A new model to predict the extent of crushing around a blasthole is presented. The model is based on the back-analysis of a comprehensive experimental program that included the direct measurement of the zone of crushing from 92 blasting tests on concrete blocks using two commercial explosives. The concrete blocks varied from low, medium to high strength and measured 1.5 in in length, 1.0 m in width and 1.1 m in height. A dimensionless parameter called the crushing zone index (CZI) is introduced. This index measures the crushing potential of a charged blasthole and is a function of the borehole pressure, the unconfined compressive strength of the rock material, dynamic Young's modulus and Poisson's ratio. It is shown that the radius of crushing is a function of the CZI and the blasthole radius. A good correlation between the new model and measured results was obtained. A number of previously proposed models could not approximate the conditions measured in the experimental work and there are noted discrepancies between the different approaches reviewed, particularly for smaller diameter holes and low strength rock conditions. The new model has been verified with full scale tests reported in the literature. Results from this validation and model evaluations show its applicability to production blasting. Regeneration of osseous defects by a tissue-engineering approach provides a novel means of treatment utilizing cell biology, materials science, and molecular biology. In this study the concept of tissue engineering was tested with collagen type I matrices seeded with cells with osteogenic potential and implanted into sites where osseous damage had occurred. Explant cultures of cells from human alveolar bone and gingiva were established. When seeded into a three-dimensional type I collagen-based scaffold, the bone-derived cells maintained their osteoblastic phenotype as monitored by mRNA and protein levels of the bone-related proteins including bone sialoprotein, osteocalcin, osteopontin, bone morphogenetic proteins 2 and 4, and alkaline phosphatase. These in vitro-developed matrices were implanted into critical-size bone defects in skulls of immunodeficient (SCID) mice. Wound healing was monitored for up to 4 weeks. When measured by microdensitometry the bone density within defects filled with osteoblast-derived matrix was significantly higher compared with defects filled with either collagen scaffold alone or collagen scaffold impregnated with gingival fibroblasts. New bone formation was found at all the sites treated with the osteoblast-derived matrix at 28 days, whereas no obvious new bone formation was identified at the same time point in the control groups. In situ hybridization for the human-specific Alu gene sequence indicated that the newly formed bone tissue resulted from both transplanted human osteoblasts and endogenous mesenchymal stem cells. The results indicate that cells derived from human alveolar bone can be incorporated into bioengineered scaffolds and synthesize a matrix, which on implantation can induce new bone formation.

Caustis blakei is an attractive cut foliage plant harvested from the wild in Australia and marketed under the name of koala fern. Previous attempts to propagate large numbers of this plant have been unsuccessful. The effect of four light irradiances on organogenesis from compact and friable callus of C. blakei was studied for 21 wk. Both callus types produced numerous primordial shoots but many failed to develop into green plantlets. However, significantly more primordial shoots and green plantlets developed on the friable callus than on the compact callus, and significantly more green plantlets were regenerated under the higher photon irradiances of 200 and 300 mumol m(-2) s(-1) than under the lower irradiances of 100 and 150 mumol m(-2) s(-1). The compact callus produced its maximum number of green plantlets early in the experiment (after 9 wk), while the friable callus continued to produce primordial shoots and green plantlets throughout the period of the experiment, and reached its maximum production of green plantlets at 21 wk under the irradiance of 300 mumol m(-2) s(-1). Organogenesis from friable callus under high irradiance (300 mumol m(-2) s(-1)) offers an efficient propagation method for C. blakei.

Early pregnancy factor (EPF) is a secreted protein with growth regulatory and immunomodulatory properties. It is an extracellular form of the mitochondrial matrix protein chaperonin 10 (Cpn10), a molecular chaperone. An understanding of the mechanism of action of EPF and an exploration of therapeutic potential has been limited by availability of purified material. The present study was undertaken to develop a simple high-yielding procedure for preparation of material for structure/function studies, which could be scaled up for therapeutic application. Human EPF was expressed in Sf9 insect cells by baculovirus infection and in Escherichia coli using a heat inducible vector. A modified molecule with an additional N-terminal alanine was also expressed in E coli. The soluble protein was purified from cell lysates via anion exchange (negative-binding mode), cation exchange, and hydrophobic interaction chromatography, yielding similar to42 and 36 mg EPF from 300 ml bacterial and I L Sf9 cultures, respectively. The preparations were highly purified ( greater than or equal to99% purity on SDS-PAGE for the bacterial products and greater than or equal to97% for that of insect cells) and had the expected mass and heptameric structure under native conditions, as determined by mass spectrometry and gel permeation chromatography, respectively. All recombinant preparations exhibited activity in the EPF bioassay, the rosette inhibition test, with similar potency both to each other and to the native molecule. In two in vivo assays of immuno suppressive activity, the delayed-type hypersensitivity reaction and experimental autoimmune encephalomyelitis, the insect cell and modified bacterial products, both with N-terminal additions (acetylation or amino acid), exhibited similar levels of suppressive activity, but the bacterial product with no N-terminal modification had no effect in either assay. Studies by others have shown that N-terminal addition is not necessary for Cpn10 activity. By defining techniques for facile production of molecules with and without immunosuppressive properties, the present studies make it possible to explore mechanisms underlying the distinction between EPF and Cpn10 activity. This article focuses on how US professional sports utilize the New International Division of Cultural Labor to supplement an overly costly local labor pool and over-supplied local market. We argue that while the classic problem of over-production is slowly eroding the sealed-off nature of US culture, the forces of its hyper-protectionist capitalism continue to characterize sports, precluding equal exchange.

The two steps of nitrification, namely the oxidation of ammonia to nitrite and nitrite to nitrate, often need to be considered separately in process studies. For a detailed examination, it is desirable to monitor the two-step sequence using online measurements. In this paper, the use of online titrimetric and off-gas analysis (TOGA) methods for the examination of the process is presented. Using the known reaction stoichiometry, combination of the measured signals (rates of hydrogen ion production, oxygen uptake and carbon dioxide transfer) allows the determination of the three key process rates, namely the ammonia consumption rate, the nitrite accumulation rate and the nitrate production rate. Individual reaction rates determined with the TOGA sensor under a number of operation conditions are presented. The rates calculated directly from the measured signals are compared with those obtained from offline liquid sample analysis. Statistical analysis confirms that the results from the two approaches match well. This result could not have been guaranteed using alternative online methods. As a case study, the influences of pH and dissolved oxygen (DO) on nitrite accumulation are tested using the proposed method. It is shown that nitrite accumulation decreased with increasing DO and pH. Possible reasons for these observations are discussed. (C) 2003 Elsevier Science Ltd. All rights reserved.

Raw milk samples from two different sources were stored at 2degreesC, 4degreesC and 7degreesC for 10 days and the growth of psychrotrophic bacteria, production of proteinase and proteolysis in the milks were measured during storage. Peptide analyses by the fluorescamine method and RP-HPLC were used in determination of proteolysis and proteinase activity. The average times taken for the psychrotroph counts to reach 10(7) cfu/mL at 2degreesC, 4degreesC and 7degreesC were approximately 9, 7 and 4 days, although there was considerable variation in growth rates in the different milks. There was little correlation between psychrotroph counts and either proteolysis or proteinase activity levels. At 2degreesC, no milk stored showed significant proteolysis by the fluorescamine method after 10 days' storage, but significant proteinase activity could be measured in some of these milks at 8 and 10 days. RP-HPLC analysis was a more sensitive means of detecting peptides than the fluorescamine method.

Nodal shoot cultures of 'Clone 003', a selected Australian papaya cultivar, were cultured on modified De Fossard medium supplemented with chemicals that either promote ethylene evolution or inhibit action while in culture. Nodal shoot cultures grown in the presence of 1-aminocyclopropane carboxylic acid (ACC, 1.0 mM) resulted in a significant reduction in percent fresh and dry weights, shoot length, leaf area, petiole length and chlorophyll content, but leaf development was significantly increased. In contrast, nodal cultures grown in the presence of silver thiosulphate (STS, 0.5 mM) significantly produced the highest percentage of fresh and dry weights, shoot length, leaf production, leaf area expansion, petiole length and leaf chlorophyll content. Nodal cultures and rooted whole plantlets placed in medium-sized (125 mL) culture vessels had significantly better growth than those cultures placed in small (70 mL) or in large (250 mL) vessels. Cultures grown in medium-sized vessels had higher fresh and dry weights, longer shoots, more leaves and larger leaf area than those cultures placed in smaller or larger vessels. Similarly, values for said growth parameters and for chlorophyll content of the nodal and rooted whole plantlets were higher when they were incubated under high light intensity of 120 mumol m(-2)s(-1) at a prevailing temperature of either 20+/-1 C or 25+/-1 C.

Low temperature during panicle development in rice increases spikelet sterility. This effect is exacerbated by high rates of nitrogen (N) application in the field. Spikelet sterility induced by low temperature and N fertilisation was examined in glasshouse experiments to clarify the mechanisms involved. In two glasshouse experiments, 12-h periods of low (18/13degreesC) and high (28/23degreesC) day/night temperatures were imposed over periods of 5-7 days during panicle development, to determine the effects of low temperature and N fertilisation on spikelet sterility. In one experiment, 50% sunlight was imposed together with low temperature to investigate the additive effects of reduced solar radiation and low temperature. The effect of increased tillering due to N fertilisation was examined by a tiller removal treatment in the same experiment. Pollen grain number and spikelet sterility were recorded at heading and harvest, respectively. Although there was no significant effect of low temperature on spikelet sterility in the absence of applied N, low temperature greatly increased spikelet sterility as a result of a reduction in the number of engorged pollen grains per anther in the presence of applied N. Spikelet sterility was strongly correlated with the number of engorged pollen grains per anther. Low temperature during very early ( late stage of spikelet differentiation-pollen mother cell stage) and peak ( second meiotic division stage-early stage of extine formation) microspore development caused a severe reduction in engorged pollen production mainly as a result of reduced total pollen production. Unlike low temperature, the effect of shading was rather small. The increased tillering due to application of high rates of N, increased both spikelet number per plant and spikelet sterility under low temperature conditions. The removal of tillers as they appeared reduced the number of total spikelets per plant and maintained a large number of engorged pollen grains per anther which, in turn, reduced spikelet sterility. The number of engorged pollen grains per anther determined the numbers of intercepted and germinated pollen grains on the stigma. It is concluded that N increased tillering and spikelet number per plant and this, in turn, reduced the number of engorged pollen grains per anther, leading into increased spikelet sterility under low temperature condition.

Low temperatures impose restrictions on rice (Oryza sativa L.) production at high latitudes. This study is related to low temperature damage that can arise mid-season during the panicle development phase. The objective of this study was to determine whether low temperature experienced by the root, panicle, or foliage is responsible for increased spikelet sterility. In temperature-controlled glasshouse experiments, water depth, and water and air temperatures, were changed independently to investigate the effects of low temperature in the root, panicle, and foliage during microspore development on spikelet sterility. The total number of pollen and number of engorged pollen grains per anther, and the number of intercepted and germinated pollen grains per stigma, were measured. Spikelet sterility was then analysed in relation to the total number of pollen grains per spikelet and the efficiency with which these pollen grains became engorged, were intercepted by the stigma, germinated, and were involved in fertilisation. There was a significant combined effect of average minimum panicle and root temperatures on spikelet sterility that accounted for 86% of the variation in spikelet sterility. Total number of pollen grains per anther was reduced by low panicle temperature, but not by low root temperature. Whereas engorgement efficiency ( the percentage of pollen grains that were engorged) was determined by both root and panicle temperature, germination efficiency (the percentage of germinated pollen grains relative to the number of engorged pollen grains intercepted by the stigma) was determined only by root temperature. Interception efficiency (i.e. percentage of engorged pollen grains intercepted by the stigma), however, was not affected by either root or panicle temperature. Engorgement efficiency was the dominant factor explaining the variation in spikelet sterility. It is concluded that both panicle and root temperature affect spikelet sterility in rice when the plant encounters low temperatures during the microspore development stage.

Este estudo sistematiza e aborda a temática da formação de educadores desenvolvida nas inter-relações entre universidades brasileiras e o Movimento dos Trabalhadores Rurais Sem Terra, especificamente nos cursos de graduação em História na Universidade Federal da Paraíba e de Engenharia Agronômica na Universidade Federal de Sergipe, no período de 2004 a 2008. Tal processo desenvolve-se em um contexto difícil e complexo da luta pela reforma agrária no Brasil, principalmente pelas transformações ocorridas nos últimos anos oriundas da ampliação das lógicas de produção do agronegócio. Esta condição leva o MST a discutir e propor uma nova concepção de reforma agrária que a designa de popular em substituição à proposta de reforma agrária clássica. Por outro lado, apresenta uma visão das universidades brasileiras e dos projetos que são construídos e implementados nos últimos anos, inclusive, observando coincidências com as políticas econômicas gerais para a sociedade. Apresenta uma concepção de formação que vem sendo construída no interior das práticas do MST que também procura as universidades para firmar convênios e desenvolver processos de escolarização/formação de seus militantes educadores. Dentre as dimensões desse processo educativo/formativo, destacam-se: vínculo permanente com os processos orgânicos; a formação como um processo ético, estético, místico que trata das atitudes/comportamentos e como um processo dialógico, crítico e articulado que contempla saberes, experiências, em uma interação que busca superar as monoculturas. Captura por intermédio da pesquisa de campo: estranhamentos, entraves, sentidos da ocupação pedagógica e coletiva, alternativas, legados que permanecem tanto no MST como na universidade e apresenta o resultado do envolvimento e atuação dos egressos de ambos os cursos na atualidade. Aponta também para desafios, possibilidades outras de enfrentar a difícil mas necessária tarefa de formar educadores, militantes capazes de coletivamente levar adiante a luta por um mundo mais justo, solidário e democrático, em que a terra e o conhecimento, juntamente com os demais bens econômicos e culturais sejam profundamente democratizados. Pretende ser uma contribuição para o debate acerca da relevância dessas inter-relações entre universidade e movimentos sociais, em que essas experiências demarcam novas possibilidades de abertura e avanços democráticos e menos elitista da universidade e novos patamares de escolarização/formação para integrantes do Movimento dos Sem Terra.

Esta dissertação trata da análise da produção científica e tecnológica internacional e brasileira na área de conhecimento Engenharia Civil, por meio de indicadores bibliométricos. A área Engenharia Civil foi escolhida em razão da sua relevância para o desenvolvimento econômico do país. No entanto, em termos absolutos e relativos, está entre os setores tecnologicamente mais atrasados da economia. A bibliometria é uma disciplina com alcance multidisciplinar que estuda o uso e os aspectos quantitativos da produção científica registrada. Os indicadores de produção científica são objeto de análise de várias áreas do conhecimento, tanto para o planejamento e a execução de políticas públicas de vários setores quanto para maior conhecimento da comunidade científica sobre o sistema em que está inserida. A metodologia utilizada para a elaboração deste estudo descritivo de caráter exploratório foi a análise documental e bibliométrica, baseada em dados das publicações científicas, no período de 1970 a 2012, e tecnológicas, no período de 2001 a 2012, da área Engenharia Civil, indexadas nas bases de dados Science Citattion Index Expanded (SCI); Social Science Citation Index (SSCI); Conference Proceedings Citation Index (CPCI) e da Derwent Innovations Index (DII), que compõem a base de dados multidisciplinar da Web of Sicence (WoS). As informações foram qualificadas e quantificadas com o auxílio do software bibliométrico VantagePoint®. Os resultados obtidos confirmaram o baixo número de publicações científicas e tecnológicas na área de conhecimento Engenharia Civil de autores filiados a instituições de ensino e pesquisa brasileiras quando comparados aos dos países industrializados. Existe um conjunto de fortes condicionantes que ultrapassam o poder de decisão e de influência da academia, dificultando e limitando a disseminação das pesquisas e patentes brasileiras relacionadas a fatores de caráter sistêmico e cultural. A possibilidade de análise de indicadores de produção científica e tecnológica na Engenharia Civil contribui para criar políticas que, se utilizadas por agências de fomento, podem subsidiar investimentos mais fundamentados por parte dos governos e da iniciativa privada, a exemplo do que é feito por outros setores industriais.
