Abstract Background The p16INK4A gene product halts cell proliferation by preventing phosphorylation of the Rb protein. The p16INK4a gene is often deleted in human glioblastoma multiforme, contributing to unchecked Rb phosphorylation and rapid cell division. We show here that transduction of the human p16INK4a cDNA using the pCL retroviral system is an efficient means of stopping the proliferation of the rat-derrived glioma cell line, C6, both in tissue culture and in an animal model. C6 cells were transduced with pCL retrovirus encoding the p16INK4a, p53, or Rb genes. These cells were analyzed by a colony formation assay. Expression of p16INK4a was confirmed by immunohistochemistry and Western blot analysis. The altered morphology of the p16-expressing cells was further characterized by the senescence-associated β-galactosidase assay. C6 cells infected ex vivo were implanted by stereotaxic injection in order to assess tumor formation. Results The p16INK4a gene arrested C6 cells more efficiently than either p53 or Rb. Continued studies with the p16INK4a gene revealed that a large portion of infected cells expressed the p16INK4a protein and the morphology of these cells was altered. The enlarged, flat, and bi-polar shape indicated a senescence-like state, confirmed by the senescence-associated β-galactosidase assay. The animal model revealed that cells infected with the pCLp16 virus did not form tumors. Conclusion Our results show that retrovirus mediated transfer of p16INK4a halts glioma formation in a rat model. These results corroborate the idea that retrovirus-mediated transfer of the p16INK4a gene may be an effective means to arrest human glioma and glioblastoma.

Abstract Background Oral squamous cell carcinoma (OSCC) is a frequent neoplasm, which is usually aggressive and has unpredictable biological behavior and unfavorable prognosis. The comprehension of the molecular basis of this variability should lead to the development of targeted therapies as well as to improvements in specificity and sensitivity of diagnosis. Results Samples of primary OSCCs and their corresponding surgical margins were obtained from male patients during surgery and their gene expression profiles were screened using whole-genome microarray technology. Hierarchical clustering and Principal Components Analysis were used for data visualization and One-way Analysis of Variance was used to identify differentially expressed genes. Samples clustered mostly according to disease subsite, suggesting molecular heterogeneity within tumor stages. In order to corroborate our results, two publicly available datasets of microarray experiments were assessed. We found significant molecular differences between OSCC anatomic subsites concerning groups of genes presently or potentially important for drug development, including mRNA processing, cytoskeleton organization and biogenesis, metabolic process, cell cycle and apoptosis. Conclusion Our results corroborate literature data on molecular heterogeneity of OSCCs. Differences between disease subsites and among samples belonging to the same TNM class highlight the importance of gene expression-based classification and challenge the development of targeted therapies.

Epidote-group minerals, together with albite, quartz, fluorite, Al-poor and Fe-rich phyllosilicates, zircon, and minor oxides and sulphides, are typical hydrothermal phases in peralkaline alkali-feldspar granites from the Corupá Pluton, Graciosa Province, South Brazil. The epidote-group minerals occur as single crystals and as aggregates filling in rock interstices and miarolitic cavities. They display complex recurrent zoning patterns with an internal zone of ferriallanite-(Ce), followed by allanite-(Ce), then epidote-ferriepidote, and an external zone with allanite-(Ce), with sharp limits, as shown in BSE and X-ray images. REE patterns show decreasing fractionation degrees of LREE over HREE from ferriallanite to epidote. The most external allanite is enriched in MREE. LA-ICP-MS data indicate that ferriallanite is enriched (>10-fold) in Ti, Sr and Ga, and depleted in Mg, Rb, Th and Zr relative to the host granite. Allanite has lower Ga and Mn and higher Zr, Nb and U contents as compared to ferriallanite, while epidote is enriched in Sr, U and depleted in Pb, Zr, Hf, Ti and Ga. The formation of these minerals is related to the variable concentrations of HFSE, Ca, Al, Fe and F in fluids remaining from magmatic crystallization, in an oxidizing environment, close to the HM buffer. L-MREE were in part released by the alteration of chevkinite, their main primary repository in the host rocks.

Este estudo ilustra o impacto da calcinação de resíduo de bauxita (RB) nas propriedades de suspensões formuladas com cimento Portland, tanto no estado fresco como no endurecido. As suspensões foram avaliadas contendo uma razão constante de água-cimento e teor de resíduo variando de 5% a 20% em peso e em substituição ao cimento. As propriedades reológicas e a resistência mecânica foram alteradas em função do aumento do teor de RB, mas a calcinação não teve influência no resultado final obtido, seja no estado fresco ou no endurecido. Assim, pode-se afirmar que a utilização de resíduo de bauxita, natural ou calcinada, em formulações com cimento Portland pode reduzir o consumo de cimento, sendo uma alternativa para a utilização de uma grande quantidade deste tipo de resíduo.

The experiment was conducted to evaluate the bromatological characteristics and the in vitro digestibility of four sugarcane varieties, subjected or not to hydrolysis, with quicklime. A completely randomized design was employed with three replications arranged in a 4 × 2 factorial scheme, with four sugarcane varieties (SP 52454, RB 867515, RB 855536 and IAC 862480), hydrolyzed or not. There was significant effect on brix (p < 0.05) and industrial fiber (p < 0.05), and IAC 862480 variety had the lowest levels of industrial fiber. There were no significant difference (p > 0.05) in neutral detergent fiber, acid detergent fiber and lignin levels among the sugarcane varieties under analysis and for the sugarcanes, hydrolyzed or not. The use of sugarcane hydrolysis with 1% quicklime improves the in vitro digestibility of NDF and ADF, regardless of the variety studied. Hydrolysis with 1% quicklime did not alter the sugarcane chemical composition.

Neste estudo, buscou-se compreender os processos e materiais que contribuíram às fontes mantélicas envolvidas na gênese da Província Magmática do Paraná (PMP). O papel que a crosta, a litosfera e o manto terrestre (raso ou profundo) desempenham na geração de assinaturas químicas e isotópicas em basaltos de derrames continentais e oceânicos tem sido um tema intensamente pesquisado e muito debatido na literatura internacional (e.g., Sheth, 2005). Embora muitos estudos já tenham sido conduzidos na PMP, existe ainda uma grande controvérsia com relação à natureza das regiões-fonte dos magmas (manto litosférico continental ou manto sublitosférico), como também acerca dos mecanismos geodinâmicos que provocam o início da fusão dessas regiões-fonte (e.g., Bellieni et al., 1984; Hawkesworth et al., 1992). Nesse contexto, uma grande quantidade de análises de elementos traço (litófilos e siderófilos), como também dados isotópicos de quatro sistemas de decaimento radioativo (Rb-Sr, Sm-Nd, U-Th-Pb e Re-Os), foram obtidos em basaltos que ocorrem no norte e sul da PMP. A regionalização geoquímica e isotópica (Sr-Nd-Pb) observada nas rochas basálticas da PMP tem sido frequentemente interpretada como resultante da fusão de manto litosférico subcontinental heterogêneo. Entretanto, dados geoquímicos de elementos altamente siderófilos e de razões isotópicas 187Os/188Os, obtidos neste estudo, indicam uma fonte mantélica homogênea, já que os basaltos baixo-TiO2 e alto- TiO2 possuem razões isotópicas de ósmio muito similares (187Os/188Osi = 0,1295±0,0018), as quais são distintas daquelas de manto litosférico subcontinental antigo (Proterozoico ou Arqueano; 187Os/188Os = 0,113). Os dados isotópicos Re-Os exibem uma tendência linear (187Re/188Os versus 187Os/188Os), definindo uma isócrona de boa qualidade (idade = 131,6 ± 2,3 Ma), consistente com as idades obtidas em trabalhos anteriores utilizando o método 40Ar/39Ar. Comumente, a PMP tem sido geneticamente ligada ao hotspot Tristão da Cunha via Rio Grande Rise e Walvis Ridge. No entanto, as razões isotópicas de ósmio determinadas nas rochas da Ilha de Tristão da Cunha são muito distintas daquelas dos basaltos da PMP, reforçando que essa pluma não deve ter contribuído com matéria na gênese dessas rochas. Desta forma, para explicar todas as características isotópicas dos basaltos da PMP é necessário o envolvimento de três componentes mantélicos (Marques et al., 1999). Um deles, empobrecido e semelhante ao DMM, estaria presente na fonte dos basaltos alto-TiO2 e baixo-TiO2, dominando as composições isotópicas de ósmio. Os outros dois componentes enriquecidos (EM-I e EM-II) seriam responsáveis pelas variações nas composições isotópicas de Sr-Nd-Pb. Na gênese dos basaltos alto-TiO2 provavelmente ocorreu envolvimento de crosta continental inferior delaminada (EM-I), enquanto nas rochas baixo-TiO2 há indicação de participação de crosta continental superior reciclada (EM-II). O componente empobrecido possui composição semelhante à de peridotitos de arco, sendo que essa assinatura isotópica pode ter sido gerada por processos metassomáticos relacionados a processos de subducção neoproterozóicas, que teriam modificado o manto astenosférico (DMM). No processo de aglutinação do Gondwana esse manto astenosférico metassomatizado pode ter sido incorporado à base da litosfera, sofrendo posterior refusão, por ocasião da atividade ígnea da PMP.

A análise de elementos traço, tais como terras raras, TH, U, Ta, Hf, Ba, Rb e Ba, é uma ferramenta muito importante para estudos petrogenéticos. No intuito de estudar tais processos em diques do Enxame Serra do Mar (litoral de São Paulo e Rio de Janeiro), pertencente à Provincia Magmática do Paraná (PMP), uma das províncias de basaltos continentais mais expressivas do mundo, foram realizadas análises por ativação com nêutrons nestes diques. A técnica, empregada no Centro de Reator de pesquisa do Instituto de Pesquisas Energéticas e Nucleares, forneceu concentrações de elementos traço com os níveis de precisão de 10% e exatidão 9% os quais são adequadas para estudos petrogenéticos. Devido as baixas concentrações dos elementos analisados, a rotina experimental de preparação das amostras abrangeu processos bastantes cuidadosos para evitar contaminação. Pineal melatonin synthesis can be modulated by many peptides, including insulin. Because melatonin appears to alter leptin synthesis, in this work we aimed to investigate whether leptin would have a role on norepinephrine- (NE-)mediated melatonin synthesis in cultured rat pineal glands. According to our data, cultured rat pineal glands express leptin receptor isoform b (Ob-Rb). Pineal expression of Ob-Rb mRNA was also observed in vivo. Administration of leptin (1 nM) associated with NE ( 1 µM) reduced melatonin content as well as arylalkylamine-N-acetyl transferase (AANAT) activity and expression in cultured pineal glands. Leptin treatment per se induced the expression of STAT3 in cultured pineal glands, but STAT3 does not participate in the leptin modulation of NE-mediated pineal melatonin synthesis. In addition, the expression of inducible cAMP early repressor (ICER) was further induced by leptin challenge when associated with NE. In conclusion, leptin inhibition of pineal melatonin synthesis appears to be mediated by a reduction in AANAT activity and expression as well as by increased expression of Icer mRNA. Peptidergic signaling within the pineal gland appears to be one of the most important signals which modulates melatonin synthesis; leptin, as a member of this system, is not an exception

Low-pressure/high-temperature (LP/HT) metamorphic belts are characterised by rocks that experienced abnormal heat flow in shallow crustal levels (T > 600 °C; P < 4 kbar) resulting in anomalous geothermal gradients (60-150 °C/km). The abnormal amount of heat has been related to crustal underplating of mantle-derived basic magmas or to thermal perturbation linked to intrusion of large volumes of granitoids in the intermediate crust. In particular, in this latter context, magmatic or aqueous fluids are able to transport relevant amounts of heat by advection, thus favouring regional LP/HT metamorphism. However, the thermal perturbation consequent to heat released by cooling magmas is responsible also for contact metamorphic effects. A first problem is that time and space relationships between regional LP/HT metamorphism and contact metamorphism are usually unclear. A second problem is related to the high temperature conditions reached at different crustal levels. These, in some cases, can completely erase the previous metamorphic history. Notwithstanding this problem is very marked in lower crustal levels, petrologic and geochronologic studies usually concentrate in these attractive portions of the crust. However, only in the intermediate/upper-crustal levels of a LP/HT metamorphic belt the tectono-metamorphic events preceding the temperature peak, usually not preserved in the lower crustal portions, can be readily unravelled. The Hercynian Orogen of Western Europe is a well-documented example of a continental collision zone with widespread LP/HT metamorphism, intense crustal anatexis and granite magmatism. Owing to the exposure of a nearly continuous cross-section of the Hercynian continental crust, the Sila massif (northern Calabria) represents a favourable area to understand large-scale relationships between granitoids and LP/HT metamorphic rocks, and to discriminate regional LP/HT metamorphic events from contact metamorphic effects. Granulite-facies rocks of the lower crust and greenschist- to amphibolite-facies rocks of the intermediate-upper crust are separated by granitoids emplaced into the intermediate level during the late stages of the Hercynian orogeny. Up to now, advanced petrologic studies have been focused mostly in understanding P-T evolution of deeper crustal levels and magmatic bodies, whereas the metamorphic history of the shallower crustal levels is poorly constrained. The Hercynian upper crust exposed in Sila has been subdivided in two different metamorphic complexes by previous authors: the low- to very low-grade Bocchigliero complex and the greenschist- to amphibolite-facies Mandatoriccio complex. The latter contains favourable mineral assemblages in order to unravel the tectono-metamorphic evolution of the Hercynian upper crust. The Mandatoriccio complex consists mainly of metapelites, meta-arenites, acid metavolcanites and metabasites with rare intercalations of marbles and orthogneisses. Siliciclastic metasediments show a static porphyroblastic growth mainly of biotite, garnet, andalusite, staurolite and muscovite, whereas cordierite and fibrolite are less common. U-Pb ages and internal features of zircons suggest that the protoliths of the Mandatoriccio complex formed in a sedimentary basin filled by Cambrian to Silurian magmatic products as well as by siliciclastic sediments derived from older igneous and metamorphic rocks. In some localities, metamorphic rocks are injected by numerous aplite/pegmatite veins. Small granite bodies are also present and are always associated to spotted schists with large porphyroblasts. They occur along a NW-SE trending transcurrent cataclastic fault zone, which represents the tectonic contact between the Bocchigliero and the Mandatoriccio complexes. This cataclastic fault zone shows evidence of activity at least from middle-Miocene to Recent, indicating that brittle deformation post-dated the Hercynian orogeny. P-T pseudosections show that micaschists and paragneisses of the Mandatoriccio complex followed a clockwise P-T path characterised by four main prograde phases: thickening, peak-pressure condition, decompression and peak-temperature condition. During the thickening phase, garnet blastesis started up with spessartine-rich syntectonic core developed within micaschists and paragneisses. Coevally (340 ± 9.6 Ma), mafic sills and dykes injected the upper crustal volcaniclastic sedimentary sequence of the Mandatoriccio complex. After reaching the peak-pressure condition (≈4 kbar), the upper crust experienced a period of deformation quiescence marked by the static overgrowths of S2 by Almandine-rich-garnet rims and by porphyroblasts of biotite and staurolite. Probably, this metamorphic phase is related to isotherms relaxation after the thickening episode recorder by the Rb/Sr isotopic system (326 ± 6 Ma isochron age). The post-collisional period was mainly characterised by decompression with increasing temperature. This stage is documented by the andalusite+biotite coronas overgrown on staurolite porphyroblasts and represents a critical point of the metamorphic history, since metamorphic rocks begin to record a significant thermal perturbation. Peak-temperature conditions (≈620 °C) were reached at the end of this stage. They are well constrained by some reaction textures and mineral assemblages observed almost exclusively within paragneisses. The later appearance of fibrolitic sillimanite documents a small excursion of the P-T path across the And-Sil boundary due to the heating. Stephanian U-Pb ages of monazite crystals from the paragneiss, can be related to this heating phase. Similar monazite U-Pb ages from the micaschist combined with the lack of fibrolitic sillimanite suggest that, during the same thermal perturbation, micaschists recorded temperatures slightly lower than those reached by paragneisses. The metamorphic history ended with the crystallisation of cordierite mainly at the expense of andalusite. Consequently, the Ms+Bt+St+And+Sill+Crd mineral assemblage observed in the paragneisses is the result of a polyphasic evolution and is characterised by the metastable persistence of the staurolite in the stability fields of the cordierite. Geologic, geochronologic and petrographic data suggest that the thermal peak recorded by the intermediate/upper crust could be strictly connected with the emplacement of large amounts of granitoid magmas in the middle crust. Probably, the lithospheric extension in the relatively heated crust favoured ascent and emplacement of granitoids and further exhumation of metamorphic rocks. After a comparison among the tectono-metamorphic evolutions of the different Hercynian crustal levels exposed in Sila, it is concluded that the intermediate/upper crustal level offers the possibility to reconstruct a more detailed tectono-metamorphic history. The P-T paths proposed for the lower crustal levels probably underestimate the amount of the decompression. Apart from these considerations, the comparative analysis indicates that P-T paths at various crustal levels in the Sila cross section are well compatible with a unique geologic scenario, characterized by post-collisional extensional tectonics and magmas ascent.
Intrinsic uncoupling in the ATP synthase of Escherichia coli. Studies on WT and ε-truncated mutants

The H+/ATP ratio in the catalysis of ATP synthase has generally been considered a fixed parameter. However, Melandri and coworkers have recently shown that, in the ATP synthase of the photosynthetic bacterium Rb.capsulatus, this ratio can significantly decrease during ATP hydrolysis when the concentration of either ADP or Pi is maintained at a low level (Turina et al., 2004). The present work has dealt with the ATP synthase of E.coli, looking for evidence of this phenomenon of intrinsic uncoupling in this organism as well. First of all, we have shown that the DCCD-sensitive ATP hydrolysis activity of E.coli internal membranes was strongly inhibited by ADP and Pi, with a half-maximal effect in the submicromolar range for ADP and at 140 µM for Pi. In contrast to this monotonic inhibition, however, the proton pumping activity of the enzyme, as estimated under the same conditions by the fluorescence quenching of the ÎpH-sensitive probe ACMA, showed a clearly biphasic progression, both for Pi, increasing from 0 up to approximately 200 µM, and for ADP, increasing from 0 up to a few µM. We have interpreted these results as indicating that the occupancy of ADP and Pi binding sites shifts the enzyme from a partially uncoupled state to a fully coupled state, and we expect that the ADP- and Pi-modulated intrinsic uncoupling is likely to be a general feature of prokaryotic ATP synthases. Moreover, the biphasicity of the proton pumping data suggested that two Pi binding sites are involved. In order to verify whether the same behaviour could be observed in the isolated enzyme, we have purified the ATP synthase of E.coli and reconstituted it into liposomes. Similarly as observed in the internal membrane preparation, in the isolated and reconstituted enzyme it was possible to observe inhibition of the hydrolytic activity by ADP and Pi (with half-maximal effects at few µM for ADP and at 400 µM for Pi) with a concomitant stimulation of proton pumping. Both the inhibition of ATP hydrolysis and the stimulation of proton pumping as a function of Pi were lost upon ADP removal by an ADP trap. These data have made it possible to conclude that the results obtained in E.coli internal membranes are not due to the artefactual interference of enzymatic activities other than the ones of the ATP synthase. In addition, data obtained with liposomes have allowed a calibration of the ACMA signal by ÎpH transitions of known extent, leading to a quantitative evaluation of the proton pumping data. Finally, we have focused our efforts on searching for a possible structural candidate involved in the phenomenon of intrinsic uncoupling. The ε-subunit of the ATP-synthase is known as an endogenous inhibitor of the hydrolysis activity of the complex and appears to undergo drastic conformational changes between a non-inhibitory form (down-state) and an inhibitory form (up-state)(Rodgers & Wilce, 2000; Gibbons et al., 2000). In addition, the results of Cipriano & Dunn (2006) indicated that the C-terminal domain of this subunit played an important role in the coupling mechanism of the pump, and those of Capaldi et al. (2001), Suzuki et al. (2003) were consistent with the down-state showing a higher hydrolysis-to-synthesis ratio than the up-state. Therefore, we decided to search for modulation of pumping efficiency in a C-terminally truncated ε mutant. A low copy number expression vector has been built, carrying an extra copy of uncC, with the aim of generating an ε-overexpressing E.coli strain in which normal levels of assembly of the mutated ATP-synthase complex would be promoted. We have then compared the ATP hydrolysis and the proton pumping activity in membranes prepared from these ε-overexpressing E.coli strains, which carried either the WT ε subunit or the ε88-stop truncated form. Both strains yielded well energized membranes. Noticeably, they showed a marked difference in the inhibition of hydrolysis by Pi, this effect being largely lost in the truncated mutant. However, pre-incubation of the mutated enzyme with ADP at low nanomolar concentrations (apparent Kd = 0.7nM) restored the hydrolysis inhibition, together with the modulation of intrinsic uncoupling by Pi, indicating that, contrary to wild-type, during membrane preparation the truncated mutant had lost the ADP bound at this high-affinity site, evidently due to a lower affinity (and/or higher release) for ADP of the mutant relative to wild type. Therefore, one of the effects of the C-terminal domain of ε appears to be to modulate the affinity of at least one of the binding sites for ADP. The lack of this domain does not appear so much to influence the modulability of coupling efficiency, but instead the extent of this modulation. At higher preincubated ADP concentrations (apparent Kd = 117nM), the only observed effects were inhibition of both hydrolysis and synthesis, providing a direct proof that two ADP-binding sites on the enzyme are involved in the inhibition of hydrolysis, of which only the one at higher affinity also modulates the coupling efficiency.

Zusammenfassung:Mit Hilfe einer neuen Formel für die Minerale der Pyrochlor-Gruppe werden sämtliche Endglieder der Na-Ca-Mikrolithe und der Ba-haltigen Mikrolithe aus der Pegmatit-Provinz Nazareno beschrieben: Die Na-reichsten Proben haben nahezu die Idealzusammensetzung eines idealen Pyrochlors, d.h. . Die Ca-reichsten Varietäten weisen maximal auf, wobei der Besetzungsanteil des Ca am A2+ ca. 93% beträgt. Die Ba-haltigen Mikrolithe sind durch eine Defektstruktur gekennzeichnet, wobei für das mögliche Endglied kein Beispiel in den Daten vorliegt. Das Endglied mit dem geringsten Defektcharakter hat folgende Stöchiometrie:

Tertiäre Vulkanite aus dem Eckfelder Maar, dem Hillscheider Diatrem und dem Hillscheid Basalt (Schlot) wurden petrologisch und geochemisch untersucht. Bis auf tonige Klasten aus dem Bohrkern des Eckfelder Maares handelt es sich bei allen weiteren Proben um undifferenzierte basische Vulkanite. Die tonigen Klasten aus dem Bohrkern müssen der ehemaligen Landoberfläche vor der Eruption des Eckfelder Maares zugerechnet werden, in dessen Krater sie während der Eruption hineingefallen sind. Bis auf die Proben des Hillscheid Basaltes sind die Proben alteriert. Die Alteration zeigt sich an der Bildung von Zeolithen und Calcitmineralisationen, die primär und sekundär gebildete Hohlräume aufgefüllt haben oder an einer vertonten Grundmasse der Proben, die daneben Mineraleinschlüsse (Spinell) und kantige Fremdgesteinsbruchstücke enthalten können. Bei den Proben mit vertonter Grundmasse handelt es sich um Palagonite, Umwandlungsprodukte aus Sideromelan (basaltischem Glas). Geochemische Analysen an Grundmassepräparaten der alterierten bis vertonten Proben zeigen, dass außer den immobilen Elementen Ti, Nb, Zr, Y alle weiteren Elemente teilweise bis vollständig abgereichert worden sind. Eine Ausnahme bildet Barium (Ba), welches z.T. in beträchtlichen Mengen in Zeolithen (Harmotom) angereichert wurde. Bei den Proben aus dem Eckfelder Maar kann die Alteration bis Vertonung der Proben alleine mit der Palagonitisierung und Verwitterung erklärt werden. Es gibt keine Hinweise auf Materialzufuhr und damit für sich anschließende hydrothermale Prozesse. Die Proben des Hillscheider Diatrem sind wesentlich geringer alteriert (glasige Grundmasse). Neuste Erkenntnisse aus einer Bohrung im Sommer 1999 im vermuteten Zentrum des Hillscheider Diatrems beschränken das Diatrem maximal auf einen kleineren Bereich im Nordosten der bisherigen Lokation. Bei der Bohrung stieß man nach 20 Meter auf Anstehendes. Im Hangschutt darüber fand man Blöcke des Hillscheid Basaltes. Eine geringere Größe der Lokation zusammen mit der geringen Alteration könnten auf deren Entstehung mit einer initialen Maarphase gefolgt von Schlackentätigkeit hinweisen. Die Schlacken könnten die ersten Ablagerungen vor Verwitterung geschützt haben. Allerdings gibt es keine Funde die eine Schlackentätigkeit belegen. Beim sogenannten 'Hillscheider Diatrem' könnte es sich aber auch um Hangschutt aus der Randbreccie des Hillscheider Basaltes handeln. Zusammen mit Bruchstücken aus dem Schlot des Hillscheid Basaltes wären die Palagonite des sogenannten 'Hillscheider Diatrem' erst in jüngster Zeit im Bereich einer Uferböschung zur Ablagerung gekommen. Dies würde allerdings das sogenannte 'Hillscheider Diatrem' in seiner Existenz in Frage stellen. Vergleiche der Proben des Hillscheid Basaltes mit basischen Hocheifelvulkaniten deuten auf kogenetische Beziehung aller Proben untereinander und ordnen die Proben des Hillscheid Basaltes geochemisch dem Hocheifelvulkanismus zu. REE- und weitere Spurenelementgehalte und auch deren Elementverhältnisse weisen für alle tertiären Eifelvulkanite gemeinsam auf Mantelschmelzen aus dem Bereich eines Granatperidotits mit niedrigen Aufschmelzgraden um ein Prozent hin. Vergleich der Elementverhältnisse hochinkompatibler Elemente im Bezug auf die Bildung mafischer Schmelzen mit primitivem Mantel deuten darauf hin, dass der Mantel im Bereich der Hocheifel verarmt ist an K, Rb, Sr und angereichert an Ba und eventuell an Nb. Ursachen für diese von typisch primären Mantelzusammensetzung abweichenden Verhältnisse könnten durch Mischungen von Mantelschmelzen mit lithosphärischem Mantel (K-Anomalie) und durch Anreicherungen mit fluiden Phasen (Ba-Anomalie) oder auch Schmelzen aus einem tieferliegenden Plume (Kelberger Hoch) verursacht worden sein. Caveolin-1 (Cav-1), the essential structural constituent of caveolae, which are flask-shaped invaginations of the plasma membrane, has been found to play a key role in the modulation of cell proliferation and cancer development. It seems to act as an oncosuppressor or a promoter of growth, depending on the histotype, stage and grade of each tumour. The aim of this study was to analyze the effects of Caveolin-1 gene silencing on the proliferation of human lung cancer and osteosarcoma in vitro. Our data show that Cav-1 silencing blocks the growth in both metastatic lung cancer cell lines analyzed, suggesting a proliferation promoting action of the protein in these cells. A marked decrease of phospho-Akt, phospho-ERK, STAT3, cyclin D1, CDK4 and consequently of phospho-Rb expression was evident in the cells treated with Cav-1 siRNA. With regards to osteosarcoma, we demonstrated that the suppression of Cav-1 results in the blocking of MG-63 and in the slowing down of HOS proliferation, suggesting a role for Cav-1 as a promoter of tumour growth in these cell lines. A marked decrease of phospho-Akt, cyclin E, CDK2 and phospho-Rb and an increase of p21 expression levels were evident in the cells treated with Cav-1 siRNA. Our results suggest two new cell cycle inhibiting pathways, mediated by Cav-1 knock-down, and provide new insights into the molecular mechanisms underlying the tumour-promoting role of Cav-1 in lung cancer and osteosarcoma. In this work we also investigated the role of estrogens in lung cancer and the functional cross-talk between Cav-1 and estrogens/estrogen receptors in it. Our results show that 17β-estradiol induces proliferation either in RAL or in SCLC-R1 cells and that both cell lines are sensitive to 4-OHT antiproliferative effect. The sensitivity to estrogen stimulation seems to be gender- and/or histological type-independent in metastatic lung cancer in vitro.

In dieser Arbeit werden geochronologische und isotopen-geochemische Daten zur Entwicklung der Zentralen Westlichen Karpathen präsentiert. Die Karpathen bilden die östliche Fortsetzung der Alpen und können in drei Alpine Grundgebirgsdecken unterteilt werden, von denen zwei, die Veporische und die Gemerische, bearbeitet wurden. In der Veporischen Einheit wurden polymetamorphe Grundgebirgseinheiten untersucht, um deren genaue Altersstellung zu definieren und sie isotopengeochemisch zu klassifizieren. Dagegen wurde in der der Gemerischen Einheit, welche die Veporische Einheit überlagert, ein spezialisierter S-Typ Granit im Detail untersucht, um die petrogenetischen Prozesse, die zur magmatischen Entwicklung dieses Granits geführt haben, zu identifizieren. U-Pb Datierungen an Zirkonen der Veporischen Grundgebirgseinheiten zeigen für die gesamte Veporische Einheit ordovizische Entsehungsalter an (440-470 Ma). Diese Datierungen revidieren publizierte kambrische Entstehungsalter dieses Grundgebirges. Die Isotopensignatur (epsilon Nd und 87Sr/86Sr) der ordovizischen Grundgebirgseinheiten, bestehend aus stark überprägten Amphiboliten und Gneissen, ist von der Signatur der sich im Norden anschliessenden Tatrischen Einheit gut unterscheidbar. Die Bleiisotopenzusammensetzung dieser Gesteine ist stark krustal geprägt und überschneidet sich mit der der Tatrischen Einheit. Zusammen mit den T-DM Altern sind diese Einheiten vergleichbar mit prävariskischen Einheiten der Alpen. Somit kann das ordovizische Grundgebirge zu den peri-Gondwana Terranen gezählt werden, die an einem aktiven Kontinentalrand im Norden von Gondwana gebildet wurden. In den Gesteinen der Veporischen Einheit wurde im Weiteren eine starke metamorphe überprägung und intensiver felsischer Magmatismus karbonischen Alters erkannt (320-350 Ma). Dieses Ereignis ist zeitgleich mit dem Magmatismus, welcher hauptsächlich in der sich im Norden anschliessenden Tatrischen Einheit beobachtet wird. Dieser gehört der variskischen Orogenese an. Intensive alpine Deformation und Metamorphose konnte in der südlichen Veporischen Einheit anhand der Einzelzirkondatierungen und der Isotopendaten der ordovizischen Einheiten nachgewiesen werden. Am Dlha Dolina Granit in der Gemerischen Einheit können starke Fraktionierungs- und Auto-Metasomatose-Effekte beobachtet werden. Durch die magmatische Fraktionierung wird eine Anreicherung der SEE erzeugt, wogegen die Metasomatose die SEE stark verarmt. Es kommt sogar zur Ausbildung eines Tetraden Effektes im SEE Muster, welche den starken Einfluss von Fluiden während der spät-magmatischen Phase belegt. Gesamtgesteins Pb-Pb Daten beschränken das minimale Intrusionsalter dieses Granites auf 240 Ma. Dieses Alter ist in guter übereinstimmung mit den Sr-Isotopendaten der magmatisch dominierten Gesteine, wohingegen die stark metasomatisch geprägten Gesteine ein zu radiogenes 87Sr/86Sri aufweisen. Während dieser Arbeit wurde intensiv mit der Blei-Isotopenzusammensetzung von Gesamtgesteinsproben gearbeitet. Um die Auswertung dieser Daten optimieren zu können wurde ein Computerscript für das GPL Programm Octave erstellt. Die Hauptaufgabe dieses Scripts besteht darin, Regressionen für geochronologische Anwendungen gemäss York (1969) zu berechnen. Ausserdem können mu und kappa-Werte für diese Regressionen berechnet und eine Hauptkomponentenanalyse, welche hilfreich für den Vergleich von zwei Datensätzen ist, durchgeführt werden. Am Ende der vorliegenden Arbeit wird die analytische Methode für einen Mikrowellen beschleunigten Säureaufschluss von granitoidem Material zur Bestimmung der Sr- und Nd-Isotopenzusammensetzung und der Elementkonzentrationen vorgestellt. Diese kombinierte Methode nutzt ein TIMS für die Sr und Nd Isotopenmessungen und eine Einzelkollektor-ICPMS zur Bestimmung der SEE, Rb und Sr Konzentrationen, welche mithilfe von relativen Sensitivitätsfaktoren gegenüber einem internen Standard quantifiziert werden. Diese Methode wird durch Messungen von internationalen Referenzmaterialien bewertet. Die Ergebnisse zeigen eine Reproduzierbarkeit von <10% für die Elementkonzentrationen und von <5% für Elementverhältnisse.
