Strong mechanical forces can, obviously, disrupt cell-cell and cell-matrix adhesions, e.g., cyclic uniaxial stretch induces instability of cell adhesion, which then causes the reorientation of cells away from the stretching direction. However, recent experiments also demonstrated the existence of force dependent adhesion growth (rather than dissociation). To provide a quantitative explanation for the two seemingly contradictory phenomena, a microscopic model that includes both integrin-integrin interaction and integrin-ligand interaction is developed at molecular level by treating the focal adhesion as an adhesion cluster. The integrin clustering dynamics and integrin-ligand binding dynamics are then simulated within one unified theoretical frame with Monte Carlo simulation. We find that the focal adhesion will grow when the traction force is higher than a relative small threshold value, and the growth is dominated by the reduction of local chemical potential energy by the traction force. In contrast, the focal adhesion will rupture when the traction force exceeds a second threshold value, and the rupture is dominated by the breaking of integrin-ligand bonds. Consistent with the experiments, these results suggest a force map for various responses of cell adhesion to different scales of mechanical force. PMID: 20542514

Seven groups of fingerling rainbow trout (S. gairdneri ) were fed for 10 weeks on 0%, 10%, 20% and 30% of cassava or rice in isonitrogenous diets. Optimum growth and food utilization was at 20% dietary cassava. High fiber content of the control diet did not suppress protein digestibility in this group. Rather, at all levels, protein digestibility was good and remained between 84.4% and 90.1%. However, in the control group, carbohydrate digestibility was very poor. The cassava diets which had the highest digestible energy as carbohydrate produced the best growth performance, food utilization and protein sparing. This thesis explored the relationship between hydrological variability and associated changes in fish communities in the upper Salado river lakes (Pampa plain, Argentina). The sampling design included five sites along the river connected lakes being explored for fish, hydrological and environmental data during different hydrological conditions. The temporal dynamic of main environmental characteristics of these lakes show that hydrology largely regulates some of the most important factors influencing fish ecology. Changes in fish communities associated with this hydrological and environmental dynamic allow to speculate a first approach towards the functioning of the system as a whole. Following oscillation between droughts and floods, study lakes have shown significant changes on abundance of major fish species, as well as on their recruitment success, which finally leaded to marked changes in fish community structure. Interestingly, trophic structure of communities did not change as much. iOdontesthes bonariensis/i was more abundant during droughts and in saltier sites but also displayed an improvement in recruitment success during these harsh abiotic conditions. Conversely, the abundance of iParapimelodus valenciennis, Cyphocharax voga/i and iCyprinus carpio/i as well as its recruitment success, were largely favoured by lower water residence times and total salinity. This dichotomy is mainly based on different life history strategies of these species against flor and environmental variability and it support the existence of different functional groups among the fish species of upper Salado river lakes. iOligosarcus jenynsii/i did not showed as evident functional response. In conclusion, hydrological and environmental variability can be considered as one of the main factors regulating the functioning and structure of fish communities in these very shallow lowland river lakes of the Pampa plain. Following these results some implications for an eventual regulation of the river regime are discussed.

In our previous work, bone cell networks with controlled spacing and functional intercellular gap junctions had been successfully established by using microcontact printing and self assembled monolayers technologies [Guo, X. E., E. Takai, X. Jiang, Q. Xu, G. M. Whitesides, J. T. Yardley, C. T. Hung, E. M. Chow, T. Hantschel, and K. D. Costa. Mol. Cell. Biomech. 3:95-107, 2006]. The present study investigated the calcium response and the underlying signaling pathways in patterned bone cell networks exposed to a steady fluid flow. The glass slides with cell networks were separated into eight groups for treatment with specific pharmacological agents that inhibit pathways significant in bone cell calcium signaling. The calcium transients of the network were recorded and quantitatively evaluated with a set of network parameters. The results showed that 18 alpha-GA (gap junction blocker), suramin (ATP inhibitor), and thapsigargin (depleting intracellular calcium stores) significantly reduced the occurrence of multiple calcium peaks, which were visually obvious in the untreated group. The number of responsive peaks also decreased slightly yet significantly when either the COX-2/PGE(2) or the NOS/nitric oxide pathway was disrupted. Different from all other groups, cells treated with 18 alpha-GA maintained a high concentration of intracellular calcium following the first peak. In the absence of calcium in the culture medium, the intracellular calcium concentration decreased slowly with fluid flow without any calcium transients observed. These findings have identified important factors in the flow mediated calcium signaling of bone cells within a patterned network.

Abstract: Experiments to determine the horizontal static bearing capacity are carried out first. The static bearing capacity is a reference for choosing the amplitudes of dynamic load. Then a series of experiments under dynamic horizontal load are carried out in laboratory to study the influences of factors, such as the scales of bucket, the amplitude and frequency of load, the density of soils etc.. The responses of bucket foundations in calcareous sand under horizontal dynamic load are analyzed according to the experimental results. The displacements of bucket and sand layer are analyzed.

The configuration of semisubmersibles consisting of pontoons and columns and their corresponding heave motion response in incident progressive waves are examined. The purpose of the present study is to provide a theoretical approach to estimating the effects of volumetric allocation on natural period and response amplitude operator (RAO) in heave motion. We conclude that the amplitude of heave motion response can be considerably suppressed by appropriately adjusting volumetric allocation so that the natural heave period keeps away from the range of wave energy. The theoretical formulae are found in good agreement with the corresponding computational results by WAMIT.

The growth response of Clarias gariepinus was investigated in various types of fertilizers. Fertilizer type was found to influence plankton abundance which in turn determine the growth and well being of C. gariepinus. The best weight increase recorded was in cow dung /NPK (1.37~c1.01g) followed by poultry (0.49~c0.31g), NPK/poultry (0.05~c0.25g) and NPK(0.03~c0.57g) The survival rate in these treatments were cow dung/NPK (100%), poultry (100%), NPK/cow/poultry (33%) and NPK (8%)

This thesis presents a technique for obtaining the stochastic response of a nonlinear continuous system. First, the general method of nonstationary continuous equivalent linearization is developed. This technique allows replacement of the original nonlinear system with a time-varying linear continuous system. Next, a numerical implementation is described which allows solution of complex problems on a digital computer. In this procedure, the linear replacement system is discretized by the finite element method. Application of this method to systems satisfying the one-dimensional wave equation with two different types of constitutive nonlinearities is described. Results are discussed for nonlinear stress-strain laws of both hardening and softening types.

Cells in the lateral intraparietal cortex (LIP) of rhesus macaques respond vigorously and in spatially-tuned fashion to briefly memorized visual stimuli. Responses to stimulus presentation, memory maintenance, and task completion are seen, in varying combination from neuron to neuron. To help elucidate this functional segmentation a new system for simultaneous recording from multiple neighboring neurons was developed. The two parts of this dissertation discuss the technical achievements and scientific discoveries, respectively.
Technology. Simultanous recordings from multiple neighboring neurons were made with four-wire bundle electrodes, or tetrodes, which were adapted to the awake behaving primate preparation. Signals from these electrodes were partitionable into a background process with a 1/f-like spectrum and foreground spiking activity spanning 300-6000 Hz. Continuous voltage recordings were sorted into spike trains using a state-of-the-art clustering algorithm, producing a mean of 3 cells per site. The algorithm classified 96% of spikes correctly when tetrode recordings were confirmed with simultaneous intracellular signals. Recording locations were verified with a new technique that creates electrolytic lesions visible in magnetic resonance imaging, eliminating the need for histological processing. In anticipation of future multi-tetrode work, the chronic chamber microdrive, a device for long-term tetrode delivery, was developed.
Science. Simultaneously recorded neighboring LIP neurons were found to have similar preferred targets in the memory saccade paradigm, but dissimilar peristimulus time histograms, PSTH). A majority of neighboring cell pairs had a difference in preferred directions of under 45° while the trial time of maximal response showed a broader distribution, suggesting homogeneity of tuning with het erogeneity of function. A continuum of response characteristics was present, rather than a set of specific response types; however, a mapping experiment suggests this may be because a given cell's PSTH changes shape as well as amplitude through the response field. Spike train autocovariance was tuned over target and changed through trial epoch, suggesting different mechanisms during memory versus background periods. Mean frequency-domain spike-to-spike coherence was concentrated below 50 Hz with a significant maximum of 0.08; mean time-domain coherence had a narrow peak in the range ±10 ms with a significant maximum of 0.03. Time-domain coherence was found to be untuned for short lags (10 ms), but significantly tuned at larger lags (50 ms).

The growth response, feed conversion ratio and cost benefits of hybrid catfish, Heterobranchus longifilis x Clarias gariepinus fed five maggot meal based diets were evaluated for 56 days in outdoor concrete tanks. Twenty-five fingerlings of the hybrid fish were stocked in ten outdoor concrete tanks of dimension 1.2mx0.13mx0.18m and code MM sub(1)-MM sub(5) in relation to their diet name. Five isonitrogenous and isocaloric maggot meal based diets namely MM sub(1)-0% maggot meal, MM sub(2)-25% maggot meal, MM sub(3)-50% maggot meal, MM sub(4-)75% maggot meal and MM sub(5-) 100% maggot meal were used for the experiment. The higher the proportion of maggot in the meal, the higher the ether extract and crude fiber. No significance difference P>0.05 exists between ash content of the experimental diets. Diet MM sub(2) had the best growth performance and highest MGR with a significant difference P<0.05 with other diets fed fish. No significance differences P>0.05 exists between the growth parameters for diets MM sub(1), MM sub(3), and MM sub(4). A positive correlation (r=1.0) exists (P<0.05, 0.25) between the growth parameters for the different experimental diets. Highest correlation r super(2)=0.9981 exists P<0.05 between MGR within the treatments. However, there no significant (P>0.05) difference in expenditure but there is between the profit indices and incidence of cost between the trials. MM sub(2) has the best yield cost and net profit. Without any reservation, inclusion of maggot based meal diet is recommended as feed of hybrid catfish to 75% inclusion for growth and profit incidence

The growth response of F1 hybrid fry of female Heterobranchus longifilis and male Clarias gariepinus were investigated under laboratory conditions in glass aquaria glass tanks and plastic basins. The larvae were produced artificially after inducement with Ovarptim. The hatching percentage was very high. Weekly mean length and weight were monitored for 6 weeks. The average length increase was higher in aquaria glass tanks than in plastic basins. However, there were depressed and irregular weight increases in both types of rearing troughs while significant weight increase (P<0.05) was recorded at week 6 in the plastic basin. Generally, the growth rate and survival in both containers were not significantly different
