A panel of monoclonal antibodies specific of alpha-tubulin (TU-01, TU-09) and beta-tubulin (TU-06, TU-13) subunits was used to study the location of N-terminal structural domains of tubulin in adult mouse brain. The specificity of antibodies was confirmed b immunoblotting experiments. Immunohistochemical staining of vibratome sections from cerebral cortex, cerebellum, hippocampus, and corpus callosum showed that antibodies TU-01, TU-09, and TU-13 reacted with neuronal and glial cells and their processes, whereas the TU-06 antibody stained only the perikarya. Dendrites and axons were either unstained or their staining was very weak. As the TU-06 epitope is located on the N-terminal structural domain of beta-tubulin, the observed staining pattern cannot be interpreted as evidence of a distinct subcellular localization of beta-tubulin isotypes or known post-translational modifications. The limited distribution of the epitope could, rather, reflect differences between the conformations of tubulin molecules in microtubules of somata and neurites or, alternatively, a specific masking of the corresponding region on the N-terminal domain of beta-tubulin by interacting protein(s) in dendrites and axons.

In this paper we included a very broad representation of grass family diversity (84% of tribes and 42% of genera). Phylogenetic inference was based on three plastid DNA regions rbcL, matK and trnL-F, using maximum parsimony and Bayesian methods. Our results resolved most of the subfamily relationships within the major clades (BEP and PACCMAD), which had previously been unclear, such as, among others the: (i) BEP and PACCMAD sister relationship, (ii) composition of clades and the sister-relationship of Ehrhartoideae and Bambusoideae + Pooideae, (iii) paraphyly of tribe Bambuseae, (iv) position of Gynerium as sister to Panicoideae, (v) phylogenetic position of Micrairoideae. With the presence of a relatively large amount of missing data, we were able to increase taxon sampling substantially in our analyses from 107 to 295 taxa. However, bootstrap support and to a lesser extent Bayesian inference posterior probabilities were generally lower in analyses involving missing data than those not including them. We produced a fully resolved phylogenetic summary tree for the grass family at subfamily level and indicated the most likely relationships of all included tribes in our analysis.

To resolve the share of limited resources, animals often compete through exchange of signals about their relative motivation to compete. When two competitors are similarly motivated, the resolution of conflicts may be achieved in the course of an interactive process. In barn owls, Tyto alba, in which siblings vocally compete during the prolonged absence of parents over access to the next delivered food item, we investigated what governs the decision to leave or enter a contest, and at which level. Siblings alternated periods during which one of the two individuals vocalized more than the other. Individuals followed turn-taking rules to interrupt each other and momentarily dominate the vocal competition. These social rules were weakly sensitive to hunger level and age hierarchy. Hence, the investment in a conflict is determined not only by need and resource-holding potential, but also by social interactions. The use of turn-taking rules governing individual vocal investment has rarely been shown in a competitive context. We hypothesized that these rules would allow individuals to remain alert to one another's motivation while maintaining the cost of vocalizing at the lowest level.

Primary sensory neurons display various neuronal phenotypes which may be influenced by factors present in central or peripheral targets. In the case of DRG cells expressing substance P (SP), the influence of peripheral or central targets was tested on the neuronal expression of this neuropeptide. DRG cells were cultured from chick embryo at E6 or E10 (before or after establishment of functional connections with targets). Preprotachykinin mRNA was visualized in DRG cell cultures by either Northern blot or in situ hybridization using an antisense labeled riboprobe, while the neuropeptide SP was detected by immunostaining with a monoclonal antibody. In DRG cell cultures from E10, only 60% of neurons expressed SP. In contrast, DRG cell cultures performed at E6 showed a significant hybridization signal and SP-like immunoreactivity in virtually all the neurons (98%). The addition of extracts from muscle, skin, brain or spinal cord to DRG cells cultured at E6 reduced by 20% the percentage of neurons which express preprotachykinin mRNA and SP-like immunoreactivity. Our results indicate that factors issued from targets inhibit SP-expression by a subset of primary sensory neurons and act on the transcriptional control of preprotachykinin gene.

La rétinopathie diabétique touche un nombre croissant de personnes, soit quatre millions en Europe, ce chiffre va probablement doubler d'ici 2030. Si l'on considère que 25-30% de ces patients sont atteints de rétinopathie diabétique, un dépistage et un traitement précoce permettent d'éviter les complications oculaires sévères telles que l'oedème maculaire cystoïde où la rétinopathie diabétique proliférative. Un résumé des dernières données de la littérature ophtalmologique est présenté en insistant sur le rôle de l'anti-VEGF (vascular endothelial growth factor) et des implants intravitréens de corticostéroïdes pour une pathologie oculaire récemment classée dans les pathologies inflammatoires. Diabetic retinopathy affects an increasing number of persons, about 4 millions in Europe, a number that will probably double until 2030. If we consider that 25-30% of patients are affected by diabetic retinopathy, an ophthalmologic screening and early therapy will allow a better visual prognosis and avoid severe ocular complications such as diabetic macular edema and proliferative diabetic retinopathy. A summary of current ophthalmologic literature was performed and was focused on the role of anti-VEGF (vascular endothelial growth factor) therapies and intraocular drug delivery of corticosteroids in a pathology that was recently classified in inflammatory pathologies.

Colour polymorphism is widespread among vertebrates and plays important roles in prey-predator interactions, thermoregulation, social competition, and sexual selection. However, the genetic mechanisms involved in colour variation have been studied mainly in domestic mammals and birds, whereas information on wild animals remains scarce. Interestingly, the pro-opiomelanocortin gene (POMC) gives rise to melanocortin hormones that trigger melanogenesis (by binding the melanocortin-1-receptor; Mc1r) and other physiological and behavioural functions (by binding the melanocortin receptors Mc1-5rs). Owing to its pleiotropic effect, the POMC gene could therefore account for the numerous covariations between pigmentation and other phenotypic traits. We screened the POMC and Mc1r genes in 107 wild asp vipers (Vipera aspis) that can exhibit four discrete colour morphs (two unpatterned morphs: concolor or melanistic; two patterned morphs: blotched or lined) in a single population. Our study revealed a correlation between a single nucleotide polymorphism situated within the 3-untranslated region of the POMC gene and colour variation, whereas Mc1r was not found to be polymorphic. To the best of our knowledge, we disclose for the first time a relationship between a mutation at the POMC gene and coloration in a wild animal, as well as a correlation between a genetic marker and coloration in a snake species. Interestingly, similar mutations within the POMC 3-untranslated region are linked to human obesity and alcohol and drug dependence. Combined with our results, this suggests that the 3-untranslated region of the POMC gene may play a role in its regulation in distant vertebrates.

From toddler to late teenager, the macroscopic pattern of axonal projections in the human brain remains largely unchanged while undergoing dramatic functional modifications that lead to network refinement. These functional modifications are mediated by increasing myelination and changes in axonal diameter and synaptic density, as well as changes in neurochemical mediators. Here we explore the contribution of white matter maturation to the development of connectivity between ages 2 and 18 y using high b-value diffusion MRI tractography and connectivity analysis. We measured changes in connection efficacy as the inverse of the average diffusivity along a fiber tract. We observed significant refinement in specific metrics of network topology, including a significant increase in node strength and efficiency along with a decrease in clustering. Major structural modules and hubs were in place by 2 y of age, and they continued to strengthen their profile during subsequent development. Recording resting-state functional MRI from a subset of subjects, we confirmed a positive correlation between structural and functional connectivity, and in addition observed that this relationship strengthened with age. Continuously increasing integration and decreasing segregation of structural connectivity with age suggests that network refinement mediated by white matter maturation promotes increased global efficiency. In addition, the strengthening of the correlation between structural and functional connectivity with age suggests that white matter connectivity in combination with other factors, such as differential modulation of axonal diameter and myelin thickness, that are partially captured by inverse average diffusivity, play an increasingly important role in creating brain-wide coherence and synchrony.

En este proyecto se tiene como objetivo comparar una EDAR específica con otras tres, desde el punto de vista ambiental, y establecer diferentes alternativas. En particular, se ha evaluado la mejor alternativa en la obtención de energía eléctrica para la propia utilización de la planta de tratamiento de aguas residuales, a partir del biogás generado en el digestor en la línea de lodos. En este tipo de instalaciones, entre las alternativas tanto en su uso actual como en fase de desarrollo, el motor de cogeneración de electricidad y el calor es el más utilizado para obtener simultáneamente la electricidad necesaria para las instalaciones y el calor necesario para mantener el digestor de lodos a la temperatura de trabajo (36ºC aproximadamente). Las otras alternativas evaluadas en este estudio son las pilas de biogás de membrana electrolítica polimérica (en inglés Polymeric Electrolyte Membrane, PEM) y las pilas de óxidos sólidos (en inglés Solid Oxide Fuel Cells, SOFC) con una turbina de gas (sistema híbrido SOFC-GT). Por otro lado, se estudian las características de los materiales que componen los dispositivos MEC (microbial electrolysis cell) y las pilas PEM y SOFC, así como las ventajas e inconvenientes de usar estas nuevas tecnologías en el tratamiento de aguas residuales, así como la evaluación del impacto ambiental de la EDAR objeto de estudio, que se ha llevado a cabo utilizando el análisis de ciclo de vida (ACV). El ACV es una herramienta que permite comparar diferentes procesos o productos que tengan la misma función, y así evaluar la alternativa que conlleve una mejora en el medio ambiental. La metodología de ACV pretende evaluar en detalle el ciclo de vida completo de un producto o proceso. Un ACV se suele definir de tipo "cradle to grave" o "desde la cuna hasta la tumba" o bien de tipo "gate to gate", o "de puerta a puerta". En el primer caso el estudio analiza el ciclo de vida completo del sistema, dese el origen hasta el final, mientras que en el segundo caso el ACV no tiene en cuenta su disposición final (vertedero, reciclaje, etc.). Un estudio de ACV del primer tipo conlleva hacer un estudio muy detallado, que en la práctica puede resultar muy largo y laborioso por la dificultad de encontrar todos los datos necesarios. Por ello, muchos estudios de ACV que se encuentran en la literatura suelen ser del tipo "gate to gate". Además, hay que esablecer las fronteras del sistema a estudiar, ya que hay procesos que tienen muy poca contribución a las categorías de impacto ambiental. En una EDAR los principales procesos considerados en el ACV llevado a cabo son el consumo de productos químicos, de electricidad, la producción de lodos y su utilización como composta, el biogás y su utilización para producir electricidad, los residuos sólidos y las distintas emisiones al medio producidas por el propio funcionamiento de la EDAR. Las operaciones relacionadas indirectamente como el transporte de los lodos, de productos químicos, de los residuos sólidos y la infraestructura con una vida media de 30 años no influyen significativamente en los resultados, por ejemplo el transporte de los lodos con un camión a 30km contribuyen en menos de 1% en todas las categorías de impacto. De acuerdo con las normativas ISO series 14040 que regulan las pautas de un ACV, se establece una unidad funcional apropiada, o sea habitante equivalente, ya que es la más apropiada por tener en cuenta la carga contaminante en el agua a tratar, parámetro imprescindible para comparar EDARs. Redefiniendo las fronteras, se realiza un ACV del depósito del biogás sin tener en cuenta el resto de la instalación y se toma como unidad funcional m3 de biogás, en el caso concreto de obtener biogás mediante un dispositivo MEC, que maximiza la cantidad de hidrógeno en detrimento de la cantidad de metano contenido en el biogás, observándose que la contribución de un biogás con un alto contenido en hidrógeno y, por tanto bajo en metano, produce una mejora ambiental. Las categorías de impacto ambiental que tienen contribución son el calentamiento global y la oxidación fotoquímica; el dispositivo MEC hace quela contribución a estas categorías de impacto sea de un orden de magnitud inferior con respecto al biogás generado en un digestor. Además, si se produce la combustión del biogás, la única categoría de impacto que tiene contribución es la de calentamiento global; para una dispositivo MEC la contribución sigue siendo un orden de magnitud inferior con respecto al biogás de un digestor de lodos.
