Amphioxus is a crucial organism for the study of vertebrate evolution. Although a genomic BAC library of Branchiostoma floridae has been constructed, we report here another BAC library construction of its distant relative species Branchiostoma belcheri. The amphioxus BAC library established in present study consists of 45,312 clones arrayed in one hundred and eighteen 384-well plates. The average insert fragment size was 120 kb estimated by Pulsed Field Gel Electrophoresis (PFGE) analysis of 318 randomly selected clones. The representation of the library is about 12 equivalent to the genome, allowing a 99.9995% probability of recovering any specific sequence of interest. We further screened the library with 4 single copied Amphi-Pax genes and identified total of 26 positive clones with average of 6.5 clones for each gene. The result indicates this library is well suited for many applications and should also serve as a useful complemental resource for the scientific community.

Attempts were made to quantify the environmental impacts of the basement walls of two commercial buildings in London. Four different retaining wall options were designed based on steel and concrete systems for each of the sites. It was considered that excavation would take place with the aid of a one or two anchors system. Evaluation of embodied energy (EE) and CO2 emissions for each of the wall designs and anchoring systems were compared. Results show that there are notable differences in EE between different wall designs. Using the averaged set of Embodied Energy Intensity (EEI) values, the use of recycled steel over virgin steel would reduce the EE of the wall significantly. The difference in anchor designs is relatively insignificant, and therefore the practicality of the design for the specific site should be the deciding factor for anchor types. Generally, the scale of environmental impacts due to constructions is large compared to other aspects in life as demonstrated with the comparisons to car emissions and household energy consumption. We constructed a high redundancy bacterial artificial chromosome library of a seriously endangered Old World Monkey, the Yunnan snub-nosed monkey (Rhinopithecus bieti) from China. This library contains a total of 136 320 BAC clones. The average insert size of BAC clones was estimated to be 148 kb. The percentage of small inserts (50-100 kb) is 2.74%, and only 2.67% non-recombinant clones were observed. Assuming a similar genome size with closely related primate species, the Yunnan snub-nosed monkey BAC library has at least six times the genome coverage. By end sequencing of randomly selected BAC clones, we generated 201 sequence tags for the library. A total of 139 end-sequenced BAC clones were mapped onto the chromosomes of Yunnan snub-nosed monkey by fluorescence in-situ hybridization, demonstrating a high degree of synteny conservation between humans and Yunnan snub-nosed monkeys. Blast search against human genome showed a good correlation between the number of hit clones and the size of the chromosomes, an indication of unbiased chromosomal distribution of the BAC library. This library and the mapped BAC clones will serve as a valuable resource in comparative genomics studies and large-scale genome sequencing of nonhuman primates. The Fort Canning Tunnel is the first road tunnel in Singapore to be built using the sprayed concrete lining (SCL) method. The major technical challenge of this was to construct a 15m wide tunnel by mining in soft ground under a shallow overburden of 3m to 9m. This paper describes the geotechnical investigations and monitoring controls for the safe and progressive execution of the works, such as soil investigations, trial forepoling works, surface settlement monitoring, tunnel settlement monitoring, face movement monitoring, and the observational approach to construction. The monitored field data showed the volume loss to range from 0.4% to 2.1%, and the observed surface settlement trough was found to agree well with the theoretical Gaussian trough. Other observations made include substantial surface settlements induced by the stress relief at and ahead of the tunnel face in spite of the forepoling umbrella, and the higher volume losses associated with higher overburden. Tunnel face movements were observed during installation of forepoling. These observations are of interest to engineers planning future SCL tunnels in similar conditions.

Histo-blood group antigens CD173 (H2) and CD174 (Lewis Y) are known to be developmentally regulated carbohydrate antigens which are expressed to a varying degree on many human carcinomas. We hypothesized that they might represent markers of cancer-initiating cells (or cancer stem cells, CSC). In order to test this hypothesis, we examined the co-expression of CD173 and CD174 with stem cell markers CD44 and CD133 by flow cytometry analysis, immunocytochemistry, and immunohistochemistry on cell lines and tissue sections from breast cancer. In three breast cancer cell lines, the percentage of CD173(+)/CD44(+) cells ranged from 17% to > 60% and of CD174(+)/CD44(+) from 21% to 57%. In breast cancer tissue sections from 15 patients, up to 50% of tumor cells simultaneously expressed CD173, CD174, and CD44 antigens. Co-expression of CD173 and CD174 with CD133 was also observed, but to a lesser percentage. Co-immunoprecipitation and sandwich ELISA experiments on breast cancer cell lines suggested that CD173 and CD174 are carried on the CD44 molecule. The results show that in these tissues CD173 (H2) and CD174 (LeY) are associated with CD44 expression, suggesting that these carbohydrate antigens are markers of cancer-initiating cells or of early progenitors of breast carcinomas.

Modern fishing boats have to be built not only on perfect lines but also with sound and strong construction materials that will ensure a long lasting trouble free service commensurate with the heavy capital investment involved. Choice of construction materials for fishing boats need careful scrutiny as they have to perform too well under most aggressive environments-sea-water and marine atmosphere. A number of alternative boat-building materials are now available whose comparative merits and demerits as well as comparative costs are brought out in this paper.

The study was conducted in collaboration with the ECFC project of the FAO (BGD/97/017) in Cox's Bazar to develop a low cost solar tunnel dryer for the production of high quality marine dried fish. The study areas were Kutubdiapara, Maheshkhali and Shahparirdip under Cox's Bazar district. Three different models of low cost solar dryer were constructed with locally available materials such as bamboo, wood, bamboo mat, hemp, canvas, wire, nails, rope, tin, polythene and net. Size of the dryers were: 20x4x3 ft ; 30x3x3 ft and 65x3x3 ft with the costs of Tk. 3060, 3530, 9600 for dryer 1, 2 and 3, respectively having different models. The drying capacities were 50, 150, 500 kg for dryer 1, 2 and 3 respectively. The average temperature range inside the dryers were 29-43°C, 34-51°C and 37-57°C for dryer 1, 2 and 3 respectively as recorded at 8:30h to 16:30h. The relative humidity were in the ranges of 22-42%, 27-39% and 24-41 % in dryer 1, 2 and 3 respectively. The fish samples used were Bombay duck, Silver Jew fish and Ribbon fish. The total drying time was in the range of 30-42, 28-38 and 24-34 hours to reach the moisture content of 12.3-14.5, 11.8-14.3, and 11.6-14.1% in dryer 1, 2 and 3 respectively. Among these three fish samples the drying was faster in Silver Jew fish followed by Bombay duck and Ribbon fish in all the three dryer.
