The in situ saphenous vein bypass has been introduced in our department since 1989. A total of 26 bypasses in 22 patients have been followed prospectively. Indications for revascularisation have been severe arterial insufficiency in 73% of the cases (stage III or IV). With the exception of one postoperative death (myocardial infarction), all the patients have recovered uneventfully, with a regression to stage I. No amputation has been necessary. Morbidity has been 30%, with mainly minor local complications. The primary patency rate is 83% at one year and 78% after 2 and 3 years, whereas the secondary patency rate is 91% at one year, and remains constant thereafter up to 3 years. Considering our results and those from the literature, we believe that the in situ technique is very valuable, especially for below-knee vascular reconstruction. Technical difficulties of the method are analysed.

The complexing capacity of synthetic (0.011 M tartrate in 13.5% ethanol) and real wine (Raimat Abadia) in titrations with added total Zn concentrations up to 0.03 M has been determined following the free Zn concentrations with AGNES (absence of gradients and Nernstian equilibrium stripping) technique. A correction to find the preconcentration factor or gain (Y1) really applied at each one of the ionic strengths reached due to Zn additions along the titration has been applied. The standard implementation of AGNES to real wine led to the observation of two anomalous behaviors: (a) an increasingly negative current in the deposition stage (labeled as “HER” effect) and (b) a minimum in the currents of the stripping stage plot (labeled as the “dip” effect). A practical strategy to apply AGNES avoiding the dip effect has been developed to quantify properly free Zn concentrations. The van den Berg–Ružic–Lee linearization method (assuming the existence of just 1:1 complexes) has been adapted to consider the dilution effect and the ionic strength changes. Aggregated stability constants and total ligand concentrations have been calculated from synthetic and wine titration data. The found complexing capacity in the studied wine (cT,L = 0.0179 ± 0.0007 M) indicates the contribution of ligands other than tartrate (which is confirmed to be the main one).

Solid tumor growth triggers a wound healing response. Similar to wound healing, fibroblasts in the tumor stroma differentiate into myofibroblasts (also referred to as cancer-associated fibroblasts) primarily, but not exclusively, in response to transforming growth factor-β (TGF-β). Myofibroblasts in turn enhance tumor progression by remodeling the stroma. Among proteases implicated in stroma remodeling, matrix metalloproteinases (MMPs), including MMP-9, play a prominent role. Recent evidence indicates that MMP-9 recruitment to the tumor cell surface enhances tumor growth and invasion. In the present work, we addressed the potential relevance of MMP-9 recruitment to and activity at the surface of fibroblasts. We show that recruitment of MMP-9 to the fibroblast cell surface occurs through its fibronectin-like (FN) domain and that the molecule responsible for the recruitment is lysyl hydroxylase 3 (LH3). Functional assays suggest that both pro- and active MMP-9 trigger α-smooth muscle actin expression in cultured fibroblasts, reflecting myofibroblast differentiation, possibly as a result of TGF-β activation. Moreover, the recombinant FN domain inhibited both MMP-9-induced TGF-β activation and α-smooth muscle actin expression by displacing MMP-9 from the fibroblast cell surface. Together our results uncover LH3 as a new docking receptor of MMP-9 on the fibroblast cell surface and demonstrate that the MMP-9 FN domain is essential for the interaction. They also show that the recombinant FN domain inhibits MMP-9-induced TGF-β activation and fibroblast differentiation, providing a potentially attractive therapeutic reagent toward attenuating tumor progression where MMP-9 activity is strongly implicated.

Due to the low genetic variability reported in the commercial plantations of papaya (Carica papaya L.), the objective of this study was analyze the genetic diversity of 32 genotypes including cultivars, landraces, inbred lines, and improved germplasm using the AFLP technique (Amplified Fragment Length Polymorphism). The genetic distance matrix was obtained using the Nei and Li genetic distance and clustering was performed using the unweighted pair-method with arithmetic mean (UPGMA). Using 11 combinations of EcoRI/MseI primers, 383 polymorphic bands were obtained. On average, 34.8 polymorphic bands were obtained per primer combination. Five clusters were formed. The traditional cultivar 'Sunrise' and the inbred line CMF-L30-08 were the closest genotypes, and the improved germplasm (CMF041) and landrace (CMF233) the most distant. The main papaya cultivars commercially grown in Brazil, as well as four inbred lines and three improved germplasm, were clustered together, however, were not grouped in the same branch. The genetic distance between the Sunrise and Golden cultivars was 0.329, and even arising from mutation and selection within the Sunrise variety, the Golden stores considerable genetic variability. Additional variability was observed in the inbred lines derived from papaya breeding program at Embrapa Cassava and Fruits.

Tämä työ käsittelee tuotekehitysprojektia, jonka tarkoituksena on suunnitella uusi nelitietyöntömastotrukki asiakaslähtöisesti QFD-laatutyökalua hyväksi käyttäen. Työ esittelee QFD-laatutyökalun, sekä varsinkin sen nelivaiheisen ASI:n toimintamallin. Työ antaa myös perusvalmiudet ASI:n toimintamallin käyttöön tuotekehitysprojektissa. Työssä keskitytään tarkastelemaan uuden ohjausjärjestelmän mekaanista kehittelyä. Suunnittelutyössä käytetään hyväksi ASI:n (American Supplier Institute) nelivaiheista QFD-laatutyökalua, joka on autoteollisuuden toimintatapoihin perustuva QFD-tekniikka. QFD tulee sanoista Quality Funcktion Deployment ja vapaasti suomennettuna se merkitsee asiakaslähtöinen tuotesuunnittelu. Nelivaiheinen QFD-malli koostuu seuraavista matriiseista: 1. Tuotteen suunnittelu, 2. osien suunnittelu ja kehittäminen, 3. prosessin suunnittelu sekä 4. tuotannon suunnittelu. Matriisin tulokset muodostavat aina seuraavan matriisin lähtökohdat. Ensimmäisen matriisin lähtökohdat muodostuvat asiakastarvekuvauksesta, joka saadaan haastatteluista, tutkimuksista yms. Tuotekehitysprojektissa kehitetty TEM-monitietrukki, jossa rajattomasti ympäripyöriviä tukipyöriä ohjataan CAN-väylän kautta älykkäällä ohjauslogiikalla, on markkinoilla erittäin kilpailukykyinen sekä ominaisuuksiensa että hintansa puolesta. QFD yhdessä huolellisesti analysoidun asiakastarvekuvauksen kanssa selkeyttää ja priorisoi tuotteen spesifikaation määrittelyä sekä ohjaa tuotteen kehittelyä ja tuotannollistamista. Tuotekehitysprojekti pysyy aikataulussaan ja tuotannollistamisen kynnyksellä tehtävät muutokset vähenevät selkeän jäsentelyn ansiosta.
