Los análisis de fiabilidad representan una herramienta adecuada para contemplar las incertidumbres inherentes que existen en los parámetros geotécnicos. En esta Tesis Doctoral se desarrolla una metodología basada en una linealización sencilla, que emplea aproximaciones de primer o segundo orden, para evaluar eficientemente la fiabilidad del sistema en los problemas geotécnicos. En primer lugar, se emplean diferentes métodos para analizar la fiabilidad de dos aspectos propios del diseño de los túneles: la estabilidad del frente y el comportamiento del sostenimiento. Se aplican varias metodologías de fiabilidad — el Método de Fiabilidad de Primer Orden (FORM), el Método de Fiabilidad de Segundo Orden (SORM) y el Muestreo por Importancia (IS). Los resultados muestran que los tipos de distribución y las estructuras de correlación consideradas para todas las variables aleatorias tienen una influencia significativa en los resultados de fiabilidad, lo cual remarca la importancia de una adecuada caracterización de las incertidumbres geotécnicas en las aplicaciones prácticas. Los resultados también muestran que tanto el FORM como el SORM pueden emplearse para estimar la fiabilidad del sostenimiento de un túnel y que el SORM puede mejorar el FORM con un esfuerzo computacional adicional aceptable. Posteriormente, se desarrolla una metodología de linealización para evaluar la fiabilidad del sistema en los problemas geotécnicos. Esta metodología solamente necesita la información proporcionada por el FORM: el vector de índices de fiabilidad de las funciones de estado límite (LSFs) que componen el sistema y su matriz de correlación. Se analizan dos problemas geotécnicos comunes —la estabilidad de un talud en un suelo estratificado y un túnel circular excavado en roca— para demostrar la sencillez, precisión y eficiencia del procedimiento propuesto. Asimismo, se reflejan las ventajas de la metodología de linealización con respecto a las herramientas computacionales alternativas. Igualmente se muestra que, en el caso de que resulte necesario, se puede emplear el SORM —que aproxima la verdadera LSF mejor que el FORM— para calcular estimaciones más precisas de la fiabilidad del sistema. Finalmente, se presenta una nueva metodología que emplea Algoritmos Genéticos para identificar, de manera precisa, las superficies de deslizamiento representativas (RSSs) de taludes en suelos estratificados, las cuales se emplean posteriormente para estimar la fiabilidad del sistema, empleando la metodología de linealización propuesta. Se adoptan tres taludes en suelos estratificados característicos para demostrar la eficiencia, precisión y robustez del procedimiento propuesto y se discuten las ventajas del mismo con respecto a otros métodos alternativos. Los resultados muestran que la metodología propuesta da estimaciones de fiabilidad que mejoran los resultados previamente publicados, enfatizando la importancia de hallar buenas RSSs —y, especialmente, adecuadas (desde un punto de vista probabilístico) superficies de deslizamiento críticas que podrían ser no-circulares— para obtener estimaciones acertadas de la fiabilidad de taludes en suelos. Reliability analyses provide an adequate tool to consider the inherent uncertainties that exist in geotechnical parameters. This dissertation develops a simple linearization-based approach, that uses first or second order approximations, to efficiently evaluate the system reliability of geotechnical problems. First, reliability methods are employed to analyze the reliability of two tunnel design aspects: face stability and performance of support systems. Several reliability approaches —the first order reliability method (FORM), the second order reliability method (SORM), the response surface method (RSM) and importance sampling (IS)— are employed, with results showing that the assumed distribution types and correlation structures for all random variables have a significant effect on the reliability results. This emphasizes the importance of an adequate characterization of geotechnical uncertainties for practical applications. Results also show that both FORM and SORM can be used to estimate the reliability of tunnel-support systems; and that SORM can outperform FORM with an acceptable additional computational effort. A linearization approach is then developed to evaluate the system reliability of series geotechnical problems. The approach only needs information provided by FORM: the vector of reliability indices of the limit state functions (LSFs) composing the system, and their correlation matrix. Two common geotechnical problems —the stability of a slope in layered soil and a circular tunnel in rock— are employed to demonstrate the simplicity, accuracy and efficiency of the suggested procedure. Advantages of the linearization approach with respect to alternative computational tools are discussed. It is also found that, if necessary, SORM —that approximates the true LSF better than FORM— can be employed to compute better estimations of the system’s reliability. Finally, a new approach using Genetic Algorithms (GAs) is presented to identify the fully specified representative slip surfaces (RSSs) of layered soil slopes, and such RSSs are then employed to estimate the system reliability of slopes, using our proposed linearization approach. Three typical benchmark-slopes with layered soils are adopted to demonstrate the efficiency, accuracy and robustness of the suggested procedure, and advantages of the proposed method with respect to alternative methods are discussed. Results show that the proposed approach provides reliability estimates that improve previously published results, emphasizing the importance of finding good RSSs —and, especially, good (probabilistic) critical slip surfaces that might be non-circular— to obtain good estimations of the reliability of soil slope systems.

In this work we review some earlier distributed algorithms developed by the authors and collaborators, which are based on two different approaches, namely, distributed moment estimation and distributed stochastic approximations. We show applications of these algorithms on image compression, linear classification and stochastic optimal control. In all cases, the benefit of cooperation is clear: even when the nodes have access to small portions of the data, by exchanging their estimates, they achieve the same performance as that of a centralized architecture, which would gather all the data from all the nodes.

Many computer vision and human-computer interaction applications developed in recent years need evaluating complex and continuous mathematical functions as an essential step toward proper operation. However, rigorous evaluation of this kind of functions often implies a very high computational cost, unacceptable in real-time applications. To alleviate this problem, functions are commonly approximated by simpler piecewise-polynomial representations. Following this idea, we propose a novel, efficient, and practical technique to evaluate complex and continuous functions using a nearly optimal design of two types of piecewise linear approximations in the case of a large budget of evaluation subintervals. To this end, we develop a thorough error analysis that yields asymptotically tight bounds to accurately quantify the approximation performance of both representations. It provides an improvement upon previous error estimates and allows the user to control the trade-off between the approximation error and the number of evaluation subintervals. To guarantee real-time operation, the method is suitable for, but not limited to, an efficient implementation in modern Graphics Processing Units (GPUs), where it outperforms previous alternative approaches by exploiting the fixed-function interpolation routines present in their texture units. The proposed technique is a perfect match for any application requiring the evaluation of continuous functions, we have measured in detail its quality and efficiency on several functions, and, in particular, the Gaussian function because it is extensively used in many areas of computer vision and cybernetics, and it is expensive to evaluate.

La mecanización de las labores del suelo es la causa, por su consumo energético e impacto directo sobre el medio ambiente, que más afecta a la degradación y pérdida de productividad de los suelos. Entre los factores de disminución de la productividad se deben considerar la compactación, la erosión, el encostramiento y la pérdida de estructura. Todo esto obliga a cuidar el manejo agrícola de los suelos tratando de mejorar las condiciones del suelo y elevar sus rendimientos sin comprometer aspectos económicos, ecológicos y ambientales. En el presente trabajo se adecuan los parámetros constitutivos del modelo de Drucker Prager Extendido (DPE) que definen la fricción y la dilatancia del suelo en la fase de deformación plástica, para minimizar los errores en las predicciones durante la simulación de la respuesta mecánica de un Vertisol mediante el Método de Elementos Finitos. Para lo cual inicialmente se analizaron las bases teóricas que soportan este modelo, se determinaron las propiedades y parámetros físico-mecánicos del suelo requeridos como datos de entrada por el modelo, se determinó la exactitud de este modelo en las predicciones de la respuesta mecánica del suelo, se estimaron mediante el método de aproximación de funciones de Levenberg-Marquardt los parámetros constitutivos que definen la trayectoria de la curva esfuerzo-deformación plástica. Finalmente se comprobó la exactitud de las predicciones a partir de las adecuaciones realizadas al modelo. Los resultados permitieron determinar las propiedades y parámetros del suelo, requeridos como datos de entrada por el modelo, mostrando que su magnitud está en función su estado de humedad y densidad, además se obtuvieron los modelos empíricos de estas relaciones exhibiendo un R2>94%. Se definieron las variables que provocan las inexactitudes del modelo constitutivo (ángulo de fricción y dilatancia), mostrando que las mismas están relacionadas con la etapa de falla y deformación plástica. Finalmente se estimaron los valores óptimos de estos ángulos, disminuyendo los errores en las predicciones del modelo DPE por debajo del 4,35% haciéndelo adecuado para la simulación de la respuesta mecánica del suelo investigado. ABSTRACT The mechanization using farming techniques is one of the main factors that affects the most the soil, causing its degradation and loss of productivity, because of its energy consumption and direct impact on the environment. Compaction, erosion, crusting and loss of structure should be considered among the factors that decrease productivity. All this forces the necessity to take care of the agricultural-land management trying to improve soil conditions and increase yields without compromising economic, ecological and environmental aspects. The present study was aimed to adjust the parameters of the Drucker-Prager Extended Model (DPE), defining friction and dilation of soil in plastic deformation phase, in order to minimize the error of prediction when simulating the mechanical response of a Vertisol through the fine element method. First of all the theoretic fundamentals that withstand the model were analyzed. The properties and physical-mechanical parameters of the soil needed as input data to initialize the model, were established. And the precision of the predictions for the mechanical response of the soil was assessed. Then the constitutive parameters which define the path of the plastic stress-strain curve were estimated through Levenberg-Marquardt method of function approximations. Lastly the accuracy of the predictions from the adequacies made to the model was tested. The results permitted to determine those properties and parameters of the soil, needed in order to initialize the model. It showed that their magnitude is in function of density and humidity. Moreover, the empirical models from these relations were obtained: R2>94%. The variables producing inaccuracies in the constitutive model (angle of repose and dilation) were defined, and there was showed that they are linked with the plastic deformation and rupture point. Finally the optimal values of these angles were established, obtaining thereafter error values for the DPE model under 4, 35%, and making it suitable for the simulation of the mechanical response of the soil under study.

Leyendo distintos artículos en la Revista de Obras Públicas (Jiménez Salas, 1945) uno recuerda a las grandes figuras como Coulomb (1773), Poncelet (1840), Rankine (1856), Culmann (1866), Mohr (1871), Boussinesq (1876) y otros muchos, que construyeron la base de un conocimiento que poco a poco irían facilitando la complicada tarea que suponía la construcción. Pero sus avances eran aproximaciones que presentaban notables diferencias frente al comportamiento de la naturaleza. Esas discrepancias con la naturaleza llegó un momento que se hicieron demasiado patentes. Importantes asientos en la construcción de los modernos edificios, rotura de presas de materiales sueltos y grandes corrimientos de tierras, por ejemplo durante la construcción del canal de Panamá, llevaron a la Sociedad Americana de Ingenieros Civiles (ASCE) a crear un comité que analizase las prácticas de la construcción de la época. Hechos similares se producían en Europa, por ejemplo en desmontes para ferrocarriles, que en el caso de Suecia supusieron unas cuantiosas perdidas materiales y humanas. El ingeniero austriaco-americano Karl Terzaghi (1883) había podido comprobar, en su práctica profesional, la carencia de conocimientos para afrontar muchos de los retos que la naturaleza ofrecía. Inicialmente buscó la respuesta en la geología pero encontró que ésta carecía de la definición necesaria para la práctica de la ingeniería, por lo que se lanzó a una denodada tarea investigadora basada en el método experimental. Comenzó en 1917 con escasos medios, pero pronto llegó a desarrollar algunos ensayos que le permitieron establecer los primeros conceptos de una nueva ciencia, la Mecánica de Suelos. Ciencia que ve la luz en 1925 con la publicación de su libro Erdbaumechanik auf bodenphysikalischer Grundlage. Rápidamente otras figuras empezaron a hacer sus contribuciones científicas y de divulgación, como es el caso del ingeniero austriaco-americano Arthur Casagrande (1902), cuya iniciativa de organizar el primer Congreso Internacional de Mecánica de Suelos e Ingeniería de Cimentaciones proporcionó el altavoz que necesitaba esa nueva ciencia para su difusión. Al mismo tiempo, más figuras internacionales se fueron uniendo a este período de grandes avances e innovadores puntos de vista. Figuras como Alec Skempton (1914) en el Reino Unido, Ralph Peck (1912) en los Estados Unidos o Laurits Bjerrum (1918) en Noruega sobresalieron entre los grandes de la época. Esta tesis investiga las vidas de estos geotécnicos, artífices de múltiples avances científicos de la nueva ciencia denominada Mecánica de Suelos. Todas estas grandes figuras de la geotecnia fueron presidentes, en distintos periodos, de la Sociedad Internacional de Mecánica de Suelos e Ingeniería de Cimentaciones. Se deja constancia de ello en las biografías que han sido elaboradas a partir de fuentes de variada procedencia y de los datos cruzados encontrados sobre estos extraordinarios geotécnicos. Así, las biografías de Terzaghi, Casagrande, Skempton, Peck y Bjerrum contribuyen no solo a su conocimiento individual sino que constituyen conjuntamente un punto de vista privilegiado para la comprensión de los acontecimientos vividos por la Mecánica de Suelos en el segundo tercio del siglo XX, extendiéndose en algunos casos hasta los albores del siglo XXI. Las aportaciones científicas de estos geotécnicos encuentran también su lugar en la parte técnica de esta tesis, en la que sus contribuciones individuales iniciales que configuran los distintos capítulos conservan sus puntos de vista originales, lo que permite tener una visión de los principios de la Mecánica de Suelos desde su mismo origen. On reading several articles in the journal, Revista de Obras Públicas (Jiménez Salas, 1945), one recalls such leading figures as Coulomb (1773), Poncelet (1840), Rankine (1856), Culmann (1866), Mohr (1871) and Boussinesq (1876) among many others, who created the basis of scientific knowledge that would make the complicated task of construction progressively easier. However, their advances were approximations which suffered considerable discrepancies when faced with the behaviour of the forces of nature. There came a time when such discrepancies became all too evident. Substantial soil settlements when constructing modern buildings, embankment dam failures and grave landslides, during the construction of the Panama Canal for example, led the American Society of Civil Engineers (ASCE) to form a committee in order to analyse construction practices of the time. Similar incidents had taken place in Europe, for example with railway slides, which in the case of Sweden, had resulted in heavy losses in both materials and human lives. During the practice of his career, the Austrian-American engineer Karl Terzaghi (1883) had encountered the many challenges posed by the forces of nature and the lack of knowledge at his disposal with which to overcome them. Terzaghi first sought a solution in geology only to discover that this lacked the necessary accuracy for the practice of engineering. He therefore threw himself into tireless research based on the experimental method. He began in 1917 on limited means but soon managed to develop several tests, which would allow him to establish the basic fundamentals of a new science; Soil Mechanics, a science which first saw the light of day on the publication of Terzaghi’s book, Erdbaumechanik auf bodenphysikalischer Grundlage. Other figures were quick to make their own scientific contributions. Such was the case of Austrian-American engineer, Arthur Casagrande (1902), whose initiative to organize the first International Congress of Soil Mechanics and Foundation Engineering provided the springboard that this science needed. At the same time, other international figures were becoming involved in this period of great advances and innovative concepts. Figures including the likes of Alec Skempton (1914) in the United Kingdom, Ralph Peck (1912) in the United States, and Laurits Bjerrum (1918) in Norway stood out amongst the greatest of their time. This thesis investigates the lives of these geotechnical engineers to whom we are indebted for a great many scientific advances in this new science known as Soil Mechanics. Moreover, each of these eminent figures held the presidency of the International Society of Soil Mechanics and Foundation Engineering, record of which can be found in their biographies, drawn from diverse sources, and by crosschecking and referencing all the available information on these extraordinary geotechnical engineers. Thus, the biographies of Terzaghi, Casagrande, Skempton, Peck and Bjerrum not only serve to provide knowledge on the individual, but moreover, as a collective, they present us with an exceptional insight into the important developments which took place in Soil Mechanics in the second third of the 20th century, and indeed, in some cases, up to the dawn of the 21st. The scientific contributions of these geotechnical engineers also find their place in the technical part of this thesis in which the initial individual contributions which make up several chapters retain their original approaches allowing us a view of the principles of Soil Mechanics from its very beginnings.

La metodología Integrated Safety Analysis (ISA), desarrollada en el área de Modelación y Simulación (MOSI) del Consejo de Seguridad Nuclear (CSN), es un método de Análisis Integrado de Seguridad que está siendo evaluado y analizado mediante diversas aplicaciones impulsadas por el CSN; el análisis integrado de seguridad, combina las técnicas evolucionadas de los análisis de seguridad al uso: deterministas y probabilistas. Se considera adecuado para sustentar la Regulación Informada por el Riesgo (RIR), actual enfoque dado a la seguridad nuclear y que está siendo desarrollado y aplicado en todo el mundo. En este contexto se enmarcan, los proyectos Safety Margin Action Plan (SMAP) y Safety Margin Assessment Application (SM2A), impulsados por el Comité para la Seguridad de las Instalaciones Nucleares (CSNI) de la Agencia de la Energía Nuclear (NEA) de la Organización para la Cooperación y el Desarrollo Económicos (OCDE) en el desarrollo del enfoque adecuado para el uso de las metodologías integradas en la evaluación del cambio en los márgenes de seguridad debidos a cambios en las condiciones de las centrales nucleares. El comité constituye un foro para el intercambio de información técnica y de colaboración entre las organizaciones miembro, que aportan sus propias ideas en investigación, desarrollo e ingeniería. La propuesta del CSN es la aplicación de la metodología ISA, especialmente adecuada para el análisis según el enfoque desarrollado en el proyecto SMAP que pretende obtener los valores best-estimate con incertidumbre de las variables de seguridad que son comparadas con los límites de seguridad, para obtener la frecuencia con la que éstos límites son superados. La ventaja que ofrece la ISA es que permite el análisis selectivo y discreto de los rangos de los parámetros inciertos que tienen mayor influencia en la superación de los límites de seguridad, o frecuencia de excedencia del límite, permitiendo así evaluar los cambios producidos por variaciones en el diseño u operación de la central que serían imperceptibles o complicados de cuantificar con otro tipo de metodologías. La ISA se engloba dentro de las metodologías de APS dinámico discreto que utilizan la generación de árboles de sucesos dinámicos (DET) y se basa en la Theory of Stimulated Dynamics (TSD), teoría de fiabilidad dinámica simplificada que permite la cuantificación del riesgo de cada una de las secuencias. Con la ISA se modelan y simulan todas las interacciones relevantes en una central: diseño, condiciones de operación, mantenimiento, actuaciones de los operadores, eventos estocásticos, etc. Por ello requiere la integración de códigos de: simulación termohidráulica y procedimientos de operación; delineación de árboles de sucesos; cuantificación de árboles de fallos y sucesos; tratamiento de incertidumbres e integración del riesgo. La tesis contiene la aplicación de la metodología ISA al análisis integrado del suceso iniciador de la pérdida del sistema de refrigeración de componentes (CCWS) que genera secuencias de pérdida de refrigerante del reactor a través de los sellos de las bombas principales del circuito de refrigerante del reactor (SLOCA). Se utiliza para probar el cambio en los márgenes, con respecto al límite de la máxima temperatura de pico de vaina (1477 K), que sería posible en virtud de un potencial aumento de potencia del 10 % en el reactor de agua a presión de la C.N. Zion. El trabajo realizado para la consecución de la tesis, fruto de la colaboración de la Escuela Técnica Superior de Ingenieros de Minas y Energía y la empresa de soluciones tecnológicas Ekergy Software S.L. (NFQ Solutions) con el área MOSI del CSN, ha sido la base para la contribución del CSN en el ejercicio SM2A. Este ejercicio ha sido utilizado como evaluación del desarrollo de algunas de las ideas, sugerencias, y los algoritmos detrás de la metodología ISA. Como resultado se ha obtenido un ligero aumento de la frecuencia de excedencia del daño (DEF) provocado por el aumento de potencia. Este resultado demuestra la viabilidad de la metodología ISA para obtener medidas de las variaciones en los márgenes de seguridad que han sido provocadas por modificaciones en la planta. También se ha mostrado que es especialmente adecuada en escenarios donde los eventos estocásticos o las actuaciones de recuperación o mitigación de los operadores pueden tener un papel relevante en el riesgo. Los resultados obtenidos no tienen validez más allá de la de mostrar la viabilidad de la metodología ISA. La central nuclear en la que se aplica el estudio está clausurada y la información relativa a sus análisis de seguridad es deficiente, por lo que han sido necesarias asunciones sin comprobación o aproximaciones basadas en estudios genéricos o de otras plantas. Se han establecido tres fases en el proceso de análisis: primero, obtención del árbol de sucesos dinámico de referencia; segundo, análisis de incertidumbres y obtención de los dominios de daño; y tercero, cuantificación del riesgo. Se han mostrado diversas aplicaciones de la metodología y ventajas que presenta frente al APS clásico. También se ha contribuido al desarrollo del prototipo de herramienta para la aplicación de la metodología ISA (SCAIS). ABSTRACT The Integrated Safety Analysis methodology (ISA), developed by the Consejo de Seguridad Nuclear (CSN), is being assessed in various applications encouraged by CSN. An Integrated Safety Analysis merges the evolved techniques of the usually applied safety analysis methodologies; deterministic and probabilistic. It is considered as a suitable tool for assessing risk in a Risk Informed Regulation framework, the approach under development that is being adopted on Nuclear Safety around the world. In this policy framework, the projects Safety Margin Action Plan (SMAP) and Safety Margin Assessment Application (SM2A), set up by the Committee on the Safety of Nuclear Installations (CSNI) of the Nuclear Energy Agency within the Organization for Economic Co-operation and Development (OECD), were aimed to obtain a methodology and its application for the integration of risk and safety margins in the assessment of the changes to the overall safety as a result of changes in the nuclear plant condition. The committee provides a forum for the exchange of technical information and cooperation among member organizations which contribute their respective approaches in research, development and engineering. The ISA methodology, proposed by CSN, specially fits with the SMAP approach that aims at obtaining Best Estimate Plus Uncertainty values of the safety variables to be compared with the safety limits. This makes it possible to obtain the exceedance frequencies of the safety limit. The ISA has the advantage over other methods of allowing the specific and discrete evaluation of the most influential uncertain parameters in the limit exceedance frequency. In this way the changes due to design or operation variation, imperceptibles or complicated to by quantified by other methods, are correctly evaluated. The ISA methodology is one of the discrete methodologies of the Dynamic PSA framework that uses the generation of dynamic event trees (DET). It is based on the Theory of Stimulated Dynamics (TSD), a simplified version of the theory of Probabilistic Dynamics that allows the risk quantification. The ISA models and simulates all the important interactions in a Nuclear Power Plant; design, operating conditions, maintenance, human actuations, stochastic events, etc. In order to that, it requires the integration of codes to obtain: Thermohydraulic and human actuations; Even trees delineation; Fault Trees and Event Trees quantification; Uncertainty analysis and risk assessment. This written dissertation narrates the application of the ISA methodology to the initiating event of the Loss of the Component Cooling System (CCWS) generating sequences of loss of reactor coolant through the seals of the reactor coolant pump (SLOCA). It is used to test the change in margins with respect to the maximum clad temperature limit (1477 K) that would be possible under a potential 10 % power up-rate effected in the pressurized water reactor of Zion NPP. The work done to achieve the thesis, fruit of the collaborative agreement of the School of Mining and Energy Engineering and the company of technological solutions Ekergy Software S.L. (NFQ Solutions) with de specialized modeling and simulation branch of the CSN, has been the basis for the contribution of the CSN in the exercise SM2A. This exercise has been used as an assessment of the development of some of the ideas, suggestions, and algorithms behind the ISA methodology. It has been obtained a slight increase in the Damage Exceedance Frequency (DEF) caused by the power up-rate. This result shows that ISA methodology allows quantifying the safety margin change when design modifications are performed in a NPP and is specially suitable for scenarios where stochastic events or human responses have an important role to prevent or mitigate the accidental consequences and the total risk. The results do not have any validity out of showing the viability of the methodology ISA. Zion NPP was retired and information of its safety analysis is scarce, so assumptions without verification or approximations based on generic studies have been required. Three phases are established in the analysis process: first, obtaining the reference dynamic event tree; second, uncertainty analysis and obtaining the damage domains; third, risk quantification. There have been shown various applications of the methodology and advantages over the classical PSA. It has also contributed to the development of the prototype tool for the implementation of the ISA methodology (SCAIS).

The calculation of the energy spectrum and absorption coefficients of quantum dot nanostructured intermediate band solar cells using the Empiric K·P Hamiltonian method and its agreement with experimental data are summarized. The well established Luttinger Kohn Hamiltonian modified by Pikus and Bir for strained material, such as quantum dot arrays, is presented using a simplified strain field that allows for square band offsets. The energy spectrum and absorption coefficients are calculated with this new Hamiltonian. With the approximations made the energy spectrum results to be exactly the same but the absorption coefficient fits experiments less accurately. The computer time using the latter Hamiltonian is much longer than the former one.

A crítica da religião é um tema recorrente no pensamento moderno e aparece até mesmo na teologia desse período. Nesse contexto, o presente estudo procura comparar a crítica da religião no pensamento de Karl Barth e Dietrich Bonhoeffer. Para tanto, a dissertação está organizada em quatro partes principais. Inicialmente, é feita uma contextualização da concepção ocidental de religião e sua crítica moderna, inclusive no âmbito teológico. A seguir são descritas a concepção e a crítica da religião no pensamento de Barth e Bonhoeffer. Finalmente, é realizada uma comparação entre ambos, que procura delinear aproximações e distanciamentos da crítica da religião desses dois teólogos. De maneira ampla, as duas críticas apontam distorções do Cristianismo e indicam propostas de restauração. Como chave geral de comparação está a percepção de que Barth critica a religião n a perspectiva da revelação enquanto Bonhoeffer faz sua crítica na perspectiva da vida.(AU)

A prática do ioga tem se tornado cada vez mais popular, não apenas pelos benefícios físicos, mas principalmente pelo bem-estar psicológico trazido pela sua prática. Um dos componentes do ioga é o Prãnãyama, ou controle da respiração. A atenção e a respiração são dois mecanismos fisiológicos e involuntários requeridos para a execução do Prãnãyama. O principal objetivo desse estudo foi verificar se variáveis contínuas do EEG (potência de diferentes faixas que o compõem) seriam moduladas pelo controle respiratório, comparando-se separadamente as duas fases do ciclo respiratório (inspiração e expiração), na situação de respiração espontânea e controlada. Fizeram parte do estudo 19 sujeitos (7 homens/12 mulheres, idade média de 36,89 e DP = ± 14,46) que foram convidados a participar da pesquisa nas dependências da Faculdade de Saúde da Universidade Metodista de São Paulo. Para o registro do eletroencefalograma foi utilizado um sistema de posicionamento de cinco eletrodos Ag AgCl (FPz, Fz, Cz, Pz e Oz) fixados a uma touca de posicionamento rápido (Quick-Cap, Neuromedical Supplies®), em sistema 10-20. Foram obtidos valores de máxima amplitude de potência (espectro de potência no domínio da frequência) nas frequências teta, alfa e beta e delta e calculada a razão teta/beta nas diferentes fases do ciclo respiratório (inspiração e expiração), separadamente, nas condições de respiração espontânea e de controle respiratório. Para o registro do ciclo respiratório, foi utilizada uma cinta de esforço respiratório M01 (Pletismógrafo). Os resultados mostram diferenças significativas entre as condições de respiração espontânea e de controle com valores das médias da razão teta/beta menores na respiração controlada do que na respiração espontânea e valores de média da potência alfa sempre maiores no controle respiratório. Diferenças significativas foram encontradas na comparação entre inspiração e expiração da respiração controlada com diminuição dos valores das médias da razão teta/beta na inspiração e aumento nos valores das médias da potência alfa, sobretudo na expiração. Os achados deste estudo trazem evidências de que o controle respiratório modula variáveis eletrofisiológicas relativas à atenção refletindo um estado de alerta, porém mais relaxado do que na situação de respiração espontânea.

The mathematical underpinning of the pulse width modulation (PWM) technique lies in the attempt to represent “accurately” harmonic waveforms using only square forms of a fixed height. The accuracy can be measured using many norms, but the quality of the approximation of the analog signal (a harmonic form) by a digital one (simple pulses of a fixed high voltage level) requires the elimination of high order harmonics in the error term. The most important practical problem is in “accurate” reproduction of sine-wave using the same number of pulses as the number of high harmonics eliminated. We describe in this paper a complete solution of the PWM problem using Padé approximations, orthogonal polynomials, and solitons. The main result of the paper is the characterization of discrete pulses answering the general PWM problem in terms of the manifold of all rational solutions to Korteweg-de Vries equations.

CD38 ligation on mouse B cells by CS/2, an anti-mouse CD38 mAb, induced proliferation, interleukin 5 (IL-5) receptor α chain expression, and tyrosine phosphorylation of Bruton tyrosine kinase (Btk) from wild-type, but not from X chromosome-linked, immunodeficient mice. B cells from fyn-deficient (Fyn−/−) and lyn-deficient (Lyn−/−) mice showed an impaired response to mAb CS/2 for proliferation and IL-5 receptor α chain expression, and B cells from fyn/lyn double-deficient (Fyn/Lyn−/−) mice did not respond at all to mAb CS/2. The Btk activation by CD38 ligation was observed in B cells from Fyn−/− mice, and it was severely impaired in B cells from Lyn−/− and Fyn/Lyn−/− mice. CD38 expression on B cells from three mutant strains was comparable to that on control B cells. We infer from these results that both Fyn and Lyn are required and that their signals are synergistic for B cell triggering after CD38 ligation. Lyn is upstream of Btk activation in the CD38 signaling. Stimulation of B cells with IL-5 together with CD38 ligation induces not only IgM but also IgG1 secretion. Analysis of the synergistic effects of IL-5 and CD38 ligation on IgG1 secretion revealed the impaired IgG1 secretion of B cells from Lyn−/− and Fyn/Lyn−/− mice. These data imply that Lyn is involved in B cell triggering by CD38 ligation plus IL-5 for isotype switching.

Potent and selective active-site-spanning inhibitors have been designed for cathepsin K, a cysteine protease unique to osteoclasts. They act by mechanisms that involve tight binding intermediates, potentially on a hydrolytic pathway. X-ray crystallographic, MS, NMR spectroscopic, and kinetic studies of the mechanisms of inhibition indicate that different intermediates or transition states are being represented that are dependent on the conditions of measurement and the specific groups flanking the carbonyl in the inhibitor. The species observed crystallographically are most consistent with tetrahedral intermediates that may be close approximations of those that occur during substrate hydrolysis. Initial kinetic studies suggest the possibility of irreversible and reversible active-site modification. Representative inhibitors have demonstrated antiresorptive activity both in vitro and in vivo and therefore are promising leads for therapeutic agents for the treatment of osteoporosis. Expansion of these inhibitor concepts can be envisioned for the many other cysteine proteases implicated for therapeutic intervention.

The semaphorins comprise a large family of membrane-bound and secreted proteins, some of which have been shown to function in axon guidance. We have cloned a transmembrane semaphorin, Sema W, that belongs to the class IV subgroup of the semaphorin family. The mouse and rat forms of Sema W show 97% amino acid sequence identity with each other, and each shows about 91% identity with the human form. The gene for Sema W is divided into 15 exons, up to 4 of which are absent in the human cDNAs that we sequenced. Unlike many other semaphorins, Sema W is expressed at low levels in the developing embryo but was found to be expressed at high levels in the adult central nervous system and lung. Functional studies with purified membrane fractions from COS7 cells transfected with a Sema W expression plasmid showed that Sema W has growth-cone collapse activity against retinal ganglion-cell axons, indicating that vertebrate transmembrane semaphorins, like secreted semaphorins, can collapse growth cones. Genetic mapping of human SEMAW with human/hamster radiation hybrids localized the gene to chromosome 2p13. Genetic mapping of mouse Semaw with mouse/hamster radiation hybrids localized the gene to chromosome 6, and physical mapping placed the gene on bacteria artificial chromosomes carrying microsatellite markers D6Mit70 and D6Mit189. This localization places Semaw within the locus for motor neuron degeneration 2, making it an attractive candidate gene for this disease.
