Background: Primary distal renal tubular acidosis (dRTA) caused by mutations in the genes that codify for the H+ -ATPase pump subunits is a heterogeneous disease with a poor phenotype-genotype correlation. Up to now, large cohorts of dRTA Tunisian patients have not been analyzed, and molecular defects may differ from those described in other ethnicities. We aim to identify molecular defects present in the ATP6V1B1, ATP6V0A4 and SLC4A1 genes in a Tunisian cohort, according to the following algorithm: first, ATP6V1B1 gene analysis in dRTA patients with sensorineural hearing loss (SNHL) or unknown hearing status. Afterwards, ATP6V0A4 gene study in dRTA patients with normal hearing, and in those without any structural mutation in the ATP6V1B1 gene despite presenting SNHL. Finally, analysis of the SLC4A1 gene in those patients with a negative result for the previous studies. Methods: 25 children (19 boys) with dRTA from 20 families of Tunisian origin were studied. DNAs were extracted by the standard phenol/chloroform method. Molecular analysis was performed by PCR amplification and direct sequencing. Results: In the index cases, ATP6V1B1 gene screening resulted in a mutation detection rate of 81.25%, which increased up to 95% after ATP6V0A4 gene analysis. Three ATP6V1B1 mutations were observed: one frameshift mutation (c.1155dupC; p.Ile386fs), in exon 12; a G to C single nucleotide substitution, on the acceptor splicing site (c.175-1G > C; p.?) in intron 2, and one novel missense mutation (c. 1102G > A; p. Glu368Lys), in exon 11. We also report four mutations in the ATP6V0A4 gene: one single nucleotide deletion in exon 13 (c.1221delG; p. Met408Cysfs* 10); the nonsense c.16C > T; p.Arg6*, in exon 3; and the missense changes c.1739 T > C; p.Met580Thr, in exon 17 and c.2035G > T; p.Asp679Tyr, in exon 19. Conclusion: Molecular diagnosis of ATP6V1B1 and ATP6V0A4 genes was performed in a large Tunisian cohort with dRTA. We identified three different ATP6V1B1 and four different ATP6V0A4 mutations in 25 Tunisian children. One of them, c.1102G > A; p.Glu368Lys in the ATP6V1B1 gene, had not previously been described. Among deaf since childhood patients, 75% had the ATP6V1B1 gene c. 1155dupC mutation in homozygosis. Based on the results, we propose a new diagnostic strategy to facilitate the genetic testing in North Africans with dRTA and SNHL.

A survey carried out on the fishery of the northern portion of Imo basin (known as Ibu project area for the purpose of this study) provided some information on the number and type of fishermen cropping this portion of the Imo River, Nigeria. The population was found to be very mobile with many itinerant or seasonal fishermen. The type of gear in use was studied and a high number of traditional dug-out canoes recorded which shows the ability of the people to make their own canoes. The presence of freshwater shrimps crayfish and clupeids at this upper region of the Imo River was observed

ENGLISH: This report presents information on the fishery for bluefin in the northeastern Pacific Ocean derived from purse seiner logbook records and landing statistics. Information on the size composition of the fish in the catch is also presented. The logbook records cover the period 19611980; however, catch statistics and length-frequency data from previous years are also presented. SPANISH: Este informe presenta los datos sobre la pesca del T. thynnus en el Océano Pacífico nordeste, obtenidos de los cuadernos de bitácora de los barcos cerqueros y de las estadísticas de desembarque. Se presenta además la información sobre la composición de talla de los peces en la captura. Los registros de bitácora abarcan el período de 1961 a 1980; no obstante, se presentan además las estadísticas de captura y los datos de la frecuencia de talla de años anteriores. Chemical narcosing of fish was found to be one of the fishing techniques used for harvesting fish stocks in the northern Cross River, Cross River State, Nigeria. The preparation of the fish poison is described. The ichthyotoxic plants used for the fish poisoning were identified and Derris elliptica recommended to be used for the development of piscicides for the removal of unwanted fish in the fish farms

ENGLISH: The growth of northern bluefin tuna is described by a two-stanza model. For fish between 191 and 564 mm in length the Gompertz curve, with values of 581 mm and 4.32 for Loo and K (annual), respectively, is used. The fish between 564 and 1530 mm grow linearly, at the rate of 0.709 mm per day. Age-O fish tagged and released in the western Pacific Ocean have been recaptured in the western, central, and eastern Pacific. The minimum time between release in the western Pacific and recapture in the eastern Pacific is 215 days. Older fish, mostly Land 2-year olds, tagged and released in the eastern Pacific have been recaptured in the eastern and western Pacific. The minimum time between release in eastern Pacific and recapture in the western Pacific is 674 days. The coefficient of natural mortality is estimated from data on growth and ambient temperature to be 0.276 on an annual basis, with 90-percent confidence limits of 0.161 and 0.47L Spawning of northern bluefin takes place only in the western Pacific. Some of the juveniles migrate to the eastern Pacific, where they reside for several months to several years before returning to the western Pacific. The portion of fish which migrate to the eastern Pacific varies among years, and this appears to be an important cause of the annual variation in the catches in the eastern Pacific Ocean. Para los peces de entre 191 y 564 mm de talla se usa la curva de Gompertz, con valores de 581 mm y 4.32 para Loo y K (anual), respectivamente. Los peces de entre 564 y 1530 mm crecen de forma lineal, a 0.709 mm por día. Peces de edad Omarcados y liberados en el Pacífico occidental han sido recapturados en el Pacífico occidental, central, y oriental. ENGLISH: The spawning of Pacific northern bluefin tuna, Thunnus thynnus, takes place only in the western Pacific Ocean (WPO), but substantial numbers of the juveniles migrate to the eastern Pacific Ocean (EPO), where they remain for several months, or longer, and the.n return to the WPO. Lengthfrequency and tagging data show that many bluefin arrive in the EPO as 1-and 2-year olds, and remain there for one or two fishing seasons before returning to the WPO. The proportion of the fish which make the west-to-east migration varies among years. The numbers of 1-, 2-, 3-, 4, and >4 –year olds in the catches of the EPO are estimated for most years of the 1952-1991 period. Rule the World es una aplicación para móviles Android. Consiste en introducir al jugador en una realidad aumentada, mediante el uso de su localización, debe de recoger diferentes objetos para darles diferentes usos, como llevarlos equipados, usarlos para construir otros objetos o enviárselos a amigos. En el siguiente documento se muestra el completo desarrollo de este proyecto, como se ha realizado la gestión, en que partes se ha dividido, la planificación que se ha llevado para realizar el trabajo, el análisis que se hizo de la aplicación, junto con su diseño, como se ha realizado el desarrollo y las pruebas. Este proyecto ha servido para afianzar conocimientos adquiridos a lo largo del grado, como el desarrollo de bases de datos, seguridad y arquitecturas y algoritmos software. Pero también ha servido para aprender nuevas cosas, como programar para un sistema diferente, utilizar elementos poco vistos en el grado, como la geolocalización y los mapas.

A survey was carried out in September 1996 alongside the annual frame survey of Kainji Lake, Nigeria. As in the previous surveys, there were 6 main fishing gear types in use on the lake, which included gill nets, drift nets, beach seines, cast nets, longlines (baited and foul hooking) acid fishing traps. A total of 813 canoes were sampled. The canoe lengths were similar to the previous gear surveys. Altogether 466 gill nets were measured. They had an average length of 149 m, and were mostly bottom set. Of the gill nets measured 68% had a mesh size below 3 inches the minimum mesh size allowable according to the Niger and Kebbi State Fisheries Edict, 1996. Of concern was the large increase in the number of 1 inch meshed nets recorded on the lake. Drift nets had a mean length of 74 m. A total of 88% of all drift nets measured had a mesh size below 2.5 inches, the minimum mesh size stipulated in the State Edict. A total number of 102 beach seines were sampled with a mean length of 110 m, the mesh size in use on the lake remained as 0.1 inches under the Edict beach seines are banned from fishing in Kainji Lake. The decline in the lengths of the beach seines (and therefore catch rates) recorded since l995 was encouraging in the light of the destructive nature of these nets. A total of 30 cast nets were sampled, they had a mean diameter of 9.8 and a mean mesh size of 1.9 inches. All the nets had a hanging ratio of 0.5. Of the cast nets measured 40% had a mesh size below the minimum size of 2 inches recommended by the State Fisheries Edicts. Of concern was the continued rise in the number of gill nets with mesh sizes less than 2 inches. There were 2 types of longlines in operation on the lake. The baited and the foul hooking longlines. They were mostly bottom set and the hook size nos. 12-14 (Kirby sea hook) were predominant. Two types of fishing traps were identified, one was covered with a mesh of thin cane, one with netting material. The fishing traps had a mean height of 0.8 m and a mean width at the vase of 0.53 m. The mean mesh size used was 0.86 inches. Citharinus was reported as the most targeted of all fish species on the lake. Synodontis membranaceous was also targeted by most gears especially the smaller meshed nets. Labeo and Tilapia were low on the list of targeted species but their incidence remains high in the catches. (PDF contains 27 pages)

Since 1993, annual frame surveys have been conducted by the Nigerian-German Kainji Lake Fisheries Promotion Project to determine the distribution and number of fishing localities, fishing canoes and fishing gears around Kainji Lake, Nigeria. The total number of fishing localities has increased from 221 in 1993 to 286 in 1996. The fishing localities included 245 permanent fishing villages, 29 permanent fishing camps, 8 temporary fishing camps (with fishermen from Kainji Lake) and 4 temporary fishing camps (with fishermen from outside Kainji Lake area). There was an increase in the total number of fishing entrepreneurs, fishing assistants and fishing canoes over the years. A total number of 5,499 fishing entrepreneurs, 12,449 fishing assistants and 9,278 fishing canoes were recorded during the 1996 frame survey. From 1995 there was a decrease in the number of shoreline fisherfolk and a decrease in the number of transport canoes, the number of engines remained the same. During the 1996 survey, a total number of 18,655 gill nets, 1,560 drift nets, 753 beach seines, 5,548 cast nets, 7,400 longlines and 36,979 traps were recorded. The concentration of the gears (number per km shoreline) was highest in substrata 06 and 08. The total number of gill nets increased from 17,680 in 1995 to 18,655 in 1996. For the remaining 5 gear types a decrease in number was observed. Despite increasing numbers of gears on the lake, of concern is the decline recorded in all the fishing methods of the number of gears owned by individual entrepreneurs. This was most notable in the gill net and longline fisheries. These two fisheries have the lowest daily catch values and coupled with the problem of gear theft on the lake, ownership in future, may be expected to fall further. The number of larger fishing units also declined as did the number of gears new entrants enter the fishery with. The decline is particularly worrying for the beach seine fishery where diversification into other fishing methods would be beneficial in light of the present ban on seines. The group of not active fishing entrepreneurs (those who do not themselves participate in fishing activities) had the highest ownership of gears whilst the new entrants into the fishery had the lowest. Nucleic acids are most commonly associated with the genetic code, transcription and gene expression. Recently, interest has grown in engineering nucleic acids for biological applications such as controlling or detecting gene expression. The natural presence and functionality of nucleic acids within living organisms coupled with their thermodynamic properties of base-pairing make them ideal for interfacing (and possibly altering) biological systems. We use engineered small conditional RNA or DNA (scRNA, scDNA, respectively) molecules to control and detect gene expression. Three novel systems are presented: two for conditional down-regulation of gene expression via RNA interference (RNAi) and a third system for simultaneous sensitive detection of multiple RNAs using labeled scRNAs.
RNAi is a powerful tool to study genetic circuits by knocking down a gene of interest. RNAi executes the logic: If gene Y is detected, silence gene Y. The fact that detection and silencing are restricted to the same gene means that RNAi is constitutively on. This poses a significant limitation when spatiotemporal control is needed. In this work, we engineered small nucleic acid molecules that execute the logic: If mRNA X is detected, form a Dicer substrate that targets independent mRNA Y for silencing. This is a step towards implementing the logic of conditional RNAi: If gene X is detected, silence gene Y. We use scRNAs and scDNAs to engineer signal transduction cascades that produce an RNAi effector molecule in response to hybridization to a nucleic acid target X. The first mechanism is solely based on hybridization cascades and uses scRNAs to produce a double-stranded RNA (dsRNA) Dicer substrate against target gene Y. The second mechanism is based on hybridization of scDNAs to detect a nucleic acid target and produce a template for transcription of a short hairpin RNA (shRNA) Dicer substrate against target gene Y. Test-tube studies for both mechanisms demonstrate that the output Dicer substrate is produced predominantly in the presence of a correct input target and is cleaved by Dicer to produce a small interfering RNA (siRNA). Both output products can lead to gene knockdown in tissue culture. To date, signal transduction is not observed in cells; possible reasons are explored.
Signal transduction cascades are composed of multiple scRNAs (or scDNAs). The need to study multiple molecules simultaneously has motivated the development of a highly sensitive method for multiplexed northern blots. The core technology of our system is the utilization of a hybridization chain reaction (HCR) of scRNAs as the detection signal for a northern blot. To achieve multiplexing (simultaneous detection of multiple genes), we use fluorescently tagged scRNAs. Moreover, by using radioactive labeling of scRNAs, the system exhibits a five-fold increase, compared to the literature, in detection sensitivity. Sensitive multiplexed northern blot detection provides an avenue for exploring the fate of scRNAs and scDNAs in tissue culture.
