β-Site amyloid precursor protein cleaving enzyme (BACE1) is the rate-limiting enzyme for production of Aβ peptides, proposed to drive the pathological changes found in Alzheimer’s disease (AD). Reticulon 3 (RTN3) is a negative modulator of BACE1 (β-secretase) proteolytic activity, while peptidylprolyl isomerase (cyclophilin)-like 2 (PPIL2) positively regulated BACE1 gene expression in a cell-based assay. This study aimed to analyze RTN3 and PPIL2 mRNA levels in four brain regions from individuals with AD and controls. BACE1 mRNA had been previously quantified in the samples, as had glial fibrillary acidic protein (GFAP) and neuron-specific enolase (NSE), to track changing cell populations in the tissue. mRNA levels in the human post mortem brain tissue were assayed using quantitative real-time polymerase chain reaction (qPCR) and qbasePLUS, employing validated stably expressed reference genes. No differences in RTN3 or PPIL2 mRNA levels were found in individuals with AD, compared to controls. Both RTN3 and PPIL2 mRNA levels correlated significantly with BACE1 mRNA and all three showed similar disease stage-dependent changes with respect to NSE and GFAP. These findings indicated that the in vitro data demonstrating an effect of PPIL2 on BACE1 expression have functional relevance in vivo. Further research into BACE1-interacting proteins could provide a fruitful approach to the modulation of this protease and consequently Aβ production.

For those working in the humanitarian sector, achieving positive outcomes for postdisaster communities through reconstruction projects is a pressing concern. In the wake of recent natural disasters, NGOs have become increasingly involved in the permanent reconstruction of affected communities. They have encountered significant barriers as they implement reconstruction programmes and this paper argues that it is important to address the visible lack of innovation that is partially to blame. The theoretical bedrock of a current research project will be used as the starting point for this argument, the overall goal of which is to design a competency-based framework model that can be used by NGOs in post-disaster reconstruction projects. Drawing on established theories of management, a unique perspective has been developed from which a competency-based reconstruction theory emerges. This theoretical framework brings together 3 distinct fields; Disaster Management, Strategic Management and Project Management, each vital
to the success of the model. The objectives of this paper are a) to investigate the role of NGOs in post-disaster reconstruction and establish the current standard of practice b) to determine the extent to which NGOs have the opportunity to contribute to sustainable community development through reconstruction c) to outline the main factors of a theoretical framework first proposed by Von Meding et al. 2009 and d) to identify the innovative measures that can be taken by NGOs to achieve more positive outcomes in their interventions. It is important that NGOs involved in post-disaster reconstruction become familiar with concepts and strategies such as those contained in this paper. Competency-based organizational change on the basis of this theory has the potential to help define the standard of best practice to which future NGO projects might align themselves.

BACKGROUND: Glucagon-like peptide-1 (GLP-1) therapies are routinely used for glycaemic control in diabetes and their emerging cardiovascular actions have been a major recent research focus. In addition to GLP-1 receptor activation, the metabolically-inactive breakdown product, GLP-1(9-36)amide, also appears to exert notable cardiovascular effects, including protection against acute cardiac ischaemia. Here, we specifically studied the influence of GLP-1(9-36)amide on chronic post-myocardial infarction (MI) remodelling, which is a major driver of heart failure progression.

METHODS: Adult female C57BL/6 J mice were subjected to permanent coronary artery ligation or sham surgery prior to continuous infusion with GLP-1(9-36)amide or vehicle control for 4 weeks.

RESULTS: Infarct size was similar between groups with no effect of GLP-1(9-36)amide on MI-induced cardiac hypertrophy, although modest reduction of in vitro phenylephrine-induced H9c2 cardiomyoblast hypertrophy was observed. Whilst echocardiographic systolic dysfunction post-MI remained unchanged, diastolic dysfunction (decreased mitral valve E/A ratio, increased E wave deceleration rate) was improved by GLP-1(9-36)amide treatment. This was associated with modulation of genes related to extracellular matrix turnover (MMP-2, MMP-9, TIMP-2), although interstitial fibrosis and pro-fibrotic gene expression were unaltered by GLP-1(9-36)amide. Cardiac macrophage infiltration was also reduced by GLP-1(9-36)amide together with pro-inflammatory cytokine expression (IL-1β, IL-6, MCP-1), whilst in vitro studies using RAW264.7 macrophages revealed global potentiation of basal pro-inflammatory and tissue protective cytokines (e.g. IL-1β, TNF-α, IL-10, Fizz1) in the presence of GLP-1(9-36)amide versus exendin-4.

CONCLUSIONS: These data suggest that GLP-1(9-36)amide confers selective protection against post-MI remodelling via preferential preservation of diastolic function, most likely due to modulation of infiltrating macrophages, indicating that this often overlooked GLP-1 breakdown product may exert significant actions in this setting which should be considered in the context of GLP-1 therapy in patients with cardiovascular disease.

This chapter argues that there is a gap between symbolic exclusion from the national community when it comes to the inclusion of new German citizens of Turkish or Kurdish background, and a broad claim to be a cosmopolitan society, at large. While focusing on narratives of minority key political activists in Berlin, and analysing individual stories on the background of contemporary populist xenophobic debates and hate crime of the 1990s, the chapter illustrates both, individual success and vulnerability due to institutionalised forms of anti- Muslim and anti-Turks segments in Germany.

O sucesso do recrutamento em espécies com ciclos de vida complexos, i.e. com duas ou mais fases de desenvolvimento, depende do fornecimento de larvas aos locais apropriados do assentamento (mecanismos de pré-assentamento) e na consequente sobrevivência destas larvas até aos estágios bentónicos iniciais (mecanismos de pós-assentamento). O estudo dos principais processos físicos que controlam a variabilidade intra-anual do recrutamento do caranguejo Carcinus maenas foi possível com à obtenção em séries diárias da abundância de megalopas no plâncton e do seu assentamento em substratos artificiais de assentamento, e de parâmetros físicos relacionados com a amplitude de maré e o upwelling, durante os meses da época reprodutiva deste caranguejo. Os nossos resultados demonstraram um padrão cíclico de fornecimento de megalopas de C. maenas aos estuários na costa ocidental de Portugal, relacionado com a amplitude de maré e favorecido pelos ventos de sul. O assentamento de megalopas nos substratos artificiais de assentamento mostrou-se desacoplado do fornecimento nos dois estuários. Os dados obtidos sugerem que as megalopas são transportadas para a costa após a acção de ventos de downwelling, e o fornecimento para os estuários ocorre por transporte selectivo por corrente de maré. A utilização de substratos artificiais para medir densidades de juvenis de Carcinus maenas deve ser ponderada, uma vez que se verificou que em habitats estruturalmente menos complexos os juvenis utilizam os colectores como refúgio; em habitats mais complexos, os juvenis preferiram refugiar-se nas ervas marinhas existentes. No entanto, a estimativa de abundâncias diárias medidas nos colectores permitiu a descrição das abundâncias dos vários estádios de desenvolvimento juvenis, bem como as respectivas taxas de crescimento. As capacidades locomotoras de diferentes classes de tamanho de juvenis de C. maenas foram estimadas mediante o cálculo de um índice de locomoção. Os nossos resultados sugerem que os juvenis mais pequenos estão relativamente protegidos em habitats com vegetação densa, provavelmente porque este tipo de habitats inibe os movimentos dos juvenis de maiores dimensões. Foram também encontradas evidências da existência de segregação temporal na actividade locomotora das diferentes classes de juvenis, que funcionará como um mecanismo que permite reduzir o canibalismo e, consequentemente, aumentar a capacidade de suporte dos habitats juvenis. Foi realizado um estudo sobre o canibalismo nos juvenis de C. maenas na Ria de Aveiro, in situ e utilizando densidades reais. Verificou-se que as presas mais pequenas eram as mais vulneráveis, sendo os juvenis de maiores dimensões os predadores mais eficazes. Por outro lado, as taxas de canibalismo foram menores em habitats com elevadas densidades de refúgio (Zostera noltii). A estimativa das taxas diárias de mortalidade devidas ao canibalismo são aproximadamente metade das taxas diárias de mortalidade devidas aos processos de pré-assentamento, indicando que esta população de C. maenas será regulada pelo fornecimento larvar, i.e., será uma população regulada essencialmente por mecanismos préassentamento.

This thesis aims at improving the knowledge on the post-fire vegetation regeneration. For that, forests and shrublands were studied, after forest fires and experimental fires. Maritime Pine (Pinus pinaster) recruitment after fire was studied. Fire severity was evidenced as a major effect on this process. High crown fire severity can combust the pines, destroying the seed bank and impeding post fire pine recruitment. However, crown combustion also influences the post-fire conditions on the soil surface, since high crown combustion (HCC) will decrease the postfire needle cast. After low crown combustion (LCC) (scorched rather than torched crowns), a considerable needle cover was observed, along with a higher density of pine seedlings. The overall trends of post-fire recruitment among LCC and HCC areas could be significantly attributed to cover by needles, as well by the estimation of fire severity using the diameters of the burned twigs (TSI). Fire increased the germination from the soil seed bank of a Pinus pinaster forest, and the effects were also related with fire severity. The densities of seedlings of the dominant taxa (genus Erica and Calluna vulgaris) were contrastingly affected in relation to the unburned situation, depending on fire severity, as estimated from the degree of fire-induced crown damage (LCC/HCC), as well as using a severity index based on the diameters of remaining twigs (TSI). Low severity patches had an increase in germination density relatively to the control, while high severity patches suffered a reduction. After an experimental fire in a heathland dominated by Pterospartum tridentatum, Erica australis and E. umbellata, no net differences in seedling emergence were observed, in relation to the pre-fire situation. However, rather than having no effect, the heterogeneity of temperatures caused by fire promoted caused divergent effects over the burned plot in terms of Erica australis germination – a progressive increased was observed in the plots were maximum temperature recorded ranged from 29 to 42.5ºC and decreased in plots with maximum temperature ranging from 51.5 to 74.5ºC. In this heathland, the seed density of two of the main species (E. australis and E. umbellata) was higher under their canopies, but the same was not true for P. tridentatum. The understory regeneration in pine and eucalypt stands, 5 to 6 years post fire, has been strongly associated with post-fire management practices. The effect of forest type was, comparatively, insignificant. Soil tilling, tree harvesting and shrub clearance, were linked to lower soil cover percentages. However, while all these management operations negatively affected the cover of resprouters, seeders were not affected by soil tilling. A strong influence of biogeographic region was identified, suggesting that more vulnerable regions may suffer higher effects of management, even under comparatively lower management pressure than more productive regions. This emphasizes the need to adequate post-fire management techniques to the target regions.

Cell cycle and differentiation are two highly coordinated processes during organ development. Recent studies have demonstrated that core cell cycle regulators also play cell cycle-independent functions in post-mitotic neurons, and are essential for the maintenance of neuronal homeostasis. CDC25 phosphatases are well-established CDK activators and their activity is mainly associated to proliferating tissues. The expression and activity of mammalian CDC25s has been reported in adult brains. However, their physiological relevance and the potential substrates in a non-proliferative context have never been addressed. string (stg) encodes the Drosophila CDC25 homolog. Previous studies from our group showed that stg is expressed in photoreceptors (PRs) and in lamina neurons, which are two differentiated cell types that compose the fly visual system. The aims of this work are to uncover the function of stg and to identify its potential neuronal substrates, using the Drosophila visual system as a model. To gain insight into the function of stg in a non-dividing context we used the GAL4/UAS system to promote downregulation of stg in PR-neurons, through the use of an RNAi transgene. The defects caused by stg loss-of-function were evaluated in the developing eye imaginal disc by immunofluorescence, and during adult stages by scanning electron microscopy. This genetic approach was combined with a specific proteomic method, two-dimensional difference gel electrophoresis (2D-DIGE), to identify the potential substrates in PR-cells. Our results showed that stg downregulation in PRs affects the well-patterned retina organization, inducing the loss of apical maintenance of PR-nuclei on the eye disc, and ommatidia disorganization. We also detected an abnormal accumulation of cytoskeletal proteins and a disruption of the axon structure. As a consequence, the projection of PR-axons into the lamina and medulla neuropils of the optic lobe was impaired. Upon stg downregulation, we also detected that PR-cells accumulate Cyclin B. Although the rough eye phenotype observed upon stg downregulation suggests neurodegeneration, we did not detect neuronal death during larval stages, suggesting that it likely occurs during pupal stages or during adulthood. By 2D-DIGE, we identified seven proteins which were differentially expressed upon stg downregulation, and are potential neuronal substrates of Stg. Altogether, our observations suggest that Stg phosphatase plays an essential role in the Drosophila visual system neurons, regulating several cell components and processes in order to ensure their homeostasis.

Post-synthetic modification (PSM) of metal-organic frameworks encompassing the chemical transformation of the linker present is a promising new route for engineering optical centres and tuning the light emission properties of materials, both in the visible and in the near infrared (NIR) spectral regions. Here, PSM of isoreticular metal-organic framework-3 (IRMOF-3) with ethyl oxalyl monochloride, ethyl acetoacetate, pentane-2,4-dione, 3-(2- hydroxyphenyl)-3-oxopropanal, 2-chloroacetic acid, glyoxylic acid, methyl vinyl ketone and diethyl (ethoxymethylene)malonate followed by chelation of trivalent lanthanide ions afforded intriguing near infrared (Nd3+) and visible (Eu3+, Tb3+) light emitters. IRMOF-3 was used as a case in point due to both its highly porous crystalline structure and the presence of non-coordinating amino groups on the benzenedicarboxylate (bdc) linker amenable to modification. The materials were characterised by elemental analysis, powder X-ray diffraction, optical, scanning and transmission electron microscopy, Fourier transform infrared spectroscopy, and liquid and solid-state nuclear magnetic resonance. The solid-state luminescence properties of Ln-modified-IRMOF-3 were investigated at room temperature. The presence of the bdc aromatic ring, β– diketonate and oxalate enhanced the Ln3+ sensitization via ligand-to-metal energy transfer (anthena effect). As far as photocalysis is concerned, we have synthesized metal−organic frameworks (Cr-MIL-125-AC, Ag-MIL-125-AC) by a green method (solid–vapors reactions). The resulting functionalized materials show a photocatalytic activity for methylene blue degradation up to 6.52 times larger than that of the commercial photocatalyst hombikat UV-100. These findings open the door for further research for improving the photocatalytic performance of metal-organic frameworks.

The years between 1945 and the early 1980s are the most celebrated in Italy’s design history. From the rhetoric of reconstruction to the postmodern provocations of the Memphis design collective, Italy’s architects played a vital role in shaping the country’s encounter with post-war modernity. Yet as often as this story has been told, it is incomplete. Craft was vital to the realisation of post-war Italian design, and an area of intense creativity in its own right, and yet has been marginalised and excluded in design historiography.

This paper examines the roles of research in teacher education across the four nations of the United Kingdom. Both devolution and on-going reviews of teacher education are facilitating a greater degree of cross-national divergence. England is becoming a distinct outlier, in which the locus for teacher education is moving increasingly away from Higher Education Institutions and towards an ever-growing number of school-based providers. While the idea of teaching as a research-based profession is increasingly evident in Scotland, Northern Ireland and Wales, it seems that England, at least in respect of the political rhetoric, recent reforms and explicit definitions, is fixed on a contrastingly divergent trajectory towards the idea of teaching as a craft-based occupation, with a concomitant emphasis on a (re)turn to the practical. It is recommended that research is urgently needed to plot these divergences and to examine their consequences for teacher education, educational research and professionalism.
