Photodynamic therapy has been described as an effective therapeutic option in selected cases of anogenital lichen sclerosus that are refractory to first-line treatments. However, procedure-related pain is a limiting factor in patient adherence to treatment. The authors report the case of a 75-year-old woman with highly symptomatic vulvar lichen sclerosus, successfully treated with photodynamic therapy. An inhaled 50% nitrous oxide/oxygen premix was administered during sessions, producing a pain-relieving, anxiolytic, and sedative effect without loss of consciousness. This ready-to-use gas mixture may be a well-tolerated and accepted alternative to classical anesthetics in Photodynamic therapy, facilitating patients' adherence to illumination of pain-prone areas.

use of additives (Mg/P and nitrification inhibitor dicyandiamide - DCD), on nitrous oxide emission during swine slurry composting. The experiment was run in duplicate; the gas was monitored for 30 days in different treatments (control, DCD, Mg/P and DCD + Mg/P). Nitrous oxide emissions rate (mg of N2O-N.day-1) and the accumulated emissions were calculated to compare the treatments. Results has shown that emissions of N-N2O were reduced by approximately 70, 46 and 96% through the additions of DCD, MgCl2.6H2O + H3PO4 and both additives, respectively, compared to the control. The aim of this investigation was to determine nitric oxide metabolite levels in saliva samples from hepatitis C virus-positive patients in an attempt to test the hypothesis if increased levels of nitric oxide metabolites correlates with the presence of HCV-RNA in saliva. Saliva of 39 HCV-positive patients and 13 HCV-negative patients, without clinical or laboratorial evidence of liver disease were tested for nitric oxide metabolites. HCV-RNA was detected in serum and saliva by a RT-PCR method and nitric oxide level was determined by evaluation of its stable degradation products, nitrate and nitrite. No differences were found between the concentration of nitrite in saliva from HCV patients and controls, in despite of the presence or not of HCV RNA in saliva. Patients with HCV and cirrhosis had higher concentrations of nitrite but not significantly different from the control group or the groups of anti-HCV patients without cirrhosis. Increased levels of nitrite were not detected in anti-HCV positive patients, an indirect indication that chronic sialoadenitis are infrequent in these patients or occurs with low intensity not sufficient to increase nitric oxide metabolite levels in saliva.

A Parte I teve como objectivo a pesquisa de novos constituintes bioactivos da planta Solanum cernuum Vell. Identificaram-se, pela primeira vez no género Solanum, três triterpenos com um esqueleto de 31-norcicloartanona((+)-cicloeucalenona, (+)-24-oxo-31 norcicloartanona e (+)-24(1,3-dioxetano)-31-norcicloartanona) e quatro alcalóides com uma unidade (2-aminopirrolidin-1-il)carboxamidina acilada com o ácido isoferúlico assumindo configurações E e Z (cernumidina,isocernumidina, cernumidina B e isocernumidina B). A (+)-24-oxo-31-norcicloartanona apresentou actividade anti-neoplásica selectiva para as células de pulmão NCIH460, actividade anti-inflamatória relevante e uma inibição acentuada da expressão da COX-2. A (+)-24(1,3-dioxetano)-31-norcicloartanona apresentou também actividade para as células de leucemia K562. A cernumidina provocou inibição da interleuquina-8 (IL-8) em células do carcinoma do cólon HT-29 superior a 50%. Foram também identificados quercitrina, afzelina, hiperina, ácido ferúlico e Nacetildopamina. Na Parte II desenvolveram-se estudos sobre metodologias de síntese de novos cromóforos derivados da 7-amino-4-metilcumarina. Foi realizado um estudo da reacção de Buchwald-Hartwig para a introdução de grupo(s) electrodoador(es) na posição 7 e de um grupo electroatractor na posição 3 com rendimentos razoáveis. Foram obtidos os compostos 7-(4-metoxifenilamino)-4-metil-2H-cromen-2-ona, 4-metil-7-(fenilamino)- 2H-cromen-2-ona, 3-bromo-7-(4-metoxi-fenilamino)-4-metil-2H-cromen-2-ona e 3- bromo-4-metil-7-(fenilamino)- 2H-cromen-2-ona. Os espectros de absorção destes quatro cromóforos apresentaram c.d.o. máximos desviados para a zona do vermelho. Para a expansão do sistema π pela introdução de um grupo etinileno como ponte conjugada na posição 3 foi necessário a activação desta posição. Para tal, procedeu-se à bromação da N-(4-metil-2-oxo-2H-cromen-7-il)octanamida seguida da reacção de Sonogashira-Hagihara com o álcool propagílico. A oxidação posterior ao aldeído formou a N-(4-metil-2-oxo-3-(3-oxoprop-1- inil)2H-cromen-7-il)octanamida. Numa aproximação convergente foi testada a síntese do ácido 2-cianopent-2-en-4-óico (sem sucesso) para posterior reacção de Sonogashira-Hagihara com 3-bromo-N-(4-metil-2-oxo-2Hcromen-7-il)octanamida.

The growing demand for materials and devices with new functionalities led to the increased inter-est in the field of nanomaterials and nanotechnologies. Nanoparticles, not only present a reduced size as well as high reactivity, which allows the development of electronic and electrochemical devices with exclusive properties, when compared with thin films. This dissertation aims to explore the development of several nanostructured metal oxides by sol-vothermal synthesis and its application in different electrochemical devices. Within this broad theme, this study has a specific number of objectives: a) research of the influence of the synthesis parameters to the structure and morphology of the nanoparticles; b) improvement of the perfor-mance of the electrochromic devices with the application of the nanoparticles as electrode; c) application of the nanoparticles as probes to sensing devices; and d) production of solution-pro-cessed transistors with a nanostructured metal oxide semiconductor. Regarding the results, several conclusions can be exposed. Solvothermal synthesis shows to be a very versatile method to control the growth and morphology of the nanoparticles. The electrochromic device performance is influenced by the different structures and morphologies of WO3 nanoparticles, mainly due to the surface area and conductivity of the materials. The dep-osition of the electrochromic layer by inkjet printing allows the patterning of the electrodes without wasting material and without any additional steps. Nanostructured WO3 probes were produced by electrodeposition and drop casting and applied as pH sensor and biosensor, respectively. The good performance and sensitivity of the devices is explained by the high number of electrochemical reactions occurring at the surface of the na-noparticles. GIZO nanoparticles were deposited by spin coating and used in electrolyte-gated transistors, which promotes a good interface between the semiconductor and the dielectric. The produced transistors work at low potential and with improved ON-OFF current ratio, up to 6 orders of mag-nitude. To summarize, the low temperatures used in the production of the devices are compatible with flexible substrates and additionally, the low cost of the techniques involved can be adapted for disposable devices.

Introduction The immune response caused by Mycobacterium leprae is a risk factor for the development of oxidative stress (OS) in leprosy patients. This study aimed to assess OS in leprosy patients before the use of a multidrug therapy. Methods We evaluated the nitric oxide (NO) concentration; antioxidant capacity; levels of malondialdehyde, methemoglobin and reduced glutathione; and the activity of catalase and superoxide dismutase (SOD) in leprosy patients. Results We observed lower SOD activity in these leprosy patients; however, the NO levels and antioxidant capacity were increased. Conclusions The infectious process in response to M. leprae could primarily be responsible for the OS observed in these patients.
