The rapid increase in population in Polk County during the last decade has been accompanied by a several-fold increase in the number and complexity of problems pertaining tothe use andcontrolof the streams andlakes of the county. The increase in problems has increased the need for more information about the occurrence and movement of surface water. The primary purpose of this report is to give that information. Its secondary purpose is to give information that will promote an understanding of the nature and causes of the problems or that will aid in arriving at practical solutions. Previous simulations of potential ichthyoplankton entrainment by power generating stations on the Potomac estuary have not included the influence of lateral transport in distributing eggs and larvae over the nursery area. Therefore, two-dimensional, vertically-averaged hydrodynamic and kinematic models of passive organism transport were developed to represent advective and dispersive processes near the proposed Douglas Point Nuclear Generating Station. Although the more refined model did not substantially alter the estimate of ichthyoplankton entrainment, it did reveal that lateral inhomogeneities in hydrodynamics could engender several fold differences in entrainment probabilities on opposite sides of the estuary. Models of higher resolution and greater biological detail did not project greater total entrainment by the Douglas Point plant, because the volume of nontidal flow past the site was large in comparison to the proposed rate of cooling water withdrawal.

Microbe-Associated Molecular Patterns and virulence effectors are recognized by plants as a first step to mount a defence response against potential pathogens. This recognition involves a large family of extracellular membrane receptors and other immune proteins located in different sub-cellular compartments. We have used phage-display technology to express and select for Arabidopsis proteins able to bind bacterial pathogens. To rapidly identify microbe-bound phage, we developed a monitoring method based on microarrays. This combined strategy allowed for a genome-wide screening of plant proteins involved in pathogen perception. Two phage libraries for high-throughput selection were constructed from cDNA of plants infected with Pseudomonas aeruginosa PA14, or from combined samples of the virulent isolate DC3000 of Pseudomonas syringae pv. tomato and its avirulent variant avrRpt2. These three pathosystems represent different degrees in the specificity of plant-microbe interactions. Libraries cover up to 26107 different plant transcripts that can be displayed as functional proteins on the surface of T7 bacteriophage. A number of these were selected in a bio-panning assay for binding to Pseudomonas cells. Among the selected clones we isolated the ethylene response factor ATERF-1, which was able to bind the three bacterial strains in competition assays. ATERF-1 was rapidly exported from the nucleus upon infiltration of either alive or heat-killed Pseudomonas. Moreover, aterf-1 mutants exhibited enhanced susceptibility to infection. These findings suggest that ATERF-1 contains a microbe-recognition domain with a role in plant defence. To identify other putative pathogen-binding proteins on a genome-wide scale, the copy number of selected-vs.-total clones was compared by hybridizing phage cDNAs with Arabidopsis microarrays. Microarray analysis revealed a set of 472 candidates with significant fold change. Within this set defence-related genes, including well-known targets of bacterial effectors, are over-represented. Other genes non-previously related to defence can be associated through this study with general or strain-specific recognition of Pseudomonas.

Information on the biology and fisheries of cobia, Rachycentron canadum, is compiled and reviewed in the FAD species synopsis style. Topics include taxonomy, morphology, distribution, reproduction, pre-adult and adult stages, food, growth, migration, population characteristics, and various aspects of exploitation. Data and information were obtained from unpublished as well as published sources. Cobia, the only species in the family Rachycentridae, is a migratory pelagic fish that occurs in tropical and subtropical seas of the world, except in the central and eastern Pacific Ocean. In the western Atlantic Ocean, spawning occurs during the warm months. Eggs and larvae are planktonic. Females grow faster than males: at 1 year, females are 36 cm FL and 0.4 kg; at 4 years, 99 cm and 11 kg; and at 8 years, 137 cm and 31 kg. Comparable data for males are: at 1 year, 31 cm and 0.3 kg; 4 years, 82 cm and 6 kg; and 8 years, 108 cm and 15 kg. Sexual maturity is attained by males at about 52 cm FL in their second year and by females at about 70 cm in their third year. Fecundity for females 100-125 cm FL varies from 1.9 to 5.4 million eggs. Cobia favor crustaceans for food, but will feed on other invertebrates and fishes as well. They attain a maximum size of over 60 kg. Cobia are fished both commercially and recreationally. Commercially, they are usually caught incidentally in both hook-and-Iine and net fISheries. In the United States, which ranks behind Pakistan, Mexico, and the Philippines in commercial production of cobia, recreational landings exceed commercial landings by more than ten-fold. [EN] The aims of this work were (i) to evaluate the potential of nanostructured lipid carriers (NLCs) as a tool to 24 enhance the oral bioavailability of poorly soluble compounds using saquinavir (SQV), a BCS class IV drug 25 and P-gp substrate as a model drug, and (ii) to study NLC transport mechanisms across the intestinal barrier. 26 Three different NLC formulations were evaluated. SQV transport across Caco-2 monolayers was enhanced up 27 to 3.5-fold by NLCs compared to SQV suspension. M cells did not enhance the transport of NLCs loaded with 28 SQV. The size and amount of surfactant in the NLCs influenced SQV's permeability, the transcytosis pathway 29 and the efflux of SQV by P-gp. An NLC of size 247 nm and 1.5% (w/v) surfactant content circumvented P-gp 30 efflux and used both caveolae- and clathrin-mediated transcytosis, in contrast to the other NLC formulations, 31 which used only caveolae-mediated transcytosis. By modifying critical physicochemical parameters of the 32 NLC formulation, we were thus able to overcome the P-gp drug efflux and alter the transcytosis mechanism 33 of the nanoparticles. These findings support the use of NLCs approaches for oral delivery of poorly 34 water-soluble P-gp substrates.

More than a decade has passed since the passage of the Marine Mammal Protection Act of 1972. During that time the U.S. tuna purse seine neet reduced its incidental porpoise mortality rate more than 10-fold. This was made possible through the development of gear and techniques aimed at reducing the frequency of many low probability events that contribute to the kill. Porpoise are killed by becoming entangled or entrapped in folds and canopies of the net and suffocating. The configuration of the net, both before and during the backdown release procedure, is a major determinant of the number of porpoise killed. Speedboats can be used to tow on the corkllne to prevent net collapse and also to adjust the net configuration to reduce net canopies prior to backdown. Deepening a net can reduce the probability of porpoise being killed by prebackdown net collapse. The effects of environmental conditions and mechanical failures on net configuration can result in high porpoise mortality unless mitigated by skilled vessel maneuvers or prevented by the timely use of speedboats to adjust the net. The backdown procedure is the only means to effectively release captured porpoise from a purse seine. It is also the time during the set when most of the mortality occurs. The use of small mesh safety panels and aprons in the backdown areas of nets reduces porpoise entanglement, and Increases the probability of an effective release. The tie-down points on the net for preparing the backdown channel must be properly located in order to optimize porpoise release. A formula uses the stretched depth of the net to calculate one of these points, making it a simple matter to locate the other. Understanding the dynamics of the backdown procedure permits a thorough troubleshooting of performance, thus preventing the repetition of poorly executed backdowns and thereby reducing mortality. Porpoise that cannot be released must be rescued by hand. A rescuer in a rigidly inflated raft can rescue porpoise effectively at any time during a net set. Hand rescue can make the difference between above average kill and zero kill sets. In all circumstances, the skill and motivation of the captain and his crew are the final determinants in the prevention of incidental porpoise mortality in tuna seining. Shortnose sturgeon (Acipenser brevirostrum), an endangered species, has experienced a several-fold increase in abundance in the Hudson River in recent decades. This population growth followed a substantial improvement in water quality during the 1970s to a large portion (c. 40%) of the species' summertime nursery area. Age structure and growth were investigated to evaluate the hypothesis that improvements in water quality stimulated population recovery through increased survival of young of the year juveniles. Specimens were captured using gill nets bi-monthly from November 2003 to November 2004 (n = 596). Annuli in fin spine sections were used to generate estimates of sturgeon age. Based upon a marginal increment analysis, annuli were determined to form at an annual rate. Age determinations yielded a catch composed of age 5-30 years for sizes 49-105cm Total Length (n = 554). Individual growth rate (von Bertalanffy coefficients: TL, = 1045mm, K = 0.07) for the population was similar to previous growth estimates within the Hudson River as well as proximal estuaries. Hindcast year-class strengths, based upon a recent stock assessment (Bain et al. 2000) and corrected for gill net mesh selectivity and cumulative mortality indicated high recruitments (28,000-43,000 yearlings)during 1986-1992, which were preceded and succeeded by c.5-year periods of lower recruitment (5,000-1 5,000 yearlings). Recruitment patterns were corroborated by trends in shortnose sturgeon bycatch from a Hudson utilities-sponsored monitoring program. Results indicated that Hudson River shortnose sturgeon abundance increased due to the formation of several strong year-classes occurring about five years subsequent to improved water quality in important nursery and forage habitats in the upper Hudson River estuary. In this essay, three lines of evidence are developed that sturgeons in the Chesapeake Bay and elsewhere are unusually sensitive to hypoxic conditions: 1. In comparison to other fishes, sturgeons have a limited behavioral and physiological capacity to respond to hypoxia. Basal metabolism, growth, and consumption are quite sensitive to changes in oxygen level, which may indicate a relatively poor ability by sturgeons to oxyregulate. 2. During summertime, temperatures >20 C amplify the effect of hypoxia on sturgeons and other fishes due to a temperature*oxygen "squeeze" (Coutant 1987)- In bottom waters, this interaction results in substantial reduction of habitat; in dry years, nursery habitats in the Chesapeake Bay may be particularly reduced or even eliminated. 3. While evidence for population level effects by hypoxia are circumstantial, there are corresponding trends between the absence of Atlantic sturgeon reproduction in estuaries like the Chesapeake Bay where summertime hypoxia predominates on a system-wide scale. Also, the recent and dramatic recovery of shortnose sturgeon in the Hudson River (4-fold increase in abundance from 1980 to 1995) may have been stimulated by improvement of a large portion of the nursery habitat that was restored from hypoxia to normoxia during the period 1973-1978. Fish assemblage structure of Maryland's coastal lagoon complex was analyzed for spatial and seasonal patterns for the period 1991-2000. Data was made available by Maryland Department of Natural Resources from their MD Coastal Bays Finfish Survey. Dominant species from separate trawl and wiw surveys included blue crab Callinectes sapidus (erroneously included here as a "fish" due to its dominance and commercial importance), bay anchovy Anchoa mitchilli, spot Leiostomous xanthurus, silver perch Bairdiella ehrysoura, and Atlantic menhaden Brevwrtia tyrannus. Ninety-four fish species were identified in the two surveys, a diversity substantially higher than other survey records for Middle Atlantic Bight estuarine and lagoon systems (richness=26 to 78 species). Total species richness for the trawl survey was highest in Chincoteague and lowest in Assawoman and Sinepuxent. On the other hand, mean richness per tow (-area) and related Shannon Weiner Diversity Index were significantly higher in the northern two bays (Assawoman and Isle of Wight Bays) than in the two southern bays (Chincoteague or Sinepuxent Bays). For the seine survey, effort-adjusted diversity indices were significantly lower for Chincoteague Bay than for the other three bays. Higher relative abundances were observed in the northern bays than in the southern bays. The trawl survey exhibited the lowest catch-per-site in Sinepuxent Bay and the highest in Assawoman Bay. The seine survey had the lowest catch-per-site in Chincoteague Bay while the other three embayments were of similar magnitude. There was clear seasonality in assemblage structure with peak abundance and diversity in the summer compared to other seasons. Blue crabs in particular showed a c. 2-fold decline in relative abundance from early summer to fall, which is likely attributable to harvest removals (i.e., an exploitation rate of c. 50%). Seagrass coverage, although increasing over the course of the 10 year survey, did not have obvious effects on species diversity and abundance across or within the embayments, although it did have positive associations with two important species: bay anchovy and summer flounder Pavalich thys dentatus. Atlantic menhaden were most dominant in Assawoman Bay, which could be related to higher primary production typically observed in this Bay in comparison to the other three. (PDF contains 99 pages)

Background: The recruitment of vascular stromal and endothelial cells is an early event occurring during cancer cell growth at premetastatic niches, but how the microenvironment created by the initial three-dimensional (3D) growth of cancer cells affects their angiogenesis-stimulating potential is unclear. Methods: The proangiogenic profile of CT26 murine colorectal carcinoma cells was studied in seven-day cultured 3D-spheroids of <300 mu m in diameter, produced by the hanging-drop method to mimic the microenvironment of avascular micrometastases prior to hypoxia occurrence. Results: Spheroid-derived CT26 cells increased vascular endothelial growth factor (VEGF) secretion by 70%, which in turn increased the in vitro migration of primary cultured hepatic sinusoidal endothelium (HSE) cells by 2-fold. More importantly, spheroid-derived CT26 cells increased lymphocyte function associated antigen (LFA)-1-expressing cell fraction by 3-fold; and soluble intercellular adhesion molecule (ICAM)-1, given to spheroid-cultured CT26 cells, further increased VEGF secretion by 90%, via cyclooxygenase (COX)-2-dependent mechanism. Consistent with these findings, CT26 cancer cells significantly increased LFA-1 expression in non-hypoxic avascular micrometastases at their earliest inception within hepatic lobules in vivo; and angiogenesis also markedly increased in both subcutaneous tumors and hepatic metastases produced by spheroid-derived CT26 cells. Conclusion: 3D-growth per se enriched the proangiogenic phenotype of cancer cells growing as multicellular spheroids or as subclinical hepatic micrometastases. The contribution of integrin LFA-1 to VEGF secretion via COX-2 was a micro environmental-related mechanism leading to the pro-angiogenic activation of soluble ICAM-1-activated colorectal carcinoma cells. This mechanism may represent a new target for specific therapeutic strategies designed to block colorectal cancer cell growth at a subclinical micrometastatic stage within the liver.

Deformation twins are often observed to meet each other to form multi-fold twins in nanostructured face-centered cubic (fcc) metals.Here we propose two types of mechanism for the nucleation and growth of four different single and multiple twins. These mechanisms provide continuous generation of twinning partials for the growth of the twins after ucleation. A relatively high stress or high strain rate is needed to activate these mechanisms, making them more prevalent in nanocrystalline materials than in their coarse-grained counterparts.Experimental observations that support the proposed mechanisms are presented.

As a basic tool of modern biology, sequence alignment can provide us useful information in fold, function, and active site of protein. For many cases, the increased quality of sequence alignment means a better performance. The motivation of present work is to increase ability of the existing scoring scheme/algorithm by considering residue–residue correlations better. Based on a coarse-grained approach, the hydrophobic force between each pair of residues is written out from protein sequence. It results in the construction of an intramolecular hydrophobic force network that describes the whole residue–residue interactions of each protein molecule, and characterizes protein's biological properties in the hydrophobic aspect. A former work has suggested that such network can characterize the top weighted feature regarding hydrophobicity. Moreover, for each homologous protein of a family, the corresponding network shares some common and representative family characters that eventually govern the conservation of biological properties during protein evolution. In present work, we score such family representative characters of a protein by the deviation of its intramolecular hydrophobic force network from that of background. Such score can assist the existing scoring schemes/algorithms, and boost up the ability of multiple sequences alignment, e.g. achieving a prominent increase (50%) in searching the structurally alike residue segments at a low identity level. As the theoretical basis is different, the present scheme can assist most existing algorithms, and improve their efficiency remarkably.
