The production and puriWcation of gilthead sea bream recombinant parathyroid hormone related protein [sbPTHrP(1–125)] using an Escherichia coli system and one step puriWcation process with continuous elution gel electrophoresis is reported. The cDNA encoding sbPTHrP(1–125) was cloned into a prokaryotic expression vector pET-11a. The recombinant plasmid was used to transfect E. coli BL21(DE3) pLysS and sbPTHrP(1–125) synthesis was induced by addition of 1mM isopropyl- -D-thiogalactopyranoside. The rapid one step isolation method gave pure sbPTHrP(1–125) as judged by SDS–PAGE and yielded up to 40mg/L of culture medium (3.3mg protein/g of bacteria). The bioactivity of recombinant sbPTHrP(1–125) assessed using an in vitro scale bioassay was found to be equipotent to PTHrP(1–34) in stimulating cAMP accumulation. Assessment of the immunological reactivity of the isolated protein by Western blot revealed it cross-reacts with antisera speciWc for the N-terminal and C-terminal region of PTHrP. In a radioimmunoassay speciWc for piscine N-terminal (1–34 aa) PTHrP, the recombinant sbPTHrP(1–125) was equipotent with PTHrP(1–34) in displacing labelled 125I-PTHrP(1–36) PTHrP from the antisera. The availability of recombinant sbPTHrP will allow the development of region speciWc assays and studies aimed at deWning post-secretory processing of this protein and its biological activity in Wsh.

Gonadotrophin-releasing hormone (GnRH) is the main neurohormone controlling gonadotrophin release in all vertebrates, and in teleost fish also of growth hormone and possibly of other adenohypophyseal hormones. Over 20 GnRHs have been identified in vertebrates and protochoordates and shown to bind cognate G-protein couple receptors (GnRHR). We have searched the puffer fish, Fugu rubripes, genome sequencing database, identified five GnRHR genes and proceeded to isolate the corresponding complementary DNAs in European sea bass, Dicentrachus labrax. Phylogenetic analysis clusters the European sea bass, puffer fish and all other vertebrate receptors into two main lineages corresponding to the mammalian type I and II receptors. The fish receptors could be subdivided in two GnRHR1 (A and B) and three GnRHR2 (A, B and C) subtypes. Amino acid sequence identity within receptor subtypes varies between 70 and 90% but only 50–55% among the two main lineages in fish. All European sea bass receptor mRNAs are expressed in the anterior and mid brain, and all but one are expressed in the pituitary gland. There is differential expression of the receptors in peripheral tissues related to reproduction (gonads), chemical senses (eye and olfactory epithelium) and osmoregulation (kidney and gill). This is the first report showing five GnRH receptors in a vertebrate species and the gene expression patterns support the concept that GnRH and GnRHRs play highly diverse functional roles in the regulation of cellular functions, besides the ‘‘classical’’ role of pituitary function regulation.

In this study we describe the isolation and characterisation of the parathyroid hormone-related protein (PTHrP) gene from the teleost Fugu rubripes. The gene has a relatively simple structure, compared with tetrapod PTHrP genes, composed of three exons and two introns, encompassing 2.25 kb of genomic DNA. The gene encodes a protein of 163 amino acids, with a putative signal peptide of 37 amino acids and a mature peptide of 126 amino acids. The overall homology with known tetrapod PTHrP proteins is low (36%), with a novel sequence inserted between positions 38 and 65, the absence of the conserved pentapeptide (TRSAW) and shortened C-terminal domain. The N-terminus shows greater conservation (62%), suggesting that it may have a hypercalcaemic function similar to that of tetrapod PTHrP. In situ localisation and RT–PCR have demonstrated the presence of PTHrP in a wide range of tissues with varying levels of expression. Sequence scanning of overlapping cosmids has identified three additional genes, TMPO, LDHB and KCNA1, which map to human chromosome 12, with the latter two mapping to 12p12-11.2. PTHrP in human also maps to this chromosome 12 sub-region, thus demonstrating conservation of synteny between human and Fugu.

A perturbação da personalidade anti-social, suscita particular interesse entre a comunidade técnica e cientifica porque são sujeitos agressivos que atentam contra si próprios e contra os que os rodeiam. Estudos recentes, revelam que indivíduos com esta perturbação apresentam, entre muitas outras características, um fraco entendimento de estímulos sociais e emocionais. Desta forma, investigadores dedicados ao estudo do processamento de emoções têm sugerido dificuldades destes sujeitos no reconhecimento, expressão e categorização de determinadas emoções, sobretudo no que se refere a emoções negativas. Esta incapacidade em decifrar signos emocionais promove uma fraca cognição social e uma fraca adesão às normas sociais. Dada a pertinência do tema, tivemos como principal interesse analisar de que forma sujeitos com perturbação anti-social da personalidade categorizam estados emocionais básicos e sociais e posteriormente compará-los com um grupo de sujeitos saudáveis. Para o efeito, foram avaliados 15 sujeitos com perturbação anti-social da personalidade, seleccionados a partir de uma base de dados do CAT de Olhão e um grupo de controlo constituído por 15 sujeitos saudáveis recolhidos ao acaso por conveniência. Foram avaliadas as características de personalidade e sintomatologia através do MCMI-III e aplicada uma prova de classificação de cartões “Card Sorting”. Os resultados sugerem que os sujeitos pertencentes ao grupo clinico têm maior dificuldade em categorizar emoções básicas e sociais, comparativamente ao grupo de controlo, sobretudo no que se refere a emoções com valência positiva. Os resultados serão discutidos à luz da literatura atual.

Os sistemas de veiculação de fármacos, presentemente, apresentam-se como uma alternativa viável às terapias convencionais. De entre os diversos sistemas de transporte passíveis de associar substâncias farmacologicamente activas, destacam-se os de base lipídica, em particular, os lipossomas, os quais constituem um dos sistemas mais estudados e com maior sucesso, comprovado pelo número de produtos em fase clínica ou já aprovados para utilização em humanos. Os lipossomas são estruturas constituídas por membranas lipídicas organizadas em bicamadas, fechadas e concêntricas, que permitem a encapsulação de moléculas hidrofílicas no espaço interno aquoso e hidrofóbicas na bicamada lipídica. No presente trabalho, foram desenvolvidas metodologias com vista à encapsulação em lipossomas do aminoglicosídeo, a Paromomicina (PRM). Este fármaco está indicado para o tratamento de doenças infecciosas nomeadamente parasitárias e bacterianas. Algumas das principais desvantagens resultantes da sua utilização em clínica são, o reduzido tempo de circulação na corrente sanguínea, rápida excreção renal e consequentemente insuficiente concentração intracelular do fármaco. Como forma de ultrapassar algumas das desvantagens apresentadas, foram desenvolvidas formulações lipossomais de PRM com vista a melhorar o desempenho deste antibiótico. Para tal, foram preparadas e caracterizadas diversas formulações lipossomais de PRM com vista à selecção daquelas que apresentem maiores valores de eficácia de encapsulação (E.E.), e superior estabilidade. Com as formulações seleccionadas foram realizados estudos in vitro de interacção lipossoma-célula, utilizando uma linha celular humana monocítica leucémica, THP-1. De entre as formulações desenvolvidas e seleccionadas para os estudos in vitro de a formulação DPPC:DPPG, foi uma das que apresentou uma E.E. superior a 80% e valores de internalização celular superiores a 90%.

This article examines education reform under the first government of Northern Ireland (1921–5). This embryonic period offered the Ulster Unionist leadership a chance to construct a more inclusive society, one that might diminish sectarian animosities, and thereby secure the fledgling state through cooperation rather than coercion. Such aspirations were severely tested by the ruling party’s need to secure the state against insurgency, and, more lastingly, to assuage the concerns of its historic constituency. The former led to a draconian security policy, the latter to a dependency on populist strategies and rhetoric. It is argued here, however, that this dependency was not absolute until July 1925. Before that, the Belfast government withstood growing pressure from populist agitators to reverse controversial aspects of its education reforms, only relenting when Protestant disaffection threatened the unity of the governing party and the existence of the state.

Objectives: This paper reports on the acceptability and effectiveness of the FITS (Focussed Intervention Training and Support) into Practice Programme. This intervention was scaled up from an earlier cluster randomised-controlled trial that had proven successful in significantly decreasing antipsychotic prescribing in care homes. Method: An in depth 10-day education course in person-centred care was delivered over a three-month period, followed by six supervision sessions. Participants were care-home staff designated as Dementia Care Coaches (DCCs) responsible for implementing interventions in 1 or 2 care homes. The course and supervision was provided by educators called Dementia Practice Development Coaches (DPDCs). Effectiveness data included monitoring antipsychotic prescriptions, goal attainment, knowledge, attitudes and implementation questionnaires. Qualitative data included case studies and reflective journals to elucidate issues of implementation. Results: Of the 100 DCCs recruited, 66 DCCs completed the programme. Pre-post questionnaires demonstrated increased knowledge and confidence and improved attitudes to dementia. Twenty per cent of residents were prescribed antipsychotics at baseline which reduced to 14% (31% reduction) with additional dose reductions being reported alongside improved personalised goal attainment. Crucial for FITS into Practice to succeed was the allocation and protection of time for the DCC to attend training and supervision and to carry out implementation tasks in addition to their existing job role. Evaluation data showed that this was a substantial barrier to implementation in a small number of homes. Discussion and conclusions: The FITS into practice programme was well evaluated and resulted in reduction in inappropriate anti-psychotic prescribing. Revisions to the intervention are suggested to maximise successful implementation.
