研究了Nd(naph)_3-Al(i-Bu)_2H-t-BuCl(或allyl-Cl)催化体系制得的顺丁橡胶(BR)的结构与性能。结果表明,[η]=2.7~3.7dL/g,分子量分散指数小于4,[η]<1低分子级分含量小于50%,顺式-1,4含量大于或等于95%时,其生胶、混炼胶和硫化胶的性能均良好,硫化胶的综合性能与一般稀土体系BR和镍系BR相当甚至超过。

提出了一类以五取代茂基稀土金属配合物与活性碱金属氢化物组成的加氢催化体系。实验表明:(C_5Me_4R)_2LnCl_2Li(THF)_2+MH(Ln=Nd,Yb,R=Pr;Ln=Gd,R=Me,Pr;M=K,Na)体系都能催化α-烯烃加氢,其活性较高,如(C_5Me_5)_2GdCl_2Li-NaH体系对1-己烯加氢TON可达40min~(-1)。碱金属氢化物的种类、THF以及温度等对其催化活性均有不同的影响。这类催化剂失活很快,其原因与反应体系中的烯烃有关。

本工作发现,用DSC测定从熔体等速降温时辐照样品的结晶温度T_c随辐照剂量R增加而线性变低,即辐照前后样品的△T_c=T_c_o-T_c_R=KR。依据Charlesby-Pinner方程,导出一新关系式,S+S~(1/2)=A+B/△T_c。已经证明,式中的T_c只与聚合物的交联度有关,与辐射交联方法和过程无关。因此,该方程可用于结晶聚合物的强化辐射交联,以获得非无规交联的瞬时G(c,1)值。本文为定量研究结晶聚合物的无规交联与非无规交联提供了一个简便的方法。

本文研究了端乙烯基聚氨酯预聚物(VTPU)同乙烯类单体甲基丙烯酸甲酯(MMA)、苯乙烯(St)、醋酸乙烯酯(VAc)、和甲基丙烯酸丁酯(BMA)聚合得到的双组份网状共聚物(BCN′s)的动态力学性能及形态结构。四种BCN′s试样两组份在动态力学谱上均有半相容的特征。体系的交联密度增大,混合熵增加,相容性改善,试样中可溶性组份含量随BCN′s的组成而变化。溶胶组份可改善体系中两组份的相容性。

Tumor necrosis factor receptor-associated factor 6 (TRAF6), a key signaling adaptor molecule common to the TNFR superfamily and IL-IR/TLR family, is important not only for a diverse array of physiological processes functions of the TNFR superfamily, but also is involved in adaptive immunity and innate immunity. In this report, the first bivalve TRAF6 (named as CfTRAF6) gene is identified and characterized from Zhikong scallop Chlamys farreri. The full-length cDNA of CfTRAF6 is of 2510 bp, consisting of a 5'-terminal untranslated region (UTR) of 337 bp, a 3'-terminal UTR of 208 bp with a canonical polyadenylation signal sequence AATAAA and a poly (A) tail, and an open reading frame (ORF) encoding a polypeptide of 655 amino acids. The predicted amino acid sequence of CfTRAF6 comprises characteristic motifs of the TRAF proteins, including a Zinc finger of RING-type, two Zinc fingers of TRAF-type, a coiled-coil region, and a MATH (the meprin and TRAF homology) domain. The overall amino acid sequence identity between CfTRAF6 and other TRAF6s is 28-68%. Phylogenetic analyses of CfTRAF6 sequence with TRAF sequences from other organisms indicate that CfTRAF6 is a true TRAF6 orthologue. The mRNA expression of CfTRAF6 in various tissues is measured by Real-time RT-PCR. The mRNA transcripts are constitutively expressed in tissues of haemocyte, muscle, mantle, heart, gonad and gill, but the highest expression is observed in the gonad. The temporal expressions of CfTRAF6 mRNA in the mixed primary cultured haemocytes are recorded after treatment with 20 mu g mL(-1) and 0.5 mu g mL(-1) peptido-glycan (PGN). The expression level of CfTRAF mRNA is down-regulated from 1.5 h to 3 h after the treatment with 0.5 mu g mL(-1) PGN, and then recovers to the original level. While the expression of CfTRAF6 is obviously decreased after treatment with 20 mu g mL(-1) PGN, and reach the lowest point (only about 1/9 times to control) at 3 h. Myeloid differentiation factor 88 (MyD88) is a universal and essential adapter for the TLR/IL-1R family. In this report, the first mollusk Myd88 ortholog (named as CfMyd88) was cloned from Zhikong scallop (Chlamys farreri). The full-length cDNA of CfMyd88 was of 1554 bp, including a 5 '-terminal untranslated region (UTR) of 427 bp, a polyA tail, and an open reading frame (ORF) of 1104 bp encoding a polypeptide of 367 amino acids containing the typical TLR and IL-1R-related (TIR) domain and death domain (DD). Homology analysis revealed that the predicted amino acid sequence of CfMyd88 was homologous to a variety of previously identified Myd88s with more than 30% identity. The temporal expressions of CfMyd88 mRNA in the mixed primary cultured haemocytes stimulated by lipopolysaccharide (LPS) and peptidoglycans (PGN) were measured by real-time RT-PCR system. The mRNA expression of CfMyd88 decreased after stimulation with both LPS and PGN, and the lowest level was about 1/3 times (at 6 h) and 1/10 times (at 9 h) to that in the control group, respectively. The expression then recovered and was upregulated to two-fold at 9 h after LPS stimulation or to the original level at 12 It after PGN stimulation. The unicellular green alga Haematococcus pluvialis accumulates a commercially valuable astaxanthin, with levels reaching up to 4% dry weight under environmental stress. In recent years, much effort has been devoted to understanding the molecular mechanisms regulating astaxanthin biosynthetic pathways. Beta-carotene ketolase (bkt), with control being exhibited at the transcription level, plays an important role in astaxanthin biosynthesis by H. pluvialis. Here we demonstrate the presence of two separate 5'-flanking regions [1.5 kilobase (kb) and 2 kb] of bkt (bkt1 and bkt2) that possess regulatory elements similar to those of known stress-responsive genes in plants. Results of 5'-deletion constructs and transient beta-galactosidase expression assays demonstrate that there may be positive regulatory elements governing expression in the shorter promoter at -1060/-900 from the 1.5 kb 5' region, and in the longer promoter at -1838/-1219 and at -1046/ -734 from the 2 kb 5' region relative to each homologous ATG start codon. Furthermore, our present studies reveal that the first intron (+371/+497) downstream from the 1.5 kb 5' untranslated region of bkt1 may function as a negative regulatory element to regulate its own promoter.

A large number of polymorphic simple sequence repeats (SSRs) or microsatellites are needed to develop a genetic map for shrimp. However, developing an SSR map is very time-consuming, expensive, and most SSRs are not specifically linked to gene loci of immediate interest. We report here on our strategy to develop polymorphic markers using expressed sequence tags (ESTs) by designing primers flanking single or multiple SSRs with three or more repeats. A subtracted cDNA library was prepared using RNA from specific pathogen-free (SPF) Litopenaeus vannamei juveniles (similar to 1 g) collected before (0) and after (48 h) inoculation with the China isolate of white spot syndrome virus (WSSV). A total of 224 clones were sequenced, 194 of which were useful for homology comparisons against annotated genes in NCBI nonredundant (nr) and protein databases, providing 179 sequences encoded by nuclear DNA, 4 mitochondrial DNA, and 11 were similar to portions of WSSV genome. The nuclear sequences clustered in 43 groups, 11 of which were homologous to various ESTs of unknown function, 4 had no homology to any sequence, and 28 showed similarities to known genes of invertebrates and vertebrates, representatives of cellular metabolic processes such as calcium ion balance, cytoskeleton mRNAs, and protein synthesis. A few sequences were homologous to immune system-related (allergens) genes and two were similar to motifs of the sex-lethal gene of Drosophila. A large number of EST sequences were similar to domains of the EF-hand superfamily (Ca2+ binding motif and FRQ protein domain of myosin light chains). Single or multiple SSRs with three or more repeats were found in approximately 61 % of the 179 nuclear sequences. Primer sets were designed from 28 sequences representing 19 known or putative genes and tested for polymorphism (EST-SSR marker) in a small test panel containing 16 individuals. Ten (53%) of the 19 putative or unknown function genes were polymorphic, 4 monomorphic, and 3 either failed to satisfactorily amplify genomic DNA or the allele amplification conditions need to be further optimized. Five polymorphic ESTs were genotyped with the entire reference mapping family, two of them (actin, accession #CX535973 and shrimp allergen arginine kinase, accession #CX535999) did not amplify with all offspring of the IRMF panel suggesting presence of null alleles, and three of them amplified in most of the IRM F offspring and were used for linkage analysis. EF-hand motif of myosin light chain (accession #CX535935) was placed in ShrimpMap's linkage group 7, whereas ribosomal protein S5 (accession #CX535957) and troponin I (accession #CX535976) remained unassigned. Results indicate that (a) a large number of ESTs isolated from this cDNA library are similar to cytoskeleton mRNAs and may reflect a normal pathway of the cellular response after im infection with WSSV, and (b) primers flanking single or multiple SSRs with three or more repeats from shrimp ESTs could be an efficient approach to develop polymorphic markers useful for linkage mapping. Work is underway to map additional SSR-containing ESTs from this and other cDNA libraries as a plausible strategy to increase marker density in ShrimpMap.

Elemental sulfur and hydrogen sulfide emitted offshore of northeastern Taiwan known to local fishermen for generations, but never studied until recently, are found to have originated from a cluster of shallow (< 30 m depth) hydrothermal vents. Among the mounds is a massive 6 m high chimney with a diameter of 4 m at the base composed of almost pure sulfur and discharging hydrothermal fluid containing sulfur particles. The sulfur in the chimney has a delta(34)S= 1.1 parts per thousand that is isotopically lighter than seawater. A yellow smoker at shallow depths with such characteristics has never been reported on anywhere else in the world. Gas discharges from these vents are dominated by CO2 (> 92%) with small amounts of H2S. Helium isotopic ratios 7.5 times that of air indicate that these gases originate from the mantle. High temperature hydrothermal fluids have measured temperatures of 78-116 degrees C and pH (25 degrees C) values as low as 1.52, likely the lowest to be found in world records. Low temperature vents (30-65 degrees C) have higher pH values. Continuous temperature records from one vent show a close correlation with diumal tides, suggesting rapid circulation of the hydrothermal fluids. (c) 2005 Elsevier B.V. All rights reserved.

Numerous cores and dating show the Yangtze River has accumulated about 1.16 x 10(12) t sediment in its delta plain and proximal subaqueous delta during Holocene. High-resolution seismic profiling and coring in the southern East China Sea during 2003 and 2004 cruises has revealed an elongated (similar to 800 km) distal subaqueous mud wedge extending from the Yangtze River mouth southward off the Zhejiang and Fujian coasts into the Taiwan Strait. Overlying what appears to be a transgressive sand layer, this distal clinoform thins offshore, from similar to 40 in thickness between the 20 and 30 m water depth to < 1-2 in between 60 and 90 m water depth, corresponding to an across shelf distance of less than 100 km. Total volume of this distal mud wedge is about 4.5 x 10(11) m(3), equivalent to similar to 5.4 x 10(11) t of sediment. Most of the sediment in this mud wedge comes from the Yangtze River, with some input presumably coming from local smaller rivers. Thus, the total Yangtze-derived sediments accumulated in its deltaic system and East China Sea inner shelf have amounted to about 1.7 x 10(12) t. Preliminary analyses suggest this longshore and across-shelf transported clinoform mainly formed in the past 7000 yrs after postglacial sea level reached its mid-Holocene highstand, and after re-intensification of the Chinese longshore current system. Sedimentation accumulation apparently increased around 2000 yrs BP, reflecting the evolution of the Yangtze estuary and increased land erosion due to human activities, such as fanning and deforestation. The southward-flowing China Coastal Current, the northward-flowing Taiwan Warm Current, and the Kuroshio Current appear to have played critical roles in transporting and trapping most of Yangtze-derived materials in the inner shelf, and hence preventing the sediment escape into the deep ocean. 本文研究了胶州湾内1988年5月至1989年5月,小拟哲水蚤[Paracalanus parvus(Claus)]、双刺纺锤水蚤[Acartia bifilosa (Giesbrecht)]、太平洋纺锤水蚤(A. pacifica steuer)和1982年5月至1983年4月中华哲水蚤(Calanus sinicus Brodsky)的种群结构和种群数量的季节变化。双剌纺锤水蚤和太平纺锤水蚤在胶州湾是季节性出现的种类,它们能产生休眠卵,本文对其休眠卵数量的季节变化以及影响休眠卵产生和孵化的因素进行了初步的探讨。本文还研究了体长的季节变化以及温度和食物条件对体长的影响。

扇贝是我国海水养殖的重要品种,但自1994年以来,养殖扇贝陆续爆发的大规模死亡,不但造成了巨大的经济损失,而且直接威胁到现有产业的生存和发展。引起扇贝大规模死亡原因是多方面的,其主要原因是养殖环境恶化、扇贝种质衰退和抗病力下降。因此,深入研究扇贝免疫防御机制,探讨提高机体抗病力的有效途径和方法,改良种质和培育抗病品系,无疑是解决目前困扰扇贝养殖业健康可持续发展的必经之路。 Toll样受体(TLRs)家族是新近发现的模式识别受体(PRRs),参与识别病原体相关的分子模式(PAMPs),在天然免疫系统中起着非常重要的作用。哺乳动物中Toll样受体信号通路还参与诱导树枝状细胞成熟、参与免疫耐受、参与凋亡发生发展、介导非感染性因素的识别等,被视为联系天然免疫和获得性免疫的桥梁。同时果蝇的Toll信号通路也是不具备获得性免疫的果蝇赖以抵御病毒、细菌和真菌感染,介导天然免疫反应的重要信号通路。 本研究采用大规模EST测序方法,结合Genome Walker库的构建和cDNA末端快速扩增技术,从栉孔扇贝克隆得到CfToll-1、CfMyd88、CfTRAF6和CfCactus这四个Toll样受体信号通路基因的全长cDNA,同时用荧光实时定量PCR技术检测了这些基因的组织分布及在脂多糖(LPS)和肽聚糖(PGN)刺激下的表达规律。 栉孔扇贝Toll样受体(CfToll-1)的cDNA序列全长4308 bp,包含5’非翻译区(UTR)211 bp,3597 bp的开放阅读框,500 bp的3’UTR,最后为18个腺嘌呤的ploy A 尾巴。开放阅读框编码1198个氨基酸的多肽,该多肽的估计分子量为137.41kd,估计的等电点为5.62,该多肽有信号肽,具有一个预测的跨膜区,因此是一种跨膜蛋白。经BLAST比对,CfToll-1基因与节肢动物多种Toll蛋白高度的相似性。SMART(Simple Modular Architecture Research Tool)软件分析,CfToll-1包含典型的Toll样受体的结构:富含亮氨酸的重复序列的胞外区(leucine-rich repeats, LRR),一段跨膜结构域,以及胞内区的TIR结构域(Toll/IL-1 receptor homologous region)。利用Real-time RT-PCR发现CfToll-1mRNA在扇贝体内普遍存在于血细胞、肌肉、外套膜、心、性腺和鳃组织中。利用体外培养的原代血细胞系研究不同浓度LPS刺激后CfToll-1的表达变化,结果显示低剂量(100ng.mL-1 )LPS 使CfToll-1 mRNA表达量减小,该变化在1.5h、3h 和9h组差异显著,虽然在6h组表达量稍有恢复,但尚未达到对照水平;用1μg.mL-1LPS处理细胞时, 6h组CfToll-1表达量明显上调,约为对照水平的2倍。证实细菌结构脂多糖对CfToll-1基因的表达有影响,且这种影响有剂量依赖效应。 栉孔扇贝Myd88同源基因(CfMyd88)的cDNA序列全长1554bp,包含5’UTR 427 bp,1101bp的开放阅读框,最后为18个腺嘌呤的ploy A 尾。CfMyd88的开放阅读框可编码367个氨基酸的多肽,该多肽的估计分子量为42.37kD,估计的等电点为5.71。利用SMART程序分析发现CfMyd88编码了Death和TIR结构域, 这两个结构域是Myd88特征结构。BLAST程序发现扇贝的序列与数据库哺乳动物的Myd88基因高度同源。原代培养的扇贝血细胞在受到PGN刺激后,CfMyd88 mRNA表达在1.5小时开始下调,直到9小时下调至对照表达量的1/10,证实肽聚糖结构对CfMyd88基因的表达有影响。 栉孔扇贝TRAF6同源基因(CfTRAF6)的cDNA序列全长2510bp,包含5’UTR 337 bp,1965bp的开放阅读框,3’UTR 208bp,最后为21 个腺嘌呤的ploy A 尾巴。CfTRAF6开放阅读框编码655个氨基酸的多肽,该多肽的估计分子量为74.09kD,估计的等电点为6.01。InterPro Scan在线分析发现CfTRAF6有典型的TRAF蛋白家族的特征结构,包括的一个指环结构,两个锌指结构,一个MATH (the meprin and TRAF homology)结构域以及Coiled-coil区域。CfTRAF6的序列与数据库多物种的TRAF6高度同源,同源性最高的是乌贼序列(Identity=68)和鼠类(Identity=45%)。利用Real-time RT-PCR,发现CfTRAF6在各组织普遍存在,在性腺中的表达最高。原代培养的扇贝血细胞在受到不同浓度PGN刺激后,与CfMyd88的情况一样,CfTRAF6的表达量变化减少,且这种变化随剂量的增加更加明显。 栉孔扇贝Cactus同源基因(CfCactus)的cDNA序列全长2488bp,包含5’UTR 181 bp,840bp的开放阅读框, 3’UTR 1467bp,最后为19个腺嘌呤的ploy A 尾巴。CfCactus的开放阅读框编码279个氨基酸的多肽,该多肽的估计分子量为31.37 kD;估计的等电点为4.74,与果蝇的Cactus基因的等电点相近(4.5)。利用SMART程序分析发现CfCactus主要编码了ANK结构域(ankyrin repeats)。Cactus基因为哺乳动物NF-κB抑制蛋白IκB的同源分子,BLAST 程序发现扇贝的序列与数据库多物种的Cactus或IκB基因高度同源。同源性最高的是太平洋牡蛎(Identity=35%)和圆尾鲎(Identities = 44%)。对CfTCactus mRNA在扇贝的血细胞、性腺、 肠的组织表达进行分析,并同时与CfTRAF6和CfMyd88的表达量进行了对比,发现CfCactus的表达水平明显高于这两个基因,而且CfTRAF6的基因表达量也高于CfMyd88,表现出级联放大效应。正常情况下,三个基因在性腺的表达量最高,推测这条通路可能和发育等功能密切相关。 通过本研究我们首次在双壳类软体动物找得到与果蝇Toll蛋白家族高度同源的CfToll-1基因,同时发现其他三个在Toll样受体信号传递过程中起重要作用的基因,其中包括在软体动物中获得的第一个Toll样受体的接头分子-CfMyd88基因,该结果直接证明软体动物具有与哺乳动物和节肢动物高度类似Myd88依赖的Toll样受体信号通路。同时通过这些基因组织分布的研究以及细菌结构LPS和PGN对这条通路上基因表达的影响,证明扇贝Toll信号通路可能与在果蝇中一样,参与扇贝的发育和免疫防御等多种功能。
