Ocean Drilling Program Site 975 is located near the base of the Menorca Rise in the South Balearic Basin of the western Mediterranean Sea. Coring at this site penetrated the Pliocene/Miocene boundary and recovered a sequence of sediments that represent the final stages of salt deposition and the transition from evaporitic to open marine conditions at the end of the Miocene (Messinian). Detailed petrographic observations and bulk mineralogical analyses by X-ray diffraction form the basis for preliminary interpretations of depositional environments for this section. Gypsum is thought to have been deposited in an evaporating basin below wave base. Cycles consisting of a clay layer overlain by gypsiferous chalk, laminated gypsum, and finally pinch-and-swell gypsum suggest upsection increases in salinity. The gypsum section is overlain by two exotic sand layers thought to mark events of fresher water (marine or meteoric) inflow to the basin. Gypsum deposition terminated and was replaced by inorganic precipitation of micritic calcite with periodic, variable dilution by fine-grained terrigenous sediment. The micritic sediments have fine, slightly wavy, laminations indicating either an algal/microbial mat origin, or varve-like fluctuations in deposition, perhaps in a deep basin. The Pliocene/Miocene boundary falls within an interval of banded micritic silty clays that reflect the final environmental fluctuations during the transition to the open marine conditions of the Pliocene.

Abundant iron-titanium (Fe-Ti) oxide gabbro, olivine gabbro, and troctolite were drilled at Hole 735B adjacent to the Atlantis II Fracture Zone of the Southwest Indian Ridge during Leg 118. The Fe-Ti oxide gabbro occurs as intrusive bodies into olivine gabbro with very sharp intrusive contacts. The size of the intrusive bodies varies from a millimeter to a few tens of meters. Mineralogical parameters, such as anorthite content of plagioclase and Mg/(Mg+Fe) ratios of mafic minerals exhibit bimodal distributions corresponding to olivine and Fe-Ti oxide gabbros, respectively. When the two major gabbro types are looked at separately, several downhole mineralogical cycles are recognized. The Fe-Ti oxide gabbros exhibit two such cycles with plagioclase becoming more sodic and mafic minerals becoming more iron-rich downward in the drill core. The olivine gabbros and troctolites, however, exhibit two cycles showing an upward increase in sodium in plagioclase and iron in mafic minerals. The mineralogical variations of these gabbros and the intrusive contact relationships probably resulted from downward intrusion of evolved magma into underlying solid or almost solidified olivine gabbros and troctolite. The dense evolved melt at the top of the cumulus pile probably formed from the crystallization of olivine gabbro cumulates followed by extreme fractional crystallization of residual melt in an isolated, ephemeral magma chamber. The interlayered occurrence of evolved and primitive gabbros from Hole 735B represents a typical section of lower ocean crust formed at a very slow spreading ridge.

The guinea pig may represent an animal model for research on ovarian infertility and improvement of the in vitro maturation (IVM) conditions is needed in this species. The aim of the present work was to immunolocalize the Epidermal Growth Factor (EGF)-Receptor in the guinea pig ovaries and to study the effect of EGF on meiotic and cytoplasmic maturation, and apoptotic rate in cumulus-oocyte-co mplexes (COCs). Immunohistochemistry was performed in paraffined ovaries using a rabbit polyclonal antibody EGF-R (1:100; Santa Cruz Biotechnology) and the ABC Vector Elite kit (Vector Laboratories). For the IVM, COCs were collected by aspiration of follicles >700μm under a stereoscopic microscope.

The results of this study challenge the widely held view that growth hormone (GH) acts only during the postnatal period. RNA phenotyping shows transcripts for the GH receptor and GH-binding protein in mouse preimplantation embryos of all stages from fertilized eggs (day 1) to blastocysts (day 4). An antibody specific to the cytoplasmic region of the GH receptor revealed receptor protein expression, first in two-cell embryos, the stage of activation of the embryonic genome (day 2), and in all subsequent stages. In cleavage-stage embryos this immunoreactivity was localized mainly to the nucleus, but clear evidence of membrane labeling was apparent in blastocysts. GH receptor immunoreactivity was also observed in cumulus cells associated with unfertilized oocytes but not in the unfertilized oocytes. The blastocyst receptor was demonstrated to be functional, exhibiting the classic bell-shaped dose–response curves for GH stimulation of both 3-O-methyl glucose transport and protein synthesis. Maximal stimulation of 40–50% was seen for both responses at less than 1 ng/ml recombinant GH, suggesting a role for maternal GH. However mRNA transcripts for GH were also detected from the morula stage (day 3) by using reverse transcription–PCR, and GH immunoreactivity was seen in blastocysts. These observations raise the possibility of a paracrine/autocrine GH loop regulating embryonic development in its earliest stages.

Growth differentiation factor-9 (GDF-9), an oocyte-secreted member of the transforming growth factor β superfamily, progesterone receptor, cyclooxygenase 2 (Cox2; Ptgs2), and the EP2 prostaglandin E2 (PGE2) receptor (EP2; Ptgerep2) are required for fertility in female but not male mice. To define the interrelationship of these factors, we used a preovulatory granulosa cell culture system in which we added recombinant GDF-9, prostaglandins, prostaglandin receptor agonists, or cyclooxygenase inhibitors. GDF-9 stimulated Cox2 mRNA within 2 h, and PGE2 within 6 h; however, progesterone was not increased until 12 h after addition of GDF-9. This suggested that Cox2 is a direct downstream target of GDF-9 but that progesterone synthesis required an intermediate. To determine whether prostaglandin synthesis was required for progesterone production, we analyzed the effects of PGE2 and cyclooxygenase inhibitors on this process. PGE2 can stimulate progesterone synthesis by itself, although less effectively than GDF-9 (3-fold vs. 6-fold increase over 24 h, respectively). Furthermore, indomethacin or NS-398, inhibitors of Cox2, block basal and GDF-9-stimulated progesterone synthesis. However, addition of PGE2 to cultures containing both GDF-9 and NS-398 overrides the NS-398 block in progesterone synthesis. To further define the PGE2-dependent pathway, we show that butaprost, a specific EP2 agonist, stimulates progesterone synthesis and overrides the NS-398 block. In addition, GDF-9 stimulates EP2 mRNA synthesis by a prostaglandin- and progesterone-independent pathway. Thus, GDF-9 induces an EP2 signal transduction pathway which appears to be required for progesterone synthesis in cumulus granulosa cells. These studies further demonstrate the importance of oocyte–somatic cell interactions in female reproduction.

Maintenance of female reproductive competence depends on the actions of several hormones and signaling factors. Recent reports suggest roles for bone morphogenetic proteins (BMPs) in early stages of folliculogenesis. A role for the type I BMP receptor BmprIB as a regulator of ovulation rates in sheep has been described recently, but little is known about the roles of BMP signaling pathways in other aspects of reproductive function. We report here that BMPRIB is essential for multiple aspects of female fertility. Mice deficient in BmprIB exhibit irregular estrous cycles and an impaired pseudopregnancy response. BmprIB mutants produce oocytes that can be fertilized in vitro, but defects in cumulus expansion prevent fertilization in vivo. This defect is associated with decreased levels of aromatase production in granulosa cells. Unexpectedly, levels of mRNA for cyclooxygenase 2, an enzyme required for cumulus expansion, are increased. BmprIB mutants also exhibit a failure in endometrial gland formation. The expression of BmprIB in uterine linings suggests that these defects are a direct consequence of loss of BMP signaling in this tissue. In summary, these studies demonstrate the importance of BMP signaling pathways for estrus cyclicity, estradiol biosynthesis, and cumulus cell expansion in vivo and reveal sites of action for BMP signaling pathways in reproductive tissues.

HLA-G is a nonclassical class I major histocompatibility complex molecule with a restricted pattern of expression that includes the placental extravillus cytotrophoblast cells in direct contact with maternal tissues. Circumstantial evidence suggests that HLA-G may play a role in protection of the semiallogeneic human fetus. We examined whether HLA-G is expressed during the critical period of preimplantation human development and whether expression of this molecule could be correlated with the cleavage rate of embryos. Using reverse transcription PCR on surplus human embryos and unfertilized oocytes from patients undergoing in vitro fertilization we detected HLA-G heavy chain mRNA in 40% of 148 of blastocysts tested. The presence of HLA-G mRNA was also detected in unfertilized oocytes and in early embryos, but not in control cumulus oophorus cells. beta 2-Microglobulin mRNA was also found in those embryos expressing HLA-G. In concordance with our mRNA data, a similar proportion of embryos stained positive for HLA-G utilizing a specific monoclonal antibody. Interestingly, expression of HLA-G mRNA was associated with an increased cleavage rate, as compared to embryos lacking HLA-G transcript. Thus, HLA-G could be a functional homologue of the mouse Qa-2 antigen, which has been implicated in differences in the rate of preimplantation embryo development. To our knowledge, the presence of HLA-G mRNA and protein in human preimplantation embryos and oocytes has not been reported previously. The correlation of HLA-G mRNA expression with cleavage rate suggests that this molecule may play an important role in human pre-embryo development.

A reciclagem de misturas asfálticas usando técnicas de misturas mornas atende às exigências da sustentabilidade, limitando o consumo de energia e de novos materiais. A reciclagem de revestimentos asfálticos fresados (RAP) é praticada há mais de 40 anos, por meio de usinagem a quente do RAP adicionado a agregados e ligante novos. Todavia, o uso de elevadas taxas de RAP (acima de 25% no total), é um grande desafio até o presente, pois limita a perda de durabilidade da mistura asfáltica final. O RAP contém ligante asfáltico envelhecido aderido aos agregados, porém uma parte desse ligante pode ser remobilizada durante a reciclagem pelo aumento da temperatura no processo de usinagem. Esta tese busca compreender o processo de remobilização do ligante envelhecido durante a reciclagem do RAP e as consequências da magnitude de temperatura empregada no processo de reaquecimento deste material envelhecido. Atualmente, tem-se buscado o uso da tecnologia de usinagem morna, permitindo a redução de temperatura total no processo de fabricação. Um dos problemas possíveis da combinação das técnicas reciclagem morna é a ocorrência do fenômeno de recobrimento duplo, que pode impactar na reologia do novo material: o ligante do RAP e o ligante novo não se misturariam totalmente, devido à redução das temperaturas de fabricação, formando camadas superpostas. Este fenômeno seria a causa de disfuncionamentos mecânicos da mistura asfáltica final. Questiona-se então a qualidade do recobrimento dos agregados do RAP, já recobertos, e a falta de um meio de caracterização para a observação da interface. Neste estudo, a qualidade do recobrimento do RAP foi avaliada em escala microscópica e macroscópica. No nível microscópico, foi proposto o uso de uma ferramenta de micro-espectroscopia infravermelha para avaliar a interface entre o RAP e o ligante novo, e seguir a distribuição espacial desses dois componentes na mistura asfáltica. Para tanto, foi realizado um desenvolvimento experimental detalhado que permitisse o uso de um acessório de imagem infravermelha em misturas asfálticas recicladas. Graças à indentificação prévia de marcadores internos, foi possível acompanhar a mobilização parcial do ligante envelhecido do RAP. Em paralelo, no nível macroscópico, foi desenvolvido um protocolo de ensaio com o objetivo de avaliar a durabilidade das misturas asfálticas recicladas com taxas elevadas de RAP (50%), com o objetivo princial de compreender se há benefícios nas propriedades mecânicas de misturas mornas recicladas. O ensaio de fadiga é um dos ensaios que permitem melhor avaliar e comparar diferentes misturas asfálticas para comporem uma camada de pavimento. Porém, este ensaio não pode ser diretamente transposto à previsibilidade de campo por ter restrições, tais como o ligante das amostras ensaiadas não teve tempo de envelhecer; e as misturas asfálticas, dependendo da quantidade de RAP e da temperatura de usinagem, podem envelhecer em velocidades diferentes em campo. No presente trabalho, foi proposta a adição de uma etapa de envelhecimento previamente aos ensaios mecânicos. Foi suposto que o comportamento real do material estaria situado entre o do material não envelhecido e do envelhecido. Os principais resultados mostram que uma mistura reciclada morna apresenta resultados satisfatórios de comportamento mecânico que atestam seu uso como camada de revestimento asfáltico, exceto que ela tem a tendência de ser mais sensível à fadiga que uma mistura com a mesma composição de agregados e de teor de ligante, fabricada a quente ou morna, mas sem a adição de RAP.

No presente artigo procurar-se-á, depois de uma breve abordagem sobre a concretização do Processo de Bolonha, nos países da união europeia, compreender em que fase se encontra a adequação dos cursos de Artes Visuais e em que medida têm sido acolhidas as orientações emanadas das diferentes conferências de ministros que tutelam o ensino superior, quanto à estrutura de graus, à duração dos ciclos, à implementação do sistema de garantia da qualidade, entre outros aspectos. Advirta-se, contudo, que para efeitos da presente caracterização apenas se consideraram as instituições de ensino superior com sítios disponíveis na internet, tendo sido efectuada a recolha de dados durante o ano de 2008 e o primeiro trimestre de 2009. O levantamento das instituições foi feito junto das associações internacionais, consideradas interlocutoras credíveis como a ELIA (The European League of Institutes of ARTS) e a Cumulus Association Of Universities and Colleges of Art, Design and Media. Em Portugal, esse levantamento foi feito junto da Direcção Geral do Ensino Superior e em Espanha junto da ANECA (Agencia Nacional de Evaluacion de la Calidad Y Acreditacion).
