The histamine releasing properties of glucose (mannose)-specific lectins isolated from Brazilian beans was examined. The Canavalia brasiliensis, Dioclea rostrata, and Dioclea virgata lectins induced histamine release in rat peritoneal mast cells similar to concanavalin A. Less potency and efficacy was observed for Canavalia maritima, Dioclea guianensis, and Dioclea violacea while very low activities were seen for the lectins from Dioclea grandiflora, Canavalia bonariensis, and Cratylia floribunda. The histamine releasing effect was quenched by higher doses of D. virgata lectin similar to what was reported for concanavalin A. This effect was abrogated by increasing the concentration of calcium in the incubating medium. As these above proteins have sites that bind calcium, higher doses of the lectins might withdraw the calcium which is essential for the mast cell secretion.

The specific heat, thermal conductivity and density of passion fruit juice were experimentally determined from 0.506 to 0.902 (wet basis) water content and temperatures from 0.4 to 68.8C. The experimental results were compared with existing and widely used models for the thermal properties. In addition, based on empiric equations from literature, new simple models were parameterized with a subset of the total experimental data. The specific heat and thermal conductivity showed linear dependency on water content and temperature, while the density was nonlinearly related to water content. The generalized predictive models were considerably good for this product but the empiric, product-specific models developed in the present work yield better predictions. Even though the existing models showed a moderate accuracy, the new simple ones would be preferred, because they constitute an easier and direct way of evaluating the thermal properties of passion fruit juice, requiring no information about the chemical composition of the product, and a reduced time of the estimation procedure, as the new empiric models are described in terms of only two physical parameters, the water content and the temperature. © Copyright 2005, Blackwell Publishing All Rights Reserved.

The recent appreciation of the role played by endogenous counterregulatory mechanisms in controlling the outcome of the host inflammatory response requires specific analysis of their spatial and temporal profiles. In this study, we have focused on the glucocorticoid-regulated anti-inflammatory mediator annexin 1. Induction of peritonitis in wild-type mice rapidly (4 h) produced the expected signs of inflammation, including marked activation of resident cells (e.g., mast cells), migration of blood-borne leukocytes, mirrored by blood neutrophilia. These changes subsided after 48-96 h. In annexin 1null mice, the peritonitis response was exaggerated (∼40% at 4 h), with increased granulocyte migration and cytokine production. In blood leukocytes, annexin 1 gene expression was activated at 4, but not 24, h postzymosan, whereas protein levels were increased ai both time points. Locally, endothelial and mast cell annexin 1 gene expression was not detectable in basal conditions, whereas it was switched on during the inflammatory response. The significance of annexin 1 system plasticity in the anti-inflammatory properties of dexamethasone was assessed. Clear induction of annexin 1 gene in response to dexamethasone treatment was evident in the circulating and migrated leukocytes, and in connective tissue mast cells; this was associated with the steroid failure to inhibit leukocyte trafficking, cytokine synthesis, and mast cell degranulation in the annexin 1null mouse. In conclusion, understanding how inflammation is brought under control will help clarify the complex interplay between pro- and anti-inflammatory pathways operating during the host response to injury and infection. Brazil is one of the main centers of genetic variability dispersion of the Passiflora genera. Its self incompatibility as well as disease incidence in its leaves and root system and, deforestation and monocultivation, promote loss of genetic material. Considering the risk of genetic erosion, the conservation of the variability in germplasm banks, which is of great interest in plant breeding, is necessary. Studies regarding the type of expiant and concentration of the culture media are necessary in order to determine protocols of establishment and in vitro conservation of passion-fruit germplasm. The objective of the present work was to evaluate the influence of the salt and nutrient concentration in the MS culture medium and types of expiants in the establishment and growth of the Passion fruit species: Passiflora giberti N. E.Brown, P. edulis Sims and P. laurifolia L. Each Passiflora species presented its own characteristics regarding in vitro development. The complete MS medium and nodal segments the second axilliary bud promoted better development of the genotypes studied.

It was studied the effect of passion fruit genotypes on Dione juno juno (Cramer) (Lepidoptera: Nymphalidae) development. The experiment was carried out in a laboratory, under controlled conditions (temperature: 26 ± 1° C, RH = 60 ± 10% and photophase of 14 hours). Newly-hatched larvae were fed with leaves from different passion fruit genotypes: Passiflora edulis Sims., P. alata Dryand., P. serrato-digitata L., P. edulis f. flavicarpa Deg. ('Sul Brasil'), P. edulis f. flavicarpa, P. edulis f. flavicarpa ('Maguary FB-100') and P. foetida L. Fifty larvae from eggs collected in the field were used per genotype. Larvae were kept on passion fruits branches inside PVC tubes until pupation. Daily observations were performed and branches were replaced whenever necessary. The following parameters were evaluated: duration and viability of larval and pupal phases, larval and pupal weight and adult longevity. The experiment was arranged in randomized blocks design with seven treatments and ten replications. Data were subjected to an ANOVA and means were compared by Tukey test at 5% of probability. The least adequate genotypes for D. juno juno development were P. alata, P. serrato-digitata and P. foetida, showing a high level of antibiosis, while P. edulis, P. edulis f. flavicarpa, 'Maguary FB-100' and 'Sul Brasil' were the most suitable.

Objective: The objective of this study was to investigate the mediators and the resident peritoneal cells involved in the neutrophil migration (NM) induced by mineral trioxide aggregate (MTA) in mice. Study design: MTA (25 mg/cavity) was injected into normal and pretreated peritoneal cavities (PC) with indomethacin (IND), dexamethasone (DEX), BWA4C, U75302, antimacrophage inflammatory protein-2 (MIP-2), and anti-interleukin-1β (IL-1β) antibodies and the NM was determined. The role of macrophage (MO) and mast cells (MAST) was determined by administration of thioglycollate 3% or 48/80 compound, respectively. The concentration of IL-1β and MIP-2 exudates was measured by ELISA. Results: MTA induced dose- and time-dependent NM into mice PC, with the participation of MO and MAST. NM was inhibited by DEX, BWA4C, and U75302, as well as anti-MIP-2 and anti-IL-1β antibodies. In the exudates, IL-1β and MIP-2 were detected. Conclusions: This study suggests that MTA induces NM via a mechanism dependent on MAST and MO mediated by IL-1β, MIP-2, and LTB4. © 2008 Mosby, Inc. All rights reserved.

This study's objective is to evaluate the photosynthates partitioning and source-sink relationships, in vegetative and reproductive twigs of sweet passion fruit, using the carbon-13, stable isotope of carbon. The leaves of vegetative and reproductive twigs were placed in a sealed chamber and 13CO 2 were injected for 30 minutes. After six hours, the different organs of the twig were collected and immersed in liquid nitrogen (-196°C). All plant samples were oven-dried, powdered and combusted in an elemental analyzer. The results showed that the methodology was efficient to evaluate the partitioning of photosynthates. In the vegetative twigs, the new open leaves, the closed leaves and growth meristem were the principal sink for the source leaves enriched with 13CO 2. In the reproductive twigs with only flowers buds, the source-sink relationship was changed and the flowers buds turn to be the principal sink and the growing leaves were secondary sinks. The presence of young fruits changed again the source-sink relationship of the twig, and they were the principal sinks. The apical leaves, with 60% of final leaf area were also a sink for photosynthates.

Four novel peptides were isolated from the venoms of the solitary eumenine wasps Eumenes rubrofemoratus and Eumenes fraterculus. Their sequences were determined by MALDI-TOF/TOF (matrix assisted laser desorption/ionization time-of-flight mass spectrometry) analysis, Edman degradation and solid-phase synthesis. Two of them, eumenitin-R (LNLKGLIKKVASLLN) and eumenitin-F (LNLKGLFKKVASLLT), are highly homologous to eumenitin, an antimicrobial peptide from a solitary eumenine wasp, whereas the other two, EMP-ER (FDIMGLIKKVAGAL-NH 2) and EMP-EF (FDVMGIIKKIAGAL-NH 2), are similar to eumenine mastoparan-AF (EMP-AF), a mast cell degranulating peptide from a solitary eumenine wasp. These sequences have the characteristic features of linear cationic cytolytic peptides; rich in hydrophobic and basic amino acids with no disulfide bond, and accordingly, they can be predicted to adopt an amphipathic α-helix secondary structure. In fact, the CD (circular dichroism) spectra of these peptides showed significant α-helical conformation content in the presence of TFE (trifluoroethanol), SDS (sodium dodecylsulfate) and asolectin vesicles. In the biological evaluation, all the peptides exhibited a significant broad-spectrum antimicrobial activity, and moderate mast cell degranulation and leishmanicidal activities, but showed virtually no hemolytic activity. © 2011 Elsevier Ltd.

The experiment was conducted in a greenhouse from May 1 to July 31, 2008, in Areia county, Paraiba State, PB, Brazil, in order to evaluate the effects of irrigation water salinity on initial growth of the passionfruit seedlings in non-saline substrate with and without bovine biofertilizer. The treatments were distributed in a completely randomized design, with three replications and twelve plants per plots, in a factorial arrangement 5 × 2 × 2, corresponding the former to the levels of salinity in the irrigation water: 0.5; 1.0; 2.0; 3.0 and 4.0 dS m-1, in soils with and without bovine biofertilizer applied at two moments (25 and 65 days after seedling emergence). The growth of the seedlings and the soil electrical conductivity were evaluated at the end of the experiment. The biofertilizer was diluted in a low saline water at a 1:1 ratio and was applied once two days before sowing, corresponding to 10% of the substrates volume. The increase in water salinity inhibited the growth in height of plants, leaf area and root length, but always to a lesser extent in the treatments with bovine biofertilizer. The increase in electrical conductivity of the irrigation water elevated the soil salinity, independently of the addition of biofertilizer.

Melanocytes are found in various organs of ectothermic animals, playing a protective role against bacteria and free radicals. It is known that pigment cells from hematopoietic organs have immune functions. However, the role of visceral melanocytes is not well understood. Cutaneous melanocytes are responsive to α-melanocyte stimulating hormone (α-MSH), which is associated with the dispersion of melanin granules within melanocytes. α-MSH has also been reported to inhibit most forms of inflammatory responses by decreasing the pro-inflammatory cytokines and neutrophil migration. The present study evaluated the influence of an α-MSH analog (Nle4, D-Phe7-α-MSH) and lipopolysaccharides (LPS) from Escherichia coli on the liver and testicular tissues of the anuran Eupemphix nattereri. The tested hypotheses were: (i) the pigmented area will increase following hormone and LPS administration, (ii) pre-treatment with α-MSH will decrease the number of mast cells, and (iii) the hormone will have protective effects against LPS-induced responses. We found that hormone administration did not change hepatic pigmentation, but increased testicular pigmentation. Testicular pigmentation quickly increased after LPS administration, whereas there was a late response in the liver. The response of enhanced pigmentation was delayed and the number of mast cells decreased in animals previously treated with the α-MSH analog when compared to the LPS group. Hemosiderin and lipofuscin were found in melanomacrophages, but not in testicular melanocytes. Although both the liver and the testes of E. nattereri have pigmented cells, these are distinct in morphology, embryonic origin, and pigmentary substances. These differences may be responsible for the different responses of these cells to the α-MSH analog and LPS administration. © 2013 Elsevier GmbH.
