The purpose of this study is to develop and evaluate techniques that improve the spatial resolution of the channels already selected in the preliminary studies for Geostationary Observatory for Microwave Atmospheric Soundings (GOMAS). Reference high resolution multifrequency brightness temperatures scenarios have been derived by applying radiative transfer calculation to the spatially and microphysically detailed output of meteorological events simulated by the University of Wisconsin - Non-hydrostatic Model System (UW-NMS). Three approaches, Wiener filter, Super-Resolution and Image Fusion have been applied to some representative GOMAS frequency channels to enhance the resolution of antenna temperatures. The Wiener filter improved resolution of the largely oversampled images by a factor 1.5- 2.0 without introducing any penalty in the radiometric accuracy. Super-resolution, suitable for not largely oversampled images, improved resolution by a factor ~1.5 but introducing an increased radiometric noise by a factor 1.4-2.5. The image fusion allows finally to further increase the spatial frequency of the images obtained by the Wiener filter increasing the total resolution up to a factor 5.0 with an increased radiometric noise closely linked to the radiometric frequency and to the examined case study.

Brown rot caused by Monilinia laxa and Monilinia fructigena is considered one of the most important diseases affecting Prunus species. Although some losses can result from the rotten fruits in the orchard, most of the damage is caused to fruits during the post-harvest phase. Several studies reported that brown rot incidence during fruit development highly varies; it was found that at a period corresponding to the the pit hardening stage, fruit susceptibility drastically decreases, to be quickly restored afterwards. However the molecular basis of this phenomenon is still not well understood. Furthermore, no difference in the rot incidence was found between wound and un-wound fruits, suggesting that resistance associated more to a specifc biochemical response of the fruit, rather than to a higher mechanical resistance. So far, the interaction Monilinia-peach was analyzed through chemical approaches. In this study, a bio-molecular approach was undertaken in order to reveal alteration in gene expression associated to the variation of susceptibility. In this thesis three different methods for gene expression analysis were used to analyze the alterations in gene expression occurring in peach fruits during the pit hardening stage, in a period encompassing the temporary change in Monilinia susceptibility: real time PCR, microarray and cDNA AFLP techniques. In 2005, peach fruits (cv.K2) were weekly harvested during a 19-week long-period, starting from the fourth week after full bloom, until full maturity. At each sampling time, three replicates of 5 fruits each were dipped in the M.laxa conidial suspension or in distilled water, as negative control. The fruits were maintained at room temperature for 3 hours; afterwards, they were peeled with a scalpel; the peel was immediately frozen in liquid nitrogen and transferred to -80 °C until use. The degree of susceptibility of peach fruit to the pathogen was determined on 3 replicates of 20 fruits each, as percentage of infected fruits, after one week at 20 °C. Real time PCR analysis was performed to study the variation in expression of those genes encoding for the enzymes of the phenylpropanoid pathway (phenylalanine ammonia lyase (PAL), chalcone synthase (CHS), cinnamate 4-hydroxylase (C4H), leucoanthocyanidine reductase (LAR), hydroxycinnamoyl CoA quinate hydroxycinnamoyl transferase (HQT) and of the jasmonate pathway, such as lipoxygenase (LOX), both involved in the production of important defense compounds. Alteration in gene expression was monitored on fruit samples of a period encompassing the pit hardening stage and the corresponding temporary resistance to M.laxa infections, weekly, from the 6thto the 12th week after full bloom (AFB) inoculated with M. laxa or mock-inoculated. The data suggest a critical change in the expression level of the phenylpropanoid pathway from the 7th to the 8th week AFB; such change could be directly physiologically associated to the peach growth and it could indirectly determine the decrease of susceptibility of peach fruit to Monilinia rot during the subsequent weeks. To investigate on the transcriptome variation underneath the temporary loss of susceptibility of peach fruits to Monilinia rot, the microarray and the cDNA AFLP techniques were used. The samples harvested on the 8th week AFB (named S, for susceptible ones) and on the 12th week AFB (named R, for resistant ones) were compared, both inoculated or mock-inoculated. The microarray experiments were carried out at the University of Padua (Dept. of Environmental Agronomy and Crop Science), using the μPEACH1.0 microarray together with the suited protocols. The analysis showed that 30 genes (corresponding to the 0.6% of the total sequences (4806) contained in the μPeach1.0 microarray) were found up-regulated and 31 ( 0.6%) down regulated in RH vs. SH fruits. On the other hand, 20 genes (0.4%) were shown to be up-regulated and 13 (0.3%) down-regulated in the RI vs. SI fruit. No genes were found differentially expressed in the mock-inoculated resistant fruits (RH) vs. the inoculated resistant ones (RI). Among the up-regulated genes an ATP sulfurylase, an heat shock protein 70, the major allergen Pru P1, an harpin inducing protein and S-adenosylmethionine decarboxylase were found, conversely among the down-regulated ones, cinnamyl alcohol dehydrogenase, an histidine- containing phosphotransfer protein and the ferritin were found. The microarray experimental results and the data indirectly derived, were tested by Real Time PCR analysis. cDNA AFLP analysis was also performed on the same samples. 339 transcript derived fragments considered significant for Monilinia resistance, were selected, sequenced and classified. Genes potentially involved in cell rescue and defence were well represented (8%); several genes (12.1%) involved in the protein folding, post-transductional modification and genes (9.2%) involved in cellular transport were also found. A further 10.3% of genes were classified as involved in the metabolism of aminoacid, carbohydrate and fatty acid. On the other hand, genes involved in the protein synthesis (5.7%) and in signal transduction and communication (5.7%) were found. Among the most interesting genes found differentially expressed between susceptible and resistant fruits, genes encoding for pathogenesis related (PR) proteins were found. To investigate on the association of Monilinia resistance and PR biological function, the major allergen Pru P1 (GenBank accession AM493970) and its isoform (here named Pru P2), were expressed in heterologous system and in vitro assayed for their anti-microbial activity. The ribonuclease activity of the recombinant Pru P1 and Pru P2 proteins was assayed against peach total RNA. As the other PR10 proteins, they showed a ribonucleolytic activity, that could be important to contrast pathogen penetration. Moreover Pru P1 and Pru P2 recombinant proteins were checked for direct antimicrobial activity. No inhibitory effect of Pru P1 or Pru P2 was detected against the selected fungi.

I rifiuti come oggetti impegnano tutte le istituzioni umane in una lotta di definizione del posto che occupano e quindi del valore che assumono. In tale dinamica la gestione dei rifiuti diventa un fatto sociale totale che coinvolge tutte le istituzioni umane in una lotta di definizione territorializzata. La storia del movimento ambientalista ci mostra come partendo dal disagio nei confronti dell’oggetto si è passati ad un disagio nei confronti delle idee che lo generano. Modernizzazione ecologica e modernizzazione democratica sembrano andare per un certo periodo d’accordo. Nei casi di conflittualità recente, e nello studio di caso approfondito di un piano provinciale della gestione rifiuti, il carattere anticipatore dell’attivismo ambientalista, sta rendendo sempre più costosi e incerti, investimenti e risultati strategici . Anche i principi delle politiche sono messi in discussione. La sostenibilità è da ricercare in una relativizzazione dei principi di policy e degli strumenti tecnici di valutazione (e.g. LCA) verso una maggiore partecipazione di tutti gli attori. Si propone un modello di governance che parta da un coordinamento amministrativo territoriale sulle reti logistiche, quindi un adeguamento geografico degli ATO, e un loro maggior ruolo nella gestione del processo di coordinamento e pianificazione. Azioni queste che devono a loro volta aprirsi ai flussi (ecologici ed economici) e ai loro attori di riferimento: dalle aziende multiutility agli ambientalisti. Infine è necessario un momento di controllo democratico che può avere una funzione arbitrale nei conflitti tra gli attori o di verifica. La ricerca si muove tra la storia e la filosofia, la ricerca empirica e la riflessione teorica. Sono state utilizzate anche tecniche di indagine attiva, come il focus group e l’intervista.

La tesi si pone come finalità quella di analizzare un comprensorio territoriale dal punto di vista insediativo, cercando di coglierne le peculiarità e i mutamenti nel lungo periodo attraverso un uso incrociato di fonti scritte e archeologiche. La ricerca ha preso avvio dall’analisi del Saltopiano, uno dei distretti di ambito rurale che si ritrova nelle fonti tra IX-XII secolo, in passato già affrontato dalla storiografia specialmente in relazione all’organizzazione istituzionale delle aree rurali tra Longobardia e Romania durante i secoli altomedievali. Attraverso l’esame delle fonti scritte edite si è cercato di ricostruire il quadro dell’organizzazione territoriale, partendo dalla disamina dei centri di potere laici ed ecclesiastici che a questa area avevano rivolto il proprio interesse patrimoniale e politico, ma proponendo in modo analitico i dati che forniscono indicazioni dirette in relazione all’organizzazione insediativa e quindi alla gestione del territorio dal punto di vista socioeconomico. E’ stato posto in risalto il carattere di un insediamento rurale a maglie larghe, secondo la scansione in fundi e la presenza di poli di accentramento importanti come pievi, castra, vici e con una compresenza, per quanto ristretta a pochi esempi significativi, di altre forme di organizzazione come la curtis e la massa. Con la prosecuzione dello studio del territorio in senso diacronico, prima la scomparsa del riferimento al Saltopiano, poi la progressiva conquista del contado da parte del Comune di Bologna ha determinato un vero e proprio mutamento nell’approccio di analisi. E’ stato dato spazio all’analisi di fondi inediti (conservati principalmente all’Archivio di Stato di Bologna) e specificamente legati alla realtà territoriale studiata. In primo luogo, sono stati esaminati gli estimi del contado (Galliera e Massumatico), una fonte già frequentata in passato da altri studiosi, soprattutto con un interesse dal punto di vista demografico e economico. Nel caso specifico, sono stati estrapolati dalle prime rilevazioni fiscali del 1235 e del 1245 e poi da quelle di primo Trecento i dati che restituiscono l’organizzazione del territorio in modo concreto. Partendo dalle riflessioni di studi svolti in passato, che avevano considerato il fondamentale inserimento di importanti famiglie cittadine nella gestione sempre più ampia dei beni agricoli nel contado è stata avviata l’analisi di un altro fondo inedito, quello dei registri del Vicariato di Galliera (in particolare quelli concernenti la denuncia dei “danni dati” sulle proprietà agricole), da cui emerge in modo evidente la presenza di famiglie come i Guastavillani, i Caccianemici, i Lambertini. Tali dati, in associazione a quelli tratti dagli estimi, hanno fornito elementi essenziali per la comprensione del territorio rurale nel suo complesso e nei rapporti di interdipendenza tra le diverse componenti sociali. Una terza parte della tesi è dedicata nella sua totalità all’analisi delle fonti materiali che forniscono dati per lo studio dell’insediamento medievale nel territorio compreso tra gli attuali comuni di S. Pietro in Casale e Galliera. Partendo da alcune ricerche preliminari compiute negli anni ’90 del secolo scorso, è stato impostato un progetto di ricerca archeologica articolatosi in due campagne di ricognizione di superficie e in una prima campagna di scavo tramite sondaggio presso la torre di Galliera, al fine di ricavare dati di prima mano in un’area pressoché inesplorata dal punto di vista archeologico. Nonostante i limiti riscontrati dal punto di vista pratico, a causa del terreno fortemente alluvionato, si sono raccolti dati specifici che aiutano a inquadrare questo comprensorio e a confrontarlo con altre aree della regione e in particolare del comitato bolognese studiate in ricerche analoghe, mettendone in evidenza le specificità e le caratterizzazioni. Inoltre, alcune importanti persistenze materiali (un sistema di torri di cui rimangono alcuni esempi ancora conservati in alzato) hanno permesso di gettare luce sul valore commerciale e quindi strategico dell’area, soprattutto in funzione del passaggio delle merci lungo una via fluviale fondamentale tra XIII-XIV secolo nel collegare Ferrara a Bologna.

Many potential diltiazem related L-VDCC blockers were developed using a multidisciplinary approach. This current study was to investigate and compare diltiazem with to the newly developed compounds by mouse Langendorff-perfused heart, Ca2+-transient and on recombinant L-VDCC. Twenty particular compounds were selected by the ligand-based virtual screening procedure (LBVS). From these compounds, five of them (5b, M2, M7, M8 and P1) showed a potent and selective inotropic activity on guinea-pig left atria driven 1 Hz. Further assays displayed an interesting negative inotropic effect of M2, M8, P1 and M7 on guinea pig isolated left papillary muscle driven at 1 Hz, a relevant vasorelaxant activity of 5b, M2, M7, M8 and P1 on K+-depolarized guinea-pig ileum longitudinal smooth muscle and a significant inhibition of contraction of 5b, M2, M8 and P1 on carbachol stimulated ileum longitudinal smooth muscle. Wild-type human heart and rabbit lung α1 subunits were expressed (combined with the regulatory α2δ and β3 subunits) in Xenopus Leavis oocytes using a two-electrode voltage clamp technique. Diltiazem is a benzothiazepine Ca2+ channel blocker used clinically for its antihypertensive and antiarrhythmic effects. Previous radioligand binding assays revealed a complex interaction with the benzothiazepine binding site for M2, M7 and M8. (Carosati E. et al. J. Med Chem. 2006, 49; 5206). In agreement with this findings, the relative order of increased rates of contraction and relaxation at lower concentrations s(≤10-6M) in unpaced hearts was M7>M2>M8>P1. Similar increases in Ca2+ transient were observed in cardiomyocytes. Diltiazem showed negative inotropic effects whereas 5b had no significant effect. Diltiazem blocks Ca2+current in a use-dependent manner and facilitates the channel by accelerating the inactivation and decelerating the recovery from inactivation. In contrast to diltiazem, the new analogs had no pronounced use-dependence. Application of 100 μM M8, M2 showed ~ 10% tonic block; in addition, M8, M2 and P1 shifted the steady state inactivation in hyperpolarized direction and the current inactivation time was significantly decreased compared with control (219.6 ± 11.5 ms, 226 ± 14.5 vs. 269 ± 12.9 vs. 199.28 ± 8.19 ms). Contrary to diltiazem, the recovery from the block by M8 and M2 was comparable to control. Only P1 showed a significantly decrease of the time for the recovery from inactivation. All of the compounds displayed the same sensitivity on the Ca2+ channel rabbit lung α1 except P1. Taken together, these findings suggest that M8, M2 and P1 might directly decrease the binding affinity or allow rapid dissociation from the benzothiazepine binding site.

Chemistry can contribute, in many different ways to solve the challenges we are facing to modify our inefficient and fossil-fuel based energy system. The present work was motivated by the search for efficient photoactive materials to be employed in the context of the energy problem: materials to be utilized in energy efficient devices and in the production of renewable electricity and fuels. We presented a new class of copper complexes, that could find application in lighting techhnologies, by serving as luminescent materials in LEC, OLED, WOLED devices. These technologies may provide substantial energy savings in the lighting sector. Moreover, recently, copper complexes have been used as light harvesting compounds in dye sensitized photoelectrochemical solar cells, which offer a viable alternative to silicon-based photovoltaic technologies. We presented also a few supramolecular systems containing fullerene, e.g. dendrimers, dyads and triads.The most complex among these arrays, which contain porphyrin moieties, are presented in the final chapter. They undergo photoinduced energy- and electron transfer processes also with long-lived charge separated states, i.e. the fundamental processes to power artificial photosynthetic systems.

The fundamental goal of this thesis is the determination of the isospin dependence of the Ar+Ni fusion-evaporation cross section. Three Ar isotope beams, with energies of about 13AMeV, have been accelerated and impinged onto isotopically enriched Ni targets, in order to produce Pd nuclei, with mass number varying from 92 to 104. The measurements have been performed by the high performance 4pi detector INDRA, coupled with the magnetic spectrometer VAMOS. Even if the results are very preliminary, the obtained fusion-evaporation cross sections behaviour gives a hint at the possible isospin dependence of the fusion-evaporation cross sections.

Leaf rust caused by Puccinia triticina is a serious disease of durum wheat (Triticum durum) worldwide. However, genetic and molecular mapping studies aimed at characterizing leaf rust resistance genes in durum wheat have been only recently undertaken. The Italian durum wheat cv. Creso shows a high level of resistance to P. triticina that has been considered durable and that appears to be due to a combination of a single dominant gene and one or more additional factors conferring partial resistance. In this study, the genetic basis of leaf rust resistance carried by Creso was investigated using 176 recombinant inbred lines (RILs) from the cross between the cv. Colosseo (C, leaf rust resistance donor) and Lloyd (L, susceptible parent). Colosseo is a cv. directly related to Creso with the leaf rust resistance phenotype inherited from Creso, and was considered as resistance donor because of its better adaptation to local (Emilia Romagna, Italy) cultivation environment. RILs have been artificially inoculated with a mixture of 16 Italian P. triticina isolates that were characterized for virulence to seedlings of 22 common wheat cv. Thatcher isolines each carrying a different leaf rust resistance gene, and for molecular genotypes at 15 simple sequence repeat (SSR) loci, in order to determine their specialization with regard to the host species. The characterization of the leaf rust isolates was conducted at the Cereal Disease Laboratory of the University of Minnesota (St. Paul, USA) (Chapter 2). A genetic linkage map was constructed using segregation data from the population of 176 RILs from the cross CL. A total of 662 loci, including 162 simple sequence repeats (SSRs) and 500 Diversity Arrays Technology markers (DArTs), were analyzed by means of the package EasyMap 0.1. The integrated SSR-DArT linkage map consisted of 554 loci (162 SSR and 392 DArT markers) grouped into 19 linkage blocks with an average marker density of 5.7 cM/marker. The final map spanned a total of 2022 cM, which correspond to a tetraploid genome (AABB) coverage of ca. 77% (Chapter 3). The RIL population was phenotyped for their resistance to leaf rust under artificial inoculation in 2006; the percentage of infected leaf area (LRS, leaf rust susceptibility) was evaluated at three stages through the disease developmental cycle and the area under disease progress curve (AUDPC) was then calculated. The response at the seedling stage (infection type, IT) was also investigated. QTL analysis was carried out by means of the Composite Interval Mapping method based on a selection of markers from the CL map. A major QTL (QLr.ubo-7B.2) for leaf rust resistance controlling both the seedling and the adult plant response, was mapped on the distal region of chromosome arm 7BL (deletion bin 7BL10-0.78-1.00), in a gene-dense region known to carry several genes/QTLs for resistance to rusts and other major cereal fungal diseases in wheat and barley. QLr.ubo-7B.2 was identified within a supporting interval of ca. 5 cM tightly associated with three SSR markers (Xbarc340.2, Xgwm146 e Xgwm344.2), and showed an R2 and an LOD peak value for the AUDPC equal to 72.9% an 44.5, respectively. Three additional minor QTLs were also detected (QLr.ubo-7B.1 on chr. 7BS; QLr.ubo-2A on chr. 2AL and QLr.ubo-3A on chr. 3AS) (Chapter 4). The presence of the major QTL (QLr.ubo-7B.2) was validated by a linkage disequilibrium (LD)-based test using field data from two different plant materials: i) a set of 62 advanced lines from multiple crosses involving Creso and his directly related resistance derivates Colosseo and Plinio, and ii) a panel of 164 elite durum wheat accessions representative of the major durum breeding program of the Mediterranean basin. Lines and accessions were phenotyped for leaf rust resistance under artificial inoculation in two different field trials carried out at Argelato (BO, Italy) in 2006 and 2007; the durum elite accessions were also evaluated in two additional field experiments in Obregon (Messico; 2007 and 2008) and in a green-house experiment (seedling resistance) at the Cereal Disease Laboratory (St. Paul, USA, 2008). The molecular characterization involved 14 SSR markers mapping on the 7BL chromosome region found to harbour the major QTL. Association analysis was then performed with a mixed-linear-model approach. Results confirmed the presence of a major QTL for leaf rust resistance, both at adult plant and at seedling stage, located between markers Xbarc340.2, Xgwm146 and Xgwm344.2, in an interval that coincides with the supporting interval (LOD-2) of QLr.ubo-7B.2 as resulted from the RIL QTL analysis. (Chapter 5). The identification and mapping of the major QTL associated to the durable leaf rust resistance carried by Creso, together with the identification of the associated SSR markers, will enhance the selection efficiency in durum wheat breeding programs (MAS, Marker Assisted Selection) and will accelerate the release of cvs. with durable resistance through marker-assisted pyramiding of the tagged resistance genes/QTLs most effective against wheat fungal pathogens.

The advent of distributed and heterogeneous systems has laid the foundation for the birth of new architectural paradigms, in which many separated and autonomous entities collaborate and interact to the aim of achieving complex strategic goals, impossible to be accomplished on their own. A non exhaustive list of systems targeted by such paradigms includes Business Process Management, Clinical Guidelines and Careflow Protocols, Service-Oriented and Multi-Agent Systems. It is largely recognized that engineering these systems requires novel modeling techniques. In particular, many authors are claiming that an open, declarative perspective is needed to complement the closed, procedural nature of the state of the art specification languages. For example, the ConDec language has been recently proposed to target the declarative and open specification of Business Processes, overcoming the over-specification and over-constraining issues of classical procedural approaches. On the one hand, the success of such novel modeling languages strongly depends on their usability by non-IT savvy: they must provide an appealing, intuitive graphical front-end. On the other hand, they must be prone to verification, in order to guarantee the trustworthiness and reliability of the developed model, as well as to ensure that the actual executions of the system effectively comply with it. In this dissertation, we claim that Computational Logic is a suitable framework for dealing with the specification, verification, execution, monitoring and analysis of these systems. We propose to adopt an extended version of the ConDec language for specifying interaction models with a declarative, open flavor. We show how all the (extended) ConDec constructs can be automatically translated to the CLIMB Computational Logic-based language, and illustrate how its corresponding reasoning techniques can be successfully exploited to provide support and verification capabilities along the whole life cycle of the targeted systems.
