No Brasil, as macrófitas aquáticas submersas, Egeria densa e Egeria najas, têm causado prejuízos aos usos múltiplos da água. Hydrilla verticillata foi recentemente introduzida, mas tem histórico como planta problemática nos EUA, no México e na Austrália. O objetivo deste trabalho foi avaliar as suscetibilidades relativas dessas três macrófitas aquáticas ao diquat e os riscos da utilização desse herbicida para o guaru (Phallocerus caudimaculatus). Para isso, foram instalados ensaios em condições de laboratório, a fim de avaliar a suscetibilidade relativa das três macrófitas por meio da manutenção de ponteiros dessas plantas em soluções contendo 0,0; 0,2; 0,4; 0,8; e 1,6 mg L-1 de diquat (Reward®) por 14 dias. A avaliação foi realizada pela variação do acúmulo de matéria fresca e do comprimento dos ponteiros no período de exposição ao herbicida. H. verticillata mostrou maior sensibilidade ao diquat em comparação com as duas macrófitas do gênero Egeria, mesmo em baixas concentrações do herbicida. Nas maiores concentrações, E. densa mostrou maior sensibilidade que E. najas. O risco da aplicação do diquat para P. caudimaculatus foi estimado pela toxicidade aguda. Alevinos de P. caudimaculatus de 0,4 ± 0,2 mg foram expostos a soluções de 0,0; 1,0; 5,0; 10,0; 15,0; 20,0; 25,0; e 30,0 mg L-1 de diquat. A concentração letal de 50% (CL(I) (50;96h)) do diquat estimada para P. caudimaculatus foi de 7,17 mg L-1. Para P. caudimaculatus, a toxicidade aguda foi superior à concentração recomendada para o controle de macrófitas aquáticas submersas, indicando risco muito baixo para esse peixe.

O experimento foi desenvolvido em casa de vegetação, com o objetivo de verificar a capacidade das sementes de Mucuna aterrima, Mucuna cinerea e Mucuna deeringiana para superar diferentes profundidades do solo acrescido de densidades de palha de cana-de-açúcar. O delineamento utilizado foi o inteiramente casualizado; os tratamentos foram distribuídos em esquema fatorial 3 x 3 x 3, com quatro repetições. Os fatores foram constituídos por camada de palha (0, 5 e 10 cm), espécies de Mucuna (M. aterrima, M. cinerea e M. deeringiana) e profundidade de semeadura (1, 4 e 8 cm). As espécies de Mucuna superaram até 23 t ha-1 de palha (10 cm) e até 8 cm de profundidade de semeadura, expondo as partes aéreas à superfície a partir dos 10 dias após emergência (DAS); elas foram capazes também de desenvolver a altura (>96 cm por planta), a área foliar (>723 cm-2 por planta) e o acúmulo de massa seca (>2,09 g por planta) até os 35 DAS.

The aim of this study was to examine the dormancy behavior of Euphorbia dracunculoides and Astragalus spp., weeds of arid chickpea. The dormancy breaking treatments were: Gibberalic acid (GA3) and Thiourea each at 50, 100, 150, 200, 250, and 300 ppm and Potassium nitrate (KNO3) at 5,000, 10,000, 15,000, 20,000, 25,000, and 30,000 ppm (24 h soaking). Germination (G) percentage and germination energy (GE) of E. dracunculoides was maximum (89 and 22, respectively) at 250 ppm concentration of GA3 and 81.50 and 11.50 at 15000 ppm concentration of KNO3. Thiourea at 250 and 300 ppm resulted in maximum G percentage (51) and GE (25.50) of E. dracunculoides, whereas the G percentage and GE of Astragalus spp. were maximum (28 and 19, respectively) at the lowest concentration of GA3 (50 ppm). On the other hand, 5000 ppm and 150 ppm concentration of KNO3 and Thiourea showed maximum GE (19.5) and G percentage (28) of Astragalus spp., respectively. Overall, effective dormancy breaking chemical against E. dracunculoides was GA3 (250 ppm) while in Astragalus spp. none of chemicals showed very impressive results. These results showed that both weeds' seeds have dormancy in their habit. Hot water treatment and the above mentioned chemicals (best concentrations) when used with 4, 8, and 12 hours soaking showed ineffective results.

Trinexapac-ethyl and sulfometuron-methyl are the most widely used ripeners in sugarcane. The application is performed by airborne spraying. Thus, if weather conditions are unfavorable, spray drift to neighboring areas may occur. The objective of this study was to assess the selectivity of the plant growth regulators trinexapac-ethyl and sulfometuron-methyl, used as sugarcane ripeners, to eucalyptus (Eucalyptus urograndis) young plants. The experiment was installed in an eucalyptus commercial yield area, in the municipality of Tamba50;, state of São Paulo, Brazil, and arranged in a 2 x 8 factorial design in randomized blocks with four replications. The treatments studied were trinexapac-ethyl and sulfometuron-methyl, sprayed in eight doses, 0; 1.0; 2.5; 5.0; 10; 25; 50 and 100% of the dose used in sugarcane as ripeners (200 g ha-1 of trinexapac-ethyl and 15 g ha-1 of sulfometuron-methyl). Chemical ripeners were applied on eucalyptus plants with 48 cm in height on average; 10.1 branches; 4.5 mm of stem diameter and 44.3 cm of crown diameter, at 46 days after seeding. Trinexapac-ethyl was selective to eucalyptus and stimulated crown diameter growth. At higher doses, sulfometuron-methyl promoted severe noticeable injuries in eucalyptus plants, such as apical bud death. However, during the assessment period the plants recovered and the visual symptoms of phytotoxicity and growth alterations were not observed at 60 days after application. The plant growth regulators trinexapac-ethyl and sulfometuron-methyl were selective to eucalyptus young plants.

Phytotoxic effects of invasive weed Parthenium hysterophorus were studied by using whole plant, leaf and root aqueous extracts at 0, 2.5, 5.0, 7.5 and 10% (w/v) concentrations against germination and early seedling growth of wheat and canola. Studies were carried out both in Petri plates with filter paper as substratum placed in controlled conditions and soil-filled plastic pots placed in open environments. Pronounced variation was noted for phytotoxic activity of different plant parts of parthenium, aqueous extract concentrations, test species, and bioassay techniques. Aqueous parthenium extracts either inhibited or delayed the germination and suppressed seedling growth of test species over control. For both test species, all the germination attributes were suppressed to a greater extent in Petri plates than in plastic pots. Leaf extracts were more suppressive to germination of test species than whole plant and root extracts. Increasing extract concentration beyond 2.5% caused significant reduction in seedling dry biomass of both test species. Aqueous parthenium extract diminished chlorophyll contents of wheat and canola by 32-63% and 29 69%, respectively. Nevertheless, an increase of 9-172% and 22-60% in phenolic contents of wheat and canola was recorded. Canola appeared to be more susceptible than wheat at all extract concentrations. Present study concluded that bioassays conducted under controlled condition using filter paper as substratum may be misleading due to over estimation of allelopathic response and variation in potential of receiver and donor species. Furthermore, it implies that threshold concentrations of allelochemicals for test species in Petri plates are rarely reached under field conditions.

Neste trabalho, foram estudadas a composição florística e a estrutura fitossociológica da vegetação em um fragmento de floresta ribeirinha, ao longo do rio Passa Cinco, Ipeúna, SP. Foram amostradas 157 parcelas contínuas de 5 x 10 m, em uma área de 0,785 ha. Foram feitas medidas de diâmetro e mapeados todos os indivíduos com DAP maior ou igual a 5 cm, em 1989 e 1998. Constatou-se a presença de um mosaico vegetacional e discutiram-se os possíveis fatores definidores deste mosaico. Foram, também, verificadas as alterações florísticas e fitossociológicas na área após nove anos. As análises multivariadas da vegetação mostraram a presença de três grupos vegetacionais com grande sobreposição aos três tipos de solo identificados na área. As análises quantitativas da estrutura da comunidade mostraram diferenças em cada um destes grupos, que se mantiveram ao longo do tempo, indicando associações florísticas próprias com grande correlação à cada tipo de solo. O grupo vegetacional sobre solo aluvial, na faixa imediatamente paralela ao curso d’água, apresentou diferenças florísticas mais pronunciadas, e maior diversidade de espécies, área basal e densidade de indivíduos. Em nove anos, poucas alterações na estrutura fitossociológica foram observadas para as espécies de maior valor de importância na área, com um aumento de 2,01% do número total de indivíduos e 2,80% na área basal total.

Os prováveis hormônios vegetais envolvidos no crescimento e acúmulo de proteínas de reserva em sementes de soja foram avaliados por meio do cultivo artificial dos frutos. Explantes de frutos de soja apresentando o pericarpo completamente expandido e sementes com aproximadamente 100 mg de massa fresca foram cultivados in vitro com várias concentrações de ácido naftaleno-acético (ANA), ácido giberélico (GA3), benziladenina (BA) e ácido abscísico (ABA). Esses fitorreguladores foram utilizados inicialmente isolados nas concentrações de 10-7, 10-6, 10-5 e 10-4 mol.L-1 cada um. Em seguida, foram combinados dois a dois, sendo as concentrações determinadas em função dos maiores incrementos na massa seca das sementes observados nos experimentos iniciais. O ANA foi ineficiente para o crescimento das sementes, que foi estimulado por GA3 e BA, porém apenas nas concentrações mais elevadas. O ABA inibiu o crescimento das sementes. Quando combinou-se o ANA com GA3 ou BA, houve aumento na massa seca das sementes, sugerindo que pode ocorrer interações entre hormônios vegetais. O ANA e a BA foram os fitorreguladores mais eficientes no acúmulo de proteínas, tanto isolados como combinados. O ABA, por sua vez, inibiu a síntese de proteínas de reserva nas sementes. O sistema artificial de cultivo utilizado nesse experimento mostrou-se eficiente para o estudo do crescimento e de teores protéicos em sementes de soja.

An area's innate potential to regenerate represents a crucial factor for its conservation and management. The seed rain and seed bank are important agents in the regeneration process. Seed banks are particularly important in communities where there is a high proportion of obligate seeders. Rocky outcrops are habitats where most part of the plant species depends on their seeds to reproduce and maintain viable populations. Therefore, seed banks ought to be important in this vegetation physiognomy. We test the hypotheses that the seed bank of the rocky outcrops found in the rupestrian fields of "Serra do Cipó", Brazil, is richer in species and denser than those formed on different vegetation physiognomies neighboring the outcrops. We then compared species abundance, species richness and composition in the rocky outcrops' seed banks with those of sandy and peaty bogs, forests, gallery forests, and "cerrados". Furthermore, we report on the natural regeneration potential of these soils by assessing a greenhouse study on seedling emergence. Soil samples were collected from 0 to 5 and 5 to 10 cm of depth. Rocky outcrops had the poorest in species and less dense seed bank and showed segregation in species composition. Emergence was greater in the most superficial layer. However, soils on rocky outcrops showed the greatest proportion of endemic threatened species in their seed banks, demonstrating their importance for biodiversity conservation of the "Serra do Cipó" rupestrian fields.

Neste estudo analisamos a estrutura da comunidade de plantas lenhosas em 11 fragmentos de cerrado (sensu stricto) situados no Triângulo Mineiro. Avaliamos se a densidade, diversidade e/ou composição da vegetação estavam relacionadas à área do fragmento ou ao seu grau de perturbação (medida pela cobertura de gramíneas exóticas e pela ocorrência histórica de gado e fogo). Em cada fragmento (com tamanho entre 7 a 509 ha) foram instaladas 50 parcelas de 10 x 10 m para contagem e identificação das árvores com circunferência a altura do peito (CAP) > 15 cm. Arbustos e arvoretas (CAP < 15 cm) foram amostrados em sub-parcelas de 5 x 10 m. Fragmentos maiores apresentaram maior número de indivíduos (de arbustos e arvoretas), de espécies (de árvores, arbustos e arvoretas), e maior diversidade (de árvores) do que os fragmentos menores. O tamanho do fragmento também explicou parte da variação na composição de espécies de arbustos e de arvoretas. Com exceção de uma relação negativa entre a cobertura de gramíneas e a densidade de arbustos e arvoretas, não encontramos qualquer correlação entre o grau de perturbação e a estrutura da vegetação. Entretanto, não foi possível separar os efeitos de área dos efeitos da perturbação, já que fragmentos menores tenderam a apresentar maior grau de perturbação, particularmente uma maior cobertura de gramíneas exóticas. Isto indica que fragmentos menores não apenas estão mais sujeitos aos efeitos decorrentes da fragmentação dos hábitats, mas também aos efeitos da ação do fogo, do gado e da invasão por gramíneas exóticas.

The mechanism whereby the immune system avoids self-aggression is one of the central issues of Immunology. The discovery of natural autoantibodies, mainly of IgM isotype, and of idiotypic interactions between antibodies indicates that elements of the immune system interact with self constituents and with themselves. Results of studies with soluble antibodies have indicated that the pool of circulating IgM represents the end result of a highly selective process of B cells activation and differentiation by self proteins resulting in the formation of a network. The objective of the present work was to determine the frequency of self-reacting B cells in normal mice. We were able to detect B cells that recognize self proteins present in extracts of different organs in normal adult, 2-3-month old, BALB/c and C57BL/6 mice with an ELISA spot assay. About 1% of total IgM-secreting cells among small, LPS-stimulated spleen cells reacted with organ extracts, whereas among large spleen cells the frequency was 5- to 10-fold lower. Immunization induced an increase in the frequency of IgM-secreting cells. The present results provide cellular evidence for the results of studies done at the serological level. The physiological role of these self-recognizing cells, as well as their participation in autoimmune processes, remain to be established

Ninety-six weanling male Wistar rats were fed for four weeks one of two different chows: a normal rat chow containing 55.5% (w/w) starch (control group, N = 48) or a rat chow in which starch was partially replaced by lactose, in such a way that the experimental group (N = 48) received 35.5% (w/w) starch and 20% (w/w) lactose. The gastric emptying of fluid was then studied by measuring the gastric retention of four test meals containing lactose (5% or 10%, w/v) or glucose + galactose (5% or 10%, w/v). Homogenates of the small intestine were assayed for lactase activity. The gastric retention values were obtained 15 min after orogastric infusion of the liquid meals. The median values for gastric retention of the 5% lactose solutions were 37.7% for the control group and 37.0% for the experimental group (P>0.02). For the 10% lactose solution the median values were 51.2% and 47.9% (P>0.02) for the control and experimental groups, respectively. However, for the 2.5% glucose + 2.5% galactose meal the median gastric retention was lower (P<0.02) in the group fed a lactose-enriched chow (38.5%) than in the control group (41.6%). For the 5% glucose + 5% galactose solution the median values were not statistically different between groups, 65.0% for the control group and 58.8% for the experimental group. The median values of the specific lactase activity in the small intestine homogenate was 0.74 U/g in the control group and 0.91 U/g in the experimental group. These values were not statistically different (P>0.05). These results suggest that the prolonged ingestion of lactose by young adult rats changes the gastric emptying of a solution containing 5% monosaccharides. This adaptation may reflect the desensitization of intestinal nutrient receptors, possibly by an osmotic effect of lactose present in the chow.

The objective of the present experiment was to assess ethyl alcohol (ETOH) dependence brought about by a semivoluntary intermittent intake regimen in rats. Male Wistar rats weighing 150-250 g at the onset of the experiment were assigned to the following groups: 0% ETOH (N = 11), 5% ETOH (N = 20), 20% ETOH (N = 20) and 40% ETOH (N = 18). ETOH solutions were offered at the end of the day and overnight from Monday to Friday, and throughout weekends, for 90 days. The concentration of the ETOH solutions was increased in a stepwise fashion allowing the rats to get used to the taste of alcohol. Reposition of pure water was permitted during 1-h water drinking periods in the morning. Daily volume intake (± SEM) averaged 25.4 ± 0.4 ml (0% ETOH), 23.8 ± 0.6 ml (5% ETOH), 17.6 ± 0.7 ml (20% ETOH) and 17.5 ± 0.6 ml (40% ETOH). ETOH consumption differed significantly (P<0.05) among groups, averaging 4.4 ± 0.2 g kg-1 day-1 (5% ETOH), 10.3 ± 0.3 g kg-1 day-1 (20% ETOH) and 26 ± 1.2 g kg-1 day-1 (40% ETOH). Furthermore, ETOH detection in plasma 10-12 h after offering the solution indicated that its consumption in the 40% ETOH group was sufficient to override its metabolism. Overt signs of ETOH dependence, such as increased thirst, hyperactivity, puffing, hair ruffling and startle responsiveness as well as reduced drowsiness, were significantly increased in the 20% and 40% ETOH groups compared to the 0% and 5% groups. Accordingly, the model described here proved to be a useful tool for the evaluation of subtle or moderate behavioral and physical consequences of long-term ETOH intake

In the present study, using noise-free simulated signals, we performed a comparative examination of several preprocessing techniques that are used to transform the cardiac event series in a regularly sampled time series, appropriate for spectral analysis of heart rhythm variability (HRV). First, a group of noise-free simulated point event series, which represents a time series of heartbeats, was generated by an integral pulse frequency modulation model. In order to evaluate the performance of the preprocessing methods, the differences between the spectra of the preprocessed simulated signals and the true spectrum (spectrum of the model input modulating signals) were surveyed by visual analysis and by contrasting merit indices. It is desired that estimated spectra match the true spectrum as close as possible, showing a minimum of harmonic components and other artifacts. The merit indices proposed to quantify these mismatches were the leakage rate, defined as a measure of leakage components (located outside some narrow windows centered at frequencies of model input modulating signals) with respect to the whole spectral components, and the numbers of leakage components with amplitudes greater than 1%, 5% and 10% of the total spectral components. Our data, obtained from a noise-free simulation, indicate that the utilization of heart rate values instead of heart period values in the derivation of signals representative of heart rhythm results in more accurate spectra. Furthermore, our data support the efficiency of the widely used preprocessing technique based on the convolution of inverse interval function values with a rectangular window, and suggest the preprocessing technique based on a cubic polynomial interpolation of inverse interval function values and succeeding spectral analysis as another efficient and fast method for the analysis of HRV signals

Paracoccidioidomycosis (PCM) is the most prevalent deep mycosis in Latin America and presents a wide spectrum of clinical manifestations. We established a genetically controlled murine model of PCM, where A/Sn mice develop an infection which mimics the benign disease (immune responses which favor cellular immunity) and B10.A animals present the progressive disseminated form of PCM (preferential activation of B cells and impairment of cellular immune responses). To understand the immunoregulatory phenomena associated with resistance and susceptibility in experimental PCM, A/Sn and B10.A mice were studied regarding antigen-elicited secretion of monokines (TNF-a and TGF-ß) and type-1 (IL-2 and IFN-g) and type-2 (IL-4,5,10) cytokines. Total lymph node cells from resistant mice infected ip with P. brasiliensis produced early and sustained levels of IFN-g and IL-2; type-2 cytokines (IL-4 and IL-5) started to appear 8 weeks after infection. In contrast, susceptible mice produced low levels of IFN-g concomitant with significant levels of IL-5 and IL-10 early in the infection. In the chronic phase of the disease, susceptible animals presented a transitory secretion of IL-2, and IL-4. In the pulmonary infection IL-4, IL-5 and IL-10 were preferentially detected in the lung cells washings of susceptible animals. After in vitro challenge with fungal antigens, normal peritoneal macrophages from B10.A mice secreted high levels of TGF-ß and low levels of TNF-a. In contrast, macrophages from A/Sn animals released high levels of TNF-a associated with a small production of TGF-ß. The in vivo depletion of IFN-g not only abrogated the resistance of A/Sn mice but also diminished the relative resistance of B10.A animals. The in vivo depletion of IL-4 did not alter the disease outcome, whereas administration of rIL-12 significantly enhanced resistance in susceptible animals. Taken together, these results suggest that an early secretion of high levels of TNF-a and IFN-g followed by a sustained secretion of IL-2 and IFN-g plays a dominant role in the resistance mechanisms to P. brasiliensis infection. In contrast, an early and ephemeral secretion of low levels of TNF-a and IFN-g associated with production of IL-5, IL-10 and TGF-ß characterizes the progressive disease of susceptible animals.

The survival of hemodialysis patients is likely to be influenced not only by well-known risk factors like age and comorbidity, but also by changes in dialysis technology and practices accumulated along time. We compared the survival curves, dialysis routines and some risk factors of two groups of patients admitted to a Brazilian maintenance hemodialysis program during two consecutive decades: March 1977 to December 1986 (group 1, N = 162) and January 1987 to June 1997 (group 2, N = 237). The median treatment time was 22 months (range 1-198). Survival curves were constructed using the Kaplan-Meier method and compared using the log-rank method. The Cox proportional hazard regression model was used to investigate the more important variables associated with outcome. The most important changes in dialysis routine and in patient care during the total period of observation were the progressive increase in the dose of dialysis delivered, the prohibition of potassium-free dialysate, the use of bicarbonate as a buffer and the upgrading of the dialysis equipment. There were no significant differences between the survival curves of the two groups. Survival rates at 1, 5 and 10 years were 84, 53 and 29%, respectively, for group 1 and 77, 42 and 21% for group 2. Patients in group 1 were younger (45.5 ± 15.2 vs 55.2 ± 15.9 years, P<0.001) and had a lower prevalence of diabetes (11.1 vs 27.4%, P<0.001) and of cardiovascular disease (9.3 vs 20.7%, P<0.001). According to the Cox multivariate model, only age (hazard ratio (HR) 1.04, confidence interval (CI) 1.03-1.05, P<0.001) and diabetes (HR 2.55, CI 1.82-3.58, P<0.001) were independent predictors of mortality for the whole group. Patients of group 2 had a lower prevalence of sudden death (19.1 vs 9.7%, P<0.001). After adjusting for age, diabetes and other mortality risk factors, the risk of death was 17% lower in group 2, although this difference was not statistically significant. We conclude that the negative effects of advanced age and of higher frequency of comorbidity on the survival of group 2 patients were probably offset by improvements in patient care and in the quality and dose of dialysis delivered, so that the survival curves did not undergo significant changes along time.
