Metsäteollisuudesta kertyy vuosittain suuria määriä ylijäämämateriaalia, kuten puun kuorta ja oksia.Ylimääräinen aines käytetään pääasiassa energiantuotantoon, mutta uusia soveltamismahdollisuuksia kaivataan. Kuoren on havaittu olevan potentiaalinen lähde monille bioaktiivisille yhdisteille, joille olisi käyttöä esimerkiksi lääke- ja kemianteollisuudessa sekä maa-, metsä- ja puutarhatuotannon tuholaistorjunnassa. Tutkimus on osa Euroopan Unionin rahoittamaa ForestSpeCs-projektia, jonka tarkoituksena on selvittää metsäteollisuuden ylijäämämateriaalien vaihtoehtoisia käyttötapoja. Valittujen kymmenen teollisesti merkittävän pohjoisen puulajin (Abies nephrolepis, Betula pendula, Larix decidua, L. gmelinii, L. sibirica, Picea abies, P. ajanensis, P. pumila, Pinus sylvestris, Populus tremula) kuoresta uutettujen aineiden soveltuvuutta syönninestoaineeksi testattiin kaaliperhosen (Pieris brassicae L.) ja krysanteemiyökkösen (Spodoptera littoralis Boisduval) toukilla sekä osittain sinappikuoriaisella (Phaedon cochloreae Fabricius) ja idänlehtikuoriaisella (Agelastica alni L.). Uutteet valmistettiin yhteistyössä projektin ryhmien avulla tai itsenäisesti erilaisin menetelmin. Testaukset tehtiin laboratorio-oloissa käyttäen lehtikiekkojen valintabiotestiä sekä karkeilla uutteilla että niistä erotelluilla yksittäisillä yhdisteillä. Tehdyistä mittauksista laskettiin syönninestoindeksit (FDI). Tulosten perusteella lähes kaikki testatut uutteet vaikuttivat ainakin jossain määrin kohdehyönteisen syöntikäyttäytymiseen. Hieman yli puolet kaaliperhosella testatuista 46 uutteesta aiheuttivat yli 50 % syönnineston eli kaaliperhonen suosi kontrollilehtiä uutteella käsiteltyjä todennäköisemmin. Krysanteemiyökkösellä yli 50 %:n syönnineston aiheuttivat vain seitsemän testatuista 56 uutteesta. Lisäksi kolme uutetta lisäsi käsiteltyjen kiekkojen syöntiä merkittävästi. Idänlehtikuoriaistoukat ja -aikuiset karttoivat erityisesti abietiinihapolla käsiteltyjä lehtiä. Sinappikuoriaisella testatut uutteet toimivat myös lupaavasti. Testattujen puulajien kuoresta on mahdollista uuttaa biologisesti aktiivisia yhdisteitä, mutta tuholaistorjunnan kannalta oikeiden pitoisuuksien ja tehokkaiden uuttomenetelmien löytäminen vaatii jatkotutkimuksia. Kuoren sisältämien yhdisteiden laatu ja määrä vaihtelevat monien tekijöiden, kuten ympäristön ja genetiikan vaikutuksesta. Hyönteisten sietokyky vaihtelee myös paljon lajeittain ja yksilöidenkin välillä on eroja. Uutteista valmistettavia torjunta-aineita olisi kuitenkin mahdollista sisällyttää esimerkiksi integroituun torjuntaan muiden menetelmien rinnalle tulevaisuudessa.

Background: Malaria was prevalent in Finland in the 18th century. It declined slowly without deliberate counter-measures and the last indigenous case was reported in 1954. In the present analysis of indigenous malaria in Finland, an effort was made to construct a data set on annual malaria cases of maximum temporal length to be able to evaluate the significance of different factors assumed to affect malaria trends. Methods: To analyse the long-term trend malaria statistics were collected from 1750–2008. During that time, malaria frequency decreased from about 20,000 – 50,000 per 1,000,000 people to less than 1 per 1,000,000 people. To assess the cause of the decline, a correlation analysis was performed between malaria frequency per million people and temperature data, animal husbandry, consolidation of land by redistribution and household size. Results: Anopheles messeae and Anopheles beklemishevi exist only as larvae in June and most of July. The females seek an overwintering place in August. Those that overwinter together with humans may act as vectors. They have to stay in their overwintering place from September to May because of the cold climate. The temperatures between June and July determine the number of malaria cases during the following transmission season. This did not, however, have an impact on the longterm trend of malaria. The change in animal husbandry and reclamation of wetlands may also be excluded as a possible cause for the decline of malaria. The long-term social changes, such as land consolidation and decreasing household size, showed a strong correlation with the decline of Plasmodium. Conclusion: The indigenous malaria in Finland faded out evenly in the whole country during 200 years with limited or no counter-measures or medication. It appears that malaria in Finland was basically a social disease and that malaria trends were strongly linked to changes in human behaviour. Decreasing household size caused fewer interactions between families and accordingly decreasing recolonization possibilities for Plasmodium. The permanent drop of the household size was the precondition for a permanent eradication of malaria.

As a prelude to achieving transgenesis in Bombyx mori, conditions have been established for successful microinjection of cloned foreign genes into the silk worm eggs. A sharpened metallic needle is used to pierce the thick chorion layer of the eggsheil, approaching through a droplet of DNA solution deposited on its surface. The microinjection is carried out within 2-2.5 h after oviposition and the injected eggs show 3-5% hatchability and 80-90% survival. Such larvae continuously expressed the microinjected cloned reporter gene, beta-galactosidase, placed under the control of a constitutively expressed cytoplasmic actin A3 gene promoter from B. mori. The expression is seen in different tissues, viz. the fat body, tracheae and the silk glands, till the late larval instars. The microinjected DNA sequences are retained in the adult G(o) moths.

Microfluidic devices have been developed for imaging behavior and various cellular processes in Caenorhabditis elegans, but not subcellular processes requiring high spatial resolution. In neurons, essential processes such as axonal, dendritic, intraflagellar and other long-distance transport can be studied by acquiring fast time-lapse images of green fluorescent protein (GFP)-tagged moving cargo. We have achieved two important goals in such in vivo studies namely, imaging several transport processes in unanesthetized intact animals and imaging very early developmental stages. We describe a microfluidic device for immobilizing C. elegans and Drosophila larvae that allows imaging without anesthetics or dissection. We observed that for certain neuronal cargoes in C. elegans, anesthetics have significant and sometimes unexpected effects on the flux. Further, imaging the transport of certain cargo in early developmental stages was possible only in the microfluidic device. Using our device we observed an increase in anterograde synaptic vesicle transport during development corresponding with synaptic growth. We also imaged Q neuroblast divisions and mitochondrial transport during early developmental stages of C. elegans and Drosophila, respectively. Our simple microfluidic device offers a useful means to image high-resolution subcellular processes in C. elegans and Drosophila and can be readily adapted to other transparent or translucent organisms.

A partial genomic clone of Bombyx mori homologue of the segment polarity gene Cubitus interruptus (BmCi), encoding the conserved zinc finger domain and harbouring two introns, has been characterized. BmCi was expressed in the silkglands of B. mori from embryonic to the late larval stages(3rd, 4th and 5th intermoults). The expression was confined to the anterior region of the middle silkglands, overlapping with the domain of sericin-2 expression and excluding the domains of Bm invected expression, namely the middle and posterior regions of the middle silkglands. In the wing discs, the expression was restricted to the anterior compartment, which increased from 4th to 5th larval intermoults and declined later in the pupal wing buds. In gonadal tissues (both ovaries and testes) BmCi was expressed from the larval to pupal stages. The transcripts were localized to the sperm tubes containing spermatogonia in the testis of Bombyx larvae. BmCi expression, however, was not detected in any of these tissues during the moulting stages. Expression of Ci in the wing discs and gonads is evolutionarily conserved, while the silkgland represents a novel domain. Our results imply that BmCi is involved in the specification and maintenance of micro-compartment identity within the middle silkglands.

Fruit flies that belong to the genus Bactrocera (Diptera: Tephritidae) are major invasive pests of agricultural crops in Asia and Australia. Increased transboundary movement of agricultural produce has resulted in the chance introduction of many invasive species that include Bactrocera mainly as immature stages. Therefore quick and accurate species diagnosis is important at the port of entry, where morphological identification has a limited role, as it requires the presence of adult specimens and the availability of a specialist. Unfortunately when only immature stages are present, a lacunae in their taxonomy impedes accurate species diagnosis. At this juncture, molecular species diagnostics based on COX-I have become handy, because diagnosis is not limited by developmental stages. Yet another method of quick and accurate species diagnosis for Bactrocera spp. is based on the development of species-specific markers. This study evaluated the utility of COX-I for the quick and accurate species diagnosis of eggs, larvae, pupae and adults of B. zonata Saunders, B. tau Walker, and B. dorsalis Hendel. Furthermore the utility of species-specific markers in differentiating B. zonata (500bp) and B. tau (220bp) was shown. Phylogenetic relationships among five subgenera, viz., Austrodacus, Bactrocera, Daculus, Notodacus and Zeugodacus have been resolved employing the 5' region of COX-I (1490-2198); where COX-I sequences for B. dorsalis Hendel, B. tau Walker, B. correcta Bezzi and B. zonata Saunders from India were compared with other NCBI-GenBank accessions. Phylogenetic analysis employing Maximum Parsimony (MP) and Bayesian phylogenetic approach (BP) showed that the subgenus Bactrocera is monophyletic.

Plants produce volatile organic compounds (VOCs) in a variety of contexts that include response to abiotic and biotic stresses, attraction of pollinators and parasitoids, and repulsion of herbivores. Some of these VOCs may also exhibit diel variation in emission. In Ficus racemosa, we examined variation in VOCs released by fig syconia throughout syconium development and between day and night. Syconia are globular enclosed inflorescences that serve as developing nurseries for pollinating and parasitic fig wasps. Syconia are attacked by gallers early in their development, serviced by pollinators in mid phase, and are attractive to parasitoids in response to the development of gallers at later stages. VOC bouquets of the different development phases of the syconium were distinctive, as were their day and night VOC profiles. VOCs such as alpha-muurolene were characteristic of the pollen-receptive diurnal phase, and may serve to attract the diurnally-active pollinating wasps. Diel patterns of release of volatiles could not be correlated with their predicted volatility as determined by Henry's law constants at ambient temperatures. Therefore, factors other than Henry's law constant such as stomatal conductance or VOC synthesis must explain diel variation in VOC emission. A novel use of weighted gene co-expression network analysis (WGCNA) on the volatilome resulted in seven distinct modules of co-emitted VOCs that could be interpreted on the basis of syconium ecology. Some modules were characterized by the response of fig syconia to early galling by parasitic wasps and consisted largely of green leaf volatiles (GLVs). Other modules, that could be characterized by a combination of syconia response to oviposition and tissue feeding by larvae of herbivorous galler pollinators as well as of parasitized wasps, consisted largely of putative herbivore-induced plant volatiles (HIPVs). We demonstrated the usefulness of WGCNA analysis of the volatilome in making sense of the scents produced by the syconia at different stages and diel phases of their development.

1. Plant reproductive phenology is generally viewed as an individual's strategy to maximize gamete exchange and propagule dispersal and is often considered largely dependent on patterns of floral initiation. Reproductive phenology, however, can be affected by proximate responses to pollinators, parasites and herbivores which could influence floral longevity or fruit development time. 2. We examined the influence of insect interactants on within-plant reproductive phenology in the fig-fig wasp nursery pollination mutualism in Ficus racemosa (Moraceae). Most figs support a wasp community comprised of a mutualistic pollinator, with several host-plant-specific non-pollinating herbivorous gallers and parasitoids. These wasps reproduce within enclosed inflorescences called syconia, which develop into fruit after pollination. While different wasp species oviposit into syconia at varying times during its ontogeny, all wasp progeny are constrained to exit syconia simultaneously just prior to fruit ripening. Developing larvae of early-ovipositing wasps may hasten syconium ontogeny through formation of earlier and larger nutrient sinks, whereas larvae of late-arriving parasites may lengthen syconium ontogeny to complete their development successfully. Seeds are also important nutrient sinks. The number of seeds and the type and number of developing wasps may therefore be expected to influence syconium development times, thereby affecting the reproductive synchrony of syconia on a plant. 3. Observations on naturally pollinated and parasitized syconia indicated that their seed and wasp content affected syconium development time. Experimental manipulations of syconia to produce only seeds or various combinations of wasps confirmed this finding. Early-ovipositing galler progeny reduced syconium development times, while gallers ovipositing concurrently with pollinators had no effect on syconium development. Late-ovipositing parasitoid progeny, the presence of only seeds within the syconium, or delayed pollination increased syconium development time. The differential development of syconia, which was influenced by mutualistic or parasitic progeny, accordingly contributed to within-tree reproductive asynchrony. 4. Synthesis. Individual reproductive units in fig trees called syconia, which also function as brood sites for pollinating and parasitic fig wasps, have plastic development durations dependent on pollination timing and species of wasps developing within them. Syconium development times are a likely compromise between conflicting demands from developing seeds and different wasp species.

Measuring forces applied by multi-cellular organisms is valuable in investigating biomechanics of their locomotion. Several technologies have been developed to measure such forces, for example, strain gauges, micro-machined sensors, and calibrated cantilevers. We introduce an innovative combination of techniques as a high throughput screening tool to assess forces applied by multiple genetic model organisms. First, we fabricated colored Polydimethylsiloxane (PDMS) micropillars where the color enhances contrast making it easier to detect and track pillar displacement driven by the organism. Second, we developed a semiautomated graphical user interface to analyze the images for pillar displacement, thus reducing the analysis time for each animal to minutes. The addition of color reduced the Young's modulus of PDMS. Therefore, the dye-PDMS composite was characterized using Yeoh's hyperelastic model and the pillars were calibrated using a silicon based force sensor. We used our device to measure forces exerted by wild type and mutant Caenorhabditis elegans moving on an agarose surface. Wild type C. elegans exert an average force of similar to 1 mu N on an individual pillar and a total average force of similar to 7.68 mu N. We show that the middle of C. elegans exerts more force than its extremities. We find that C. elegans mutants with defective body wall muscles apply significantly lower force on individual pillars, while mutants defective in sensing externally applied mechanical forces still apply the same average force per pillar compared to wild type animals. Average forces applied per pillar are independent of the length, diameter, or cuticle stiffness of the animal. We also used the device to measure, for the first time, forces applied by Drosophila melanogaster larvae. Peristaltic waves occurred at 0.4Hz applying an average force of similar to 1.58 mu N on a single pillar. Our colored microfluidic device along with its displacement tracking software allows us to measure forces applied by multiple model organisms that crawl or slither to travel through their environment. Predation risk can strongly constrain how individuals use time and space. Grouping is known to reduce an individual's time investment in costly antipredator behaviours. Whether grouping might similarly provide a spatial release from antipredator behaviour and allow individuals to use risky habitat more and, thus, improve their access to resources is poorly known. We used mosquito larvae, Aedes aegypti, to test the hypothesis that grouping facilitates the use of high-risk habitat. We provided two habitats, one darker, low-risk and one lighter, high-risk, and measured the relative time spent in the latter by solitary larvae versus larvae in small groups. We tested larvae reared under different resource levels, and thus presumed to vary in body condition, because condition is known to influence risk taking. We also varied the degree of contrast in habitat structure. We predicted that individuals in groups should use high-risk habitat more than solitary individuals allowing for influences of body condition and contrast in habitat structure. Grouping strongly influenced the time spent in the high-risk habitat, but, contrary to our expectation, individuals in groups spent less time in the high-risk habitat than solitary individuals. Furthermore, solitary individuals considerably increased the proportion of time spent in the high-risk habitat over time, whereas individuals in groups did not. Both solitary individuals and those in groups showed a small increase over time in their use of riskier locations within each habitat. The differences between solitary individuals and those in groups held across all resource and contrast conditions. Grouping may, thus, carry a poorly understood cost of constraining habitat use. This cost may arise because movement traits important for maintaining group cohesion (a result of strong selection on grouping) can act to exaggerate an individual preference for low-risk habitat. Further research is needed to examine the interplay between grouping, individual movement and habitat use traits in environments heterogeneous in risk and resources. El presente estudio realizado en el municipio de San Dionisia Departamento de Matagalpa, a travéz de un estudio retrospectivo del Banco de datos de seguimiento a fincas que PRODESSA venía realizando desde 1988 en 6 comunidades, permitió caracterizar el ganado manejado en pequeñas fincas encontrándose, un rango promedio de producción de leche de 3 a 5.3 litros y un rango promedio de lactación entre 7.7 a 11.1 meses. La alimentación principal es a base de pasto jaragua (Hyparrhenia ruffa), rastrojos de csecha, leguminosas naturales, en menor grado el pasto Taiwán (Pennisetum purpureum), pasto estrella, (Cynodon nlenfuensis), y suministro de sal común. En salud animal las actividades realizadas son: vacunación contra Ántrax y Pierna Negra, la realizan 2 veces en el año el 100% de productores, control de ectoparásitos 12 baños promedio anuales el 100% de productores, desparasitación interna, 2 veces en el año el 24% de productores, una vez al año el 57% y el 11% no lo realizan, la vitaminación, el 60% realiza una por año, el 12% 2 por año y el 28% de productores no vitaminan. Las pequeñas fincas presentan áreas no mayores de 5 mz. de tierra el 46.75%, el 33.76% con áreas no mayores de 10 mz. de terreno y el 19.43% con áreas mayores de 10 mz. de tierra. Además se definieron siete prácticas de manejo y se evaluó su efecto en vacas con anestro obteniéndose los siguientes resultados: la primera práctica consistió en la suplementación alimenticia a base de maíz triturado (A), desparasitación interna con levamisol (D) y masajes estimulantes (M) a 33 vacas representando el 100% de los animales, manifestando celo al final de la práctica el 9.09%. La segunda práctica se aplicó a un total de 16 vacas que, representan el 48.48% del total inicial, a estas se les suministró vitamina AD,E (V), de las cuales presentaron celo el 21.21%. La tercera práctica se realizó con 14 vacas que representan el 42.42% del cohorte, a las que se les suministró minerales (Mi) aplicando Tonofosfan, presentó celo el 24.24% del total inicial. La cuarta práctica se aplicó a 9 vacas que representan el 27.27% del cohorte, que posterior a la aplicación de la primera y la segunda práctica se les suministró minerales, presentando celo el 12.12% del total inicial. La quinta práctica se aplicó a un total de 6 vacas que representaron el 18.18% del total, a estas se suministró vitamina AD,E, presentó celo el 15.15% del total inicial. La sexta práctica se ejecutó con 5 vacas representando el 15.15%, se les suministro hormonas, a 4 de ellas por presencia de cuerpo lúteo persistente, se les aplicó la hormona Prosolvin y a una Por presencia de quiste folicular se le aplicó la hormona Foligon, de las cuales presentó celo el 3.03%. La séptima práctica se realizó con una vaca, que representa el 3.03% del total inicial se le aplicó la hormona Foligon por presencia de quiste folicular, no logrando esta vaca presentar celo con esta última práctica. Al comparar las prácticas entre ellas se obtuvieron los siguientes resultados: Se encontraron diferencias significativas (P<0.05) al comparar la primera práctica con la segunda, tercera, cuarta y quinta práctica, mayoritariamente hacia la segunda, tercera, cuarta y quinta práctica, de igual manera al comparar la quinta con la sexta y séptima práctica se encontraron diferencias significativas (P<0.05) con mayor efecto a la quinta práctica.

En el presente estudio se compararon cuatro dietas con diferentes niveles de energía 2,940, 3,040, 3,140 y 3,240 (Kcal EM/Kg de alimento), los cuales fueron ajustados a los requerimientos nutricionales en cuanto a la relación caloría proteína recomendados por la Hubbard Farm, Inc. (1991). Durante un periodo de 56 días los broilers fueron evaluados en la granja avícola "Buenos Aires" que pertenece a la Tip Top Industrial S.A. se utilizaron 803 pollos de engorde de un día de nacidos de la línea Hubbard, los que fueron distribuidos aleatoriamente en tres unidades experimentales proporcionales a los cuatro tratamientos existentes, estos distribuidos en un diseño completamente al azar y sometidos a la prueba de Tukey el que proporciona la superioridad existente entre tratamientos. Se encontraron diferencias significativas (P<0.05) entre tratamientos en las variables peso vivo, ganancia de peso y conversión alimenticia pero no así para el consumo alimenticio. Los tratamientos T1 T2, T3 y T4 presentaron mortalidad al final del periodo de 2.57, 4.41, 7.88 y 10.89 (%) respectivamente y rendimiento en la canal del 84.45 % para todos los tratamientos. Se concluyó que las dietas más energéticas proporcionaban las mejores conversiones alimenticias y las mayores ganancias de peso. Y a medida que se incrementaba el nivel energético en las dietas los costos alimenticios también se incrementaban. La fórmula más económica es el T1 (2940 Kcal EM/Kg de alimento, 21.63 y 18.89 % de proteína para inicio y finalización) durante todas las semanas de estudio. El momento óptimo para la matanza es la sexta semana para el T 1 (2940 Kcal EM/Kg de alimento, 21.63 y 18.89 % de proteínas) y T2 (3040 Kcal EM/Kg de alimento, 22.36 y 19.15 % de proteínas) por efecto de los costos alimenticios.

En el periodo de los meses Enero-Febrero de 1973, se llevo a cabo el estudio del crecimiento de larvas de Heliothis zea en dieta artificial, en el Laboratorio de Biología de la Universidad Nacional Autónoma de Nicaragua (con sede en el Departamento de Leon). Habiendose antes hecho pruebas preliminares en el Laboratorio de Biología de la Escuela Nacional de Agricultura y Ganadería (con sede en el Departamento de Managua). Los objetivos eran: a) determinar el numero de mudas; b) la duración de su ciclo larval y c) la curva de crecimiento de Heliothis zea. Para ello se trabajo en el Laboratorio, con crianza del insecto, haciendo uso de la dieta artificial de Shorey modificada. Los datos a tomar fueron: a) longitud del cuerpo en mms.; b) tiempo transcurrido al tiempo de la muda; c) largo de capsula cefálica; d) ancho de capsula cefálica. De los resultados obtenidos, se detectaron seis mudas en el desarrollo de su ciclo larval. La duración de su ciclo larval vario desde 13 días hasta 21 días, encontrándose un promedio de 16.07 días como resultado de 28 observaciones que se tomaron. En la duración de su estado pupal se obtuvo un promedio de 10.06 días, resultado obtenido de 18 observaciones los análisis se efectuaron en el Instituto Interamericano de Ciencias Agrícolas, obteniéndose los siguientes resultados: a) A medida que aumentaba la edad de la larva, los periodos entre muda son mas variables. b) Siendo la variación del tiempo y longitud del cuerpo, máxima en la sexta muda. c) Los tamaños de las larvas son bastantes constante durante las dos primeras mudas; y se incrementa la variabilidad en las cuatro ultimas. d) Se logro ajuste a la ecuación logística en promedio de 99.10% y con los parámetros B0, B1, B2, se hizo curvas de crecimiento promedio de larvas de Heliothis zea. e) Análisis de correlación fueron efectuados, para conocer el grado de asociación de las variables largo del cuerpo vs. ancho de capsula cefálicas, no encontrándose correlación alguna.
