Human MxA protein belongs to the superfamily of dynamin-like large GTPases that are involved in intracellular membrane trafficking. MxA is induced by interferons-alpha/beta (IFN-alpha/beta) and is a key component of the antiviral response against RNA viruses. Here, we show that MxA localizes to membranes that are positive for specific markers of the smooth endoplasmic reticulum, such as Syntaxin17, but is excluded from other membrane compartments. Overexpression of MxA leads to a characteristic reorganization of the associated membranes. Interestingly, Hook3, mannose-6-phosphate receptor, and Lamp-1, which normally accumulate in cis-Golgi, endosomes, and lysosomes, respectively, also colocalized with MxA, indicating that these markers were redistributed to the MxA-positive compartment. Functional assays, however, did not show any effect of MxA on endocytosis or the secretory pathway. The present results demonstrate that MxA is an IFN-induced antiviral effector protein that resembles the constitutively expressed large GTPase family members in its capacity to localize to and reorganize intracellular membranes.

The role of the small GTP-binding protein Rho in the process of smooth muscle cell (SMC) phenotypic modulation was investigated using cultured rabbit aortic SMCs. Both Rho transcription and Rho protein expression were high for the first 3 days of culture (contractile state cells), with expression decreasing after change to the synthetic state and peaking upon return to the contractile phenotype. Activation of Rho (indicated by translocation to the membrane) also peaked upon return to the contractile state and was low in synthetic state SMCs. Transient transfection of synthetic state rabbit SMCs with constitutively active Rho (val14rho) caused a dramatic decrease in cell size and reorganization of cytoskeletal proteins to resemble those of the contractile phenotype; alpha-actin and myosin adopted a tightly packed, highly organized arrangement, whereas vimentin localized to the immediate perinuclear region and focal adhesions were enlarged. Conversely, specific inhibition of endogenous Rho, by expression of C3 transferase, resulted in the complete loss of actin and myosin filaments without affecting the distribution of vimentin. Focal adhesions were reduced in number. Thus, Rho plays a key role in regulating SMC phenotypic expression.

Smooth muscle cell (SMC) phenotypic modulation from the mature ’contractile’ to a less differentiated ’synthetic’ phenotype involves not only altered expression but also a reorganisation of contractile and cytoskeletal proteins. Objective: To investigate the role of RhoA, a known regulator of the actin cytoskeleton, in SMC phenotypic regulation. Methods: Rho transcription (RT-PCR), expression (Western analysis) and activation (membrane translocation or Rho ’pull-down’ assay) was investigated in cultured rabbit aortic SMC during phenotypic modulation, and under the influence of known SM-regulatory proteins (thrombin, heparin and TGF- β). Rho’s effect on cell morphology was examined by transient transfection of ’synthetic’ state SMC with either constitutively active Rho (Val14RhoA) or its inhibitor, C3 transferase. Results: RhoA transcription was elevated in the first 3 days of primary culture, and protein expression peaked at 2 days post-confluence when SMC return to a more ’contractile’ state. However, RhoA showed augmented activation at three time-points in primary culture: the transition point when SMCs enter logarithmic growth and are highly motile, upon reaching quiescence, and when they return to a more ’contractile’ state. Thrombin, heparin and TGF-β activated RhoA in ’synthetic’ state SMCs. Transfection with Val14RhoA caused a dramatic decrease in SMC size and a reorganization of cytoskeletal proteins, reminiscent of the ’contractile’ phenotype. Specific inhibition of endogenous Rho by C3 transferase resulted in an almost complete loss of contractile proteins. Conclusion: These data indicate that Rho is an important determining factor of SMC functional state.

A presente dissertação teve como objetivo verificar as razões pelas quais a empresa Metal Leve cedeu o controle acionário a uma empresa concorrente de porte internacional e comparar a situação existente à época da sua desnacionalização e o estágio em que se encontra a nova controladora Mahle Metal Leve S. A. em termos de produção e gestão, após a reorganização produtiva. Esse processo de internacionalização produtiva acarretou modificações na empresa ensejando uma reestruturação da produção e da gestão e um novo círculo vicioso, constituindo as bases de um novo crescimento econômico, com um projeto estratégico de longo prazo, associado ao seu poder econômico, a sua capacidade gerencial e a sua tradição. O estudo está fundamentado em um conjunto de informações sobre os dois momentos, focalizando os resultados financeiros, aspectos gerenciais, liderança, capacidade competitiva e a evolução ao longo desses dois momentos comparados. Concluindo que não apenas o fenômeno da globalização da economia internacional tornou inevitável a cessão do controle acionário. Porém, também faltou visão estratégica para perceber as mudanças que inevitavelmente ocorriam a sua volta e que a nova controladora, a Mahler Metal Leve, trouxe uma competência gerencial que resultou em ganhos de produtividade e melhorou sua competitividade

A presente tese propõe uma interpretação exegética da profecia de Jr 30-31 na perspectiva social. Jr 30-31 forma uma unidade literária no livro de Jeremias, composta por subunidades que podem ser designadas de perícopes. Grande parte das expectativas salvíficas deste trecho literário devem ser atribuídas à literatura originária do livro, e provém das articulações sociais engendradas por Jeremias no fim do século 7 e início do século 6 a.C. em prol dos empobrecidos da antiga sociedade palestina Israel/Norte e de Judá/Sul. Os primeiros ditos salvíficos de Jr 30-31 surgiram na época de Josias (Jr 30,10-11.18-21; 31,2-5). Nessa época, originaram-se as expectativas de salvação dirigidas para as populações israelitas do Norte. Outro intenso surgimento das expectativas salvíficas aconteceu nos anos imediatamente posteriores à queda de Judá, em 587 a.C., quando Jeremias novamente direcionou uma palavra de esperança aos pobres do Israel/Norte, e incluiu também em sua mensagem aqueles que permaneceram na terra de Judá/Sul depois do saque babilônico. Nesse cenário podem ser localizadas as seguintes perícopes: 30,3.5-7.12-17; 31,15.16-20.21-22.27-28.31-34. A presente tese supõe que, de modo geral, Jr 30-31 seja uma reconfirmação da desmilitarização e da desurbanização de Jerusalém ocorridas naquele período, já que esse novo cenário político e econômico favoreceu os desprestigiados da Palestina. O tribalismo é o moto das expectativas salvíficas da literatura jeremiana original. No engendramento de uma nova sociedade, retribalizada, livre do jugo monárquico e dos imperialismos, Jr 30-31 defendem a posse da terra aos camponeses que sofreram espoliações do império assírio e dos reis judaítas. Com a queda do Estado de Judá, os empobrecidos poderiam retomar suas vidas e possuir a terra como meio de produção e subsistência. A relação entre as palavras de salvação e o tribalismo também pode ser notado em outros trechos do livro de Jeremias. A estruturação verbal proponente de destruição e reconstrução de 31,28 pode ser encontrada em Jr 1,10; 18,7.9; 24,6; 42,10 e 45,4. As promessas de salvação contidas em Jr 1,10, 31,27-28 e 42,10 anunciam a continuidade da vida na terra de Judá depois da catástrofe de 587 a.C. Essa ideia também pode ser percebida em Jr 23,5-6, 30,8-9. Em Jr 24,6, por sua vez, lê-se uma promessa para os exilados de Judá, que viviam na Babilônia sob o sistema tribal. Em Jr 3,6-13.19-25; 4,1-2, as expectativas salvíficas de Jeremias apresentam o caminho para a reorganização social através da conversão para Javé.

O livro de Malaquias apresenta oráculos que conservam informações relevantes do período pós exílico sob o domínio da Pérsia e, especialmente, os problemas religiosos focalizando a displicência dos sacerdotes no cumprimento de suas funções e os problemas sociais apontando para um amplo movimento de opressão externa e interna. Esta pesquisa buscou verificar, no primeiro capítulo, como a mensagem de Malaquias através de sua forma e conteúdos visava aplacar as insatisfações internas dos grupos que habitavam o território de Judá. Neste capítulo, foram abordadas as discussões histórico-literárias sobre data, autoria, destinatários, forma do anúncio. Ainda procurou-se realizar levantamentos históricos verificando o sistema e as estruturas de opressão dos persas no território de Judá e também as faces da reorganização da sociedade de Judá. O segundo capítulo enfatizou, mais propriamente, o trabalho exegético no quarto oráculo (2,17-3,5). Neste pode-se verificar uma síntese dos temas centrais da mensagem de Malaquias: O juízo divino que viria pela negligência religiosa e que exigia compromissos com a justiça prática na vida do povo. A partir das discussões exegéticas buscou-se verificar como o profeta apresentou ao povo e aos sacerdotes quais seriam os agentes e as ações transformadoras que Javé promoveria para restaurar a justiça e o culto purificado. O terceiro capítulo apresenta um tema recorrente nos oráculos do livro de Malaquias: a justiça. Para isso, foram analisadas três perícopes que tratam o tema da justiça enfocadamente, as influências do sistema judicial persa na prática da justiça cotidiana em Judá e a realidade de opressão nos sistema de parentesco entre as famílias (clãs) que habitavam o território de Judá - (2,10-16; 2,17-3,5 e 3,13-21). A pesquisa de Malaquias aponta para um profeta engajado política e socialmente. O Mensageiro manteve os ideais proféticos e desejou reacender os valores da aliança entre o povo e fortalecer a confiança na ação de Javé que restauraria a justiça na prática cotidiana.(AU)

As várias teorias acerca da estrutura de capital despertam interesse motivando diversos estudos sobre o assunto sem, no entanto, ter um consenso. Outro tema aparentemente pouco explorado refere-se ao ciclo de vida das empresas e como ele pode influenciar a estrutura de capital. Este estudo teve como objetivo verificar quais determinantes possuem maior relevância no endividamento das empresas e se estes determinantes alteram-se dependendo do ciclo de vida da empresa apoiada pelas teorias Trade Off, Pecking Order e Teoria da Agência. Para alcançar o objetivo deste trabalho foi utilizado análise em painel de efeito fixo sendo a amostra composta por empresas brasileiras de capital aberto, com dados secundários disponíveis na Economática® no período de 2005 a 2013, utilizando-se os setores da BM&FBOVESPA. Como resultado principal destaca-se o mesmo comportamento entre a amostra geral, alto e baixo crescimento pelo endividamento contábil para o determinante Lucratividade apresentando uma relação negativa, e para os determinantes Oportunidade de Crescimento e Tamanho, estes com uma relação positiva. Para os grupos de alto e baixo crescimento alguns determinantes apresentaram resultados diferentes, como a singularidade que resultou significância nestes dois grupos, sendo positiva no baixo crescimento e negativa no alto crescimento, para o valor colateral dos ativos e benefício fiscal não dívida apresentaram significância apenas no grupo de baixo crescimento. Para o endividamento a valor de mercado foi observado significância para o Benefício fiscal não dívida e Singularidade. Este resultado reforça o argumento de que o ciclo de vida influência a estrutura de capital

Aims: To explore newly diagnosed Type 2 diabetes patients' views about Scottish diabetes services at a time when these services are undergoing a major reorganization. To provide recommendations to maximize opportunities brought by the devolvement of services from secondary to primary healthcare settings. Methods: Qualitative panel study with 40 patients newly diagnosed with Type 2 diabetes, recruited from hospital clinics and general practices in Lothian, Scotland. Patients were interviewed three times over 1 year. The study was informed by grounded theory, which involves concurrent data collection and analysis. Results: Patients were generally satisfied with diabetes services irrespective of the types of care received. Most wanted their future care/review to be based in general practice for reasons of convenience and accessibility, although they dis-liked it when appointments were scheduled for different days. Many said they lacked the knowledge/confidence to know how to manage their diabetes in particular situations, and needed access to healthcare professionals who could answer their questions promptly. Patients expressed a need for primary care professionals who had diabetes expertise, but who had more time and were more accessible than general practitioners. Patients who had encountered practice lead nurses for diabetes spoke particularly positively of these professionals. Conclusions: Nurses with diabetes training are particularly well placed to provide information and support to patients in primary care. Ideally, practices should run 'one-stop' diabetes clinics to provide structured care, with easily accessible dietetics, podiatry and retinopathy screening. Newly diagnosed patients may benefit from being made more aware of specific services provided by charitable organizations such as Diabetes UK. © 2005 Diabetes UK.

This paper takes a practice perspective on organizing, re-conceptualizing coordination mechanisms as dynamic activities that are under continuous construction and modification in order to socially accomplish intra-organizational relationships and activities. The paper is based on the case of Servico, an organization undergoing a major reorganization of its value chain in response to a change in government regulation. We examine the specific performances through which the ostensive and abstract character of a coordination mechanism, ‘end-to-end management’, is defined and refined into a set of activities that actors can use to effect the re-organization of relationships between two divisions during the delivery of a critical regulatory goal. We find six cycles of iteration between the ostensive and performative nature of end-to-end, which progressively help to organize three phases in the reorganization of Servico; absence, presence and formalization. The discussion examines the processual evolution of these cycles and phases and their implications for the way that reorganization occurred. We draw these findings together in a process model that makes contributions to the literature on organizing, on ostensive and performative routines, and on organizational restructuring.

The demands towards the contemporary information systems are constantly increasing. In a dynamic business environment an organization has to be prepared for sudden growth, shrinking or other type of reorganization. Such change would bring the need of adaptation of the information system, servicing the company. The association of access rights to parts of the system with users, groups of users, user roles etc. is of great importance to defining the different activities in the company and the restrictions of the access rights for each employee, according to his status. The mechanisms for access rights management in a system are taken in account during the system design. In most cases they are build in the system. This paper offers an approach in user rights framework development that is applicable in information systems. This work presents a reusable extendable mechanism that can be integrated in information systems.

Abstract Various lubricating body fluids at tissue interfaces are composed mainly of combinations of phospholipids and amphipathic apoproteins. The challenge in producing synthetic replacements for them is not replacing the phospholipid, which is readily available in synthetic form, but replacing the apoprotein component, more specifically, its unique biophysical properties rather than its chemistry. The potential of amphiphilic reactive hypercoiling behaviour of poly(styrene-alt-maleic acid) (PSMA) was studied in combination with two diacylphosphatidylcholines (PC) of different chain lengths in aqueous solution. The surface properties of the mixtures were characterized by conventional Langmuir-Wilhelmy balance (surface pressure under compression) and the du Noüy tensiometer (surface tension of the non-compressed mixtures). Surface tension values and 31P NMR demonstrated that self-assembly of polymer-phospholipid mixtures were pH and concentration-dependent. Finally, the particle size and zeta potential measurements of this self-assembly showed that it can form negatively charged nanosized structures that might find use as drug or lipids release systems on interfaces such as the tear film or lung interfacial layers. The structural reorganization was sensitive to the alkyl chain length of the PC.

The utilization of solar energy by photovoltaic (PV) systems have received much research and development (R&D) attention across the globe. In the past decades, a large number of PV array have been installed. Since the installed PV arrays often operate in harsh environments, non-uniform aging can occur and impact adversely on the performance of PV systems, especially in the middle and late periods of their service life. Due to the high cost of replacing aged PV modules by new modules, it is appealing to improve energy efficiency of aged PV systems. For this purpose, this paper presents a PV module reconfiguration strategy to achieve the maximum power generation from non-uniformly aged PV arrays without significant investment. The proposed reconfiguration strategy is based on the cell-unit structure of PV modules, the operating voltage limit of gird-connected converter, and the resulted bucket-effect of the maximum short circuit current. The objectives are to analyze all the potential reorganization options of the PV modules, find the maximum power point and express it in a proposition. This proposition is further developed into a novel implementable algorithm to calculate the maximum power generation and the corresponding reconfiguration of the PV modules. The immediate benefits from this reconfiguration are the increased total power output and maximum power point voltage information for global maximum power point tracking (MPPT). A PV array simulation model is used to illustrate the proposed method under three different cases. Furthermore, an experimental rig is built to verify the effectiveness of the proposed method. The proposed method will open an effective approach for condition-based maintenance of emerging aging PV arrays.

The urgent need for teachers led the Florida legislature in 1887 to establish the Florida State Normal College at DeFuniak Springs. The college closed in 1905 with passage of the Buckman Act, which mandated a complete reorganization of state-supported higher education and ended coeducation for white students. This small college, open for eighteen years, was uniquely situated in time and place to examine larger questions in American educational history as well as contribute to the history of higher education in Florida, which developed differently than in other states.^ This historical case study used archival sources to examine this institution, and contribute to the history of the origins of Florida's system of higher education. Key questions guiding the research were the nature of the students, fundamental aspects of school life, the impact of the school on the students, and the role of the school in the development of higher education in Florida. Original sources included the Catalogs, Register and Minutes of the school. The census of 1900 was used to develop information on the backgrounds of the students. Findings were: DeFuniak Springs was chosen for the school because of the Florida Chautauqua; the school was coeducational and had few rules but the internalized social codes of the students resulted in almost no difficulties with discipline; the students, a majority of whom were women, were from middle-class southern families; the college compared favorably in faculty, facilities and curriculum to institutions elsewhere; although few students graduated, alumni played a key role in shaping Florida's common schools; and, the Buckman Act entirely changed the nature of higher education in Florida.^ Implications were: The coeducational nature of the college a hundred years ago significantly changes the picture of Florida's higher education; the school was small, but its influence far outlasted the institution; and, the school struggled with issues which continue to trouble modern educators such as finances, the legislature, student retention, underpreparedness, and the proper structuring of a curriculum, which indicates the persistence of these issues. ^

The purpose of this research was to explore the effects of a reform that took place in an elementary school during 2000/2001 as a result of a failure rating on the Florida Comprehensive Assessment Test on the structure and the personnel of the organization. ^ The exploration took place over a period of 10 months starting in August 2000 until June 2001. It focused on the effect of the failure rating on the: (a) structure and operation of the school; (b) morale, beliefs, behaviors, and daily lives of teachers and the principal; and (c) the effect of the reform effort on the leadership style of the principal, whether she became a transactional or a transformative leader. ^ The researcher assumed the role of a participant observer. Data sources were her personal recollections of major events that took place during the year of the reform, interviews, observations, and school documents. The sample included 15 teachers present during the time of the reform. Ten taught second through fifth grade. The remaining five participants were the music teacher, the counselor, and the writing, reading and technology specialists. Together they represented the instructional team or represented special education areas. ^ The findings indicated that the reform effort had an effect on the structure and the operation of the school. The changes included reorganization of the physical set up, changes in curriculum and instruction, changes in the means of communication among the staff, and the addition of new staff members including an official agent of change. The reform had a greater effect on the daily lives of teachers and their morale than on their beliefs and behaviors. Teachers reported that during the effort their daily lives were stressful and their morale very low due to the enormous expectations that they had to meet. On the other hand, the reform effort had a positive effect on the daily life, morale, beliefs, and behaviors of the principal. It energized her. She spoke positively about the change. She functioned as an effective, positive, resilient transactional leader who did what was necessary in order to enable the teachers to cope with the complex situation. ^
