Introduction: The primary somatosensory cortex (SI) contains Brodmann areas (BA) 1, 2, 3a, and 3b. Research in non-human primates showed that BAs 3b, 1, and 2 each contain one full representation of the hand with separate representations for each finger. This research also showed that the finger representation in BA3b has larger and clearer finger somatotopy than BA1 and 2. Although several efforts to map finger somatotopy in SI by fMRI have been made at 1.5 and 3T these studies have yielded variable results and were not able to detect single subject finger somatotopy, probably due to the limited spatial extent of the cortical areas representing a digit (close to the resolution in most fMRI experiments), complications due to acquisition of consistent maps for individual subjects (Schweizer et al 2008), or inter-individual variability in sulcal anatomy impeding group studies. Here, we used 7T fMRI to investigate finger somatotopy in SI, some of its functional characteristics, and its reproducibility. Methods: Eight right-handed male subjects were scanned on a 7T scanner (Siemens Medical, Germany) with an 8-channel Tx/Rx rf-coil (Rapid Biomedical, Germany). 1.3x1.3x1.3mm3 resolution fMRI data were acquired using a sinusoidal readout EPI sequence (Speck et al, 2008) and FOV=210mm, TE/TR=27ms/2.5s, GRAPPA=2. Each volume contained 28 transverse slices covering SI. A single EPI volume with 64 slices was acquired to aid coregistration. 1x1x1mm3 anatomical data were acquire using the MP2RAGE sequence (Marques et al, 2009; TE/TR/TI1,2/TRmprage=2.63ms/7.2ms/0.9,3.2s/5s). Subjects were positioned supine in the scanner with their right arm comfortably against the magnet bore. An experimenter was positioned at the entrance of the bore where he could easily reach and stroke successively the two distal phalanxes of each digit. The order of stroked digit was D1 (thumb)-D3-D5-D2-D4, with 20s ON, 10s OFF alternated. This sequence was repeated four times per run and two functional runs were acquired per subject. Realignment, smoothing (FWHM 2 mm), coregistration of the anatomical to the fMRI data and calculation of t-statistics were done using SPM8. An SI mask was obtained via an F-contrast (p<0.001) over all digits. Within the mask, voxels were labeled with the number of the digit demonstrating the highest t-value for that particular voxel. Results: For all subjects, areas corresponding to the five digits were identified in contralateral SI. BA3b showed the most consistent somatotopic finger representation (see an example in Fig.1). The five digits were localized in a consecutive order in the cortex, with D1 most anterior, inferior and distal and D5, most posterior, superior and medial (mean distance between centres of mass of digit representations ±stderr: 4.2±0.7mm; see Fig. 2). The analysis of average beta values within each finger representation region revealed the specificity of the somatotopic region to the tactile input for each tested finger (except digit 4 and 5). Five of these subjects also presented an orderly and consecutive representation of the five digits in BA1 and 2. Conclusions: Our data reveal that the increased BOLD sensitivity at 7T and the high spatial resolution used in this study allow consistent somatotopic mapping using human touch as a stimulus and that human SI contains at least three separate regions that contain five separate representations of all single contralateral fingers. Moreover, adjacent fingers were represented at adjacent cortical regions across the three SI regions. The spatial organization of SI as reflected in individual subject topography corresponds well with previous electrophysiological data in non-human primates. The small distance between digit representations highlights the need for the high spatial resolution available at 7T.

Aquest treball té com a objecte l’anàlisi de la implantació de la hipertextualitat, la multimedialitat, la interactivitat i la publicitat com a elements vertebradors de la premsa digital internacional. Primerament, es descriuen de forma teòrica els meritats elements. A continuació, es presenten els resultats de l’estudi empíric desenvolupat, analitzant els anteriors factors, sobre quatre capçaleres digitals internacionals. Finalment, concloem que la implantació de les variables estudiades es troba en fase d'expansió, tot i que encara lluny del nivell òptim d'explotació; també que els diaris europeus són més proclius a fomentar l'ús de la hipertextualitat, mentre els americans prefereixen la multimedialitat; que els principals problemes a superar seran les formes tradicionals de pensar i treballar així com la falta d'inversió generalitzada i, per acabar, que el futur passarà per continuar en la línia de treball sense oblidar-se d'evolucionar.

Este trabalho reporta a utilização de ferramentas de um SIG (Sistema de Informação Geográfica) para o delineamento das geoformas e pedopaisagens em um estudo de caso em microbacia do município de Viçosa (MG), a partir dos dados da carta topográfica (escala 1:50.000) da região. Trabalhos de campo permitiram aferir uma exatidão global de 72 % na classificação das formas da paisagem e correlacionar as classes obtidas com as classes de solos dominantes. Após ajustes, elaborou-se um mapa síntese preliminar de distribuição de pedoformas, que auxilia no mapeamento de solos. O método de classificação das formas da paisagem baseado na geração de um MNT (Modelo Numérico do Terreno) e no cálculo dos atributos do relevo (elevação, declividade, curvatura) mostra-se uma alternativa rápida e econômica em comparação ao delineamento manual a partir da utilização de análise estereoscópica de fotografias aéreas.

BACKGROUND: The nuclear receptors are a large family of eukaryotic transcription factors that constitute major pharmacological targets. They exert their combinatorial control through homotypic heterodimerisation. Elucidation of this dimerisation network is vital in order to understand the complex dynamics and potential cross-talk involved. RESULTS: Phylogeny, protein-protein interactions, protein-DNA interactions and gene expression data have been integrated to provide a comprehensive and up-to-date description of the topology and properties of the nuclear receptor interaction network in humans. We discriminate between DNA-binding and non-DNA-binding dimers, and provide a comprehensive interaction map, that identifies potential cross-talk between the various pathways of nuclear receptors. CONCLUSION: We infer that the topology of this network is hub-based, and much more connected than previously thought. The hub-based topology of the network and the wide tissue expression pattern of NRs create a highly competitive environment for the common heterodimerising partners. Furthermore, a significant number of negative feedback loops is present, with the hub protein SHP [NR0B2] playing a major role. We also compare the evolution, topology and properties of the nuclear receptor network with the hub-based dimerisation network of the bHLH transcription factors in order to identify both unique themes and ubiquitous properties in gene regulation. In terms of methodology, we conclude that such a comprehensive picture can only be assembled by semi-automated text-mining, manual curation and integration of data from various sources.

High blood pressure (BP) is more prevalent and contributes to more severe manifestations of cardiovascular disease (CVD) in African Americans than in any other United States ethnic group. Several small African-ancestry (AA) BP genome-wide association studies (GWASs) have been published, but their findings have failed to replicate to date. We report on a large AA BP GWAS meta-analysis that includes 29,378 individuals from 19 discovery cohorts and subsequent replication in additional samples of AA (n = 10,386), European ancestry (EA) (n = 69,395), and East Asian ancestry (n = 19,601). Five loci (EVX1-HOXA, ULK4, RSPO3, PLEKHG1, and SOX6) reached genome-wide significance (p < 1.0 × 10(-8)) for either systolic or diastolic BP in a transethnic meta-analysis after correction for multiple testing. Three of these BP loci (EVX1-HOXA, RSPO3, and PLEKHG1) lack previous associations with BP. We also identified one independent signal in a known BP locus (SOX6) and provide evidence for fine mapping in four additional validated BP loci. We also demonstrate that validated EA BP GWAS loci, considered jointly, show significant effects in AA samples. Consequently, these findings suggest that BP loci might have universal effects across studied populations, demonstrating that multiethnic samples are an essential component in identifying, fine mapping, and understanding their trait variability.

Chloride channels represent a group of targets for major clinical indications. However, molecular screening for chloride channel modulators has proven to be difficult and time-consuming as approaches essentially rely on the use of fluorescent dyes or invasive patch-clamp techniques which do not lend themselves to the screening of large sets of compounds. To address this problem, we have developed a non-invasive optical method, based on digital holographic microcopy (DHM), allowing monitoring of ion channel activity without using any electrode or fluorescent dye. To illustrate this approach, GABA(A) mediated chloride currents have been monitored with DHM. Practically, we show that DHM can non-invasively provide the quantitative determination of transmembrane chloride fluxes mediated by the activation of chloride channels associated with GABA(A) receptors. Indeed through an original algorithm, chloride currents elicited by application of appropriate agonists of the GABA(A) receptor can be derived from the quantitative phase signal recorded with DHM. Finally, chloride currents can be determined and pharmacologically characterized non-invasively simultaneously on a large cellular sampling by DHM.

L'explosió de la informació digital en les col·leccions bibliotecàries d'aquesta dècada passada ens ha ofert molts beneficis, especialment en relació al seu accés, emmagatzematge i ús. No obstant això, queden tota una sèrie de qüestions referents a la preservació a llarg termini d'aquests materials que encara no tenen resposta. Aquest article explora alguns dels aspectes principals, com ara les pròpies dades que cal preservar; les amenaces derivades de l'obsolescència dels mitjans físics d'emmagatzematge, els formats, el programari i el maquinari; les tècniques proposadesmactualment per a la preservació a llarg termini; la importància de conservar també les metadades, i els tipus d'institucions que estan implicades -o que haurien d'estar-hi- en la cerca de les solucions necessàries.

A finales del pasado septiembre de 2005 la Comisión Europea publicó una comunicación con su estrategia sobre el patrimonio impreso y gráfico europeo y las bibliotecas digitales. Esta iniciativa, i2010: digital libraries, pretende garantizar el acceso a estos recursos mediante su digitalización y puesta en línea. Además, el programa hace un llamamiento a la preservación.

Desde 1996 varios países han llevado a cabo estrategias para garantizar el acceso permanente a la producción digital propia: las páginas web y el resto de recursos digitales publicados en internet. Estas estrategias están orientadas a asegurar en la medida de las posibilidades tecnológicas actuales a la adecuación del ciclo documental clásico a las páginas web: la compilación, el procesamiento, la preservación y el acceso permanente a la producción digital. Las bibliotecas nacionales han sido a menudo impulsoras de estas acciones y en el caso español, la Biblioteca de Catalunya (BC) ha puesto en marcha el proyecto Padicat (Patrimonio Digital de Cataluña), dedicado al archivo de la web catalana. Este artículo presenta el proyecto Padicat.

Se presenta el formato de libro multimedia Daisy que permite integrar texto, sonido, imágenes y vídeo de forma sincronizada y con una navegación avanzada por el texto y el audio que lo hace altamente usable para lecturas en digital. También se presenta su componente principal DTBook xml, disposición estructurada del texto que permite generar formatos alternativos. El artículo detalla las diferencias de Daisy respecto a otros audiolibros, los dispositivos de lectura existentes y su adopción en el mercado y en la enseñanza. Si se abaratan los costes de los aparatos lectores y se soluciona la gestión de derechos digitales, tiene muchas posibilidades de convertirse en estrella para editar libros digitales.

Se presenta una panorámica de las acciones de investigación y desarrollo en preservación digital en el ámbito internacional, incluyendo un apartado dedicado a la situación en España. La mayor atención se da en las aplicaciones de preservación digital en bibliotecas y archivos, pero también se presenta someramente la situación en otros entornos como los repositorios institucionales, los archivos administrativos, los archivos personales, la industria aeroespacial y los museos. El análisis de los protagonistas de la investigación lleva a destacar el papel preeminente de algunas bibliotecas y archivos nacionales, de determinados países y una incipiente entrada de empresas de software. En cuanto a las líneas de investigación destaca la integración de herramientas para generar sistemas de preservación integrados.
