World consumption of vegetable oils has increased in recent years because of its application in food, chemical, pharmaceutical and, more recently, energy industry. However, oilseeds, which these oils are extracted, have low viability, affecting the cultivation and productivity of these species. The aim of this study was to analyze the effect of aging on the coordination of catalase (CAT) and ascorbate peroxidase (APX) antioxidant systems in safflower and sunflower. . Therefore, seeds were subjected to accelerated aging for 3, 6 and 9 days and grown in moistened paper towel for 72 hours. Additionally, before accelerated aging, sunflower seeds were pretreated by osmopriming with 10 mM ascorbate (ASC) or 3 amino 1,2,4 triazol (3-AT), a specific inhibitor of CAT activitie. The method of artificial aging used was efficient in both species, because it caused a decrease in germination, seedling development and growth, especially in safflower. The aging caused inhibition of CAT activity for both species and to compensate for such inhibition , sunflower increased mRNA expression of this enzyme , while safflower mobilized over the activity of APX. Analysis of the expression of malate synthase and sugar content demonstrated that sunflower seeds consumes lipid reserves in quiescent state, while the safflower is more dependent on carbohydrate. Pretreatment with 3-AT inhibited CAT activity and stimulated the APX, though with ASC acted reverse on these systems. None of the treatments recovered the physiological decline aging. It is concluded that aging change the oilseeds antioxidant metabolism, despite interspecies variations in response to this process, the depletion of the CAT antioxidant system was common. Because of this we propose that the measurement of CAT activity can be used to identify aging seed lots.

Campomanesia adamantium and Campomanesia pubescens are morphologically similar, occur in the same regions of the Cerrado, are difficult to differentiate and exhibit naturally slow growth and development. The objectives of this study were to analyze fruit and seed biometric data, emergence capacity and seedling growth characteristics. Fruit from both species was collected and used to measure biometric data of the fruit and seeds and to set up two emergence and two seedling growth experiments. C. adamantium fruit is round and has wider, pale yellow seeds while C. pubescens fruit is ellipsoidal to pyriform with yellow gold seeds. C. adamantium’s greater fresh fruit mass and biometric variability favors selection of promising material for commercialization. Seed drying reduced the speed and rate of seedling emergence of C. adamantium, but had no effect on C. pubescens. Leaf number, height, shoot dry weight and the ratio of shoot dry weight to root dry weight were greater in C. adamantium than in the slower growing C. pubescens. Increases in substrate volume favor seedling development. Slow-release fertilizer application at 1g per 115 cm3 substrate increased leaf formation, plant height and shoot dry weight of seedlings of both species and lateral budding in C. adamantium. Until 120 days after transplant, lateral budding was not observed in C. pubescens seedlings. For all traits evaluated in this experiment, C. adamantium seedling growth was greater than that of C. pubescens.

Molybdenum is one of the essential micronutrients for soybeans, acting directly on nitrogen metabolism as enzyme cofactor of nitrogenase. Usually, this nutrient is supplied to the plants through seed treatment or foliar application. The aim of this study was to evaluate the molybdenum effects by foliar in the physiological potential of soybean seeds and verify its interference in the enzyme activities involved in nitrogen metabolism. Soybean seeds of BMX Turbo cultivar were used, produced in Erechim, RS, harvest 2013, from plants treated with the following Mo concentrations: 0; 25; 50 and 75 g ha-1, supplied through two commercial products (Biomol and Molybdate) and stored during 0 and 6 months in uncontrolled conditions. The first experiment was conducted in Seedtes Seed Analysis Laboratory in Pato Branco, PR. The used design was completely randomized in a factorial analysis 4 x 2 x 2 with four replications each. The physiological potential of the seeds was evaluated by the germination test, seedling growth, accelerated aging and emergence on the soil. The second experiment was conducted in a greenhouse, where the seeds derived from treatments with different concentrations of Mo: 0; 25; 50 and 75 g ha-1 supplied through two commercial products (Biomol and Molybdate) were grown in vases. The used design was completely randomized in a factorial analysis 4 x 2 with four replications. Evaluations were performed when the plants reached the R1 phenological stage concerning the nodulation, dry matter of root and shoot of the plants and the determination of the activity of the enzymes glutamine synthetase and glutamate synthetase and the content of total soluble proteins. The data were submitted to variance analysis and when significant they were assessed by Tukey’s test for comparison of products and seed storage and with regression study to the concentrations at 5% probability. Analyses were performed using SISVAR statistical software. The soybean seed storage under uncontrolled conditions affected the seed vigour produced with Mo, regardless of the commercial product used during production. The application of Mo through foliar positively influences the production of soya beans which presented increasing responses in the germination and vigour with the application of Mo above 25 g ha-1 . The enrichment of Mo through foliar did not affect the nodulation of plants of the next generation, however, the use of Mo above 25 g ha-1 provided an increase in the activity of enzymes involved in nitrogen metabolism as well as on the total protein content.

The scientific research in seed technology is based on techniques that aim the reduction of costs and time, standardization, improvement and establishment of analytical methods while maintaining a high level of reliability of the results. This study sought to elucidate the reliability of electrical conductivity and pH of the exudate compared to the classic germination test, which was developed in two separate studies, however interrelated with each other, as to their final goals. The experimental material of this study consisted of seeds of the species Aspidosperma parvifolium (guatambu), Aspidosperma polyneuron (peroba-rosa), Cabralea canjerana (canjerana), Cariniana legalis (jequitibá), Gallesia integrifolia (pau-d'alho), Handroanthus chrysotrichus (ipê-amarelo), Lonchocarpus campestris (rabo-de-bugio) and Pterogyne nitens (amendoim-do-campo). The physiological quality of the studied seed species was evaluated through the electrical conductivity and pH test of the exudate by mass and individual methods being compared and correlated with the results obtained in the germination test. In addition to the tested methods, imbibition periods of the seeds were evaluated for conductivity and pH, which corresponded to 2, 4, 6, 8, 24 and 48 hours. The electrical conductivity test was efficient in both of the used methods to evaluate the physiological quality of the studied seed species when compared to the standard germination test. The pH test of the exudate applied by the individual method was more efficient and thorough to evaluate the physiological quality of the studied seed species, than the mass method. For the species Gallesia integrifolia, Cariniana legalis and Lonchocarpus campestris the pH tests of the exudate tests were not efficient due to poor or absent correlation between germination and pH.

The use of inputs containing phosphites have been presenting results in many studies, taking on importance to the control of diseases in some cultures and demonstrating the resistance induction in seedlings, with ability to activate defense mechanisms, conferring protection to plants against microorganisms. The soybean crop is recognized for its importance in providing grains and derivatives for human consumption, animal, production of biofuels, pharmaceuticals, among others. Positive results obtained through studies based on resistance inducers in some cultures arouse the interest for further study. The objective of this study was to evaluate the effect of potassium phosphites on the resistance induction and treatment of soybean seeds. Therefore were conducted four laboratory studies at the Federal Technological University of Paraná, Campus of Dois Vizinhos. In the first study it was evaluated the quality attributes of the seeds and the resistance induction as seed treatment. Then it was verified that phosphites have action upon the seedlings metabolism in due to seed treatment, having the phosphite Reforce® contributed to seed quality attributes and phosphites FitofosK® and Fitofos K Plus® induced the resistance increasing the activity of β-1,3-glucanase. In the second study it was evaluated the the resistance induction in soybean cotyledons, in which the phosphites demonstrated induction potential of phytoalexin gliceolin. In the third study It was evaluated the soybean seed health treated with potassium phosphites.. it was observed that the phosphites reduced the incidence of many fungi on seeds, especially of storage fungi like Aspergillus sp. and Fusarium semitectum. In the fourth study it was evaluated the in vitro effect of potassium phosphites on pathogenic fungi of the culture. And it was found direct action of phosphites on the mycelial growth of Fusarium semitectum, Pythium sp. and Sclerotinia sclerotiorum. Based on these results, we concluded that potassium phosphites have potential in seeds treatment, as resistance inducer and on in vitro control of phytopathogens.

The use of biopolymers that help to fix pesticides efficiently and degrade easily without harming the environment, and still improve the physiological performance of field soybean seed may bring contributions to the soybean yield. This study aimed to evaluate the effect of cassava starch polymers (AM), sodium alginate (ALG) and polyvinyl alcohol (PVOH), in the concentrations 2, 4 and 6 g / 100 ml of solution, in the physiological attributes of seeds soy, seed speed soaking and performance of soybean seeds after three months of storage. The soybean variety used was the NK 7059 RR. The experimental design used for the three studies was a factorial with 48 experimental units: 3 polymers (AM, ALG and PVOH), 4 different concentrations (0%, 2%, 4% and 6%), with four replications, in a completely randomized design. It was observed the level of significance of the factors and their interactions, applying the test F. The polymers were evaluated by the Tukey test at 5% probability, and the concentrations were evaluated by polynomial regression. The witness obtained better results for most variables studied. Among the polymers, the best coating was observed PVOH because it was the less viscous polymer and visually not served as a substrate for microorganisms. However, also, satisfactory results were obtained for the AM and ALG polymers at a concentration of 2%. There was not interference of the polymers studied with regard to reduction of imbibition rate of soybean seeds. The hydrophilicity of polymers, mainly the AM and ALG accelerated soaking seeds harming germination at concentrations 4% and 6%. In general, the higher the concentration of polymers tended to worse results.

Sesbania punicea (Cav.) Benth.

The seed size used for seeding has caused doubts among soybean producers. The study aimed to determine whether there may be differences between seed size with respect to depth of fertilizer deposition. The field experiment was conducted at the Experimental Area UTFPR Campus Pato Branco, using a precision seeder for direct seeding. The design was a randomized blocks, with five repetitions. The treatments were composed by the combination of two seed sizes (large seed with 6,5 mm and 5,5 mm with small seed) and two fertilizer deposition depths in relation to the seed (fertilizer near the seed with about 3 cm away and fertilizer distant from the seeds with about 10 cm). Data were subjected to analysis of variance. When the test value F was significant at 5% probability was applied to the Duncan test for comparison of means. The shallower depth of fertilizer deposition provided larger number of pods per plant and increased number of grains per plant. Already the largest depth of fertilizer deposition provided greater plant height at 30 days after sowing and R2 stage, greater ground area mobilized, higher plant population in all periods, greater depth of deposition of seeds and a higher rate of emergency speed.

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulphate fractionation, affinity chromatography on immobilized Trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named ITC, had Mr of 32.5 kDa by SDS-PAGE and was composed by two subunits with 27.7 and 5.6 kDa linked by disulphide bridges, a typical characteristic of Kunitz-Inhibitor family. ITC was stable until 50°C, and at 100°C its residual activity was of about 60%. Also, ITC was stable at pHs 2 to 12. The inhibition of trypsin by ITC was non-competitive, with a Ki of 8,8 x 10-7M. ITC inhibits weakly other serine proteinases such as chymotrypsin and elastase. The inhibition of papain (44% of inhibition), a cysteine proteinase was an indicative of the bi-functionality of ITC. In vitro assays against digestive proteinases from several Lepdoptera, Diptera and Coleoptera pests were made. ITC inhibited in 100% digestive enzymes of Ceratitis capitata (fruit fly), Spodoptera frugiperda and Alabama argillacea, the last one being a cotton pest. It also inhibited in 74.4% Callosobruchus maculatus (bean weevil) digestive enzymes, a Coleoptera pest. ITC, when added in artificial diet models, affected weakly the development of C. capitata larvae and it had a WD50 of 2.65% to C. maculatus larvae
