impliziten Körperbilder liegen den Darstellungen zu Grunde? Welche kulturellen und politischen Normen werden über den Körper konstruiert? Welche Rolle spielt diese Konstruktion des Körpers für die Darstellung von „Realität“ in den entsprechenden Formaten? Was ist der größere gesellschaftliche Zusammenhang dieser Darstellung? rnTrotz der Vielzahl und Komplexität der Darstellungsweisen des Körpers soll anhand verschiedener repräsentativ ausgewählter Reality-TV-Formate eine Annäherung an die heterogene kulturelle sowie politische Tragweite derselben geleistet werden. Hierzu werden insbesondere Formate bevorzugt, die explizit die Transformation des Körpers thematisieren. Dies kann durch kosmetische, d.h. nicht-operative Veränderungen (Kleidung, Make-up, Frisur), Fitness und Ernährung bis hin zu medizinischen Eingriffen wie etwa plastischer Chirurgie erfolgen.rnAls erstes wird der Untersuchungsgegenstand genauer eingegrenzt, wobei sich zeigt, dass sich exakte Grenzziehungen aufgrund des schwer greifbaren Reality-TV-Genres und der transmedialen Eigenschaft des Makeover-Fensehtextes als Herausforderung erweisen. Danach werden die kulturwissenschaftlichen Annahmen, die dieser Arbeit zugrunde liegen, ausgeführt. Darüber hinaus wird das revolutionäre Forschungsfeld der Fat Studies eruiert und auch der körpertheoretische Zugang erläutert. Abschließend wird näher auf den filmnarratologischen Zugang, der den Analysen als theoretische, aber auch methodische Grundlage dient, eingegangen. Zudem wird behandelt wie der Körper in den behandelten Makeover-Formaten zunächst erzählbar gemacht und erzählt wird. Die Körper der Teilnehmer werden zunächst in einen ökonomischen Diskurs der Maße und Zahlen überführt, um im weiteren Verlauf die „Transformation“ auch diskursiv dramatisieren zu können. Die über die Ökonomisierung vorbereitete, zumeist als märchenhaft dargestellte, Verwandlung der Teilnehmer, kulminiert immer in der Gegenüberstellung des „Vorher“-Bildes und des „Nachher“-Bildes. Diese Ökonomisierung ist allerdings nur die narrative Grundlage einer viel umfassenderen, in den Sendungen meist implizit vermittelten Selbstregierungstechnologie, der „Gouvernementalität“, die kontrovers im Hinblick auf Vertreter einer Affekttheorie, die die Vermittlung der besagten Gouvernementalität in Zweifel ziehen, diskutiert wird. Die Kernthese der vorliegenden Arbeit, die den Körper als entscheidendes, die „Realität“ der Makeover-Formate affirmierendes Element versteht, ist ferner eng mit dem Begriff der „Authentizität“ verknüpft. „Authentische“ Effekte sind oft das selbsterklärte Ziel der Einsätze des Körpers im Reality-TV und äußern sich in unterschiedlichen Erzähltechniken wie den autobiografischen Elementen der Formate. Die bereits im Genre des Reality-TV angelegte Selbstreflexivität sowohl auf Inhalts- als auch auf Produktionsseite wird abschließend kontextualisiert und bewertet. Letztendlich stellt sich die Frage, welche kulturellen Widerstände und Spielarten trotz der sehr dogmatisch wirkenden Inhalte der Makeover-Serien erhalten bleiben.

The purpose of this work is to introduce to the Italian audience one of the best known works written by the Russian novelist Aleksej Ivanov. The title of the book we refer to is Geograf glubus propil, [The geography drank his globe away]. The novel features peculiarities and themes that can be interesting also for non-Russian readers, being also an example of how Russian contemporary literature has evolved through the years. Nevertheless, it has already been translated in French and Serbian. In last years, the author has become increasingly famous in Russia: he has won many prices and has published a large number of books, many of which were awarded with different literature prizes. Moreover, a film based on the plot of the novel analyzed has been released in 2013, carrying the same name of the book. The film received positive feedback from both cinema critics and mass audience. The following work has been divided into two parts: an introductory part where the author and his book are presented, together with the main character and his peculiarities. It is then explained the context in which the book was created, the purpose of the author and how the book was perceived by readers. In addition to that, I listed some of the problems I had to face while translating , and the solutions I found in order to solve them. The second part is fully dedicated to the translation itself.
