This thesis examines Paul Austers extremely neglected early work: his poetry. Five books were published: Unearth (1974), Wall writing (1976), Effigies (1977), Fragments from cold (1977) and Facing the music (1980), available only at antiquarians and restrective universities libraries in the United States, as well as at the New York Public Library. Studies around Austers poetic oeuvre are restricted to papers, reviews, translators introduction, and a thesis that focus on his poetry to produce new analyses and interpretations of Austers novelistic works. The aim of this thesis is to gather this scattered material and provide new parameters of study around his poetry. Divided into three chapters, the first one maps the literary magazines where Auster published his poems at first, the translations of his poems and critical fortune; the second examines Austers five books according to three specific topics authorship, language, I and other themes related to them; the third analyzes White Spaces, text considered by the author as the bridge that leads him to prose and in this thesis as a singular writing in which Auster consolidates his literary project, since poetry, previous to prose, yet to come. Maurice Blanchot, Karlheinz Stierle and, principally, Auster, lay the foundation of the investigation. Other theorists who contribute to the understanding of the subject will be called to build the sui generis comparativism put into effect here

Esta pesquisa tem em vista a influência geográfica do Cinema Guaraci, um artefato geográfico, colaborador do desenvolvimento econômico e na formação sociocultural da população de Rocha Miranda. Localizado na zona norte da cidade do Rio de Janeiro, descortinando esta parte da cidade pouco estudada nos estudos citadinos, constitui este o recorte espacial sob o marco temporal compreendendo o período de sua inauguração de 1953 a 2013, quando persistem os esforços em prol da reabertura desta sala de espetáculos. Através da relação de significações e topofilia, de moradores e frequentadores do lugar, para com este testemunho geográfico, foram escolhidos para esta investigação indivíduos de diversas gerações. Ressalta-se a importância deste símbolo, para a organização espacial do bairro, bem como a obstinada estima pela reabertura do Cine. Para alcançar a profundidade das expressões simbólica e geográfica deste bem, serão realizadas entrevistas e mesmo conversas informais com seus residentes e frequentadores. De igual modo, no processo de elaboração desta pesquisa ainda constam coleta de dados e material bibliográfico com respeito a informações sobre o bairro obtidos nos órgãos de pesquisa competentes, sede do Grupo Pró-Cine Guaraci, instituições como a Biblioteca Nacional, o Arquivo Geral da Cidade, a Fundação Parques e Jardins, o Instituto Pereira Passos, as bibliotecas da Universidade do Estado do Rio de Janeiro, além do enriquecimento de obras de autores como Yi-Fu Tuan, David Seamon, Roberto L. Corrêa, Zeny Rosendahl, João B. F. Mello, Alice Gonzaga, Ecléa Bosi, entre outros. Desta feita, ao analisar a evolução de um símbolo, desde o seu surgimento, de como este influenciou na dinâmica e crescimento econômico de um bairro, testifica-se o quanto é fundamental sua permanência, especialmente quando sua influência distende na formação cultural de sua gente.

Esta dissertação trata das transformações ocorridas na cultura política mexicana do final do século XIX até o início do XX, enfocando o acervo imagético de um dos principais ilustradores mexicanos, José Guadalupe Posada. Está inserida na Linha de Pesquisa Política e Cultura do Programa de Pós-Graduação em História da UERJ. Os procedimentos teórico-metodológicos nortearam-se pelos aportes da História Cultural e as fontes documentais, em sua maioria ilustrações, foram pesquisadas em arquivos públicos digitais e bibliotecas. O ilustrador viveu entre 1852 e 1913 e foi considerado um dos responsáveis pelo fortalecimento da arte gráfica na América Latina, além de se destacar como um dos intelectuais mais expressivos de sua época e testemunha ocular dos acontecimentos que levaram à Revolução Mexicana. Através daquelas ilustrações analisamos o contexto histórico do México ao longo do governo de Porfírio Díaz. Foram destacadas as culturas políticas que nortearam o percurso do país, a primeira entre a metade e o fim do século XIX, a outra na primeira década do século XX. As obras de José Guadalupe Posada são uma das mais ricas fontes históricas sobre o período e contribuíram para a disseminação de ideias e valores políticos que circularam no México durante o Porfiriato.

Amphibian skin is a rich resource of antimicrobial peptides, like maximins and maximin Hs from frog Bombina maxima. Novel cDNA clones encoding a precursor protein, which comprises a novel maximin peptide (maximin 9) and reported maximin H3, were isolated from two constructed skin cDNA libraries of B. maxima. The predicted primary structure of maximin 9 is GIGRKFLGGVKTTFRCGVKDFASKHLY-NH2. A surprising substitution is at position 16, with a free cysteine in maximin 9 rather than usual conserved glycine in other reported maximins. Maximin 9, the homodimer form and its Cys(16) to Gly(16) mutant were synthesized and their antimicrobial activities were evaluated. Unlike previously reported maximin 3, the tested bacterial and fungal strains were resistant to maximin 9, its homodimer and the Cys(16) to Gly(16) mutant (with MICs > 100 mu M). On the other hand, interestingly, while eight clinical Mollicutes strains were generally resistant to maximin 9 homodimer and its Cys(16) to Gly(16) mutant, most of them are sensitive to maximin 9 at a peptide concentration of 30 mu M, especially in the presence of dithiothreitol. These results indicate that the presence of a reactive Cys residue in maximin 9 is important for its antimycoplasma activity. The diversity of antimicrobial peptide cDNA structures encountered in B. maxima skin cDNA libraries and the antimicrobial specificity differences of the peptides may reflect well the species' adaptation to the unique microbial environments. Antimicrobial peptides secreted by the skin of many amphibians play an important role in innate immunity. From two skin cDNA libraries of two individuals of the Chinese red belly toad (Bombina maxima), we identified 56 different antimicrobial peptide cDNA sequences, each of which encodes a precursor peptide that can give rise to two kinds of antimicrobial peptides, maximin and maximin H. Among these cDNA, we found that the mean number of nucleotide substitution per non-synonymous site in both the maximin and maximin H domains significantly exceed the mean number of nucleotide substitution per synonymous site, whereas the same pattern was not observed in other structural regions, such as the signal and propiece peptide regions, suggesting that these antimicrobial peptide genes have been experiencing rapid diversification driven by Darwinian selection. We cloned and sequenced seven genes amplified from skin or liver genomic DNA. These genes have three exons and share the same gene structure, in which both maximin and maximin H are encoded by the third exon. This suggests that alternative splicing and somatic recombination are less likely to play a role in creating the diversity of maximins and maximin Hs. The gene trees based on different domain regions revealed that domain shuffling or gene conversion among these genes might have happened frequently.

The growing interest in innovative reactors and advanced fuel cycle designs requires more accurate prediction of various transuranic actinide concentrations during irradiation or following discharge because of their effect on reactivity or spent-fuel emissions, such as gamma and neutron activity and decay heat. In this respect, many of the important actinides originate from the 241Am(n,γ) reaction, which leads to either the ground or the metastable state of 242Am. The branching ratio for this reaction depends on the incident neutron energy and has very large uncertainty in the current evaluated nuclear data files. This study examines the effect of accounting for the energy dependence of the 241Am(n,γ) reaction branching ratio calculated from different evaluated data files for different reactor and fuel types on the reactivity and concentrations of some important actinides. The results of the study confirm that the uncertainty in knowing the 241Am(n,γ) reaction branching ratio has a negligible effect on the characteristics of conventional light water reactor fuel. However, in advanced reactors with large loadings of actinides in general, and 241Am in particular, the branching ratio data calculated from the different data files may lead to significant differences in the prediction of the fuel criticality and isotopic composition. Moreover, it was found that neutron energy spectrum weighting of the branching ratio in each analyzed case is particularly important and may result in up to a factor of 2 difference in the branching ratio value. Currently, most of the neutronic codes have a single branching ratio value in their data libraries, which is sometimes difficult or impossible to update in accordance with the neutron spectrum shape for the analyzed system.

The pathogenic process of highly pathogenic avian influenza virus (HPAIV) infection is poorly understood. To explore the differential expression of kidney genes as a result of HPAIV infection, two cDNA libraries were constructed from uninfected and infected kidneys by suppression subtractive hybridization (SSH). Fifteen genes including IFN-stimulated genes (ISG12), lymphocyte antigen 6 complex locus E gene (LY6E), matrix Gla protein gene (MGP), lysozyme gene, haemopoiesis related membrane protein I gene, KIAA1259, MGC68696, G6pe-prov protein gene (G6PC), MGC4504, alcohol dehydrogenase gene (ADH), glutathione S-transferase gene (GST), sodium-dependent high-affinity dicarboxylate transporter gene (SDCT), Synaptotagmin XV (SytXV) and two novel genes were found significantly up-regulated or dramatically suppressed. Differential expression of these genes was further identified by Northern blot. Functional analysis indicated that the regulation of their expression might contribute to the pathogenic process of HPAIV infection. In contrast, the increased expression of three IFN-stimulated genes named ISG12, LY6E, and haemopoiesis related membrane protein 1 gene might reflect host defense responses. Further study showed that ISG12 protein failed to directly interact with NS1 protein of HPAIV which expressed simultaneously in the organs where HPAIV replication occurred, by use of BacterioMatch two-hybrid system. Therefore, our findings may provide new insights into understanding the molecular mechanism underlying the pathophysiological process of HPAIV infection in chicken. (c) 2007 Elsevier Ltd. Cross-species nuclear transfer (NT) has been used to retain the genetic viability of a species near extinction. However, unlike intra-species NT, most embryos produced by cross-species NT were unable to develop to later stages due to incompatible nucleocytoplasmic interactions between the donor nuclei and the recipient cytoplasm from different species. To study the early nucleocytoplasmic interaction in cross-species NT, two laboratory fish species (zebrafish and rare minnow) from different subfamilies were used to generate cross-subfamily NT embryos in the present study. Suppression subtractive hybridization (SSH) was performed to screen out differentially expressed genes from the forward and reverse subtractive cDNA libraries. After dot blot and real-time PCR analysis, 80 of 500 randomly selective sequences were proven to be differentially expressed in the cloned embryos. Among them, 45 sequences shared high homology with 28 zebrafish known genes, and 35 sequences were corresponding to 22 novel expressed sequence tags (ESTs). Based on functional clustering and literature mining analysis, up-and down-regulated genes in the cross-subfamily cloned embryos were mostly relevant to transcription and translation initiation, cell cycle regulation, protein binding, etc. To our knowledge, this is the first report on the determination of genes involved in the early development of cross-species NT embryos of fish. We present the Unified Form Language (UFL), which is a domain-specific language for representing weak formulations of partial differential equations with a view to numerical approximation. Features of UFL include support for variational forms and functionals, automatic differentiation of forms and expressions, arbitrary function space hierarchies formultifield problems, general differential operators and flexible tensor algebra. With these features, UFL has been used to effortlessly express finite element methods for complex systems of partial differential equations in near-mathematical notation, resulting in compact, intuitive and readable programs. We present in this work the language and its construction. An implementation of UFL is freely available as an open-source software library. The library generates abstract syntax tree representations of variational problems, which are used by other software libraries to generate concrete low-level implementations. Some application examples are presented and libraries that support UFL are highlighted. © 2014 ACM.

Immunological methods have been developed for the diagnosis of Myxobolus rotundus but their use has been limited for the prevention and therapy of this serious parasitic pathogen. Phage display antibody libraries are a powerful technique for the development of antibodies to molecules of interest and have advantages over traditional hybridroma approaches. In the present study, four antigen fractions related to M. rotundus were prepared and a combined phage display single-chain antibody fragments (ScFv) library was constructed against this parasite. Preliminary analysis indicated that a combined antibody library of about 2.08 X 10(5) individual clones and high diversity was generated. After four rounds of screening (bio-panning) against soluble spore protein prepared from lysed, intact, mature M rotundus spores, a strain monoclonal phage display ScFv, termed pCAN-6H9, with better affinity, was isolated. The pCAN-6H9 gene fragment was sequenced and analysed. The specificity of pCAN-6H9 was further demonstrated by dot-blot. In competition enzyme-linked immunosorbent assay, both the original and enriched phage-displayed ScFv repertoire showed significant inhibition of mouse anti-M rotundus serum binding to coated antigen, while the inhibition rate of monoclonal pCAN-6H9 phage particles was only 11.83%.

From (GATA)(n) and (AAAG)(n) enriched genomic libraries for the Chinese sturgeon (Acipenser sinensis), 50 primer pairs were developed using the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol. Forty-six primer pairs exhibited highly polymorphic with two to 11 alleles per locus, while the rest four displayed monomorphic. These markers yielded 246 alleles in a survey of eight specimens of wild A. sinensis. Average observed heterozygosity ranged from 0.13 to 1.00. These loci should provide sufficient levels of genetic diversity to allow parentage analysis for artificial stocking management and delineation of fine-scale population structure.
