The hypothesis that fatigue during prolonged exercise arises from insufficient intramuscular glycogen, which limits tricarboxylic acid cycle (TCA) activity due to reduced TCA cycle intermediates (TCAI), was tested in this experiment. Seven endurance-trained men cycled at approximately 70% of peak O(2) uptake (Vo(2 peak)) until exhaustion with low (LG) or high (HG) preexercise intramuscular glycogen content. Muscle glycogen content was lower (P < 0.05) at fatigue than at rest in both trials. However, the increase in the sum of four measured TCAI (>70% of the total TCAI pool) from rest to 15 min of exercise was not different between trials, and TCAI content was similar after 103 +/- 15 min of exercise (2.62 +/- 0.31 and 2.59 +/- 0.28 mmol/kg dry wt for LG and HG, respectively), which was the point of volitional fatigue during LG. Subjects cycled for an additional 52 +/- 9 min during HG, and although glycogen was markedly reduced (P < 0.05) during this period, no further change in the TCAI pool was observed, thus demonstrating a clear dissociation between exercise duration and the size of the TCAI pool. Neither the total adenine nucleotide pool (TAN = ATP + ADP + AMP) nor IMP was altered compared with rest in either trial, whereas creatine phosphate levels were not different when values measured at fatigue were compared with those measured after 15 min of exercise. These data demonstrate that altered glycogen availability neither compromises TCAI pool expansion nor affects the TAN pool or creatine phosphate or IMP content during prolonged exercise to fatigue. Therefore, our data do not support the concept that a decrease in muscle TCAI during prolonged exercise in humans compromises aerobic energy provision or is the cause of fatigue.

Development of civil aerospace composites is key to future “greener” aircraft. Aircraft manufacturers must improve efficiency of their product and manufacturing processes to remain viable. The aerospace industry is undergoing a materials revolution in the design and manufacture of composite airframes. The Airbus A350 and Boeing 787 (both due to enter service in the latter part of this decade) will push utilisation levels of  composite materials beyond 50% of the total airframe by weight. This  change requires massive investment in materials technology, manufacturing capability and skills development. The Quickstep process provides the ability to rapidly cure aerospace standard composite materials whilst providing enhanced mechanical properties. Utilising fluid to transfer heat to the   composite component during the curing process allows far higher heat rates than with conventional cure techniques. The rapid heat-up rates reduce the viscosity of the resin system greatly to provide a longer processing window introducing greater flexibility and removing the need for high pressure during cure. Interlaminar fracture toughness (Mode I) and Interfacial Shear Strength of aerospace standard materials cured using Quickstep have been    compared to autoclave cured laminates. Results suggest an improvement in fibre-matrix adhesion.

We present an input-output analysis of the life-cycle labor, land, and greenhouse gas (GHG) requirements of alternative options for three case studies: investing money in a new vehicle versus in repairs of an existing vehicle (labor), passenger transport modes for a trip between Sydney and Melbourne (land use), and renewable electricity generation (GHG emissions). These case studies were chosen to demonstrate the possibility of rank crossovers in life-cycle inventory (LCI) results as system boundaries are expanded and upstream production inputs are taken into account. They demonstrate that differential convergence can cause crossovers in the ranking of inventories for alternative functional units occurring at second-and higher-order upstream production layers.

Life-cycle assessment (LCA) is a method for evaluating the environmental impacts of products holistically, including direct and supply chain impacts. The current LCA methodologies and the standards by the International Organization for Standardization (ISO) impose practical difficulties for drawing system boundaries; decisions on inclusion or exclusion of processes in an analysis (the cutoff criteria) are typically not made on a scientific basis. In particular, the requirement of deciding which processes could be excluded from the inventory can be rather difficult to meet because many excluded processes have often never been assessed by the practitioner, and therefore, their negligibility cannot be guaranteed. LCA studies utilizing economic input−output analysis have shown that, in practice, excluded processes can contribute as much to the product system under study as included processes; thus, the subjective determination of the system boundary may lead to invalid results. System boundaries in LCA are discussed herein with particular attention to outlining hybrid approaches as methods for resolving the boundary selection problem in LCA. An input−output model can be used to describe at least a part of a product system, and an ISO-compatible system boundary selection procedure can be designed by applying hybrid input−output-assisted approaches. There are several hybrid input−output analysis-based LCA methods that can be implemented in practice for broadening system boundary and also for ISO compliance.

The fur seal is a mammal with an unusual ability to turn its milk production on and off without significantly altering the gross morphology of the mammary gland. This atypical lactation cycle is due to the fact that maternal foraging and infant nursing are spatially and temporally separate (Bonner, 1984). Maternal care involves the suckling of offspring over a period of at least 4 months, but lactation can extend to more than 12 months. Following a perinatal fast of approximately 1 week, females depart the breeding colony to forage at sea and, for the remainder of lactation, alternate between short periods ashore suckling their young with longer periods of up to 4 weeks foraging at sea. Whilst foraging at sea, milk production in the fur seal mammary gland either ceases or is reduced (Arnould & Boyd, 1995b).

During the period of 1990-2002 US households experienced a dramatic wealth cycle, induced by a 369% appreciation in the value of real per capita liquid stock market assets followed by a 55% decline. However, consumer spending in real terms continued to rise throughout this period. Using data from 1990-2005, traditional life-cycle approaches to estimating macroeconomic wealth effects confront two puzzles: (i) econometric evidence of a stable cointegrating relationship among consumption, income, and wealth is weak at best; and (ii) life-cycle models that rely on aggregate measures of wealth cannot explain why consumption did not collapse when the value of stock market assets declined so dramatically. We address both puzzles by decomposing wealth according to the liquidity of household assets. We find that the significant appreciation in the value of real estate assets that occurred after the peak of the wealth cycle helped sustain consumer spending from 2001 to 2005.

The objective of this research was to determine whether joint angles at critical gait events and during major energy generation/absorption phases of the gait cycle would reliably discriminate age-related degeneration during unobstructed walking. The gaits of 24 healthy adults (12 young and 12 elderly) were analysed using the PEAK Motus motion analysis system. The elderly participants showed significantly greater single (60.3% versus 62.3%, p < 0.01) and double ( p < 0.05) support times, reduced knee flexion (47.7° versus 43.0°, p < 0.05) and ankle plantarflexion (16.8° compared to 3.3°, p = 0.053) at toe off, reduced knee flexion during push-off and reduced ankle dorsiflexion (16.8° compared to 22.0°, p < 0.05) during the swing phase. The plantarflexing ankle joint motion during the stance to swing phase transition (A2) for the young group (31.3°) was about twice ( p < 0.05) that of the elderly (16.9°). Reduced knee extension range of motion suggests that the elderly favoured a flexed-knee gait to assist in weight acceptance. Reduced dorsiflexion by the elderly in the swing phase implies greater risk of toe contact with obstacles. Overall, the results suggest that joint angle measures at critical events/phases in the gait cycle provide a useful indication of age-related degeneration in the control of lower limb trajectories during unobstructed walking.

Work conducted in the Millard Biochemistry Research Laboratory examines the dual nature of molecules as carcinogens and anti-tumor agents through the molecular mechanisms of duplex DNA damage by bifunctional alkylating agents. Diepoxybutane (DEB) and epichlorohydrin (ECH) are polar molecules that form covalent DNA interstrand lesions by cross-linking the N7 position of deoxyguanosine residues. A recent experiment indicated that ECH preferentially targets nuclear DNA over mitochondrial DNA, whereas DEB shows similar rates of lesion formation for both loci. It was concluded that preferential targeting of nuclear DNA results from relatively poor uptake of ECH across the mitochondrial membrane. The objective of my honors research was to determine if the cytotoxicities of DEB and ECH vary according to the presence of the nuclear envelope in 6C2 chicken erythro-progenitor cells. The cytotoxicity of DEB and ECH was compared between cells randomly distributed throughout the cell cycle (Go/G, and S » G2/M) and cells enriched in G2/M stages. Results indicated that ECH is more cytotoxic than DEB in both unsynchronized control 6C2 cells and synchronized 6C2 cells enriched in G2/M stages of the cell cycle. Treatment with either bifunctional alkylating agent induced greater cytotoxicity in 6C2 cells enriched in G2/M stages than in unsynchronized control 6C2 cells, suggesting that the presence of the nuclear envelope-or any plasma membrane-may inhibit the reactivity of DEB and ECH.

This study describes the macrophyte assemblages of temporary floodplain wetlands situated on the floodplain of the Murray River, southeast Australia. Wetlands in the study are subject to flooding, the frequency, duration, and magnitude of which are dictated by the current, regulated river-flow regime. Our aim was to examine the influence of the existing flooding regime on macrophyte assemblages and to trial a monitoring program, based on a multiple before-after-control-impact (MBACI) design, to detect the impact of proposed environmental water allocations (EWAs) on the wetlands. Two categories of flooding regime were identified based on the flow magnitudes required for flooding to occur (flooding thresholds). In this scheme, wetlands with relatively low flooding thresholds are classed as ‘impact’ and those with higher thresholds are classed as ‘control.’ The wetlands were surveyed over a two-year period that incorporated at least one wetting-drying cycle at all wetlands. Results showed significant differences between survey times (season and year), but differences between flooding regime categories were significant only for some components of macrophyte assemblages. Differences between survey dates appear to reflect largely short-term responses to the most recent flood events. However, macrophyte differences observed between control and impact wetlands reflected the cumulative effect of flood events over several years. Differences between control and impact wetlands were strongest for post-flooding surveys based on full assemblages (using ANOSIM) and among specific taxa and functional groups (using ANOVA). Power to detect differences between control and impact wetlands was greatest for species richness and total abundance, but taxa with low variability among wetlands, and hence good power, were actually less sensitive to hydrologic change. We conclude that the MBACI design used in this study will be most effective in detecting wetland ecosystem responses to the implementation of EWAs if response variables are carefully chosen based on their sensitivity to hydrologic change.

In this paper we will draw on research conducted for the AFL which, in part, illuminated aspects of the Faustian pact that many young men enter into in order to become an elite level, professional footballer in what is increasingly a global sports entertainment industry. In order to develop an identity as an AFL footballer these young men willingly sell their body, mind and soul to one club, or to many. For varying lengths of time these pacts can have significant payoffs - in terms of a sense of self, and in monetary terms. For many though, these payoffs are limited and must be accounted for sometime in the future - an accounting that in Faustian terms, can carry significant costs to the body, mind and soul long after the cheering has stopped, and when the benefits come mainly in the form of memories. In this paper we argue that elements of these pacts can be identified and analysed via the following: understanding AFL as a sports entertainment business; using Foucault's work on the care of the self to explore what it means to be an elite level professional and the demands made by others on the body, mind and soul of players; and the idea that a career as an elite level professional footballer has a number of phases (early, mid and late) in which the nature of a professional identity - shaped by different demands on the body, mind and soul- changes.
