Este trabalho teve como objetivo avaliar o efeito de solução de cloreto de cálcio e película de alginato de sódio na conservação de laranja 'Pera' minimamente processada. A qualidade da laranja minimamente processada submetida aos tratamentos com cloreto de cálcio a 1%, alginato de sódio a 1% e o controle (sem aplicação de tratamento) foi monitorada a cada três dias, por análises físicas e químicas, microbiológicas e sensoriais por um período de 12 dias de armazenamento a 5 ºC. O tratamento com alginato de sódio (1%) apresentou menor perda de massa ao longo do período de armazenamento, porém, as amostras submetidas a este tratamento, tiveram sua qualidade prejudicada quanto aos teores de ácido ascórbico, açúcares, teor de sólidos solúveis e firmeza. No final do armazenamento, o tratamento com cloreto de cálcio (1%) apresentou a melhor eficiência na manutenção das características iniciais do fruto, preservando os teores de ácido ascórbico, acidez titulável, 'ratio', açúcares e firmeza, evidenciada pela menor solubilização de pectinas. Análise microbiológica detectou valores insignificantes para bactérias psicrotróficas, bolores e leveduras e coliformes totais em todos os tratamentos, mostrando que os cuidados tomados com as condições higiênicas levaram à obtenção de um produto com padrão microbiológico de acordo com a legislação de alimentos, apresentando ausência de Salmonella e ausência de coliformes a 45 ºC. Sensorialmente, a laranja minimamente processada tratada com cloreto de cálcio (1%) e o controle, apresentou-se em condições de consumo por nove dias de armazenamento.

As fibras alimentares são consumidas naturalmente em cereais, frutas e hortaliças, mas também são adicionadas na forma concentrada em alimentos processados. Diferentes fontes de fibras têm diferentes estruturas e composições químicas, que irão definir a sua finalidade nutricional ou tecnológica. Este trabalho tem como objetivo a caracterização de fibra alimentar do albedo da laranja submetida a dois métodos de secagem (liofilização e convencional) e divididas por intervalos granulométricos. Para as determinações de densidade, área superficial, porosidade e distribuição de tamanho de poro, utilizaram-se técnicas clássicas da tecnologia de sistema particulado e foram comparadas com imagens de microscopia eletrônica de varredura. Os resultados apontaram que o método de secagem é o que mais influenciou na redução do volume de poros do material, também visualizado nas características microestruturais reveladas pela análise de imagem. O intervalo granulométrico da fibra particulada apresentou-se inversamente proporcional à densidade e à área superficial específica do material. A fibra alimentar do albedo da laranja apresentou características estruturais que permitem a diversificação de novos produtos alimentícios com alto valor nutritivo e comercial.

The artisanal production of cachaça, a beverage obtained by the fermentation of sugar cane juice after distillation, especially by small-sized producers, has traditionally used natural ferment ("fermento caipira") which consists of sugar cane juice with crushed corn, powdered rice, or citrus fruits. In despite of the difficulties in quality control due to the high level of contaminants and longer periods of preparation, the sensorial quality of the beverage may be attributed to the physiological activities of wild yeasts and even bacteria present during fermentation when this ferment is used. In this context, the aim here was to evaluate the microbiological (yeasts) and physicochemical characteristics of sugar cane juice extracted from different parts of three different varieties (RB72454, RB835486, and RB867515) of the cane stalk (lower, medium, and upper sections) in three harvesting periods (from May to December 2007) in an area under organic management. The juice from the upper section (from the eleventh internode to the top) of the sugar cane stalk could be indicated for the preparation of the natural ferment since it is as a source of yeasts and reducing sugars, especially the variety RB867515. Due to the seasonality, the best period for using this part of the sugar cane stalk is at the beginning of harvesting when the phenolic compounds are at low concentration, but there are higher number of Saccharomyces population and other yeast species. The high acidity in this section of the plant could result in a better control of bacterial contamination. These findings explain the traditional instructions of adding the upper sections for the preparation of natural ferment and can help its management in order to get a better performance with respect to organic cachaça production.

This study aimed at evaluating compositional changes in the quality of 'Ortanique' tangor after coating with the carnauba-based waxes Aruá Tropical® or Star Light®. The storage conditions studied simulated those of local marketing (22 ± 2 °C, 60 ± 5% RH). Non-destructive analysis, mass loss, peel color, and sensory evaluation, were performed upon coating and every three days up to the fifteenth day of storage. Destructive analysis, peel moisture content, chlorophyll of the peel, pulp color, juice content, soluble solids (SS), titratable acidity (TA), pH, and soluble solids to titratable acidity ratio, were performed upon coating and every four days up to the sixteenth day of storage. The assay was conducted using an entirely randomized design, with three replications (destructive analyses) or ten replications (non-destructive analyses), in a split plot scheme. Wax-coating, especially Aruá Tropical®, maintained fruit freshness by reducing mass loss and peel dehydration and retaining green color. Peel moisture content, chlorophyll content, and juice content had lower rates in the wax coated fruits. Puncture force, soluble solids, titratable acidity, pH, and soluble solids to titratable acidity ratio varied vary little over the course of storage. Sensory evaluation showed that the application of Aruá Tropical keeps 'Ortanique' tangor fresher for 6 days longer for commercialization.

The purpose of this study was to evaluate the changes in concentrations of O2 and CO2 inside packages of minimally processed Pera orange. Previously selected oranges that were washed, sanitized, and chilled were peeled using hydrothermal treatment (immersion of fruits in water at 50 °C for 8 minutes). The peeled oranges were then packed in five different plastic packages under passive and active modified atmosphere (5% O2 + 10% CO2 + 85% N2). The fruits were stored at 6 °C and 12 °C. The package headspace gas composition was evaluated for twelve days at 6 °C and nine days at 12 °C. The polypropylene film (32 µm) promoted modified atmosphere similar to that initially injected (5% O2 + 10% CO2 + 85% N2) at 6 °C and 12 °C. With regard to the atmosphere modification system, the injection of a gas mixture anticipated achieving an equilibrium atmosphere inside the packages at 12 °C. At 6 °C, the gas composition inside the packages was kept close to that of the injection, but the equilibrium was not verified.

The effects of carnauba-based wax on the quality of 'Delta Valencia' orange produced in Ceará state, Brazil, were studied. The fruits were coated with carnauba-based wax and refrigerated (7 ± 2 ºC and 85 ± 2% R.H.) for 28 days. The quality attribute parameters assessed were weight loss, peel color (brightness, hue angle, and chromaticity), peel moisture, pH, soluble solids (SS), titratable acidity (TA), SS/TA ratio, ascorbic acid, total soluble sugars, reducing sugars, yellow flavonoids, and polyphenols. The results showed that 'Delta Valencia' oranges grown in the dry climate of Ceará state has excellent quality. The coated fruits lost mass at a lower rate than the the control fruits. No significant loss of soluble solids, titratable acidity, pH, and SS/TA ratio was observed, while ascorbic acid, soluble sugars, reducing sugars, yellow flavonoids, and polyphenols increased during storage in both the coated and control fruits. Carnauba-based wax coated fruits showed no signal of dehydration keeping their shiny green peel up to the end of the storage. The use of coating was crucial for the maintenance of visual quality by reducing mass loss, as well as keeping peel moisture.

The objective of the present study was the isolation of a yeast strain, from citrus fruit peels, able to produce a polygalacturonase by submerged fermentation with maceration activity of raw cassava roots. Among 160 yeast strains isolated from citrus peels, one strain exhibited the strongest pectinolytic activity. This yeast was identified as Wickerhamomyces anomalus by 5.8S-ITS RFLP analysis and confirmed by amplification of the nucleotide sequence. The yeast produced a polygalacturonase (PG) in Erlenmeyer shake flasks containing YNB, glucose, and citrus pectin. PG synthesis occurred during exponential growth phase, reaching 51 UE.mL-1 after 8 hours of fermentation. A growth yield (Yx/s) of 0.43 gram of cell dry weight per gram of glucose consumed was obtained, and a maximal specific growth rate (µm) of 0.346 h-1 was calculated. The microorganism was unable to assimilate sucrose, galacturonic acid, polygalacturonic acid, or citrus pectin, but it required glucose as carbon and energy source and polygalacturonic acid or citrus pectin as inducers of enzyme synthesis. The crude enzymatic extract of Wickerhamomyces anomalus showed macerating activity of raw cassava. This property is very important in the production of dehydrated mashed cassava, a product of regional interest in the province of Misiones, Argentina.

This study explores variegated means through which ports have become increasingly entangled in the planning logic of neoliberal innovation-driven economy. The research topic belongs to the academic disciplines of economics and human geography. The aim of the thesis is to analyse how the notion of innovation, adopted in a variety of supranational and national port policy documents, is deployed in operational port environment in two different ports of the Baltic Sea Region: the port of Stockholm, Sweden, and the port of Klaipeda, Lithuania. This novel innovation agenda is visible in several topics I examine in the study, that is, port governance, environmental issues, and seaport – port-city interface. The gathered primary source material on port policy documents, strategies, development planning documents and reports is analysed by utilizing the qualitative content analysis research method. Moreover, the empirical part of the case study, that is, tracing innovation practices in mundane port activities is based on collected qualitative semi-structured interviews with port authorities in Klaipeda and Stockholm, researchers and other port experts. I examine the interview material by employing the theoretical reading research method. In my analysis, I have reframed port-related policy development by tracing and identifying the port transformation from “functional terminals” to “engines for growth”. My results show that this novel innovation-oriented rhetoric imprinted in the narrative “engines for growth” is often contested in daily port practices. In other words, my analysis reveals that the port authorities’ and other port actors’ attitudes towards innovations do not necessarily correspond to the new narrative of innovation and do not always “fit” within a framework of neoliberal economic thinking that glorifies the “culture of innovations”. I argue that the ability to develop innovative initiatives in the ports of Klaipeda and Stockholm is strongly predetermined by local conditions, a port’s governance model, the way port actors perceive the importance of innovations per se, demand factors and new regulations.

INTRODUÇÃO: As intercorrências do acesso vascular têm sido a maior causa de internação entre os pacientes com estágio V da doença renal crônica (DRC) em hemodiálise (HD). Apesar de campanhas para a diminuição do uso de cateter venoso central (CVC) como via de acesso para HD, este ainda representa a principal via de acesso para crianças e adolescentes que iniciam HD. OBJETIVOS E MÉTODOS: Este estudo tem o objetivo de avaliar, por meio de um coorte retrospectivo, o tipo de acesso vascular inicial, a incidência de complicações dos acessos vasculares e as razões de falência dos acessos em crianças e adolescentes com idade entre 0 e 18 anos que iniciaram HD no período de 1997 a 2007. RESULTADOS: Foram estudados 251 acessos em 61 pacientes, sendo 97 fístulas arteriovenosas (FAV) e 154 CVC de curta permanência. Dos pacientes do estudo 51 % iniciaram HD pelo CVC. A média de idade dos pacientes no início da HD foi de 12,5 anos. A doença de base predominante foi glomerulopatia (46%). A principal causa de retirada de CVC foi infecção, em 35%. A sobrevida média do CVC foi de 40 dias. A falência primária da FAV foi detectada em 37,8% das FAV confeccionadas. Para as FAV funcionantes, a principal causa de falência foi a trombose (84%). A infecção não foi a causa de nenhuma falência de FAV. Comparando-se os tipos de acesso, constatou-se risco de infecção 34 vezes maior para os pacientes em uso de CVC em relação aos em uso de FAV. CONCLUSÃO: A infecção foi a maior causa de retirada de CVC temporário. Esse estudo sugere que o CVC temporário deve ser evitado, e, sempre que possível, substituído por FAV ou CVC de longa permanência. A trombose foi a principal causa de perda da FAV, reforçando a importância de um programa para a detecção precoce da disfunção do acesso.

ResumoAtualmente, cateteres venosos permanentes (CVCp) estão se tornando cada vez mais uma alternativa de acesso vascular de longa permanência para pacientes nos quais o acesso arteriovenoso não pode ser confeccionado, sendo a oclusão trombótica complicação mecânica comum. Essa complicação pode ocasionar mudanças frequentes dos locais de cateter, eliminando os sítios vasculares. Este estudo teve como objetivo realizar uma revisão narrativa do manejo da oclusão trombótica de CVCp na população em HD. O tratamento da trombose de CVC consiste em infusão de solução salina ou na administração de trombolíticos como plasminogênio tecidual ativado, reteplase ou uroquinase. Há poucos estudos sobre o uso de alteplase em CVCp obstruídos na população em diálise, e todos eles relatam sucesso entre 80 a 95% dos casos com uso de trombolítico na dose de 1 mg/ml. Por tratar-se de medicamento de custo elevado, estudos sugerem que a criopreservação e o fracionamento da alteplase tornam o uso financeiramente viável.

O objetivo desta pesquisa foi estudar os efeitos da secagem sobre o comportamento fisiológico das sementes de tangerina (Citrus reticulata Blanco var. Cleópatra) durante o armazenamento. Assim, sementes extraídas manualmente de frutos maduros foram lavadas em água corrente e o excesso de água drenado à sombra. Em seguida, uma amostra representativa de sementes foi removida a fim de constituir a porção com o maior grau de umidade, 48%, a ser estudado. As sementes remanescentes foram submetidas a secagem em equipamento com circulação forçada de ar a 28±2°C, visando à obtenção dos percentuais de 39, 31, 24, 14, 10 e 7 referentes aos demais graus de umidade. As sementes, embaladas em sacos de polietileno, foram armazenadas em câmaras com temperatura controlada de 10 e 20°C. No início do armazenamento, e a intervalos de quatro semanas até 48 semanas, as sementes foram submetidas às seguintes avaliações: umidade, germinação, emergência de plântulas, índice de velocidade de emergência e comprimento de plântula. Concluiu-se que, considerando o intervalo de 39 a 7% de grau de umidade e a temperatura de 10ºC, o desempenho fisiológico de sementes de tangerina 'Cleopatra' durante o armazenamento, é favorecido pela desidratação até graus de umidade de, no mínimo, 24%.
Storage of 'Swingle' citrumelo seeds in different maturation stages subjected to fungicide treatment

The establishment of citrus orchards with rootstocks from seed with a low physiological quality has been a recurring problem. Low quality seeds directly affect both the final stand and the time required for seedling production. The irregular maturation of fruits, seed recalcitrance, and the high incidence of fungi, make long term storage difficult, even at low temperatures. This study evaluated the storage potential and the use of fungicide treatments on 'Swingle' citrumelo seeds extracted from fruits collected at two maturation stages, green or ripe. The seeds were subjected to a thermal treatment, treated with Derosal, Thiram or Tecto+Captan fungicides, packaged in impermeable polyethylene bags and stored in a cold chamber for nine months. Every three months, the physiological and sanitary qualities of the seeds were evaluated from germination and sanitary tests and also from enzyme profiles. Seeds from green fruits deteriorated less than those from mature fruits; deterioration increased up to nine months of storage; treatment with the Tecto+Captan mixture gave effective pathogen control and maintained seed quality during storage. The germination of the green and ripe seeds is satisfactory (70%) after three months storage.

Diabetes mellitus is a disorder of inadequate insulin action and consequent high blood glucose levels. Type 2 diabetes accounts for the majority of cases of the disease and is characterized by insulin resistance and relative insulin deficiency resulting in metabolic deregulation. It is a complex disorder to treat as its pathogenesis is not fully understood and involves a variety of defects including ~-cell failure, insulin resistance in the classic target tissues (adipose, muscle, liver), as well as defects in a-cells and kidney, brain, and gastrointestinal tissue. Present oral treatments, which aim at mimicking the effects of insulin, remain limited in their efficacy and therefore the study of the effects of novel compounds on insulin target tissues is an important area of research both for potentially finding more treatment options as well as for increasing our knowledge of metabolic regulation in health and disease. In recent years the extensively studied polyphenol, resveratrol, has been reported to have antidiabetic effects showing that it increases glucose uptake by skeletal muscle cells and prevents fatty acid-induced insulin resistance in vitro and in vivo. Naringenin, a citrus flavonoid with structural similarities to resveratrol, is reported to have antioxidan.t, antiproliferative, anticancer, and anti-inflammatory properties. Effects on glucose and lipid metabolism have also been reported including blood glucose and lipid lowering effects. However, whether naringenin has insulinlike effects is not clear. In the present study the effects of naringenin on glucose uptake in skeletal muscle cells are examined and compared with those of insulin. Naringenin treatment of L6 myotubes increased glucose uptake in a dose- and time dependent manner and independent of insulin. The effects of naringenin on glucose uptake achieved similar levels as seen with maximum insulin stimulation and its effect was additive with sub-maximal insulin treatment. Like insulin naringenin treatment did not increase glucose uptake in myoblasts. To elucidate the mechanism involved in naringenin action we looked at its effect on phosphatidylinositol 3-kinase (PI3K) and Akt, two signalling molecules that are involved in the insulin signalling cascade leading to glucose uptake. Naringenin did not stimulate basal or insulinstimulated Akt phosphorylation but inhibition of PI3K by wortmannin partially repressed the naringenin-induced glucose uptake. We also examined naringenin's effect on AMP-activated protein kinase (AMPK), a molecule that is involved in mediating glucose uptake by a variety of stimuli. Naringenin stimulated AMPK phosphorylation and this effect was not inhibited by wortmannin. To deduce the nature of the naringenin-stimulated AMPK phosphorylation and its impact on glucose uptake we examined the role of several molecules implicated in mod.ulating AMPK activity including SIRTl, LKB 1, and ca2+ Icalmodulin-dependent protein kinase kinase (CaMKK). Our results indicate that inhibition of SIRTI did not prevent the naringeninstimulated glucose uptake Of. AMPK phosphorylation; naringenin did not stimulate LKB 1 phosphorylation; and inhibition of CaMKK did not prevent naringeninstimulated glucose uptake. Inhibition of AMPK by compound C also did not prevent naringenin-stimulated glucose uptake but effectively inhibited the phosphorylation of AMPK suggesting that AMPK may not be required for the naringenin-stimulated glucose uptake.

The interaction between local and reflexive control of skin blood flow (SkBF) is unclear. This thesis isolated the roles of rectal (Tre) and local (Tloc) temperature on forearm SkBF regulation at normal and elevated body temperatures, and to investigate the interaction between local and reflexive SkBF control. While either normothermic (Tre ~37.0°C) or hyperthermic (∆Tre +1.1°C), SkBF was assessed on the dorsal aspect of each forearm in 10 participants while Tloc was manipulated in an A-B-A-B fashion between neutral (33.0°C) and hot (38.5°C). Finally, local heating to 44°C was performed to elicit maximal SkBF. Data are presented as a percentage of maximal cutaneous vascular conductance (CVC), calculated as laser-Doppler flux divided by mean arterial pressure. Tloc manipulations performed during normothermia had significantly greater effects on CVC than during hyperthermia. The decreased modification to SkBF from the Tloc changes during hyperthermia suggests that strong reflexive vasodilation attenuates local SkBF control mechanisms.

Comparativement au génome contenu dans le noyau de la cellule de plante, nos connaissances des génomes des deux organelles de cette cellule, soit le plastide et la mitochondrie, sont encore très limitées. En effet, un nombre très restreint de facteurs impliqués dans la réplication et la réparation de l’ADN de ces compartiments ont été identifiés à ce jour. Au cours de notre étude, nous avons démontré l’implication de la famille de protéines Whirly dans le maintien de la stabilité des génomes des organelles. Des plantes mutantes pour des gènes Whirly chez Arabidopsis thaliana et Zea mays montrent en effet une augmentation du nombre de molécules d’ADN réarrangées dans les plastides. Ces nouvelles molécules sont le résultat d’une forme de recombinaison illégitime nommée microhomology-mediated break-induced replication qui, en temps normal, se produit rarement dans le plastide. Chez un mutant d’Arabidopsis ne possédant plus de protéines Whirly dans les plastides, ces molécules d’ADN peuvent même être amplifiées jusqu’à cinquante fois par rapport au niveau de l’ADN sauvage et causer un phénotype de variégation. L’étude des mutants des gènes Whirly a mené à la mise au point d’un test de sensibilité à un antibiotique, la ciprofloxacine, qui cause des bris double brin spécifiquement au niveau de l’ADN des organelles. Le mutant d’Arabidopsis ne contenant plus de protéines Whirly dans les plastides est plus sensible à ce stress que la plante sauvage. L’agent chimique induit en effet une augmentation du nombre de réarrangements dans le génome du plastide. Bien qu’un autre mutant ne possédant plus de protéines Whirly dans les mitochondries ne soit pas plus sensible à la ciprofloxacine, on retrouve néanmoins plus de réarrangements dans son ADN mitochondrial que dans celui de la plante sauvage. Ces résultats suggèrent donc une implication pour les protéines Whirly dans la réparation des bris double brin de l’ADN des organelles de plantes. Notre étude de la stabilité des génomes des organelles a ensuite conduit à la famille des protéines homologues des polymérases de l’ADN de type I bactérienne. Plusieurs groupes ont en effet suggéré que ces enzymes étaient responsables de la synthèse de l’ADN dans les plastides et les mitochondries. Nous avons apporté la preuve génétique de ce lien grâce à des mutants des deux gènes PolI d’Arabidopsis, qui encodent des protéines hautement similaires. La mutation simultanée des deux gènes est létale et les simples mutants possèdent moins d’ADN dans les organelles des plantes en bas âge, confirmant leur implication dans la réplication de l’ADN. De plus, les mutants du gène PolIB, mais non ceux de PolIA, sont hypersensibles à la ciprofloxacine, suggérant une fonction dans la réparation des bris de l’ADN. En accord avec ce résultat, la mutation combinée du gène PolIB et des gènes des protéines Whirly du plastide produit des plantes avec un phénotype très sévère. En définitive, l’identification de deux nouveaux facteurs impliqués dans le métabolisme de l’ADN des organelles nous permet de proposer un modèle simple pour le maintien de ces deux génomes.
