Second-rank tensor interactions, such as quadrupolar interactions between the spin- 1 deuterium nuclei and the electric field gradients created by chemical bonds, are affected by rapid random molecular motions that modulate the orientation of the molecule with respect to the external magnetic field. In biological and model membrane systems, where a distribution of dynamically averaged anisotropies (quadrupolar splittings, chemical shift anisotropies, etc.) is present and where, in addition, various parts of the sample may undergo a partial magnetic alignment, the numerical analysis of the resulting Nuclear Magnetic Resonance (NMR) spectra is a mathematically ill-posed problem. However, numerical methods (de-Pakeing, Tikhonov regularization) exist that allow for a simultaneous determination of both the anisotropy and orientational distributions. An additional complication arises when relaxation is taken into account. This work presents a method of obtaining the orientation dependence of the relaxation rates that can be used for the analysis of the molecular motions on a broad range of time scales. An arbitrary set of exponential decay rates is described by a three-term truncated Legendre polynomial expansion in the orientation dependence, as appropriate for a second-rank tensor interaction, and a linear approximation to the individual decay rates is made. Thus a severe numerical instability caused by the presence of noise in the experimental data is avoided. The purpose of this project was to discern the inherent tension present in narratives told by adolescents with a visual impairment as they attempted to make sense of their experiences, specifically those surrounding risk. Mediated action, based on the foundational work of Vygotsky and Bakhtin, was used as both a theoretical and methodological approach; it is the theory that there are two components that constitute any human action: the "agent," or the person who is doing the acting, and the "mediational means" that he or she is using to accomplish the action in question. Tension ensues as neither is able to fully explain human behaviour. Ten adolescents with a visual impairment participated in a narrative interview, revealing numerous counter-narratives surrounding risk-taking, including "experimentation undertaken using good judgment." Participants offered examples of how they engaged, appropriated, resisted and transformed the dominant narratives of disability and adolescence in their identity formation.

During infection, the model plant Arabidopsis thaliana is capable of activating long lasting defence responses both in tissue directly affected by the pathogen and in more distal tissue. Systemic acquired resistance (SAR) is a type of systemic defence response deployed against biotrophic pathogens resulting in altered plant gene expression and production of antimicrobial compounds. One such gene involved in plant defence is called pathogenesis-related 1 (PR1) and is under the control of several protein regulators. TGA II-clade transcription factors (namely TGA2) repress PR1 activity prior to infection by forming large oligomeric complexes effectively blocking gene transcription. After pathogen detection, these complexes are dispersed by a mechanism unknown until now and free TGA molecules interact with the non-expressor of pathogenesis-related gene 1 (NPR1) protein forming an activating complex enabling PR1 transcription. This study elucidates the TGA2 dissociation mechanism by introducing protein kinase CK2 into this process. This enzyme efficiently phosphorylates TGA2 resulting in two crucial events. Firstly, the DNA-binding ability of this transcription factor is completely abolished explaining how the large TGA2 complexes are quickly evicted from the PR1 promoter. Secondly, a portion of TGA2 molecules dissociate from the complexes after phosphorylation which likely makes them available for the formation of the TGA2-NPR1 activating complex. We also show that phosphorylation of a multiserine motif found within TGA2’s N terminus is responsible for the change of affinity to DNA, while modification of a single threonine in the leucine zipper domain seems to be responsible for deoligomerization. Despite the substantial changes caused by phosphorylation, TGA2 is still capable of interacting with NPR1 and these proteins together form a complex on DNA promoting PR1 transcription. Therefore, we propose a change in the current model of how PR1 is regulated by adding CK2 which targets TGA2 displacing it’s complexes from the promoter and providing solitary TGA2 molecules for assembly of the activating complex. Amino acid sequences of regions targeted by CK2 in Arabidopsis TGA2 are similar to those found in TGA2 homologs in rice and tobacco. Therefore, the molecular mechanism that we have identified may be conserved among various plants, including important crop species, adding to the significance of our findings.

Immobilized lipase B from Candida antarctica (N435) was investigated as a potential biocatalyst to generate silicone-based chiral polymers from monomers derived from the enzymatic dihydroxylation of bromobenzene. Several conditions and parameters have been investigated for this purpose and lipase transesterification preference to each of the free secondary alcohols in the chiral monomers was documented. The N435 was challenged with a series of substrates where the free alcohol moieties were systematically protected in order to study the substrate preference(s) for the transesterification reactions.

The primary goal was to test a mediated-moderation model in which dispositional optimism was the moderator and its role was mediated by problem-focused coping. A secondary goal was to demonstrate that posttraumatic growth could be differentiated from maturation and normal development. Two groups of participants were recruited and completed questionnaires twice with a 60-day interval: One group (Trauma), described a traumatic experience and the second group (Non-trauma), described a significant experience. Contrary to the hypothesis, only problem-focused coping and deliberate rumination predicted posttraumatic growth, and these findings were only observed in concurrent analyses. Furthermore, the results indicated that there was no significant difference between groups on growth scores at either Time 1 or Time 2. The findings suggest that the term “posttraumatic growth” may refer to the context in which growth occurs rather than to some developmental process that uniquely follows trauma.

Digital Terrain Models (DTMs) are important in geology and geomorphology, since elevation data contains a lot of information pertaining to geomorphological processes that influence the topography. The first derivative of topography is attitude; the second is curvature. GIS tools were developed for derivation of strike, dip, curvature and curvature orientation from Digital Elevation Models (DEMs). A method for displaying both strike and dip simultaneously as colour-coded visualization (AVA) was implemented. A plug-in for calculating strike and dip via Least Squares Regression was created first using VB.NET. Further research produced a more computationally efficient solution, convolution filtering, which was implemented as Python scripts. These scripts were also used for calculation of curvature and curvature orientation. The application of these tools was demonstrated by performing morphometric studies on datasets from Earth and Mars. The tools show promise, however more work is needed to explore their full potential and possible uses.

BACKGROUND: HIV-1 Vpu targets newly synthesized CD4 receptor for rapid degradation by a process reminiscent of endoplasmic reticulum (ER)-associated protein degradation (ERAD). Vpu is thought to act as an adaptor protein, connecting CD4 to the ubiquitin (Ub)-proteasome degradative system through an interaction with beta-TrCP, a component of the SCFbeta-TrCP E3 Ub ligase complex. RESULTS: Here, we provide direct evidence indicating that Vpu promotes trans-ubiquitination of CD4 through recruitment of SCFbeta-TrCP in human cells. To examine whether Ub conjugation occurs on the cytosolic tail of CD4, we substituted all four Ub acceptor lysine residues for arginines. Replacement of cytosolic lysine residues reduced but did not prevent Vpu-mediated CD4 degradation and ubiquitination, suggesting that Vpu-mediated CD4 degradation is not entirely dependent on the ubiquitination of cytosolic lysines and as such might also involve ubiquitination of other sites. Cell fractionation studies revealed that Vpu enhanced the levels of ubiquitinated forms of CD4 detected in association with not only the ER membrane but also the cytosol. Interestingly, significant amounts of membrane-associated ubiquitinated CD4 appeared to be fully dislocated since they could be recovered following sodium carbonate salt treatment. Finally, expression of a transdominant negative mutant of the AAA ATPase Cdc48/p97 involved in the extraction of ERAD substrates from the ER membrane inhibited Vpu-mediated CD4 degradation. CONCLUSION: Taken together, these results are consistent with a model whereby HIV-1 Vpu targets CD4 for degradation by an ERAD-like process involving most likely poly-ubiquitination of the CD4 cytosolic tail by SCFbeta-TrCP prior to dislocation of receptor molecules across the ER membrane by a process that depends on the AAA ATPase Cdc48/p97.

Cette recherche porte sur la lexicologie, la lexicographie et l’enseignement/apprentissage du lexique. Elle s’inscrit dans le cadre du projet Modélisation ontologique des savoirs lexicographiques en vue de leur application en linguistique appliquée, surnommé Lexitation, qui est, à notre connaissance, la première tentative d’extraction des savoirs lexicographiques — i.e. connaissances déclaratives et procédurales utilisées par des lexicographes — utilisant une méthode expérimentale. Le projet repose sur le constat que les savoirs lexicographiques ont un rôle crucial à jouer en lexicologie, mais aussi en enseignement/apprentissage du lexique. Dans ce mémoire, nous décrirons les méthodes et les résultats de nos premières expérimentations, effectuées à l’aide du Think Aloud Protocol (Ericsson et Simon, 1993). Nous expliquerons l’organisation générale des expérimentations et comment les savoirs lexicographiques extraits sont modélisés pour former une ontologie. Finalement, nous discuterons des applications possibles de nos travaux en enseignement du lexique, plus particulièrement pour la formation des maîtres.

L’hexokinase (HK) est la première enzyme du métabolisme des hexoses et catalyse la réaction qui permet aux hexoses d’entrer dans le pool des hexoses phosphates et donc par le fait même la glycolyse. Bien que le glucose soit son principal substrat, cette enzyme peut aussi phosphoryler le mannose et le fructose. Malgré son importance dans le métabolisme primaire, l’HK n’a jamais été purifiée à homogénéité sous forme native. Le but de ce travail était donc de purifier une isoforme d’HK à partir de tubercule de Solanum tuberosum et par la suite de caractériser ses propriétés cinétiques. Bien avant que je commence mon travail, un groupe de recherche avait déjà séparé et partiellement purifié trois isoformes d’HK de S. tuberosum. Un protocole d’extraction était donc disponible, mais l’HK ainsi extraite était peu stable d’où le besoin d’y apporter certaines modifications. En y ajoutant certains inhibiteurs de protéases ainsi qu’en modifiant les concentrations de certains éléments, le tampon d’extraction ainsi modifié a permis d’obtenir un extrait dont l’activité HK était stable pendant au moins 72h après l’extraction, en empêchant la dégradation. À l’aide du tampon d’extraction optimisé et d’une chromatographie sur colonne de butyl sépharose, il a été possible de séparer 4 isoformes d’HKs. Par la suite, une isoforme d’HK (HK1) a été purifiée à l’homogénéité à l’aide de 5 étapes de chromatographie supplémentaires. En plus de caractériser les propriétés cinétiques de cette enzyme, l’analyse de séquençage par MS/MS a permis de l’associer au produit du gène StHK1 de Solanum tuberosum. Avec une activité spécifique de 10.2 U/mg de protéine, il s’agit de l’HK purifiée avec l’activité spécifique la plus élevée jamais rapportée d’un tissu végétal.L’ensemble des informations recueillies lors de la purification de HK1 a ensuite été utilisée pour commencer la purification d’une deuxième isoforme (HK3). Ce travail a permis de donner des lignes directrices pour la purification de cette isoforme et certains résultats préliminaires sur sa caractérisation enzymatique.

HIV upregulates cell-surface expression of specific ligands for the activating NKG2D receptor, including ULBP-1, -2, -3, but not MICA or MICB, in infected cells both in vitro and in vivo. However, the viral factor(s) involved in NKG2D ligand expression still remains undefined. HIV-1 Vpr activates the DNA damage/stress-sensing ATR kinase and promotes G2 cell-cycle arrest, conditions known to upregulate NKG2D ligands. We report here that HIV-1 selectively induces cell-surface expression of ULBP-2 in primary CD4+ T-lymphocytes by a process that is Vpr-dependent. Importantly, Vpr enhanced the susceptibility of HIV-1-infected cells to NK cell-mediated killing. Strikingly, Vpr alone was sufficient to upregulate expression of all NKG2D ligands and thus promoted efficient NKG2D-dependent NK cell-mediated killing. Delivery of virion-associated Vpr via defective HIV-1 particles induced analogous biological effects in non-infected target cells, suggesting that Vpr may act similarly beyond infected cells. All these activities relied on Vpr ability to activate the ATR-mediated DNA damage/stress checkpoint. Overall, these results indicate that Vpr is a key determinant responsible for HIV-1-induced upregulation of NKG2D ligands and further suggest an immunomodulatory role for Vpr that may not only contribute to HIV-1-induced CD4+ T-lymphocyte depletion but may also take part in HIV-1-induced NK cell dysfunction.

Le facteur d'initiation de la traduction chez les eukaryotes eIF4E (4E) est un puissant oncogène en raison de sa capacité à faciliter l'export et/ou la traduction de certains transcripts, dont beaucoup sont eux-mêmes des oncogènes. 4E intéragit avec un grand nombre de protéines régulatrices dont la protéine 4E-T (pour 4E-Transporter). La capacité de 4E-T à modifier la localisation subcellulaire de 4E pourrait offrir un mécanisme permettant de modifier le potentiel oncogène d'une cellule. La surexpression de 4E-T dans des cellules d'ostéosarcome conduit à l’augmentation du nombre et de la taille des P-bodies, dans lesquels 4E colocalisent avec 4E-T mais pas avec la version tronquée 4E-T/Y30A. Cependant, les différentes expériences menées, permettant d’analyser les taux de transcription, la quantité de protéine, les profiles polysomiques ainsi que la distribution nucléo-cytoplasmique, montrent que la surexpression de 4E-T n'a pas d'effet sur la fonction de 4E. L'observation d’un enrichissement cytoplasmique et d’une charge réduite de ribosomes sur les transcripts codant les protéines cycline D1 et ODC (profile polysomique) dans la lignée 4E-T suggère un role de 4E-T dans la séquestration cytoplasmique de certains transcrips par un mécanisme qui reste encore à déterminer.

Nous rapportons les résultats d'une étude des désintégrations semileptoniques non-charmées B^+--> êta^(') l^+v et B^0--> pi^- l^+v, mesurés par le détecteur BABAR avec une production d'environ 464 millions de paires de mésons BBbar issues des collisions e^+e^- à la résonance Upsilon(4S). L'analyse reconstruit les événements avec une technique relâchée des neutrinos. Nous obtenons les rapports d'embranchement partiels pour les désintégrations B^+--> êta l^+v et B^0--> pi^- l^+v en trois et douze intervalles de q^2, respectivement, à partir desquels nous extrayons les facteurs de forme f_+(q^2) et les rapports d'embranchement totaux B(B^+--> êta l^+v) = (3.39 +/- 0.46_stat +/- 0.47_syst) x 10^-5 et B(B^0--> pi^- l^+v) = (1.42 +/- 0.05_stat +/- 0.08_syst) x 10^-4. Nous mesurons aussi B(B^+--> êta' l^+v) = (2.43 +/- 0.80_stat +/- 0.34_syst) x 10^-5. Nous obtenons les valeurs de la norme de l'élément |V_ub| de la matrice CKM en utilisant trois calculs différents de la CDQ.
