An uncommon disseminated Mycobacterium tuberculosis infection is described in a 12-year-old female dog presenting with fever, dyspnea, cough, weight loss, lymphadenopathy, melena, epistaxis, and emesis. The dog had a history of close contact with its owner, who died of pulmonary tuberculosis. Radiographic examination revealed diffuse radio-opaque images in both lung lobes, diffuse visible masses in abdominal organs, and hilar and mesenteric lymphadenopathy. Bronchial washing samples and feces were negative for acid-fast organisms. Polymerase chain reaction (PCR)-based species identification of bronchial washing samples, feces, and urine revealed M. tuberculosis using PCR-restriction enzyme pattern analysis-PRA. Because of public health concerns, which were worsened by the physical condition of the dog, euthanasia of the animal was recommended. Rough and tough colonies suggestive of M. tuberculosis were observed after microbiological culture of lung, liver, spleen, heart, and lymph node fragments in Löwenstein-Jensen and Stonebrink media. This study describes the comparison of three methods for genotyping of Mycobacterium tuberculosis, namely MIRU-VNTR (mycobacterial interspersed repetitive units-variable number of tandem repeats), spoligotyping and, for the first time, MLST (Multilocus Sequence Typing). In order to evaluate the discriminatory power of these methods, a total of 44 M. tuberculosis isolates obtained from sputum specimens of patients from Brazil were genotyped. Among the three methods, MLST showed the lowest discriminatory power compared to the other two techniques. MIRU-VNTR showed better discriminatory power when compared to spoligotyping, however, the combination of both methods provides the greatest level of discrimination and therefore this combination is the most useful genotyping tool to be applied to M. tuberculosis isolates. © 2013 Elsevier B.V.

Loss of response on repetitive drug exposure (i.e., tachyphylaxis) is a particular problem for the vasoconstrictor effects of medications containing oxymetazoline (OXY), an α1-adrenoceptor (AR) agonist of the imidazoline class. One cause of tachyphylaxis is receptor desensitization, usually accompanied by phosphorylation and internalization. It is well established that a1A-ARs are less phosphorylated, desensitized, and internalized on exposure to the phenethylamines norepinephrine (NE), epinephrine, or phenylephrine (PE) than are the a1B and a1D subtypes. However, here we show in human embryonic kidney-293 cells that the low-efficacy agonist OXY induces G protein-coupled receptor kinase 2-dependent a1A-AR phosphorylation, followed by rapid desensitization and internalization (∼40% internalization after 5 minutes of stimulation), whereas phosphorylation of α1A-ARs exposed to NE depends to a large extent on protein kinase C activity and is not followed by desensitization, and the receptors undergo delayed internalization (∼35% after 60 minutes of stimulation). Native α1A-ARs from rat tail artery and vas deferens are also desensitized by OXY, but not by NE or PE, indicating that thisproperty of OXY is not limited to recombinant receptors expressed in cell systems. The results of the present study are clearly indicative of agonist-directed a1A-AR regulation. OXY shows functional selectivity relative to NE and PE at a1A-ARs, leading to significant receptor desensitization and internalization, which is important in view of the therapeutic vasoconstrictor effects of this drug and the varied biologic process regulated by α1A-ARs. Background: Mycobacterium spp. is one of the most important species of zoonotic pathogens that can be transmitted from cattle to humans. The presence of these opportunistic, pathogenic bacteria in bovine milk has emerged as a public-health concern, especially among individuals who consume raw milk and related dairy products. To address this concern, the Brazilian control and eradication program focusing on bovine tuberculosis, was established in 2001. However, bovine tuberculosis continues to afflict approximately 1,3 percent of the cattle in Brazil. In the present study, 300 samples of milk from bovine herds, obtained from both individual and collective bulk tanks and informal points of sale, were cultured on Löwenstein-Jensen and Stonebrink media. Polymerase chain reaction (PCR)-based tests and restriction-enzyme pattern analysis were then performed on the colonies exhibiting phenotypes suggestive of Mycobacterium spp., which were characterized as acid-fast bacilli.Results: Of the 300 bovine milk samples that were processed, 24 were positively identified as Mycobacterium spp.Molecular identification detected 15 unique mycobacterial species: Mycobacterium bovis, M. gordonae, M. fortuitum, M. intracellulare, M. flavescens, M. duvalii, M. haemophilum, M. immunogenum, M. lentiflavum, M. mucogenicum, M. novocastrense, M. parafortuitum, M. smegmatis, M. terrae and M. vaccae. The isolation of bacteria from the various locations occurred in the following proportions: 9 percent of the individual bulk-tank samples, 7 percent of the collective bulk-tank samples and 8 percent of the informal-trade samples. No statistically significant difference was observed between the presence of Mycobacterium spp. in the three types of samples collected, the milk production profiles, the presence of veterinary assistance and the reported concerns about bovine tuberculosis prevention in the herds.Conclusion: The microbiological cultures associated with PCR-based identification tests are possible tools for the investigation of the presence of Mycobacterium spp. in milk samples. Using these methods, we found that the Brazilian population may be regularly exposed to mycobacteria by consuming raw bovine milk and related dairy products. These evidences reinforces the need to optimize quality programs of dairy products, to intensify the sanitary inspection of these products and the necessity of further studies on the presence of Mycobacterium spp. in milk and milk-based products. © 2013 Franco et al.; licensee BioMed Central Ltd.

The Amazonian brown brocket Mazama nemorivaga (Cuvier, 1817) is a small to medium-sized deer from the Amazon rainforest and ecotones. The first karyotype described was 2n=67 to 69 + 2-7 B and FN= 69-72, in which all chromosomes were acrocentric and the X chromosome was the only submetacentric chromosome. However, important aspects of the species chromosome evolution were not resolved because of the lack of information on chromosome banding. The G-banding pattern of M. nemorivaga karyotype showedthe presence of an XX/XY1Y2 sex chromosome system as a product of an X-autosome tandem fusion, which results in a basic 2n=68, FN=70 in females and 2n= 69, FN=70 in males. The fact that this karyotype only differs from that of Capreolus capreolus pygargus (Pallas, 1771; 2n=70, FN=72+B) by X-autosome tandem fusion may corroborate the basal condition of M. nemorivaga and its proximity to the ancestral karyotype of the American Odocoileini. A derived karyotype 2n=67, XY1Y2, FN=70 + 3B from the Brazilianstate of Mato Grosso (the western Amazon) may be evidence of differentiation between western and eastern populations. © Bruno Ferreto Fiorillo et al.

A multiyear solution of the SIRGAS-CON network was used to estimate the strain rates of the earth surface from the changing directions of the velocity vectors of 140 geodetic points located in the South American plate. The strain rate was determined by the finite element method using Delaunay triangulation points that formed sub-networks; each sub-network was considered a solid and homogeneous body. The results showed that strain rates vary along the South American plate and are more significant on the western portion of the plate, as expected, since this region is close to the subduction zone of the Nazca plate beneath the South American plate. After using Euler vectors to infer Nazca plate movement and to orient the velocity vectors of the South American plate, it was possible to estimate the convergence and accommodation rates of the Nazca and South American plates, respectively. Strain rate estimates permitted determination of predominant contraction and/or extension regions and to establish that contraction regions coincide with locations with most of the high magnitude seismic events. Some areas with extension and contraction strains were found to the east within the stable South American plate, which may result from different stresses associated with different geological characteristics. These results suggest that major movements detected on the surface near the Nazca plate occur in regions with more heterogeneous geological structures and multiple rupture events. Most seismic events in the South American plate are concentrated in areas with predominant contraction strain rates oriented northeast-southwest; significant amounts of elastic strain can be accumulated on geological structures away from the plate boundary faults; and, behavior of contractions and extensions is similar to what has been found in seismological studies. © 2013 Elsevier Ltd.
