Fez-se um estudo de seleção de métodos para cobre disponível, empregando-se trinta solos coletados na região de Jaboticabal, SP. Os solos utilizados pertencem aos seguintes grandes grupos: Solos Podzolizados de Lins e Marília, variação Lins e variação Marília, Latossolo Vermelho-Escuro fase arenosa, Latossolo Roxo, e Terra Roxa Estruturada. O cobre dos solos foi extraído através das seguintes soluções extratoras: CaCl2 0,5 M; Mg02 2,0 N; CH3COONH4 1,0 N1 pH 6,0 e pH 4,8; HCl 0,05 N e O,1 N; H2SO4 0,05 N; DTPA 0,005 M pH 7,3 e pH 6,0; EDTA 0,01 M + (NH4)2CO3 1,0 MpH 8,6; EDTA 0,02 M + CH3C0011 0,5 M + CH3COONH4 0,5 M pH 4,65 (AAAc-EDTA); e Na2EDTA 1,0%. Foi realizado ensaio em casa de vegetação, e o milho (Zea mays L.) foi empregado como planta teste. Excluindo-se o extrator CaCl2 0,5 M, todos os demais apresentaram coeficientes de correlação entre cobre extraído do solo e o absorvido pelo milho, significativos e próximos uns dos outros. Assim, com base também em parâmetros como o coeficiente de variação, o coeficiente angular e a praticidade do método, pode-se indicar um dos seguintes extratores para a avaliação do cobre disponível dos solos: DTPA 0,005 M pH 7,3, HCl 0,1 N e Na2EDTA 1,0%.

The efficiency of two methods for determining 3-hydroxybutyrate and 3-hydroxyvalerate content in the biopolymer (poly(3-hydroxybutyrate-co-3-hydroxyvalerate) has been evaluated. Both methods are based on (a) the hydrolysis of polymers, (b) the esterification with methyl (method A) and isoamilic alcohol (method B) and (c) analysis of the esters by gas chromatography using flame ionization detection (GC-FID). After optimization of the analytical conditions, the main validation parameters (linearity, quantification limit, detection limit, recovery, sensitivity and precision) were determined showing that both analytical procedures can be applied to control the process of poly(3-hydroxybutyrate) production. However, method B gives better analytical results than method A and has the advantage of not using chlorinated solvents.

The digital elevation model is important to determine the slope and land use capability, therefore, a proposal of methodology for acquisition of elevation data contemplating an efficient algorithm to generate a slope map was developed. Thus, it was aimed to obtain and evaluate a digital elevation model without the vetorization of the contours on planialtimetric charts. The area for acquisition of elevation data was Sao Manuel, SP. The data were collected by two methods: level contour vetorization and the gathering of elevation points on the level contour with maximum elevation points. The elevation data were analyzed by geostatistical techniques. Inspite of wide difference in the number of collected points between two methods, the variograms were adjusted to the exponential model and showed a range of approximately 1500 m, which does not justify the wide difficulty in vetorization of the planialtimetric charts, once the data points collected in the area were appropriately distributed, they represented rightly the terrain surface.

Estimating total landings of artisanal fisheries in managed areas is essential for the development of programs aimed at the establishment of sustainable production systems. However, few experiences addressed the constraints of assessments targeted at multispecific and small scale fisheries with pulverized landings. Total annual production (fish, lobsters and octopus) at the Marine Extractive Reserve of Corumbau, Bahia, Brazil, in 2006, was estimated through yields recorded by fisherfolks, in the scope of a participatory fisheries monitoring program. Three designs were employed: 1) simple random sampling, 2) stratified random sampling by month and fishing frequency, 3) stratified random sampling by community, fishing frequency and months, resulting in estimates of 349t (95% confidence interval = 305-393), 330t (289-372) and 315t (279-351), respectively. This kind of information can help design strategies for aggregation of value to small scale fisheries production, and also for the dimensioning of the corresponding infrastructure. However, these estimates shall not be used to support fishing effort increase, as there is high uncertainty regarding the state of the stocks, as well as limitations related to the asymmetry in monthly landings per fisherfolk and to the sampling coverage in the different strata.

Citrus blackfly, Aleurocanthus woglumi Ashby, is a serious pest of citrus culture and other economically important fruit crops. It is a present quarantine pest or A2 maximum alert restricting trades with other regions free of its presence. Since the frst occurrence of the citrus blackfly in Belem in 2001 its dissemination was quickly to other States and regions of citrus production in Brazil. As an exotic pest, basic knowledge is scarce in order to establish the appropriate management to the insect in Brazil. Thus, the aim of the present study was to provide information about important aspects of A. woglumi, such as: history and geographical distribution, bioecology, host plants, appropriate control methods, among others, in order to provide subsidies for futures researches about the citrus blackfly in Brazil.

The authors studied the lateral positional desviations of the mandible, in relation to the facial median line in 30 (thirty) full edentulous patients, with the purpose to verify the influence of the unstrained guided (chinpoint guidance) and deglutition methods for the determination of centric relation. According the results obtained they conclude that the unstrained guided method produced a mandibular lateral desviation with a mean value of the 0.752 mm and in the deglutition method the mean value observed was 1.109 mm. The statistical analysis of the results revealed for the unstrained guided method a proportion of the points to the right and left of the median line not statistically significant, while for the deglutition method the difference was statistically significant at a level of 5%.

The chronology of the wound healing process following tooth extraction was studied by means of two kinds of histological cuts. Two groups of 42 albino rats were employed. In the first one, the sockets were cut in a transversal way. In the second one the cuts were performed in a longitudinal way. The rats were sacrificed after 3, 6, 9, 15, 21, 24, and 28 days following the surgeries. After laboratorial outline the obtained pieces were stained by hematoxylin and eosin for histological purposes. It way be concluded that: 1. The results got from longitudinal cuts were in agreement to those described by other authors; 2. The transversal cuts allow us to detect intensive resorption of the lateral alveolar wall at the cervical thirs; 3. On the 21st day following dental extraction the incisor socket of the rat shows a great deal of areas not ossified; 4. The healing process of dental extraction wounds of the upper incisor of the rat is completed between 24 and 28 post operative days.

In order to determine the value of immunohistochemical staining methods for the morphologic diagnosis, we studied 949 histologic specimens sent for consultation to the Immunohistochemistry Laboratory of Department of Pathology of the Medical School of Botucatu in the period 1984-1989. All case were submitted to the immunoperoxidase staining with the methods PAP or ABC. Immunohistochemical stains confirmed the original morphologic diagnosis in 468 cases (49.3%); made the definitive diagnosis from a list of differential diagnostic possibilities in 244 cases (25.7%); provided contributory information in 74 cases (7.8%); were non-contributory in 114 cases (12%) and rendered an unsuspected diagnosis in 49 cases (5.2%). In some cases with non-contributory information the differences in methods of fixation might have led to suboptimal preservation of tissue antigens. The immunohistochemical staining may provide important and sometimes essential informations for definitive diagnosis. This technique was particularly useful for differential diagnosis between carcinoma, lymphoma and melanoma.

The aboveground biomass content of a region can be estimated by either direct or indirect methods. Direct methods correspond to the biomass content determination with scales and extrapolation of results to larger areas. It is a destructive and very laborious procedure. Indirect methods utilize formulas whose entrance parameters are obtained from forest inventories. Forest inventories are made with the purpose to plan exploration and land use and the inventory data are frequently not suitable for biomass estimation. Problems with both methods increase in the Amazon region, where little information is available on forest biomass. The objective of this paper is to establish, by comparing the application of the indirect and direct methods in the determination of the biomass, the more appropriate indirect formulation to represent the characteristic vegetation of a region in the amazonian forest. A 0.2 hectare area was chosen, which was part of a major forest clearing experiment conducted in Tomé Açu, a town located 250 km south of Belém, the capital of the Brazilian state of Pará. The entire biomass in the area was weighted with scales during the three weeks that followed the cut of the forest in July 1994. A detailed inventory was carried out in the area and then the indirect method was applied in the data. Seven different formulas for determining biomass were used. Comparison of the data of real mass and the mass obtained through the application of the seven formulas indicated that the more suitable for the region is given by FW = α · φβ · Hγ, where FW is total fresh weight (kg), φ is the diameter at breast height (cm), H is the total height of the tree and α, β and γ are regression coefficients (equal to 0.026, 1.529 and 1.747, respectively).

Fifteen soil samples and four extradants (DTPA, Na 2EDTA, HCl and Mehlich 1) were used to study soil extraction methods for available zinc. Each extradant was studied in three soil:solution ratios (1:2, 1:4 and 1:10) and in three extraction periods in the ratio most frequently used. The zinc extracted by DTPA, Na 2EDTA and HCl was not affected by use of soil:solution ratios 1:2 and 1:4, but the amount extracted increased with the 1:10 ratio. In this relation, the variation coefficients increased. With Mehlich, no definite tendency was observed. The increase of extraction period for DTPA extractant, in 1:2 ratio, had little effect on the amount of zinc extracted and on the precision. As to Na 2EDTA extractant in 1:10 ratio, with the increase of extraction time, both increased. As to HCl and Mehlich, both in 1:4 ratio, the increase in extraction period had no effect on the amount of zinc extracted and no definite effect on the precision. Thus, the use of 1:2 ratio for DTPA and Na 2EDTA and 1:4 ratio for acid extractants is recommended. Concerning the period of extraction, 120 min should be used with DTPA and Na 2EDTA, 15 min with HCl and 5 min with Mehlich.

The objective of this research was to evaluate the effectiveness of the extraction of Xanthomonas campestris pv. phaseoli from naturally infected dry bean seeds. Extraction methods tested included soaking whole seeds in sterilized saline phosphate buffer and crushing seeds after soaking in sterilized saline phosphate buffer. The bacterium was isolated on a semiselective agar medium. The seed crushing method was found to be more effective. The bacterium strains isolated were pathogenic to bean leaves, reacted with X. campestris pv. phaseoli antiserum, and had morphological and physiological/biochemical characteristics typical of the X. campestris pv. phaseoli.

Silages of Pennisetum purpureum Schum. cv. Guaçu prepared with 0, 8, 16 and 24% of ground ear com with husks, wheat bran and saccharin, dry weight of additive/wet weight of green chop basis. The experimental design was a randomized blocks one in split-plot; the plots were the additives and levels, and the sub-plots the sampling methods. The material was ensiled using plastic vessels in middle of which holed pvc pipes (3 inches diameter) were put. These pipes (one per vessel) had the same length as the height of the vessels, and were filled at the same time and compacted the same way as the vessels. The first method of sampling used the material ensiled inside the pvc pipe, which was lifted out from the vessel at the moment of the silo opening. The other sampling method, normaly used in digestibility trials, consisted of samples composed by daily sub-samples collected in the vessels. The pvc sampling method was more efficient because it sampled a profile of the whole silage. All of the silages showed high percentages of lactic acid and low percentages or even absence of butyric acid, though in all silages high ammoniacal-N percentages were detected.

BACKGROUND: Ultrasonography (US), Computed Tomography (CT), and Magnetic Resonance imaging (MR) were compared for the staging of renal tumors. The differences between these imaging techniques were also studied for their ability to detect adenopathies, vascular invasion, distant intra-abdominal metastases, and particularly adjacent organ invasion. METHODS: Thirty-one patients with solid or complex renal masses were prospectively studied using US, CT, and MR. Differences between the results obtained were studied using the COCHRAN G test and the McNEMAR test. The sensitivity and specificity of each diagnostic technique were compared against a gold standard of the surgical and histopathological findings. RESULTS: The following sensitivities were obtained: For the detection of adenopathy, US 63.6%, CT and MR 90.9%. For vascular invasion, US 42.8%, CT and MR 85.7%. For the adjacent organ invasion, US 28.5%, CT 85.7%, and MR 71.4%. Some of the criteria that suggest invasion of adjacent structures include: the envelopment of the adjacent structures by the tumor, tumor extension into the adjacent structures with an irregular appearance, and alterations in shape, size, and density of adjacent structures. Loss of fat planes between the tumor and adjacent structures is not a sign of tumor invasion. CONCLUSIONS: Significant differences were found in the detection capacity of US in relation to CT and MR, which were similar. All three techniques were highly sensitive and specific only in the detection of distant abdominal metastases. In addition to the accuracy of these diagnostic modalities for the detection and staging of tumors, invasiveness, risks and cost should be considered in relation to relative costs and benefits.
