As the worldwide prevalence of diabetes mellitus continues to increase, diabetic retinopathy remains the leading cause of visual impairment and blindness in many developed countries. Between 32 to 40 percent of about 246 million people with diabetes develop diabetic retinopathy. Approximately 4.1 million American adults 40 years and older are affected by diabetic retinopathy. This glucose-induced microvascular disease progressively damages the tiny blood vessels that nourish the retina, the light-sensitive tissue at the back of the eye, leading to retinal ischemia (i.e., inadequate blood flow), retinal hypoxia (i.e., oxygen deprivation), and retinal nerve cell degeneration or death. It is a most serious sight-threatening complication of diabetes, resulting in significant irreversible vision loss, and even total blindness.

Unfortunately, although current treatments of diabetic retinopathy (i.e., laser therapy, vitrectomy surgery and anti-VEGF therapy) can reduce vision loss, they only slow down but cannot stop the degradation of the retina. Patients require repeated treatment to protect their sight. The current treatments also have significant drawbacks. Laser therapy is focused on preserving the macula, the area of the retina that is responsible for sharp, clear, central vision, by sacrificing the peripheral retina since there is only limited oxygen supply. Therefore, laser therapy results in a constricted peripheral visual field, reduced color vision, delayed dark adaptation, and weakened night vision. Vitrectomy surgery increases the risk of neovascular glaucoma, another devastating ocular disease, characterized by the proliferation of fibrovascular tissue in the anterior chamber angle. Anti-VEGF agents have potential adverse effects, and currently there is insufficient evidence to recommend their routine use.

In this work, for the first time, a paradigm shift in the treatment of diabetic retinopathy is proposed: providing localized, supplemental oxygen to the ischemic tissue via an implantable MEMS device. The retinal architecture (e.g., thickness, cell densities, layered structure, etc.) of the rabbit eye exposed to ischemic hypoxic injuries was well preserved after targeted oxygen delivery to the hypoxic tissue, showing that the use of an external source of oxygen could improve the retinal oxygenation and prevent the progression of the ischemic cascade.

The proposed MEMS device transports oxygen from an oxygen-rich space to the oxygen-deficient vitreous, the gel-like fluid that fills the inside of the eye, and then to the ischemic retina. This oxygen transport process is purely passive and completely driven by the gradient of oxygen partial pressure (pO2). Two types of devices were designed. For the first type, the oxygen-rich space is underneath the conjunctiva, a membrane covering the sclera (white part of the eye), beneath the eyelids and highly permeable to oxygen in the atmosphere when the eye is open. Therefore, sub-conjunctival pO2 is very high during the daytime. For the second type, the oxygen-rich space is inside the device since pure oxygen is needle-injected into the device on a regular basis.

To prevent too fast or too slow permeation of oxygen through the device that is made of parylene and silicone (two widely used biocompatible polymers in medical devices), the material properties of the hybrid parylene/silicone were investigated, including mechanical behaviors, permeation rates, and adhesive forces. Then the thicknesses of parylene and silicone became important design parameters that were fine-tuned to reach the optimal oxygen permeation rate.

The passive MEMS oxygen transporter devices were designed, built, and tested in both bench-top artificial eye models and in-vitro porcine cadaver eyes. The 3D unsteady saccade-induced laminar flow of water inside the eye model was modeled by computational fluid dynamics to study the convective transport of oxygen inside the eye induced by saccade (rapid eye movement). The saccade-enhanced transport effect was also demonstrated experimentally. Acute in-vivo animal experiments were performed in rabbits and dogs to verify the surgical procedure and the device functionality. Various hypotheses were confirmed both experimentally and computationally, suggesting that both the two types of devices are very promising to cure diabetic retinopathy. The chronic implantation of devices in ischemic dog eyes is still underway.

The proposed MEMS oxygen transporter devices can be also applied to treat other ocular and systemic diseases accompanied by retinal ischemia, such as central retinal artery occlusion, carotid artery disease, and some form of glaucoma.

The energy spectra of tritons and Helium-3 nuclei from the reactions 3He(d,t)2p, 3H(d,3He)2n, 3He(d,3He)pn, and 3H(d,t)pn were measured between 6° and 20° at a bombarding energy of 10.9 MeV. An upper limit of 5 μb/sr. was obtained for producing a bound di-neutron at 6° and 7.5°. The 3He(d,t)2p and 3H(d,3He)2n data, together with previous measurements at higher energies, have been used to investigate whether one can unambiguously extract information on the two-nucleon system from these three-body final state reactions. As an aid to these theoretical investigations, Born approximation calculations were made employing realistic nucleon-nucleon potentials and an antisymmetrized final state wave function for the five-particle system. These calculations reproduce many of the features observed in the experimental data and indicate that the role of exchange processes cannot be ignored. The results show that previous attempts to obtain information on the neutron-neutron scattering length from the 3H(d,3He)2n reaction may have seriously overestimated the precision that could be attained.

We propose a united theory that describes the two-center recording system by taking scattering noise into account. The temporal evolution of the signal-to-noise ratio in doubly doped photorefractive crystals is described based on jointly solving material equations and coupled-wave equations with the fourth-order Runge-Kutta method. Roles of microcosmic optical parameters of dopants on the signal-to-noise ratio are discussed in detail. The theoretical results can confirm and predict experimental results. (c) 2005 Elsevier GmbH. In a tagging experiment carried out in the Kenyan waters of Lake Victoria, an annual growth increment of 29 cm yr was obtained for Lates niloticus (L.). Growth parameters obtained using the von Bertalanffy model on the growth curve fitted by eye were L (inf.) = 122 cm yr and k = 0.26 yr. Data for other species tagged were inadequate to obtain meaningful results.

The first chapter of this thesis deals with automating data gathering for single cell microfluidic tests. The programs developed saved significant amounts of time with no loss in accuracy. The technology from this chapter was applied to experiments in both Chapters 4 and 5.

The second chapter describes the use of statistical learning to prognose if an anti-angiogenic drug (Bevacizumab) would successfully treat a glioblastoma multiforme tumor. This was conducted by first measuring protein levels from 92 blood samples using the DNA-encoded antibody library platform. This allowed the measure of 35 different proteins per sample, with comparable sensitivity to ELISA. Two statistical learning models were developed in order to predict whether the treatment would succeed. The first, logistic regression, predicted with 85% accuracy and an AUC of 0.901 using a five protein panel. These five proteins were statistically significant predictors and gave insight into the mechanism behind anti-angiogenic success/failure. The second model, an ensemble model of logistic regression, kNN, and random forest, predicted with a slightly higher accuracy of 87%.

The third chapter details the development of a photocleavable conjugate that multiplexed cell surface detection in microfluidic devices. The method successfully detected streptavidin on coated beads with 92% positive predictive rate. Furthermore, chambers with 0, 1, 2, and 3+ beads were statistically distinguishable. The method was then used to detect CD3 on Jurkat T cells, yielding a positive predictive rate of 49% and false positive rate of 0%.

The fourth chapter talks about the use of measuring T cell polyfunctionality in order to predict whether a patient will succeed an adoptive T cells transfer therapy. In 15 patients, we measured 10 proteins from individual T cells (~300 cells per patient). The polyfunctional strength index was calculated, which was then correlated with the patient's progress free survival (PFS) time. 52 other parameters measured in the single cell test were correlated with the PFS. No statistical correlator has been determined, however, and more data is necessary to reach a conclusion.

Finally, the fifth chapter talks about the interactions between T cells and how that affects their protein secretion. It was observed that T cells in direct contact selectively enhance their protein secretion, in some cases by over 5 fold. This occurred for Granzyme B, Perforin, CCL4, TNFa, and IFNg. IL- 10 was shown to decrease slightly upon contact. This phenomenon held true for T cells from all patients tested (n=8). Using single cell data, the theoretical protein secretion frequency was calculated for two cells and then compared to the observed rate of secretion for both two cells not in contact, and two cells in contact. In over 90% of cases, the theoretical protein secretion rate matched that of two cells not in contact.

O gênero Pterodon pertence à família das Papilonaceas e inclui cinco espécies nativas do Brasil: P. pubescens Benth., P. emarginatus Vog., P. apparicioi Pedersoli e P. abruptus Benth., sendo a espécie objeto deste estudo a P. polygalaeflorus Benth.. Seus frutos são livremente comercializados em mercados da flora medicinal e utilizados pela medicina popular devido a propriedades anti-reumática, analgésica, antiinflamatória, dentre outros efeitos associados a esses frutos. O principal uso popular está relacionado ao efeito antiartrítico que parece se encontrar na fração oleosa do fruto. O objetivo deste trabalho foi avaliar o extrato etanólico de Pterodon polygalaeflorus (EEPpg) quanto ao seu potencial antiinflamatório crônico através do modelo de artrite induzida por colágeno (CIA) e seu efeito sobre os linfócitos in vitro, bem como sobre a expansão de células MAC-1+ induzida por adjuvante completo de Freund (AFC). A caracterização química do EEPpg foi realizada por cromatografia em camada delgada (TLC), cromatografia líquida de alta performance (HPLC) e cromatografia gasosa acoplada a espectrômetro de massa (GC-MS), através dos quais uma gama de compostos, incluindo terpenóides de polaridades variadas e flavonóides, foram observados. No modelo de CIA, o EEPpg reduziu significativamente parâmetros associados ao desenvolvimento e progressão da doença e à severidade da doença , inibindo em até 99% o seu desenvolvimento e levando a ausência de sinais clínicos evidentes após tratamento com as menores doses do extrato (0,01 mg/kg e 0,001 mg/kg). O tratamento com EEPpg também reduziu características histopatológicas típicas de articulações de animais com CIA, que também são observadas na artrite reumatóide. O EEPpg reduziu significativamente o peso dos linfonodos dos camundongos, bem como o número absoluto de segmentados, monócitos e linfócitos no sangue. In vitro, O EEPpg mostrou uma atividade anti-proliferativa dos esplenócitos estimulados com concanavalina A (Con A) ou lipopolissacarídeo (LPS) analisada através do ensaio de redução do sal de tetrazólio MTT, corroborada pelo seu efeito sobre o ciclo celular de linfócitos estimulados com Con A, onde o EEPpg nas concentrações de 5, 10 e 20 μg/mL reduziu significativamente, de maneira concentração-dependente, o número de células nas fases S+G2/M e aumentou na fase G0/G1 do ciclo celular. O efeito anti-proliferativo do EEPpg parece também estar associado ao aumento da apoptose dos linfócitos após estimulação com Con A, com aumento estatisticamente significativo no percentual de células mortas por apoptose nas maiores concentrações . O EEPpg inibiu a expansão de células Mac-1+ induzida por AFC no baço, porém não no peritônio. Esse resultado sugere um efeito inibidor do EEPpg sobre a migração celular para as articulações artríticas. Esses resultados contribuem para a validação do uso popular de P. polygalaeflorus contra doenças relacionadas a processos inflamatórios e imunes, sobretudo na artrite reumatóide, antes nunca demonstrado.

A review article detailing the background, development and functionality of the Windermere Profiler, a multi parameter environmental monitoring instrument for use in lakes, reservoirs and rivers. The article explains the requirement for regular data collection by the Freshwater Biological Association at Windermere. The article covers the requirements of a profiling instrument, the design considerations, the electronic circuitry, the computer program, the operation of the computer software, the profiler in use and further developments to the design. A number of figures and images accompany the article.

Techniques are developed for estimating activity profiles in fixed bed reactors and catalyst deactivation parameters from operating reactor data. These techniques are applicable, in general, to most industrial catalytic processes. The catalytic reforming of naphthas is taken as a broad example to illustrate the estimation schemes and to signify the physical meaning of the kinetic parameters of the estimation equations. The work is described in two parts. Part I deals with the modeling of kinetic rate expressions and the derivation of the working equations for estimation. Part II concentrates on developing various estimation techniques.

Part I: The reactions used to describe naphtha reforming are dehydrogenation and dehydroisomerization of cycloparaffins; isomerization, dehydrocyclization and hydrocracking of paraffins; and the catalyst deactivation reactions, namely coking on alumina sites and sintering of platinum crystallites. The rate expressions for the above reactions are formulated, and the effects of transport limitations on the overall reaction rates are discussed in the appendices. Moreover, various types of interaction between the metallic and acidic active centers of reforming catalysts are discussed as characterizing the different types of reforming reactions.

Part II: In catalytic reactor operation, the activity distribution along the reactor determines the kinetics of the main reaction and is needed for predicting the effect of changes in the feed state and the operating conditions on the reactor output. In the case of a monofunctional catalyst and of bifunctional catalysts in limiting conditions, the cumulative activity is sufficient for predicting steady reactor output. The estimation of this cumulative activity can be carried out easily from measurements at the reactor exit. For a general bifunctional catalytic system, the detailed activity distribution is needed for describing the reactor operation, and some approximation must be made to obtain practicable estimation schemes. This is accomplished by parametrization techniques using measurements at a few points along the reactor. Such parametrization techniques are illustrated numerically with a simplified model of naphtha reforming.

To determine long term catalyst utilization and regeneration policies, it is necessary to estimate catalyst deactivation parameters from the the current operating data. For a first order deactivation model with a monofunctional catalyst or with a bifunctional catalyst in special limiting circumstances, analytical techniques are presented to transform the partial differential equations to ordinary differential equations which admit more feasible estimation schemes. Numerical examples include the catalytic oxidation of butene to butadiene and a simplified model of naphtha reforming. For a general bifunctional system or in the case of a monofunctional catalyst subject to general power law deactivation, the estimation can only be accomplished approximately. The basic feature of an appropriate estimation scheme involves approximating the activity profile by certain polynomials and then estimating the deactivation parameters from the integrated form of the deactivation equation by regression techniques. Different bifunctional systems must be treated by different estimation algorithms, which are illustrated by several cases of naphtha reforming with different feed or catalyst composition.

Chronic diseases of the central nervous system are poorly treated due to the inability of most therapeutics to cross the blood-brain barrier. The blood-brain barrier is an anatomical and physiological barrier that severely restricts solute influx, including most drugs, from the blood to the brain. One promising method to overcome this obstacle is to use endogenous solute influx systems at the blood-brain barrier to transport drugs. Therapeutics designed to enter the brain through transcytosis by binding the transferrin receptor, however, are restricted within endothelial cells. The focus of this work was to develop a method to increase uptake of transferrin-containing nanoparticles into the brain by overcoming these restrictive processes.

To accomplish this goal, nanoparticles were prepared with surface transferrin molecules bound through various liable chemical bonds. These nanoparticles were designed to shed the targeting molecule during transcytosis to allow increased accumulation of nanoparticles within the brain.

Transferrin was added to the surface of nanoparticles through either redox or pH sensitive chemistry. First, nanoparticles with transferrin bound through disulfide bonds were prepared. These nanoparticles showed decreased avidity for the transferrin receptor after exposure to reducing agents and increased ability to enter the brain in vivo compared to those lacking the disulfide link.

Next, transferrin was attached through a chemical bond that cleaves at mildly acidic pH. Nanoparticles containing a cleavable link between transferrin and gold nanoparticle cores were found to both cross an in vitro model of the blood-brain barrier and accumulate within the brain in significantly higher numbers than similar nanoparticles lacking the cleavable bond. Also, this increased accumulation was not seen when using this same strategy with an antibody to transferrin receptor, indicating that behavior of nanoparticles at the blood-brain barrier varies depending on what type of targeting ligand is used.

Finally, polymeric nanoparticles loaded with dopamine and utilizing a superior acid-cleavable targeting chemistry were investigated as a potential treatment for Parkinson’s disease. These nanoparticles were capable of increasing dopamine quantities in the brains of healthy mice, highlighting the therapeutic potential of this design. Overall, this work describes a novel method to increase targeted nanoparticle accumulation in the brain.

O envelhecimento populacional é um fenômeno mundial, assim como o predomínio de mortes por doenças cardiovasculares. Estudos demonstram que o envelhecimento acarreta aumento da rigidez vascular e perturbações na reatividade macrovascular. O presente estudo comparou a microcirculação deste processo com adultos jovens através da pletismografia de oclusão venosa (POV) e da videocapilaroscopia do leito periungueal (VC). Para isto, desenvolveu-se um estudo transversal com dois grupos: idade entre 18 e 30 anos (n=16) e outro grupo com idade igual ou superior a 60 anos (n=21), além da subdivisão deste grupo em três subgrupos: idosas saudáveis (n=8), idosas em tratamento de hipertensão arterial (IDHAS,n=6) e idosas em tratamento de dislipidemia (IDDIS,n=6). Foram realizadas avaliações clínica, antropométrica, bioquímica e microcirculatória. Como resultados, a VC mostrou aumento dos diâmetros capilares aferente, apical e eferente e redução da relação velocidade máxima/ velocidade basal de deslocamento de hemácias para grupo de idosas e todos os subgrupos. A POV revelou diminuição da vasodilatação endotélio-dependente em todos os grupos estudados e a vasodilatação endotélio-independente alterou-se apenas em IDHAS e IDDIS. Como conclusões houve, no envelhecimento, alterações estruturais e funcionais da microcirculação assim como redução da vasodilatação endotélio-dependente. A vasodilatação endotélio-independente manteve-se inalterada, indicando que a célula muscular lisa da parede vascular permanece preservada, porém sofre alterações nos grupos IDHAS e IDDIS.

Avaliamos o efeito do consumo materno de SDG (Diglicosídeo Secoisolariciresinol) e de óleo de Linhaça+SDG sobre parâmetros bioquímicos e hormonais das ratas e das proles machos e fêmeas na lactação. As ratas lactantes foram separadas em: controle (C), ração controle cuja proteína foi caseína; (SDG): ração C com 400mg de SDG/Kg de ração; OLSDG: ração C com 400mg de SDG/Kg de ração e 7% de óleo de linhaça. No 14 e 20 dias de lactação as ratas foram ordenhadas e no 21 dia foram sacrificadas por punção cardíaca. Leite e soro foram coletados para avaliação bioquímica e hormonal. Hormônios foram quantificados por radioimunoensaio. As proles machos e fêmeas foram sacrificadas aos 14 e 21 dias de idade. Os animais foram eviscerados para análise da composição corporal. Monitoramos a ingestão alimentar e a massa corporal (MC) durante o período experimental. As ratas SDG apresentaram maior gordura corporal (GC; +39%), enquanto as OLSDG menor conteúdo mineral (-20%) e trigliceridemia (TG) (-39%). As ratas SDG e OLSDG apresentaram hiperprolactinemia (+389% e 153%, respectivamente) sem alteração na concentração de estradiol. No 14 dia de lactação, o leite das ratas OLSDG apresentou menores teores de lactose(-17%) e de proteínas (-20%) e o das ratas SDG apenas menor teor de proteína (-21%). A partir do 13 dia de lactação tanto os machos quanto as fêmeas OLSDG apresentaram menor MC (-14%, -16%, respectivamente). No 14 dia de lactação os machos SDG e OLSDG apresentaram menor gordura corporal (-24%, -55%, respectivamente ) e a prole SDG maior massa de gordura visceral (+39%). Os machos SDG apresentaram maiores concentrações de TG (+105%) e hipoprolactinemia (-41%). Os machos OLSDG também apresentaram hipoprolactinemia (-41%). As fêmeas SDG e OLSDG apresentaram maior estradiol aos 14 dias (+86% e +176%) que se normalizou aos 21 dias, maior colesterolemia (+16%) e as SDG apresentaram maior trigliceridemia (+74%). Aos 21 dias os machos e as fêmeas SDG e OLSDG apresentaram menor trigliceridemia (-48%, -54%,42% e -59%, respectivamente). Os dois componentes principais da semente de linhaça produzem alterações bioquímicas e hormonais tanto nas mães, quanto nas proles, independente do sexo. Entretanto, as alterações observadas diferem entre mães e prole e de acordo com o gênero. Entre as alterações mais importantes ressaltamos a hiperprolactinemia materna que pode ser um dos motivos para a hipoprolactinemia da prole e a hipertrigliceridemia causada pela ingestão de SDG pelas mães.

O objetivo deste estudo foi verificar se o tratamento periodontal não cirúrgico exercia alguma influência sobre o perfil lipídico, os elementos celulares das séries banca e vermelha do sangue, plaquetas e VHS de pacientes portadores de periodontite crônica generalizada. Dezoito pacientes, com média de idade de 50,6 anos ( 7,6), foram submetidos, previamente ao tratamento periodontal e 30 dias após o mesmo, à coleta de 10ml de sangue periférico, através do qual foram analisados o perfil lipídico, os elementos celulares das séries branca e vermelha, o número de plaquetas e VHS. Destes 18 pacientes, 7, com média de idade de 47,4 anos ( 5,9) também foram reavaliados 90 dias após o término do tratamento. Os parâmetros clínicos utilizados, previamente ao tratamento e nas reavaliações, foram o Índice de Placa (IP), de Silness e Löe (1964), o Índice Gengival (IG), de Löe (1967), Sangramento na Sondagem (SS), Profundidade de Bolsa à Sondagem (PBS) e Nível de Inserção (NI). Foram ainda registrados e classificados os sítios com envolvimento de furca. O tratamento periodontal consistiu de terapia básica não cirúrgica. Após 30 dias do término do tratamento periodontal todos os pacientes foram reavaliados sendo verificada melhora significativa (P<0,05) dos valores de IP, IG, SS e PBS e de NI ≥ 6mm (P=0,05). Sítios com envolvimento de furca classes II e III apresentaram também diminuição significativa (P=0,01). Os 7 pacientes submetidos às reavaliações de 30 e 90 dias pós-tratamento também mostraram melhora significativa (P<0,05) dos valores de IP, IG, SS e PBS entre estas fases. Já o NI entre 4-5mm aumentou de forma significativa (P=0,04) entre o pré-tratamento e 90 dias após o mesmo, enquanto que o NI ≥ 6mm diminui significativamente entre as reavaliações de 30 e 90 dias (P=0,01 e P=0,02, respectivamente). Quando comparados os valores de 30 com os de 90 dias resultados semelhantes aos supracitados foram observados, inclusive o aumento do NI entre 4-5mm (P=0,02). É verificado também entre estas fases um IG aumentado (P=0,07). Quanto aos valores hematológicos ocorreu uma diminuição significativa dos níveis de bastões (P=0,05) e de monócitos (P=0,03) após o tratamento periodontal (30 dias), enquanto que o colesterol total e o LDL apresentaram uma tendência ao aumento (P=0,09 para ambos). Já nos sete pacientes submetidos às duas reavaliações o colesterol total apresentou aumento significativo entre as fases pré-tratamento, 30 (P=0,04) e 90 dias (P=0,02) após terapia, assim como o LDL (P=0,04 e P=0,03, respectivamente). Quando comparados os valores plaquetários entre as fases 30 e 90 dias pós-tratamento, verifica-se uma tendência a sua diminuição (P=0,09). O Índice de Castelli II (relação colesterol/HDL) apresenta entre as fases pré e 30 dias pós-tratamento tendência a aumento (P=0,09). Através desses resultados é possível concluir que o tratamento periodontal exerceu influência sobre bastões e monócitos do sangue, caracterizada pela diminuição dessas células, e sobre o colesterol total e o LDL, representada pelo aumento de seus valores.

The high-density holographic recording parameters of a novel two dyes-sensitized photopolymer under different exposure wavelengths are studied. The results show that the maximum diffraction efficiency, exposure sensitivity, maximum refraction index modulation, dynamic range, and the exposure time constant increases with the increase of the exposure wavelength.
