In this study, the effect of anti-corrosion inhibitor addition to epoxy coating, on the disbanding rate was evaluated. First to determination of mechanism, the bare steel substrates were immersed in the 3.5% NaCl solution and the solution containing 1 mM anti corrosion. The Electrochemical Impedance Spectroscopy was performed after 5 and 24 hour. The results indicated a lower corrosion rate in the presence of inhibitor. During the time, charge transfer resistance, was decreased for the substrates immersed in NaCl solution, and increased for the substrates immersed in NaCl solution containing 1 mM anti corrosion. This result can be related to more stability of corrosion products in presence of anti-corrosion and film formation. The coated substrates, with four different concentration of anticorrosion in coating, were protected under -1.2 voltage in the 3.5% NaCl solution. After 12 and 24 hour, the EIS test and disbanding area measurement, were evaluate. The lower disbanding rate, more charge transfer resistance and less double layer capacitance for the coating containing 0.75w% inhibitor, were observed. The result of Pull-off test after 1 day immersion in 3.5% NaCl solution, showed more wet adhesion for the coating containing 0.75w% inhibitor. The images of FE-SEM electron microscope and surface analyses EDX on the coated substrate after disbanding and the bare substrate immersed in 3.5w% NaCl containing 1 mM inhibitor, were proved the formation of stabilized film.

As polyphenolic compounds isolated from plants extracts, flavonoids have been applied to various pharmaceutical uses in recent decades due to their anti-inflammatory, cancer preventive, and cardiovascular protective activities. In this study, we evaluated the effects of the flavonoid quercetin on Crotalus durissus terrificus secretory phospholipase A2 (sPLA2), an important protein involved in the release of arachidonic acid from phospholipid membranes. The protein was chemically modified by treatment with quercetin, which resulted in modifications in the secondary structure as evidenced through circular dichroism. In addition, quercetin was able to inhibit the enzymatic activity and some pharmacological activities of sPLA2, including its antibacterial activity, its ability to induce platelet aggregation, and its myotoxicity by approximately 40%, but was not able to reduce the inflammatory and neurotoxic activities of sPLA2. These results suggest the existence of two pharmacological sites in the protein, one that is correlated with the enzymatic site and another that is distinct from it. We also performed molecular docking to better understand the possible interactions between quercetin and sPLA2. Our docking data showed the existence of hydrogen-bonded, polar interactions and hydrophobic interactions, suggesting that other flavonoids with similar structures could bind to sPLA2. Further research is warranted to investigate the potential use of flavonoids as sPLA2 inhibitors. (C) 2010 Elsevier B.V. All rights reserved.

Purpose: To develop a micelle-enhanced spectrofluorimetric method for the assay of azilsartan (AZL) in bulk form and spiked human plasma without the need for derivatization procedure. Method: The proposed method was based on studying the fluorescence behavior of AZL in Cremophor RH 40 (Cr RH 40) micellar system. The fluorescence intensity was measured at 371 nm after excitation at 264 nm. The proposed procedure was validated according to International Council on Harmonization (ICH) guidelines. Results: In aqueous solution, the fluorescence intensity of AZL was greatly enhanced by more than 3- fold in the presence of Cr RH 40. The fluorescence –concentration plot was linear over the range of 10 – 500 ng.mL-1, with a limit of detection of 3.287 ngmL-1. The proposed method was successfully applied to the determination of AZL in pure powder form and spiked human plasma. The mean recovery of AZL in spiked human plasma using the proposed method was 90.54 ± 1.17 %. Conclusion: The suggested method is highly sensitive and simple, and can easily be applied for the quantification of AZL in pure powder form as well as in biological fluids such as plasma.

Purpose: To study the effect of conformal radiotherapy combined with epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) in the second-line treatment of non-small cell lung cancer (NSCLC). Methods: A total of 316 patients attending Shanghai Pulmonary Hospital affiliated to Tongji University, were divided into two groups: 106 patients were treated with conformal radiotherapy combined with EGFR-TKI (gefitinib, 250 mg/day; or erlotinib, 150 mg/day), while 210 patients were treated with EGFRTKI alone. Some factors, including adverse reactions (AR), disease control rate (DCR), progression-free survival (PFS), overall survival (OS), and one-year and two-year survival rate, were evaluated. Results: No obvious difference was observed in AR between the two groups (p > 0.05). In the combination therapy group, complete response (CR) was 5 cases, partial response (PR) 43 cases, and stable disease (SD) 47 cases, progressive disease (PD) was 11 cases, response rate (RR) was 45.3 %, and DCR 89.6 %. Median PFS in the combination therapy group and targeted therapy group was 6.5 and 5.0 months, respectively. On the other hand, median OS in the combination therapy group and targeted group was 14.1 and 12.6 months, respectively. One-year survival rate of the combination therapy group and EGFR-TKI group was 60.3 and 50.0 %, respectively, while the two-year survival rate was 26.3 and 19.0 %, respectively. Conclusion: Conformal radiotherapy combined with EGFR-TKI can be used as an effective second-line treatment for NSCLC.

An organic salt comprising of an imidazolinium cation and a 4-hydroxy cinnamate anion has been shown to be a viable inhibitor for reducing the corrosion of steel in 0.01M NaCl aqueous solutions under acidic, neutral and basic conditions. The efficiency is particularly high at pH 8 (86%). Of most significance is that the individual components of this compound do not inhibit as effectively at equivalent concentrations, particularly at a pH of 2, suggesting there is a true synergy resulting from the combination of anion and cation. The immersion studies show the efficacy of these inhibitors to stifle corrosion as observed from optical, SEM and profilometry experiments. The mechanism of inhibition appears to be dominated by anodic behavior and further surface characterization work will investigate the origin of this inhibition and the synergy observed.

Alogliptin, a dipeptidyl peptidase-4 (DPP-4) inhibitor that is a class of relatively new oral hypoglycaemic drugs used in patients with type 2 diabetes (T2DM), can be used as monotherapy or in combination with other anti-diabetic agents, including metformin, pioglitazone, sulfonylureas and insulin with a considerable therapeutic effect. Alogliptin exhibits favorable pharmacokinetic and pharmacodynamic profiles in humans. Alogliptin is mainly metabolized by cytochrome P450 (CYP2D6) and CYP3A4. Dose reduction is recommended for patients with moderate or worse renal impairment. Side effects of alogliptin include nasopharyngitis, upper-respiratory tract infections and headache. Hypoglycaemia is seen in about 1.5% of the T2DM patients. Rare but severe adverse reactions such as acute pancreatitis, serious hypersensitivity including anaphylaxis, angioedema and severe cutaneous reactions such as Stevens-Johnson syndrome have been reported from post-marketing monitoring. Pharmacokinetic interactions have not been observed between alogliptin and other drugs including glyburide, metformin, pioglitazone, insulin and warfarin. The present review aimed to update the clinical information on pharmacodynamics, pharmacokinetics, adverse effects and drug interactions, and to discuss the future directions of alogliptin.

PURPOSE: Malignant rhabdoid tumor (MRT) and atypical teratoid rhabdoid tumors (ATRT) are rare aggressive undifferentiated tumors primarily affecting the kidney and CNS of infants and young children. MRT are almost exclusively characterized by homozygous deletion or inactivation of the chromatin remodeling gene SMARCB1 SMARCB1 protein loss leads to direct impairment of chromatin remodeling and we have previously reported a role for this protein in histone acetylation. This provided the rationale for investigating the therapeutic potential of histone deactylase inhibitors (HDACi) in MRT. EXPERIMENTAL DESIGN: Whereas previously HDACis have been used at doses and schedules that induce cytotoxicity, in the current studies we have tested the hypothesis, both in vitro and in vivo, that sustained treatment of human MRT with low-dose HDACi can lead to sustained cell growth arrest and differentiation. RESULTS: Sustained low-dose panobinostat (LBH589) treatment led to changes in cellular morphology associated with a marked increase in the induction of neural, renal, and osteoblast differentiation pathways. Genome-wide transcriptional profiling highlighted differential gene expression supporting multilineage differentiation. Using mouse xenograft models, sustained low-dose LBH589 treatment caused tumor growth arrest associated with tumor calcification detectable by X-ray imaging. Histological analysis of LBH589-treated tumors revealed significant regions of ossification, confirmed by Alizarin Red staining. Immunohistochemical analysis showed increased TUJ1 and PAX2 staining suggestive of neuronal and renal differentiation, respectively. CONCLUSIONS: Low-dose HDACi treatment can terminally differentiate MRT tumor cells and reduce their ability to self-renew. The use of low-dose HDACi as a novel therapeutic approach warrants further investigation.

Soils under irrigated agriculture are a significant source of nitrous oxide (N2O) owing to high inputs of nitrogen (N) fertiliser and water. This study investigated the potential for N2O mitigation by manipulating the soil moisture deficit through irrigation scheduling in combination with, and in comparison to, using the nitrification inhibitor, 3,4-dimethylpyrazole phosphate (DMPP). Lysimeter cores planted with wheat were fitted with automated chambers for continuous measurements of N2O fluxes. Treatments included conventional irrigation (CONV), reduced deficit irrigation (RED), CONV-DMPP and RED-DMPP. The total seasonal volume of irrigation water applied was constant for all treatments but the timing and quantity in individual irrigation applications varied among treatments. 15N-labelled urea was used to track the source of N2O emissions and plant N uptake. The majority of N2O emissions occurred immediately after irrigations began on 1 September 2014. Applying RED and DMPP individually slightly decreased N2O emissions but when applied in combination (RED-DMPP) the greatest reductions in N2O emissions were observed. There was no effect of treatments on plant N uptake, 15N recovery or yield possibly because the system was not N limited. Half of the plant N and 53% to 87% of N2O was derived from non-fertiliser sources in soil, highlighting the opportunity to further exploit this valuable N pool.
