miR-21 is the most commonly over-expressed microRNA (miRNA) in cancer and a proven oncogene. Hsa-miR-21 is located on chromosome 17q23.2, immediately downstream of the vacuole membrane protein-1 (VMP1) gene, also known as TMEM49. VMP1 transcripts initiate ∼130 kb upstream of miR-21, are spliced, and polyadenylated only a few hundred base pairs upstream of the miR-21 hairpin. On the other hand, primary miR-21 transcripts (pri-miR-21) originate within the last introns of VMP1, but bypass VMP1 polyadenylation signals to include the miR-21 hairpin. Here, we report that VMP1 transcripts can also bypass these polyadenylation signals to include miR-21, thus providing a novel and independently regulated source of miR-21, termed VMP1–miR-21. Northern blotting, gene-specific RT-PCR, RNA pull-down and DNA branching assays support that VMP1–miR-21 is expressed at significant levels in a number of cancer cell lines and that it is processed by the Microprocessor complex to produce mature miR-21. VMP1 and pri-miR-21 are induced by common stimuli, such as phorbol-12-myristate-13-acetate (PMA) and androgens, but show differential responses to some stimuli such as epigenetic modifying agents. Collectively, these results indicate that miR-21 is a unique miRNA capable of being regulated by alternative polyadenylation and two independent gene promoters.

While historical studies of the Atlantic slave trade have amply demonstrated the magnitude of slave mortality during the Middle Passage, only recently have they started to examine how the captives might have endured and coped with this traumatic experience. Although it constitutes a major topos in African diasporic culture, the Middle Passage has only occasionally been represented directly and in details in novels and in films. This article examines three recent narratives of the Middle Passage, Fred D'Aguiar's novel Feeding the Ghosts (1998), Guy Deslauriers's film Passage du milieu (2000), and Stephanie Smallwood's historical study Saltwater Slavery: A Middle Passage from Africa to American Diaspora (2007). Beyond their individual poetic, aesthetic, and scholarly qualities, what is most striking about these three texts is that they all use the figure of the living dead in order to explore the captives' experience of the transatlantic journey. If the ghastly quality of the living dead powerfully captures the life-threatening material and physical conditions the captives endured on the voyage, its dual, liminal character also allows D'Aguiar, Deslauriers, and Smallwood to represent the metaphysical, psychological, social, and cultural journey they were forced to undertake. Through their use of the trope of the living dead, these three texts show that if death is indeed a central aspect of the experience of the Middle Passage, it impacts the captives in ways that go well beyond the issue of mortality.

Grx3 and Grx4, two monothiol glutaredoxins of Saccharomyces cerevisiae, regulate Aft1 nuclear localisation. We provide evidence of a negative regulation of Aft1 activity by Grx3 and Grx4. The Grx domain of both proteins played an important role in Aft1 translocation to the cytoplasm. This function was not, however, dependent on the availability of iron. Here we demonstrate that Grx3, Grx4 and Aft1 interact each other both in vivo and in vitro, which suggests the existence of a functional protein complex. Interestingly, each interaction occurred independently on the third member of the complex. The absence of both Grx3 and Grx4 induced a clear enrichment of G1 cells in asynchronous cultures, a slow growth phenotype, the accumulation of intracellular iron and a constitutive activation of the genes regulated by Aft1. The grx3grx4 double mutant was highly sensitive to the oxidising agents hydrogen peroxide and t-butylhydroperoxide but not to diamide. The phenotypes of the double mutant grx3grx4 characterised in this study were mainly mediated by the Aft1 function, suggesting that grx3grx4 could be a suitable cellular model for studying endogenous oxidative stress induced by deregulation of the iron homeostasis. However, our results also suggest that Grx3 and Grx4 might play additional roles in the oxidative stress response through proteins other than Aft1.
Disseny i desenvolupament d’un sistema de monitorització en continu de la qualitat de l’aigua de riu

Disseny i muntatge d’un sistema de monitorització a temps real de la qualitat fisico-química de l’aigua en rius mediterranis, que sigui autònom telegestionat i de baix manteniment. Aquest sistema permet realitzar un seguiment de la dinàmica de la qualitat de l’aigua de riu, essent possible detectar episodis d’empitjorament sobtat de la qualitat. El desenvolupament de sensors per mesurar la qualitat de l’aigua en continu ha estat una realitat en la darrera dècada, millorant-ne la robustesa i baixant-ne els costos. Cada instrument cobreix diferents aplicacions dins el cicle de l’aigua, des d’aigües superficials de riu, aigües residuals o aigües potables. Actualment la dificultat no està en la sensorística, sinó en el desenvolupament d’estacions de monitorització autònomes i de baix manteniment

O objetivo deste trabalho foi avaliar o número de anteras por flor, grãos de pólen por antera e capacidade germinativa do pólen de diferentes cultivares de macieiras. O trabalho foi executado no Laboratório de Fisiologia do Desenvolvimento e Genética Vegetal da Universidade Federal de Santa Catarina, e as coletas a campo foram realizadas na Epagri/Estação Experimental de Caçador-SC, em outubro de 2005. Foram utilizadas as seguintes cultivares comerciais de macieira desenvolvidas no Brasil: Primícia, Princesa, Fred Hough, Catarina, Baronesa, Lisgala, Suprema, Condessa, Daiane, Duquesa, Imperatriz e Joaquina. As cultivares de macieira Condessa, Princesa, Eva, Duquesa, Imperatriz, Gala, Fred Hough, Daiane, Baronesa e Suprema produzem pólen em quantidade suficiente e com boa capacidade germinativa. A cv. Condessa, embora apresente alta capacidade germinativa de pólen, produz menos anteras e grãos de pólen por antera que as demais. A cv. Princesa é a que apresenta o melhor perfil como polinizadora, por conjugar número de anteras/flor, número de grãos de pólen/antera e capacidade germinativa do pólen mais satisfatórios.

Nucleoside transporters (NTs) mediate the uptake of nucleosides and nucleobases across the plasma membrane, mostly for salvage purposes. The canonical NTs belong to two gene families, SLC29 and SLC28. The former encode equilibrative nucleoside transporter proteins (ENTs), which mediate the facilitative diffusion of natural nucleosides with broad selectivity, whereas the latter encode concentrative nucleoside transporters (CNTs), which are sodium-coupled and show high affinity for substrates with variable selectivity. These proteins are expressed in most cell types, exhibiting apparent functional redundancy. This might indicate that CNTs play specific roles in the physiology of the cell beyond nucleoside salvage. Here, we addressed this possibility using adenoviral vectors to restore tumor cell expression of hCNT1 or a polymorphic variant (hCNT1S546P) lacking nucleoside translocation ability. We found that hCNT1 restoration in pancreatic cancer cells significantly altered cell-cycle progression and phosphorylation status of key signal-transducing kinases, promoted poly-(ADP ribose) polymerase hyperactivation and cell death, and reduced tumor growth and cell migration. Importantly, the translocation-defective transporter triggered these same effects on cell physiology. These data predict a novel and totally unexpected biological role for the nucleoside transporter protein hCNT1 that appears to be independent of its role as mediator of nucleoside uptake by cells, thereby suggesting a transceptor function. A un mes de enero anormalmente templado, durante el cual en las comarcas próximas a Barcelona había principiado ya la brotación primaveral de muchas especies, sucedió, entre los días 2 y 23 de febrero, un período de fríos excepcionales, en que el termómetro descendió por debajo del punto de congelación durante muchas noches consecutivas. He aquí algunos de los datos obtenidos por el Observatorio Meteorológico de Barcelona (Travessera de Dalt) y publicados en el Boletín de Estadística Municipal
