A general MHC-heterozygote advantage in parasite-infected organisms is often assumed, although there is little experimental evidence for this. We tested the response of MHC-congenic mice (F2 segregants) to malaria and found the course of infection to be significantly influenced by MHC haplotype, parasite strain, and host gender. However, the MHC heterozygotes did worse than expected from the average response of the homozygotes.

Methods Ten patients with aniridia from 3 families of Egyptian origin underwent full ophthalmologic, general and neurological examination, and blood drawing. Cerebral MRI was performed in the index case of each family. Genomic DNA was prepared from venous leukocytes and direct sequencing of all the exons and intron-exon junctions of the PAX6 gene was performed after PCR amplification. Results Common features observed in the three families included absence of iris tissue, corneal pannus with different degrees of severity and foveal hypoplasia with severely reduced visual acuity. In families 2 and 3, additional findings such as lens dislocation, lens opacities or polar cataract and glaucoma were observed. We identified two novel (c.170-174delTGGGC [p.L57fs17] and c.475delC [p.R159fs47]) and one known (c.718C>T) PAX6 mutations in the affected members of the 3 families. Systemic and neurological examination was normal in all ten affected patients. Cerebral MRI showed absence of the pineal gland in all three index patients. Severe hypoplasia of the brain anterior commissure was associated to the p.L57fs17mutation, absence of the posterior commissure to both p.R159fs47 and p.R240X, and optic chiasma atrophy and almost complete agenesis of the corpus callosum to p.R240X. Conclusions We identified two novel PAX6 mutations in families with severe aniridia from Northern Egypt, an ethnic group which is not well studied. In addition to common phenotype of aniridia and despite normal neurological examination, absence of the pineal gland was observed in all 3 index patients. The heterogeneity of brain anomalies related to PAX6 mutations is underexplored and is highlighted in this study.

A few bacterial species are known to produce and excrete hydrogen cyanide (HCN), a potent inhibitor of cytochrome c oxidase and several other metalloenzymes. In the producer strains, HCN does not appear to have a role in primary metabolism and is generally considered a secondary metabolite. HCN synthase of proteobacteria (especially fluorescent pseudomonads) is a membrane-bound flavoenzyme that oxidizes glycine, producing HCN and CO2. The hcnABC structural genes of Pseudomonas fluorescens and P. aeruginosa have sequence similarities with genes encoding various amino acid dehydrogenases/oxidases, in particular with nopaline oxidase of Agrobacterium tumefaciens. Induction of the hcn genes of P. fluorescens by oxygen limitation requires the FNR-like transcriptional regulator ANR, an ANR recognition sequence in the -40 region of the hcn promoter, and nonlimiting amounts of iron. In addition, expression of the hcn genes depends on a regulatory cascade initiated by the GacS/GacA (global control) two-component system. This regulation, which is typical of secondary metabolism, manifests itself during the transition from exponential to stationary growth phase. Cyanide produced by P. fluorescens strain CHA0 has an ecological role in that this metabolite accounts for part of the biocontrol capacity of strain CHA0, which suppresses fungal diseases on plant roots. Cyanide can also be a ligand of hydrogenases in some anaerobic bacteria that have not been described as cyanogenic. However, in this case, as well as in other situations, the physiological function of cyanide is unknown.

BACKGROUND/OBJECTIVES: To assess the distribution of interleukin (IL)-1β, IL-6, tumour necrosis factor (TNF)-α and C-reactive protein (CRP) according to the different definitions of metabolically healthy obesity (MHO). SUBJECTS/METHODS: A total of 881 obese (body mass index (BMI) > or =30 kg/m2) subjects derived from the population-based CoLaus Study participated in this study. MHO was defined using six sets of criteria including different combinations of waist, blood pressure, total high-density lipoprotein cholesterol or low-density lipoprotein -cholesterol, triglycerides, fasting glucose, homeostasis model, high-sensitivity CRP, and personal history of cardiovascular, respiratory or metabolic diseases. IL-1β, IL-6 and TNF-α were assessed by multiplexed flow cytometric assay. CRP was assessed by immunoassay. RESULTS: On bivariate analysis some, but not all, definitions of MHO led to significantly lower levels of IL-6, TNF-α and CRP compared with non-MH obese subjects. Most of these differences became nonsignificant after multivariate analysis. An posteriori analysis showed a statistical power between 9 and 79%, depending on the inflammatory biomarker and MHO definition considered. Further increasing sample size to overweight+obese individuals (BMI > or =25 kg/m2, n=2917) showed metabolically healthy status to be significantly associated with lower levels of CRP, while no association was found for IL-1β. Significantly lower IL-6 and TNF-α levels were also found with some but not all MHO definitions, the differences in IL-6 becoming nonsignificant after adjusting for abdominal obesity or percent body fat. CONCLUSIONS: MHO individuals present with decreased levels of CRP and, depending on MHO definition, also with decreased levels in IL-6 and TNF-α. Conversely, no association with IL-1β levels was found.

A aplicação de gesso para tornar o Al indisponível, assim como para suprir nutrientes do solo para as plantas, pode proporcionar condições para a obtenção de elevada produtividade das culturas. Este trabalho teve como objetivo avaliar o efeito do gesso (0, 1.000, 2.000, 3.000, 4.000 e 5.000 kg ha-1) nas características químicas de duas áreas com Latossolo Vermelho eutroférrico de textura argilosa, com ou sem Al trocável, e na nutrição mineral e produtividade das culturas de trigo e soja em sistema semeadura direta. As doses de gesso foram aplicadas em superfície seis meses após a calagem. Aos seis e 12 meses após a aplicação do gesso, foram coletadas amostras de solo nas profundidades de 0-0,10; 0,10-0,20 e 0,20-0,40 m. O uso de até 5.000 kg ha-1 de gesso resultou em aumento dos teores de Ca2+ e SO4(2-) e lixiviação de K+ até 0,20 m e Mg2+ até 0,40 m de profundidade, um ano após a aplicação, sendo a dose de 3.000 kg ha-1 a que proporcionou maior redução do Al trocável. A aplicação de gesso implicou aumento dos teores foliares de Ca e Mg na cultura da soja, redução dos teores de Ca e Mg na cultura do trigo e aumento no teor de S em ambas as culturas, enquanto o teor de K aumentou até a dose de 3.170 kg ha-1 de gesso na cultura da soja. A utilização do gesso eleva a produtividade do trigo, cultivar CD 104, em solo com presença de Al trocável, mas não influencia a produtividade da soja em ambas os solos, com ou sem Al.

Antemortem demonstration of ischemia has proved elusive in head injury because regional CBF reductions may represent hypoperfusion appropriately coupled to hypometabolism. Fifteen patients underwent positron emission tomography within 24 hours of head injury to map cerebral blood flow (CBF), cerebral oxygen metabolism (CMRO2), and oxygen extraction fraction (OEF). We estimated the volume of ischemic brain (IBV) and used the standard deviation of the OEF distribution to estimate the efficiency of coupling between CBF and CMRO2. The IBV in patients was significantly higher than controls (67 +/- 69 vs. 2 +/- 3 mL; P < 0.01). The coexistence of relative ischemia and hyperemia in some patients implies mismatching of perfusion to oxygen use. Whereas the saturation of jugular bulb blood (SjO2) correlated with the IBV (r = 0.8, P < 0.01), SjO2 values of 50% were only achieved at an IBV of 170 +/- 63 mL (mean +/- 95% CI), which equates to 13 +/- 5% of the brain. Increases in IBV correlated with a poor Glasgow Outcome Score 6 months after injury (rho = -0.6, P < 0.05). These results suggest significant ischemia within the first day after head injury. The ischemic burden represented by this "traumatic penumbra" is poorly detected by bedside clinical monitors and has significant associations with outcome.

Several high-quality publications were published in 2013 and some major trials studies were started. In Guillain-Barré syndrome, events included the launch of IGOS and a better understanding of diagnostic limits, the effect of influenza vaccination, and better care, but uncertainty remains about analgesics. A new mouse model was also described. In chronic inflammatory demyelinating polyradiculoneuropathy (CIDP), diagnostic pitfalls can be recalled. Our knowledge of underlying pathophysiological processes has improved, and the value of monitoring with function and deficit scores has been demonstrated. IVIG can sometimes be effective longer than expected, but CIDP remains sensitive to corticosteroids, particularly with the long-term beneficial effects of megadose dexamethasone. The impact of fingolimod remains to be demonstrated in an ongoing trial. Advances concerning multifocal motor neuropathy, inflammatory plexopathy, and neuropathy with anti -MAG activity are discussed but treatments already recognized as effective should not be changed. O plantio direto de qualidade depende de um manejo adequado do solo que promova melhorias em sua estrutura. Isso está associado ao sistema de culturas adotado. O objetivo deste trabalho foi avaliar a contribuição de sistemas de culturas de longo prazo (18 anos) na qualidade estrutural de um Latossolo Vermelho mesoférrico argiloso sob plantio direto nos Campos Gerais do Paraná. Foram avaliados cinco sistemas de culturas: trigo-soja [Tr-So]; aveia-milho-trigo-soja [Av-Mi-Tr-So]; ervilhaca-milho-trigo-soja [Er-Mi-Tr-So]; azevém-milho-azevém-soja [Az-Mi-Az-So]; e alfafa-milho [Alf-Mi]. Amostras de solo foram coletadas nas camadas de 0-5, 5-10 e 10-20 cm, em anéis volumétricos e em blocos com estrutura preservada. Na camada de 0-5 cm, as menores densidades de solo tenderam a ocorrer nos sistemas Av-Mi-Tr-So (0,96 Mg m-3) e Er-Mi-Tr-So (0,93 Mg m-3). Nas camadas de 5-10 e 10-20 cm, as menores densidades de solo ocorreram no sistema Alf-Mi (1,14 e 1,17 Mg m-3, respectivamente). Tendência coerente foi observada para a macroporosidade, que na camada superficial foi maior nos sistemas Av-Mi-Tr-So (0,29 m³ m-3) e Er-Mi-Tr-So (0,30 m³ m-3) e, nas camadas de 5-10 e 10-20 cm, tendeu a ser maior no sistema Alf-Mi (0,19 m³ m-3). A microporosidade não apresentou tendência clara entre os sistemas. A condutividade hidráulica saturada teve relação direta com a macroporosidade, com Er-Mi-Tr-So apresentando o maior valor na camada de 0-5 cm (224 mm h-1) e Alf-Mi nas camadas de 5-10 (170 mm h-1) e 10-20 cm (147 mm h-1). O sistema Er-Mi-Tr-So apresentou o menor diâmetro médio ponderado úmido de agregados na camada de 0-5 cm (2,39 mm), e o Tr-So, o maior (3,04 mm). Os maiores valores de resistência mecânica do solo à penetração foram observados no sistema Tr-So, superando 1,5 MPa na camada de 7,5 a 22,5 cm de profundidade. O sistema Alf-Mi apresentou o menor grau de compactação (0,2 MPa cm). Os resultados são atribuídos, principalmente, à ação das raízes das espécies que constituem os sistemas de culturas e à intensidade de tráfego de máquinas em cada sistema. Considerando a camada de 0-20 cm como um todo, o sistema semiperene Alf-Mi possui maior capacidade de promover melhorias na qualidade estrutural do solo, em comparação aos sistemas baseados em cultivos de espécies anuais. Sistemas bianuais de rotação, baseados em plantas de cobertura como aveia-preta ou ervilhaca, promovem melhorias na qualidade estrutural do solo em relação à sucessão trigo-soja.

An instrument designed to measure thermal conductivity of consolidated rocks, dry or saturated, using a transient method is presented. The instrument measures relative values of the thermal conductivity, and it needs calibration to obtain absolute values. The device can be used as heat pulse line source and as continuous heat line source. Two parameters to determine thermal conductivity are proposed: TMAX, in heat pulse line source, and SLOPE, in continuous heat line source. Its performance is better, and the operation simpler, in heat pulse line-source mode with a measuring time of 170 s and a reproducibility better than 2.5%. The sample preparation is very simple on both modes. The performance has been tested with a set of ten rocks with thermal conductivity values between 1.4 and 5.2 W m¿1 K¿1 which covers the usual range for consolidated rocks.

Peatlands are soil environments that store carbon and large amounts of water, due to their composition (90 % water), low hydraulic conductivity and a sponge-like behavior. It is estimated that peat bogs cover approximately 4.2 % of the Earth's surface and stock 28.4 % of the soil carbon of the planet. Approximately 612 000 ha of peatlands have been mapped in Brazil, but the peat bogs in the Serra do Espinhaço Meridional (SdEM) were not included. The objective of this study was to map the peat bogs of the northern part of the SdEM and estimate the organic matter pools and water volume they stock. The peat bogs were pre-identified and mapped by GIS and remote sensing techniques, using ArcGIS 9.3, ENVI 4.5 and GPS Track Maker Pro software and the maps validated in the field. Six peat bogs were mapped in detail (1:20,000 and 1:5,000) by transects spaced 100 m and each transect were determined every 20 m, the UTM (Universal Transverse Mercator) coordinates, depth and samples collected for characterization and determination of organic matter, according to the Brazilian System of Soil Classification. In the northern part of SdEM, 14,287.55 ha of peatlands were mapped, distributed over 1,180,109 ha, representing 1.2 % of the total area. These peatlands have an average volume of 170,021,845.00 m³ and stock 6,120,167 t (428.36 t ha-1) of organic matter and 142,138,262 m³ (9,948 m³ ha-1) of water. In the peat bogs of the Serra do Espinhaço Meridional, advanced stages of decomposing (sapric) organic matter predominate, followed by the intermediate stage (hemic). The vertical growth rate of the peatlands ranged between 0.04 and 0.43 mm year-1, while the carbon accumulation rate varied between 6.59 and 37.66 g m-2 year-1. The peat bogs of the SdEM contain the headwaters of important water bodies in the basins of the Jequitinhonha and San Francisco Rivers and store large amounts of organic carbon and water, which is the reason why the protection and preservation of these soil environments is such an urgent and increasing need.

A influência do sistema de uso e manejo na qualidade física do solo tem sido tema de destaque em razão de seus impactos ambientais e agronômicos. O intervalo hídrico ótimo (IHO) é um moderno indicador da qualidade física que potencialmente pode apresentar os mecanismos e processos de perda ou recuperação da qualidade física do solo, por causa do seu uso e manejo. Diante disso, o objetivo deste trabalho foi quantificar a influência de diferentes sistemas de uso e manejo no IHO de um Latossolo Vermelho distrófico, textura franco-arenosa, composto por 170, 40 e 790 g kg-1 de argila, silte e areia, respectivamente. Foram selecionadas quatro áreas, sob os seguintes sistemas de uso e manejo do solo: mata nativa; pastagem cultivada por mais de 20 anos; citros por mais de 10 anos, antecedido por 10 anos de pastagens; e cultivo com culturas comerciais (milho, sorgo, aveia e mandioca), por cerca de 15 anos, após 10 anos de pastagem cultivada. Foram coletadas 48 amostras indeformadas, em cada área, no centro da camada de 0-0,10 m de profundidade. As amostras foram submetidas a potenciais de -10 a -15.000 hPa, em que foram determinadas a curva de retenção de água, a curva de resistência do solo à penetração e, posteriormente, a densidade do solo (Ds) e o IHO. A Ds foi influenciada pelos sistemas de uso e manejo do solo na sequência cultivo = citros > pastagem > mata nativa. A relação entre IHO e Ds foi linear e negativa, à exceção da mata nativa que na faixa de Ds entre 1,35-1,55 Mg m-3 apresentou relação positiva. Valores de Ds, em que o IHO = 0, associados com severa degradação física do solo, foram de 1,75 e 1,80 Mg m-3, nos solos sob citros e cultivo, respectivamente; a proporção desses valores foi de 21 % para o solo sob citros e 18 %, para o sob cultivo. A perda de qualidade física do solo foi menos acentuada no solo sob pastagem, quando comparado com os solos sob citros e cultivo, sendo a diminuição do IHO menos acentuada com o aumento da Ds. O menor valor de Ds em que iniciou a redução do IHO foi tomado como densidade do solo de alerta (Ds a). A Ds a foi de 1,55 Mg m-3 e pode ser utilizada como valor de referência no processo de recuperação da qualidade física do solo, válida para solos com textura, uso e manejo semelhante ao deste estudo.
