The rates of whole body nitric oxide (NO) synthesis, plasma arginine flux, and de novo arginine synthesis and their relationships to urea production, were examined in a total of seven healthy adults receiving an L-amino acid diet for 6 days. NO synthesis was estimated by the rate of conversion of the [15N] guanidino nitrogen of arginine to plasma [15N] ureido citrulline and compared with that based on urinary nitrite (NO2-)/nitrate (NO3-) excretion. Six subjects received on dietary day 7, a 24-hr (12-hr fed/12-hr fasted) primed, constant, intravenous infusion of L-[guanidino-15N2]arginine and [13C]urea. A similar investigation was repeated with three of these subjects, plus an additional subject, in which they received L-[ureido-13C]citrulline, to determine plasma citrulline fluxes. The estimated rates (mean +/- SD) of NO synthesis over a period of 24 hr averaged 0.96 +/- 0.1 mumol .kg-1.hr-1 and 0.95 +/- 0.1 mumol.kg-1.hr-1, for the [15N]citrulline and the nitrite/nitrate methods, respectively. About 15% of the plasma arginine turnover was associated with urea formation and 1.2% with NO formation. De novo arginine synthesis averaged 9.2 +/- 1.4 mumol. kg-1.hr-1, indicating that approximately 11% of the plasma arginine flux originates via conversion of plasma citrulline to arginine. Thus, the fraction of the plasma arginine flux associated with NO and also urea synthesis in healthy humans is small, although the plasma arginine compartment serves as a significant precursor pool (54%) for whole body NO formation. This tracer model should be useful for exploring these metabolic relationships in vivo, under specific pathophysiologic states where the L-arginine-NO pathway might be altered.

A elevada concentração de cloro das bifenilas policloradas provoca alta toxicidade do composto, o qual dificulta sua biodegradação. A contaminação de PCB no Brasil foi confirmada em estudo realizado na Bahia de Santos-São Vicente (São Paulo), o qual revelou a necessidade de um plano de ação para o controle e remoção de PCB no Brasil. Pretendeu-se assim, na realização da presente pesquisa, verificar quatro hipóteses: (1) A técnica de Microextração em fase sólida é uma metodologia eficaz para avaliação de bifenilas policloradas de amostras de reatores; (2) A condição fermentativa-metanogênica abriga comunidade resistente ao PCB, e removê-lo; (3) A condição desnitrificante abriga comunidade resistente ao PCB, e removê-lo e (4) A remoção de PCB, bem como, a composição microbiana é distinta em cada condição metabólica. Para tanto, reatores em batelada foram montados separadamente com biomassa anaeróbia proveniente de reator UASB usado no tratamento de água residuária de avicultura e biomassa de sistemas de lodos ativados de tratamento de esgoto sanitário. Os reatores operados em condição mesófila foram alimentados com meio sintético, co-substratos, sendo etanol (457 mg.L-1) e formiato de sódio (680 mg.L-1) para os reatores anaeróbios, e somente etanol (598 mg.L-1) para os reatores anóxicos, além de PCB padrão Sigma (congêneres PCBs 10, 28, 52, 153, 138 e 180) em diferentes concentrações, dependendo do objetivo do ensaio. A aplicação do método de extração por SPME com análise em cromatógrafo gasoso com detector por captura de elétrons foi adequada para a determinação dos seis congêneres de PCB. Obteve-se ampla faixa de linearidade, seletividade frente aos vários interferentes, além da robustez do método, utilidade e confiabilidade na identificação e quantificação específica dos seis congêneres de PCB. A Hipótese 1 foi aceita; ou seja, por meio da aplicação da metodologia SPME foi possível quantificar os PCB nos reatores em batelada. Apesar de ter sido comprovada a inibição metanogênica na presença de PCB, com IC50 de 0,03 mg.L

O fenômeno conhecido como Nitrificação e Desnitrificação Simultânea (SND) significa que em um mesmo reator ocorre simultaneamente a nitrificação e a desnitrificação, sob condições de operações idênticas, podendo ser justificada principalmente pela teoria de microambiente no floco ou biofilme. Assim, em um único reator, sob condições controladas de oxigênio dissolvido (OD) e elevados tempos de residênciacelular épossível que ocorra a nitrificação e a criação de zonas anóxicas no interior dos flocos ou biofilme para a ocorrência da desnitrificação. Neste sentido, a tecnologia MBBR/IFAStem como característicaelevado tempo de residência celular do biofilme formado nos meios suporte presentes no reator. Deste modo, neste estudo avaliou-se a remoção de nitrogênio via SND em um sistema IFAS quando submetido a diferentes concentrações de OD e Tempo de DetençãoHidraulica de 5,5 e 11 horas, tratando efluente sanitário e efluente sintético. Os resultados experimentais demonstraram que pode ser possível desenvolver efetiva SND com concentrações de OD média de 1,0 mg.L-1 e 1,5 mg.L-1. Sendo que, foram obtidas eficiência média de remoção de NTde cerca de 68% e concentrações médias efluente de N-NH4 de aproximadamente 5,0 mg L-1, de N-NO3 inferiores a 4,5 mg L-1 e de N-NO2 em torno de 0,1 mg L-1, e com eficiência média de remoção DQO solúvel acima de 90%, quando empregado efluente sintético. Ademais, por meio da avaliação da emissão de Óxido Nitroso (N2O), foi possível comprovar que a desnitrificação ocorreu de forma efetiva.

O uso ineficiente do nitrogênio (N) é um problema global, que pode diminuir a sustentabilidade da produção agrícola da cana-de-açúcar para fins energéticos devido às emissões de N2O oriundas da fertilização nitrogenada. A eficiência de utilização de nitrogênio (EUN) tem sido pouco estudada em genótipos de cana-de-açúcar, o que pode ser preocupante devido a importância da cultura para o país. O objetivo desse estudo foi avaliar a EUN de genótipos de cana-de-açúcar, verificando se genótipos contrastantes tem preferência de absorção por amônio ao invés de nitrato, e checar se extratos radiculares de Brachiaria humidicola e Saccharum spontaneum aumentam a eficiência de utilização de N pela cultura. Para isso três experimentos foram desenvolvidos em condições controladas em casa-de-vegetação e câmara de crescimento. No primeiro deles, 18 genótipos foram submetidos a dois níveis de N: limitante (baixo N) e suficiente (alto N). Posteriormente, quatro genótipos selecionados quanto a EUN do primeiro experimento, foram cultivados em câmara de crescimento por 69 dias, onde receberam quantidades conhecidas de N-fertilizante marcado ou no 15NH4+ ou no 15NO3-, sendo avaliados 24 e 72 horas após a aplicação do fertilizante marcado. Um terceiro ensaio foi realizado, em que extratos radiculares de Brachiaria humidicola (BCH) e Saccharum spontaneum (SCS) foram aplicados ao solo com o intuito de verificar seu potencial como inibidor da nitrificação, comparando-os com a dicianodiamida (DCD), e seu efeito para o aumento do aproveitamento do N pela cana-de-açúcar. No experimento de EUN verificou-se grande variação entre os genótipos. Dos 18 genótipos avaliados, seis foram classificados como responsivos a N e eficientes na utilização de N, três foram não eficientes e responsivos; dois foram eficientes e não responsivos, e sete não eficientes e não responsivos. Esse resultado comprova que a recomendação da adubação nitrogenada pode realmente estar pouco precisa, pois não leva em consideração a EUN de cada genótipo, sendo realizada uma aplicação genérica. As medições de fotossíntese mostraram que os genótipos mais eficientes na utilização do N, não necessariamente possuem as taxas fotossintéticas mais elevadas, estando a EUN diretamente relacionada à quantidade de raízes das plantas. No segundo trabalho, verificou-se que o N na planta proveniente do fertilizante (NPPF), 72 horas após a aplicação do N, foi 36% menor quando a fonte marcada foi NO3-, atestando que a cana-de-açúcar tem preferência de absorção por amônio em relação ao nitrato nos primeiros dias após a adubação nitrogenada. Por fim, os resultados do terceiro ensaio mostraram que a aplicação de sulfato de amônio (SA), associado ao inibidor sintético DCD, manteve os valores de N-NO3- baixos ao longo de todo período de avaliação (durante 60 dias), enquanto que com SA, SA+BCH e SA+SCS houve um aumento na concentração de nitrato no solo já a partir de 15 dia após a fertilização. O uso de DCD reduziu o fluxo médio de N2O durante o período de avaliação em relação ao uso isolado do fertilizante, o que não foi observado com o uso de extratos de raízes de Brachiaria humidicola e Saccharum spontaneum

The nitrogen cycle (N-cycle), principally supported by prokaryotes, involves different redox reactions mainly focused on assimilatory purposes or respiratory processes for energy conservation. As the N-cycle has important environmental implications, this biogeochemical cycle has become a major research topic during the last few years. However, although N-cycle metabolic pathways have been studied extensively in Bacteria or Eukarya, relatively little is known in the Archaea. Halophilic Archaea are the predominant microorganisms in hot and hypersaline environments such as salted lakes, hot springs or salted ponds. Consequently, the denitrifying haloarchaea that sustain the nitrogen cycle under these conditions have emerged as an important target for research aimed at understanding microbial life in these extreme environments. The haloarchaeon Haloferax mediterranei was isolated 20 years ago from Santa Pola salted ponds (Alicante, Spain). It was described as a denitrifier and it is also able to grow using NO3-, NO2- or NH4+ as inorganic nitrogen sources. This review summarizes the advances that have been made in understanding the N-cycle in halophilic archaea using Hfx mediterranei as a haloarchaeal model. The results obtained show that this microorganism could be very attractive for bioremediation applications in those areas where high salt, nitrate and nitrite concentrations are found in ground waters and soils.

A series of CeO2–Nb2O5 mixed oxides with different Nb content, as well as the pure oxides, have been synthesized by co-precipitation with excess urea. These materials have been used as supports for platinum catalysts, with [Pt(NH3)4](NO3)2 as precursor. Both supports and catalysts have been characterized by several techniques: N2 physisorption at 77 K, X-ray diffraction, Raman spectroscopy, X-ray photoelectron spectroscopy, UV–vis spectroscopy, scanning electron microscopy, transmission electron microscopy, temperature-programmed reduction and temperature-programmed desorption (CO and H2), and their catalytic behaviour has been determined in the PROX reaction, both with an ideal gas mixture (CO, O2 and H2) and in simulated reformate gas containing CO2 and H2O. Raman spectroscopy analysis has shown the likely substitution of some Ce4+ cations by Nb5+ to some extent in supports with low niobium contents. Moreover, the presence of Nb in the supports hinders their ability to adsorb CO and to oxidize it to CO2. However, an improvement of the catalytic activity for CO oxidation is obtained by adding Nb to the support, although the Pt/Nb2O5 catalyst shows very low activity. The best results are found with the Pt/0.7CeO2–0.3Nb2O5 catalyst, which shows a high CO conversion (85%) and a high yield (around 0.6) after a reduction treatment at 523 K. The effect of the presence of CO2 and H2O in the feed has also been determined.

While party membership figures are clearly in decline in several Western countries, different interpretations have been offered on the likely consequences of this trend. Some authors stress that members have lost most of their importance for political parties that increasingly rely on professionalized campaign techniques. Other scholars have expressed concern about the decline of party membership. They emphasize the fact that party members continue to function as an important linkage mechanism providing a structural alignment between the party and society (and thus also to potential voters). By means of an election forecasting model for Belgium, we test whether party membership figures still can be related to election results. Results show that party membership has a strong effect on election results, and furthermore, that this relation does not weaken during the period under investigation (1981-2010). The analysis also demonstrates that forecasting models can also be used in a complex multiparty system like Belgium.

Sediment porewater oxygen profiles were measured with micro and needle electrodes in sediment cores of 27 stations in the Skagerrak (northeastern North Sea). Oxygen penetration depth ranged from 3 to 20 mm depth. Fluxes estimated from the oxygen gradients varied from 3 to 18 mmol m**-2 d**-1. Oxygen penetration and flux depend on water depth, but possibly more on the hydrological conditions, related to the import of fresh organic matter by primary production in the water column. Oxygen fluxes were not related to the total organic carbon (TOC) content of the sediments. Stations in the eastern part of the Skagerrak showed high burial rates of TOC. At 6 stations porewater chemistry of Fe, Mn and NO3- was strongly associated with the oxygen distribution. The average relative contribution of terminal electron acceptors to carbon mineralisation was estimated at 85% for O2, 0.5% for Mn, 4.5% for [NO3]3-, 1% for Fe and 9% for [SO4]2-. At one station the occurrence of exceptionally high solid manganese oxyhydroxides was probably related to an active internal manganese cycle.

Biogeochemical reef studies carried out in 1981 and 1984 found low concentration of total natural and anthropogenic hydrocarbons in inshore waters. Detection of lignin in marine and bottom sediments indicates that the land has major effect on makeup of organic matter there. Comparison of compositions of organic matter in sea water, suspended matter and bottom sediments indicated that it was altered rapidly by the reef community. Thus, in the inshore zone of the island, runoff from the land is important in supplying nutrients to the reef ecosystem alongside with transport of nutrients by deep waters. Concentrations of nutri¬ents (N, P) in the inshore zone are higher than in waters of the tropical part of the ocean. Nitrogen is the limiting element in development of phytoplankton in the inshore zone.

We present sedimentary geochemical data and in situ benthic flux measurements of dissolved inorganic nitrogen (DIN: NO3-, NO2-, NH4+) and oxygen (O2) from 7 sites with variable sand content along 18°N offshore Mauritania (NW Africa). Bottom water O2 concentrations at the shallowest station were hypoxic (42 µM) and increased to 125 µM at the deepest site (1113 m). Total oxygen uptake rates were highest on the shelf (-10.3 mmol O2 /m2 d) and decreased quasi-exponentially with water depth to -3.2 mmol O2 /m2 d. Average denitrification rates estimated from a flux balance decreased with water depth from 2.2 to 0.2 mmol N /m2 d. Overall, the sediments acted as net sink for DIN. Observed increases in delta 15NNO3 and delta 18ONO3 in the benthic chamber deployed on the shelf, characterized by muddy sand, were used to calculate apparent benthic nitrate fractionation factors of 8.0 pro mille (15epsilon app) and 14.1 pro mille (18epsilon app). Measurements of delta 15NNO2 further demonstrated that the sediments acted as a source of 15N depleted NO2-. These observations were analyzed using an isotope box model that considered denitrification and nitrification of NH4+ and NO2-. The principal findings were that (i) net benthic 14N/15N fractionation (epsilon DEN) was 12.9 ± 1.7pro mille, (ii) inverse fractionation during nitrite oxidation leads to an efflux of isotopically light NO2- (-22 ± 1.9 pro mille), and (iii) direct coupling between nitrification and denitrification in the sediment is negligible. Previously reported epsilon DEN for fine-grained sediments are much lower (4-8 pro mille). We speculate that high benthic nitrate fractionation is driven by a combination of enhanced porewater-seawater exchange in permeable sediments and the hypoxic, high productivity environment. Although not without uncertainties, the results presented could have important implications for understanding the current state of the marine N cycle.

The work in this sub-project of ESOP focuses on the advective and convective transforma-tion of water masses in the Greenland Sea and its neighbouring areas. It includes observational work on the sub-mesoscale and analysis of hydrographic data up to the gyre-scale. Observations of active convective plumes were made with a towed chain equipped with up to 80 CTD sensors, giving a horizontal and vertical resolution of the hydrographic fields of a few metres. The observed scales of the penetrative convective plumes compare well with those given by theory. On the mesoscale the structure of homogeneous eddies formed as a result of deep convection was observed and the associated mixing and renewal of the intermediate layers quantified. The relative importance and efficiency of thermal and haline penetrative convection in relation to the surface boundary conditions (heat and salt fluxes and ice cover) and the ambient stratification are studied using the multi year time series of hydro-graphic data in the central Greenland Sea. The modification of the water column of the Greenland Sea gyre through advection from and mixing with water at its rim is assessed on longer time scales. The relative contributions are quantified using modern water mass analysis methods based on inverse techniques. Likewise the convective renewal and the spreading of the Arctic Intermediate Water from its formation area is quantified. The aim is to budget the heat and salt content of the water column, in particular of the low salinity surface layer, and to relate its seasonal and interannual variability to the lateral fluxes and the fluxes at the air-sea-ice interface. This will allow to estimate residence times for the different layers of the Greenland Sea gyre, a quantity important for the description of the Polar Ocean carbon cycle.

Frost flowers, intricate featherlike crystals that grow on refreezing sea ice leads, have been implicated in lower atmospheric chemical reactions. Few studies have presented chemical composition information for frost flowers over time and many of the chemical species commonly associated with Polar tropospheric reactions have never been reported for frost flowers. We undertook this study on the sea ice north of Barrow, Alaska to quantify the major ion, stable oxygen and hydrogen isotope, alkalinity, light absorbance by soluble species, organochlorine, and aldehyde composition of seawater, brine, and frost flowers. For many of these chemical species we present the first measurements from brine or frost flowers. Results show that major ion and alkalinity concentrations, stable isotope values, and major chromophore (NO3- and H2O2) concentrations are controlled by fractionation from seawater and brine. The presence of these chemical species in present and future sea ice scenarios is somewhat predictable. However, aldehydes, organochlorine compounds, light absorbing species, and mercury (part 2 of this research and Sherman et al. (2012, doi:10.1029/2011JD016186)) are deposited to frost flowers through less predictable processes that probably involve the atmosphere as a source. The present and future concentrations of these constituents in frost flowers may not be easily incorporated into future sea ice or lower atmospheric chemistry scenarios. Thinning of Arctic sea ice will likely present more open sea ice leads where young ice, brine, and frost flowers form. How these changing ice conditions will affect the interactions between ice, brine, frost flowers and the lower atmosphere is unknown.

We present in situ microelectrode measurements of sediment formation factor and porewater oxygen and pH from six stations in the North Atlantic varying in depth from 2159 to 5380 m. A numerical model of the oxygen data indicates that fluxes of oxygen to the sediments are as much as an order of magnitude higher than benthic chamber flux measurements previously reported in the same area. Model results require dissolution driven by metabolic CO2 production within the sediments to explain the pH data; even at the station with the most undersaturated bottom waters >60% of the calcite dissolution occurs in response to metabolic CO2. Aragonite dissolution alone cannot provide the observed buffering of porewater pH, even at the shallowest station. A sensitivity test of the model that accounts for uncertainties in the bottom water saturation state and the stoichiometry between oxygen consumption and CO2 production during respiration constrains the dissolution rate constant for calcite to between 3 and 30% day**-1, in agreement with earlier in situ determinations of the rate constant. Model results predict that over 35% of the calcium carbonate rain to these sediments dissolves at all stations, confirmed by sediment trap and CaCO3 accumulation data.

The Red Sea has a special place among the adjacent seas of the world. High evaporation, exclusion of its deep water from contact with the Indian Ocean proper and complete absence of continental drainage may result special conditions of the chemistry of the Red Sea. This paper aims to describe and explain the peculiarity of the hydrochemical situation. The influence of the topography, of the inflow and outflow through the straights of Bab el Mandeb, of the evaporation, of the stability of the water layers, and of the circulation will be studied. An attempt is made to estimate the apparent oxygen ultilisation in order to obtain an indication of the biological activity. A further attempt is made toward the quantitative estimation of the circulation of the nutrients and also to obtain some information about transport, dissolution, and precipitation of calcium carbonate. The basis of these investigations are mainly observations of R. V. "Meteor" during the International Indian Ocean Expedition 1964/65. The determination of dissolved oxygen, dissolved inorganic phosphate, nitrate, nitrite, ammonia, pH, alkalinity, silicate as well as salinity and temperature forms the necessary basis for such an investigation of the chemical conditions. In the first chapter the methods and some modifications for the determination of the chemical properties as applied during the I.I.O.E. cruise of R. V. "Meteor" are described. The new methods, as worked out and tested under sea going conditions during several years by the author, are described in more detail. These are the methods for nitrate, silicate, the automatic determination of dissolved inorganic phosphate and silicate, the automated determination of total phosphorus, the in situ recording of the oxygen tension, and the modification for the determination of ammonia, calcium, and dissolved oxygen. With these revised methods more than 18,000 determinations have been carried out during the Indian Ocean cruise. The complete working up of the chemical data of the Indian Ocean Expedition of R. V. "Meteor" is devided into four sections: Contributions 1) to the Chemistry of the Red Sea and the Inner Gulf of Aden, 2) to the Gulf of Aden and the Somali Coast Region, 3) to the Western Indian Coast Region, and 4) to the Persian Gulf and the Straits of Oman. This paper presents the first contribution. The special hydrographical conditions are discussed. It can be shown, that the increase of salinity in the surface waters from the south to the north of the Red Sea is only to about 30 % due to evaporation. The remaining increase is presumed to be due to the admixture of deep water to the surface layers. A special rate for the consumption of oxygen (0.114 ml/ l/a) is derived for the deep water of the Red Sea at 1500 m. Based upon the distribution of the dissolved oxygen along the axii of the Red Sea, a chematic model for the longitudinal circulation of the Red Sea is constructed. This model should be considered as a first approximation and may explain the special distribution of phosphate, nitrate, and silicate. Based upon the evaluation of the residence time of the deep water a dissolution rate for silicate is estimated as 1 mygat/a. It seems possible to calculate residence times of water masses outside the Red Sea from the silicate content. The increase of silicate and the consumption of oxygen lead to residence times of the water below the thermocine of 30 to 48 years. The distribution of oxygen in the Straits of Bab el Mandeb is described and discussed. The rate of consumption of the oxygen in the outflowing Red Sea water is estimated to 8.5 ml/ l/a. This rather high rate is explained with reference to the special conditions in the outflowing water. The Red Sea water is characterized initially by a relative high content of oxygen and a low content of nutrients. The increase in nutrients and the decrease in the oxygen content is a secondary process of the Red Sea water on its way to the Arabian Sea. Based upon the vertical distribution of the dissolved inorganic phosphate vertical exchange coefficients of 1 - 4 g/cm/sec and vertical current speeds of 10**-5 to 10**-4 cm/sec are calculated for some stations in the Red Sea. The distribution of phosphate, silicate, nitrate, nitrite and ammonia for the Red Sea and the Straits of Bab el Mandeb are discussed. The special circulation is evaluated and the balance of the nutrients is estimated by means of the brutto transport. The nutrient deficit is assumed to be balanced by sporadic inflow of intermediate water from the Gulf of Aden. An example for such an inflow has been observed and is demonstrated. The silicate-salinity relationships are a suitable way for characterizing water masses in the Red Sea. Equations for the calculation of the different components from the carbonate system, the ion activities, and the calcium carbonate saturation are evaluated. The influence of temperature and pressure is taken into account. The carbonate saturation is calculated from the determined concentrations of calcium, alkalinity, and the hydrogen ion activity. Saturation values of 320 % are found for the surface layer and of 100% ± 1 for the deep water. The extraordinary equilibrium conditions may explain the constant Ca/Cl ratio and also the sedimentation of undissolved carbonate skelecons even in greater depths. A main sedimentation rate of 2 * 10**-3cm/year is evaluated from a total sedimentation of 10 * 106 to/a of calcium carbonate in the Red Sea.
