The authors investigated various life history aspects of 19 rockfish species (Sebastes chlorostictus, S. constellatus, S. dalli, S. elongatus, S. ensifer, S. entomelas, S. flavidus, S. goodei, S. hopkinsi, S. levis, S. melanostomus, S. miniatus, S. ovalis, S. paucispinis, S. rosaceus, S. rosenblatti, S. rufus, s. saxicola, S. semicinctus) from the southern California Bight. These aspects included depth distribution, age-length relationships (of 7 species), length-weight relationships, size at first maturity, spawning season, and fecundity. Growth rates of female S. elongatus, S. hopkinsi, S. ova/is, S. saxicola, and S. semicinctus were higher than male conspecifics. Multiple spawning per season was found in 12 species. Generally, most species spawned between late winter and early summer, though there was some spawning within the genus throughout the year. Spawning season duration ranged from 2 (S. flavidus) to 10 months (S. paucispinis). Spawning seasons tended to start earlier in the year and be of longer duration in the southern California Bight, compared to published data on central California conspecifics. Males matured at a smaller length in 7 of the 17 species studied. Maximum fecundities ranged from 18,000 (S. dalll) to about 2,680,000 (S. levis). (PDF file contains 44 pages.)

Some 25 to 30 yr ago, when we as students were beginning our respective careers and were developing for the first time our awareness of marine mammals in the waters separating western North America from eastern Asia, we had visions of eventually bridging the communication gap which existed between our two countries at that time. Each of us was anxious to obtain information on the distribution, biology, and ecological relations of "our" seals and walruses on "the other side," beyond our respective political boundari~s where we were not permitted to go to study them. We were concerned that the resource management practices on the other side of the Bering and Chukchi Seas, implemented in isolation, on a purely unilateral basis, might endanger the species which we had come to know and were striving to conserve. At once apparent to both of us was the need for free exchange of biological information between our two countries and, ultimately, joint management of our shared resources. In a small way, we and others made some initial efforts to generate that exchange by personal correspondence and through vocal interchange at the annual meetings of the North Pacific Fur Seal Commission. By the enabling Agreement on Cooperation in the Field of Environmental Protection, reached between our two countries in 1972, our earlier visions at last came true. Since that time, within the framework of the Marine Mammal Project under Area V of that Agreement, we and our colleagues have forged a strong bond of professional accord and respect, in an atmosphere of free intercommunication and mutual understanding. The strength and utility of this arrangement from the beginning of our joint research are reflected in the reports contained in this, the first compendium of our work. The need for a series of such a compendia became apparent to us in 1976, and its implementation was agreed on by the regular meeting of the Project in La Jolla, Calif., in January 1977. Obviously, the preparation and publication of this first volume has been excessively delayed, in part by continuing political distrust between our governments but mainly by increasing demands placed on the time of the contributors. In this period of growing environmental concern in both countries, we and our colleagues have been totally immersed in other tasks and have experienced great difficulty in drawing together the works presented here. Much of the support for doing so was provided by the State of Alaska, through funding for Organized Research at the University of Alaska-Fairbanks. For its ultimate completion in publishable form we wish to thank Helen Stockholm, Director of Publications, Institute of Marine Science, University of Alaska, and her staff, especially Ruth Hand, and the numerous referees narned herein who gave willingly oftheir time to review each ofthe manuscripts critically and to provide a high measure of professionalism to the final product. ENGLISH: A study of the temporal and spatial distribution of larval tunas and the concomitant oceanic conditions was made in cooperation with the Direccion General de Pesca e Industrias Conexas of Mexico. Field work consisted of eight hydrographic cruises made from October 1966 through August 1967 near the entrance of the Gulf of California. From January through April, surface currents were southerly at velocities up to 20 cm/sec; currents in June were variable in direction and mostly less than 10 cm/sec; by August the surface current was northerly at 10-15 cm/sec. Surface winds were usually secondary to the distribution of mass as an influence on the surface circulation. Currents at 100 m were generally similar in direction to those at the surface, but the water moved more slowly. Between the surface and 100 m, southbound currents crossed the entrance of the Gulf at velocities of 5-10 cm/sec during January and April, forming frontal boundaries with the California Current water, which often occurred south of the entrance. From April to August, the median concentration of surface chlorophyll a increased from 0.65 to 0.97 mg/m3, while the median productivity increased from 5.6 mgC/m3/day in April to 17.8 mgC/m3/day in June before returning to 2.6 mgC/m3/day in August. Primary productivity was closely correlated with the concentration of surface chlorophyll a. Productivity was generally higher in the vicinity of the Gulf than that found for water in the open Pacific. Productivity was highest near Islas Las Tres Marias and second highest near Cabo San Lucas, both locations of local upwelling. The standing crop of phytoplankton was shown to be subjected to progressively heavier grazing pressure in the spring and summer by zooplankton. SPANISH: Un estudio de la distribución temporal y espacial de las larvas de atún y de las condiciones oceánicas concomitantes fue realizado en cooperación con la Dirección General de Pesca e Industrias Conexas de México. El trabajo experimental consistió en ocho cruceros hidrográficos realizados desde octubre 1966 hasta agosto 1967, cerca a la entrada del Golfo de California. De enero a abril, las corrientes superficiales fueron meridionales alcanzando velocidades hasta de 20 cm/seg; las corrientes en junio fueron variables en dirección y la mayoría con una velocidad de menos de 10 cm/seg; en agosto la corriente superficial fue septentrional a 10-15 cm/seg, Los vientos superficiales fueron por lo común secundarios a la dístríbucíón de la masa, como una influencia de la circulación superficial. Las corrientes a 100 m fueron generalmente similares en dirección a las de la superficie, pero el agua se movió más lentamente. Entre la superficie y los 100 m, las corrientes que se dirigen hacia el sur cruzaron la entrada del Golfo a velocidades de 5-10 cm/seg durante enero y abril formando límites frontales con el agua de la Corriente de California, que apareció a menudo al sur de la entrada. DNA charge transport (CT) involves the efficient transfer of electrons or electron holes through the DNA π-stack over long molecular distances of at least 100 base-pairs. Despite this shallow distance dependence, DNA CT is sensitive to mismatches or lesions that disrupt π-stacking and is critically dependent on proper electronic coupling of the donor and acceptor moieties into the base stack. Favorable DNA CT is very rapid, occurring on the picosecond timescale. Because of this speed, electron holes equilibrate along the DNA π-stack, forming a characteristic pattern of DNA damage at low oxidation potential guanine multiplets. Furthermore, DNA CT may be used in a biological context. DNA processing enzymes with 4Fe4S clusters can perform DNA-mediated electron transfer (ET) self-exchange reactions with other 4Fe4S cluster proteins, even if the proteins are quite dissimilar, as long as the DNA-bound [4Fe4S]3+/2+ redox potentials are conserved. This mechanism would allow low copy number DNA repair proteins to find their lesions efficiently within the cell. DNA CT may also be used biologically for the long-range, selective activation of redox-active transcription factors. Within this work, we pursue other proteins that may utilize DNA CT within the cell and further elucidate aspects of the DNA-mediated ET self-exchange reaction of 4Fe4S cluster proteins.

Dps proteins, bacterial mini-ferritins that protect DNA from oxidative stress, are implicated in the survival and virulence of pathogenic bacteria. One aspect of their protection involves ferroxidase activity, whereby ferrous iron is bound and oxidized selectively by hydrogen peroxide, thereby preventing formation of damaging hydroxyl radicals via Fenton chemistry. Understanding the specific mechanism by which Dps proteins protect the bacterial genome could inform the development of new antibiotics. We investigate whether DNA-binding E. coli Dps can utilize DNA CT to protect the genome from a distance. An intercalating ruthenium photooxidant was employed to generate oxidative DNA damage via the flash-quench technique, which localizes to a low potential guanine triplet. We find that Dps loaded with ferrous iron, in contrast to Apo-Dps and ferric iron-loaded Dps which lack available reducing equivalents, significantly attenuates the yield of oxidative DNA damage at the guanine triplet. These data demonstrate that ferrous iron-loaded Dps is selectively oxidized to fill guanine radical holes, thereby restoring the integrity of the DNA. Luminescence studies indicate no direct interaction between the ruthenium photooxidant and Dps, supporting the DNA-mediated oxidation of ferrous iron-loaded Dps. Thus DNA CT may be a mechanism by which Dps efficiently protects the genome of pathogenic bacteria from a distance.

Further work focused on spectroscopic characterization of the DNA-mediated oxidation of ferrous iron-loaded Dps. X-band EPR was used to monitor the oxidation of DNA-bound Dps after DNA photooxidation via the flash-quench technique. Upon irradiation with poly(dGdC)2, a signal arises with g = 4.3, consistent with the formation of mononuclear high-spin Fe(III) sites of low symmetry, the expected oxidation product of Dps with one iron bound at each ferroxidase site. When poly(dGdC)2 is substituted with poly(dAdT)2, the yield of Dps oxidation is decreased significantly, indicating that guanine radicals facilitate Dps oxidation. The more favorable oxidation of Dps by guanine radicals supports the feasibility of a long-distance protection mechanism via DNA CT where Dps is oxidized to fill guanine radical holes in the bacterial genome produced by reactive oxygen species.

We have also explored possible electron transfer intermediates in the DNA-mediated oxidation of ferrous iron-loaded Dps. Dps proteins contain a conserved tryptophan residue in close proximity to the ferroxidase site (W52 in E. coli Dps). In comparison to WT Dps, in EPR studies of the oxidation of ferrous iron-loaded Dps following DNA photooxidation, W52Y and W52A mutants were deficient in forming the characteristic EPR signal at g = 4.3, with a larger deficiency for W52A compared to W52Y. In addition to EPR, we also probed the role of W52 Dps in cells using a hydrogen peroxide survival assay. Bacteria containing W52Y Dps survived the hydrogen peroxide challenge more similarly to those containing WT Dps, whereas cells with W52A Dps died off as quickly as cells without Dps. Overall, these results suggest the possibility of W52 as a CT hopping intermediate.

DNA-modified electrodes have become an essential tool for the study of the redox chemistry of DNA processing enzymes with 4Fe4S clusters. In many cases, it is necessary to investigate different complex samples and substrates in parallel in order to elucidate this chemistry. Therefore, we optimized and characterized a multiplexed electrochemical platform with the 4Fe4S cluster base excision repair glycosylase Endonuclease III (EndoIII). Closely packed DNA films, where the protein has limited surface accessibility, produce EndoIII electrochemical signals sensitive to an intervening mismatch, indicating a DNA-mediated process. Multiplexed analysis allowed more robust characterization of the CT-deficient Y82A EndoIII mutant, as well as comparison of a new family of mutations altering the electrostatics surrounding the 4Fe4S cluster in an effort to shift the reduction potential of the cluster. While little change in the DNA-bound midpoint potential was found for this family of mutants, likely indicating the dominant effect of DNA-binding on establishing the protein redox potential, significant variations in the efficiency of DNA-mediated electron transfer were apparent. On the basis of the stability of these proteins, examined by circular dichroism, we proposed that the electron transfer pathway in EndoIII can be perturbed not only by the removal of aromatic residues but also through changes in solvation near the cluster.

While the 4Fe4S cluster of EndoIII is relatively insensitive to oxidation and reduction in solution, we have found that upon DNA binding, the reduction potential of the [4Fe4S]3+/2+ couple shifts negatively by approximately 200 mV, bringing this couple into a physiologically relevant range. Demonstrated using electrochemistry experiments in the presence and absence of DNA, these studies do not provide direct molecular evidence for the species being observed. Sulfur K-edge X-ray absorbance spectroscopy (XAS) can be used to probe directly the covalency of iron-sulfur clusters, which is correlated to their reduction potential. We have shown that the Fe-S covalency of the 4Fe4S cluster of EndoIII increases upon DNA binding, stabilizing the oxidized [4Fe4S]3+ cluster, consistent with a negative shift in reduction potential. The 7% increase in Fe-S covalency corresponds to an approximately 150 mV shift, remarkably similar to DNA electrochemistry results. Therefore we have obtained direct molecular evidence for the shift in 4Fe4S reduction potential of EndoIII upon DNA binding, supporting the feasibility of our model whereby these proteins can utilize DNA CT to cooperate in order to efficiently find DNA lesions inside cells.

In conclusion, in this work we have explored the biological applications of DNA CT. We discovered that the DNA-binding bacterial ferritin Dps can protect the bacterial genome from a distance via DNA CT, perhaps contributing to pathogen survival and virulence. Furthermore, we optimized a multiplexed electrochemical platform for the study of the redox chemistry of DNA-bound 4Fe4S cluster proteins. Finally, we have used sulfur K-edge XAS to obtain direct molecular evidence for the negative shift in 4Fe4S cluster reduction potential of EndoIII upon DNA binding. These studies contribute to the understanding of DNA-mediated protein oxidation within cells.

Este trabajo de fin de grado, se ha realizado sobre la "C aracterizac ión espectroscópica de nuevos BODIPYs ", u n tema actual y de gran int erés. Es un trabajo de tipo experimental, realizado en el laboratorio de Espectroscopia Molecular del departamento de Química - Física de la Facultad de Ciencia y Tecnología de la UPV - EHU , dirigido por T. Arbeloa. Se ha llevado a cabo una pequeña investigación desarrollando una serie de experimentos en el laboratorio que han sido acompañados de un estudio bibliográfico sobre la situación del tema. Se han analizado las características fotofísicas de varios colorantes,sintetizados por un grupo de investigación de síntesis orgánica, y posteriormente se ha hecho un estudio sobre los resultados más relevantes obtenidos.

O envelhecimento populacional é um fenômeno mundial, assim como o predomínio de mortes por doenças cardiovasculares. Estudos demonstram que o envelhecimento acarreta aumento da rigidez vascular e perturbações na reatividade macrovascular. O presente estudo comparou a microcirculação deste processo com adultos jovens através da pletismografia de oclusão venosa (POV) e da videocapilaroscopia do leito periungueal (VC). Para isto, desenvolveu-se um estudo transversal com dois grupos: idade entre 18 e 30 anos (n=16) e outro grupo com idade igual ou superior a 60 anos (n=21), além da subdivisão deste grupo em três subgrupos: idosas saudáveis (n=8), idosas em tratamento de hipertensão arterial (IDHAS,n=6) e idosas em tratamento de dislipidemia (IDDIS,n=6). Foram realizadas avaliações clínica, antropométrica, bioquímica e microcirculatória. Como resultados, a VC mostrou aumento dos diâmetros capilares aferente, apical e eferente e redução da relação velocidade máxima/ velocidade basal de deslocamento de hemácias para grupo de idosas e todos os subgrupos. A POV revelou diminuição da vasodilatação endotélio-dependente em todos os grupos estudados e a vasodilatação endotélio-independente alterou-se apenas em IDHAS e IDDIS. Como conclusões houve, no envelhecimento, alterações estruturais e funcionais da microcirculação assim como redução da vasodilatação endotélio-dependente. A vasodilatação endotélio-independente manteve-se inalterada, indicando que a célula muscular lisa da parede vascular permanece preservada, porém sofre alterações nos grupos IDHAS e IDDIS.

[EN] This paper reports an innovative technique for reagents storage in microfluidic devices by means of a one-step UV-photoprintable ionogel-based microarray on non-modified polymeric substrates. Although the ionogel and the ink-jet printing technology are well published, this is the first study where both are used for long-term reagent storage in lab-on-a-chip devices. This technology for reagent storage is perfectly compatible with mass production fabrication processes since pre-treatment of the device substrate is not necessary and inkjet printing allows for an efficient reagent deposition process. The functionality of this microarray is demonstrated by testing the release of biotin-647 after being stored for 1 month at room temperature. Analysis of the fluorescence of the ionogel-based microarray that contains biotin-647 demonstrated that 90% of the biotin-647 present was released from the ionogel-based microarray after pumping PBS 0.1% Tween at 37 °C. Moreover, the activity of biotin-647 after being released from the ionogel-based microarray was investigated trough the binding capability of this biotin to a microcontact printed chip surface with avidin. These findings pave the way for a novel, one-step, cheap and mass production on-chip reagents storage method applicable to other reagents such as antibodies and proteins and enzymes.

O objetivo desta tese é analisar experiências de adoecimento e seus desdobramentos na vida cotidiana, a partir do relato de mulheres que tiveram câncer de mama e realizaram tratamento médico. A maior parte da pesquisa de observação participante foi realizada numa associação de apoio durante os anos de 2005/2006. Neste período acompanhei as atividades propostas por um grupo de profissionais de saúde voluntários interessados em resgatar a auto-estima destas mulheres. Analiso o diálogo estabelecido entre tais profissionais e as freqüentadoras da associação, em sua quase totalidade constituída por mulheres de classes populares, a fim de perceber valores que norteiam comportamentos e discursos sobre a doença. Além do registro de uma relação hierarquizada entre profissionais e leigos, analiso o diálogo (mais igualitário) das mulheres entre si e suas reflexões a respeito de suas vidas, formulando concepções etiológicas acerca da doença que as atingiu, assim como sobre o momento em que suas vidas se encontravam quando da descoberta do câncer. Uma vez finalizado o percurso terapêutico, de volta para casa, as estratégias adotadas consistem em retomar sua vida como era ?antes?. As formas cotidianas da gestão das marcas do câncer no corpo são analisadas do ponto de vista de minhas informantes e tratam de temas relativos à conjugalidade e ao trabalho doméstico. Enfim, trato do sofrimento instituinte de certo tipo de feminilidade.

O objetivo desse estudo foi investigar a origem e a função da fantasia na obra de Sigmund Freud, por meio do papel que desempenha na constituição do psiquismo humano e da sua importância em produzir um sentido para a vida. A pesquisa - de cunho bibliográfico - buscou esclarecer os diferentes usos e modificações do termo "fantasia" no decorrer das descobertas realizadas por Freud ao longo de sua vida e obra, desde o período denominado de pré-psicanalítico (1886/1899) até os últimos textos publicados no Esboço de Psicanálise (1940 [1938]). A análise da investigação revelou três grandes eixos descritivos, complexos, por vezes contraditórios e submetidos a constantes redefinições que caracterizam a fantasia como um sintoma, uma criação e um enigma da transmissão geracional. A conclusão corrobora o pressuposto de que a vida não tem sentido sem a capacidade de fantasiar. A fantasia ocupa um lugar de fundamental importância no pensamento freudiano, comparecendo como um elemento essencial na constituição do psiquismo e revelando horizontes, para que os estudiosos da psicanálise pudessem ampliar essa investigação tão reveladora da psiquê.
