O nosso planeta está atualmente sobre significativa pressão ambiental antropogénica, sendo as principais consequências desta situação a perda de biodiversidade, alterações climáticas e poluição. Este trabalho parte da ideia central de que a explosão demográfica já verificada e projetada para as próximas décadas constitui a origem última e a principal causa da pressão ambiental atual e futura. Nesta sequência, apresenta-se aqui um estudo sobre a explosão demográfica da população humana desde 1950 e anos anteriores, crescimento atual e previsões até 2050 e futuras. Ao fazê-lo, são identificados os aspetos que mais significativamente contribuem para esta situação, é analisado em detalhe cada aspeto identificado e são apresentadas possíveis soluções para a mitigação dos mesmos. Apresentam-se possíveis soluções e políticas para alcançar um patamar de crescimento populacional global em valores ambientalmente sustentáveis e propõe-se um valor máximo alvo para o valor da população global que permita um desenvolvimento sustentável. O momento atual, mais do que qualquer outro na história da humanidade, é de grande mudança e definirá de forma determinante em que planeta escolhemos viver: um planeta completamente humanizado que serve uma espécie ou um planeta em que a nossa espécie vive em equilíbrio com os ecossistemas e a biodiversidade que ainda subsiste atualmente, optando por uma sustentabilidade a longo prazo. Este trabalho conclui que apenas através da promoção e reforço significativo de políticas e programas ativos e eficazes de controlo de crescimento da população global e não de mera monitorização e projeção, como é tendência dos estudos e programas atuais, se poderá efetivamente atingir uma realidade próxima do segundo cenário.

Este trabalho tem o objectivo de criar um Editor e Visualizador Web de Formas de Onda para controladores digitais especificados com modelos Redes de Petri Input-Output Place-Transition (IOPT). Após uma análise das ferramentas existentes e constatando-se a inexistência de uma ferramenta adequada a essa função, desenvolveu-se uma ferramenta denominada Wave4IOPT, que permite a visualização das formas de onda de sinais e eventos de entrada e de saída ao longo do tempo. A ferramenta permite também a visualização dos resultados do histórico de uma simulação de uma Rede de Petri IOPT, proveniente do Simulador das IOPT-Tools. Esta ferramenta incorpora funcionalidades de edição, modos de visualização e um módulo básico de identificação e correcção de erros dos valores das formas de onda. O Wave4IOPT está disponível a partir de um browser e prevê-se que venha a estar integrado no ambiente de ferramentas IOPT-Tools. Esta ferramenta foi construída utilizando tecnologias Web como HTML, JavaScript, CSS, SVG e JSON. Adicionalmente, o Wave4IOPT poderá também servir para a edição, visualização e análise de outros tipos de sinais digitais, desde que sejam preenchidos os requisitos da estrutura do ficheiro JSON que será lido pela ferramenta.

The advent of bioconjugation impacted deeply the world of sciences and technology. New biomolecules were found, biological processes were understood, and novel methodologies were formed due to the fast expansion of this area. The possibility of creating new effective therapies for diseases like cancer is one of big applications of this now big area of study. Off target toxicity was always the problem of potent small molecules with high activity towards specific tumour targets. However, chemotherapy is now selective due to powerful linkers that connect targeting molecules with affinity to interesting biological receptors and cytotoxic drugs. This linkers must have very specific properties, such as high stability in plasma, no toxicity, no interference with ligand affinity nor drug potency, and at the same time, be able to lyse once inside the target molecule to release the therapeutic warhead. Bipolar environments between tumour intracellular and extracellular medias are usually exploited by this linkers in order to complete this goal. The work done in this thesis explores a new model for that same task, specific cancer drug delivery. Iminoboronates were studied due to its remarkable selective stability towards a wide pH range and endogenous molecules. A fluorescence probe was design to validate this model by creating an Off/On system and determine the payload release location in situ. A process was optimized to synthetize the probe 8-(1-aminoethyl)-7-hydroxy-coumarin (1) through a reductive amination reaction in a microwave reactor with 61 % yield. A method to conjugate this probe to ABBA was also optimized, obtaining the iminoboronate in good yields in mild conditions. The iminoboronate model was studied regarding its stability in several simulated biological environments and each half-life time was determined, showing the conjugate is stable most of the cases except in tumour intracellular systems. The construction of folate-ABBA-coumarin bioconjugate have been made to complete this evaluation. The ability to be uptaken by a cancer cell through endocytosis process and the conjugation delivery of coumarin fluorescence payload are two features to hope for in this construct.

Search is now going beyond looking for factual information, and people wish to search for the opinions of others to help them in their own decision-making. Sentiment expressions or opinion expressions are used by users to express their opinion and embody important pieces of information, particularly in online commerce. The main problem that the present dissertation addresses is how to model text to find meaningful words that express a sentiment. In this context, I investigate the viability of automatically generating a sentiment lexicon for opinion retrieval and sentiment classification applications. For this research objective we propose to capture sentiment words that are derived from online users’ reviews. In this approach, we tackle a major challenge in sentiment analysis which is the detection of words that express subjective preference and domain-specific sentiment words such as jargon. To this aim we present a fully generative method that automatically learns a domain-specific lexicon and is fully independent of external sources. Sentiment lexicons can be applied in a broad set of applications, however popular recommendation algorithms have somehow been disconnected from sentiment analysis. Therefore, we present a study that explores the viability of applying sentiment analysis techniques to infer ratings in a recommendation algorithm. Furthermore, entities’ reputation is intrinsically associated with sentiment words that have a positive or negative relation with those entities. Hence, is provided a study that observes the viability of using a domain-specific lexicon to compute entities reputation. Finally, a recommendation system algorithm is improved with the use of sentiment-based ratings and entities reputation.

É descrito o desenvolvimento ovocitário em Macrobrachium amazonicum(Heller, 1862), um camarão dulcícola da família Palaemonidae da Amazônia Central. O seu desenvolvimento é semelhante ao de Macrobrachium acanthurus(Wiegmann, 1836). As fases iniciais do desenvolvimento ocupam a região intema do ovario, enquanto que as fases vitelogênica e madura situam-se perifericamente. São descritas ainda a morfologia dos folículos pós-ovulatórios e das células foliculares.

A área de distribuição de Leporinus pachycheilusBritski, 1976 (Teleostei, Anostomidae), descrita do rio Aripuanã, é ampliada para os rios Jamari e Machado, ambos afluentes do Madeira, assim como para os rios Uatumã, Araguari e Tocantins. Esta espécie apresenta alta variabilídade intra e inter-populacional no padrão de colorido, mas a consistência dos caracteres morfológicos indica que se trata de um grupo bem definido e não distinto do material descrito para a localidade tipo (rio Aripuanã). Uma nova espécie (Leporinus juliisp.n.) é descrita para os rios Xingu e Trombetas. Esta é muito semelhante a L. pachycheilus,mas difere basicamente pelo maior número de escamas ao redor do pedúnculo caudal (16 contra 12) e pelo padrão de colorido, formado por manchas arredondadas, ao invés de listras longitudinais. Ambas as espécies são restritas a trechos de corredeiras de rios que drenam os escudos das Guianas e do Brasil Central e são as únicas espécies de anostomídeos da Amazônia que apresentam boca totalmente inferior e dentes incisiviformes, dispostos lado a lado.

Foram determinados diversos parâmetros da biologia de Conotrachelus hutneropictus Fiedler, importante praga do cacaueiro, Theobroma cacao L. e do cupuaçuzeiro, T. grandiflorum (Will, ex Spreng.) Schum na Amazônia brasileira. A pesquisa foi realizada em laboratório, com fotofase natural de 12 horas, sob condições controladas de temperatura (27 ± 2eC) e umidade relativa (80 ± 10%). C. hutneropictus alimentado com folhas novas e frutos novos de cacueiros, apresentou em média, um ciclo de ovo a emergência do adulto do solo de 79 a 151 dias e em média, 108,80 ± 9,44 dias (Χ ±Εχ95) para machos e 156,87 ±16,19 dias para fêmeas. O período de pré-oviposição foi de 16,20 ± 0,49 dias. O periodo de oviposição foi de 80,50 ± 5,58 dias, colocando cada fêmea, de 55 a 153 ovos, em média 108,45 ± 6,21 ovos, sendo as posturas diárias e constituídas em média por 1,29 ± 0,03 ovos. No interior dos frutos de cacau, o inseto necessitou de 4,73 ± 0,06 dias e 26,64 ±0,17 dias, para o desenvolvimento embrionário e larval, respectivamente. Após esse período, um total de 20,25 ± 1,50 dias se passaram no solo, para o desenvolvimento das fases de pré-pupa (6,07 ± 0,06 dias), pupa (9,62 ±0,10 dias) e completa formação fisiológica do adulto (4,56 ±0,11 dias). Neste interim, C. humeropictus torna-se mais vulnerável ao ataque de seus inimigos naturais, principalmente fungos, dentre os quais Metarhiziutn anisopliae (Metsch.) Sorok. e Beauveria bassiana (Bals.) Vuill., únicos inimigos constatados para a espécie no decorrer da pesquisa.

The weak fixation of biomaterials within the bone structure is one of the major reasons of implants failures. Calcium phosphate (CaP) coatings are used in bone tissue engineering to improve implant osseointegration by enhancing cellular adhesion, proliferation and differentiation, leading to a tight and stable junction between implant and host bone. It has also been observed that materials compatible with bone tissue either have a CaP coating or develop such a calcified surface upon implantation. Thus, the development of bioactive coatings becomes essential for further improvement of integration with the surrounding tissue. However, most of current applied CaP coatings methods (e.g. physical vapor deposition), cannot be applied to complex shapes and porous implants, provide poor structural control over the coating and prevent incorporation of bioactive organic compounds (e.g. antibiotics, growth factors) because of the used harsh processing conditions. Layer-by-layer (LbL) is a versatile technology that permits the building-up of multilayered polyelectrolyte films in mild conditions based on the alternate adsorption of cationic and anionic elements that can integrate bioactive compounds. As it is recognized in natureâ s biomineralization process the presence of an organic template to induce mineral deposition, this work investigate a ion based biomimetic method where all the process is based on LbL methodology made of weak natural-origin polyelectrolytes. A nanostructured multilayer component, with 5 or 10 bilayers, was produced initially using chitosan and chondroitin sulphate polyelectrolyte biopolymers, which possess similarities with the extracellular matrix and good biocompatibility. The multilayers are then rinsed with a sequential passing of solutions containing Ca2+ and PO43- ions. The formation of CaP over the polyelectrolyte multilayers was confirmed by QCM-D, SEM and EDX. The outcomes show that 10 polyelectrolyte bilayer condition behaved as a better site for initiating the formation of CaP as the precipitation occur at earlier stages than in 5 polyelectrolyte bilayers one. This denotes that higher number of bilayers could hold the CaP crystals more efficiently. This work achieved uniform coatings that can be applied to any surface with access to the liquid media in a low-temperature method, which potentiates the manufacture of effective bioactive biomaterials with great potential in orthopedic applications.

Larval development of the freshwater shrimp Pseudopalaemon amazonensis Ramos-Porto was studied in the laboratory based on the offspring of ovigerous females collected in a small “terra-firme” forest stream near Manaus, Brazil. Ovigerous females with a mean total length of 36.5 ± 1.9 mm carried 13-19 eliptical, yolk-rich eggs measuring 2.55 ± 0.16 x 1.64 ± 0.11 mm. The larval period consisted of 3 benthie stages and the larvae accomplished metamorphosis after 7-8 days without feeding. The newly-hatched larva had sessile eyes and all appendages, except for the uropods; chelipeds were present as uniramous buds, but walking legs were fully developed and functional. Descriptions and illustrations of the 3 larval and first juvenile stages are presented.

Bioactive glass nanoparticles (BGNPs) promote an apatite surface layer in physiologic conditions that lead to a good interfacial bonding with bone.1 A strategy to induce bioactivity in non-bioactive polymeric biomaterials is to incorporate BGNPs in the polymer matrix. This combination creates a nanocomposite material with increased osteoconductive properties. Chitosan (CHT) is a polymer obtained by deacetylation of chitin and is biodegradable, non-toxic and biocompatible. The combination of CHT and the BGNPs aims at designing biocompatible spheres promoting the formation of a calcium phosphate layer at the nanocomposite surface, thus enhancing the osteoconductivity behaviour of the biomaterial. Shape memory polymers (SMP) are stimuli-responsive materials that offer mechanical and geometrical action triggered by an external stimulus.2 They can be deformed and fixed into a temporary shape which remains stable unless exposed to a proper stimulus that triggers recovery of their original shape. This advanced functionality makes such SMPs suitable to be implanted using minimally invasive surgery procedures. Regarding that, the inclusion of therapeutic molecules becomes attractive. \The idea that social processes develop in a cyclical manner is somewhat like a `Lorelei'. Researchers are lured to it because of its theoretical promise, only to become entangled in (if not wrecked by) messy problems of empirical inference. The reasoning leading to hypotheses of some kind of cycle is often elegant enough, yet the data from repeated observations rarely display the supposed cyclical pattern. (...) In addition, various `schools' seem to exist which frequently arrive at di erent conclusions on the basis of the same data." (van der Eijk and Weber 1987:271). Much of the empirical controversies around these issues arise because of three distinct problems: the coexistence of cycles of di erent periodicities, the possibility of transient cycles and the existence of cycles without xed periodicity. In some cases, there are no reasons to expect any of these phenomena to be relevant. Seasonality caused by Christmas is one such example (Wen 2002). In such cases, researchers mostly rely on spectral analysis and Auto-Regressive Moving-Average (ARMA) models to estimate the periodicity of cycles.1 However, and this is particularly true in social sciences, sometimes there are good theoretical reasons to expect irregular cycles. In such cases, \the identi cation of periodic movement in something like the vote is a daunting task all by itself. When a pendulum swings with an irregular beat (frequency), and the extent of the swing (amplitude) is not constant, mathematical functions like sine-waves are of no use."(Lebo and Norpoth 2007:73) In the past, this di culty has led to two di erent approaches. On the one hand, some researchers dismissed these methods altogether, relying on informal alternatives that do not meet rigorous standards of statistical inference. Goldstein (1985 and 1988), studying the severity of Great power wars is one such example. On the other hand, there are authors who transfer the assumptions of spectral analysis (and ARMA models) into fundamental assumptions about the nature of social phenomena. This type of argument was produced by Beck (1991) who, in a reply to Goldstein (1988), claimed that only \ xed period models are meaningful models of cyclic phenomena".We argue that wavelet analysis|a mathematical framework developed in the mid-1980s (Grossman and Morlet 1984; Goupillaud et al. 1984) | is a very viable alternative to study cycles in political time-series. It has the advantage of staying close to the frequency domain approach of spectral analysis while addressing its main limitations. Its principal contribution comes from estimating the spectral characteristics of a time-series as a function of time, thus revealing how its di erent periodic components may change over time. The rest of article proceeds as follows. In the section \Time-frequency Analysis", we study in some detail the continuous wavelet transform and compare its time-frequency properties with the more standard tool for that purpose, the windowed Fourier transform. In the section \The British Political Pendulum", we apply wavelet analysis to essentially the same data analyzed by Lebo and Norpoth (2007) and Merrill, Grofman and Brunell (2011) and try to provide a more nuanced answer to the same question discussed by these authors: do British electoral politics exhibit cycles? Finally, in the last section, we present a concise list of future directions.
