O paraquat, herbicida amplamente utilizado na agricultura, é um produto tóxico, pois pode causar intoxicações fatais, principalmente pela falta de antídoto eficaz para reversão do quadro clínico. Os efeitos toxicológicos são decorrentes da indução ao stress oxidativo. O órgão alvo principal é o pulmão, que pode apresentar edema, hemorragia, inflamação intersticial e fibrose, culminando com falência respiratória grave e morte. Além disso, é nefrotóxico, hepatotóxico, miotóxico e neurotóxico. O tratamento da intoxicação além de visar a diminuição da absorção e estimular a excreção do paraquat absorvido, atualmente é baseado em medidas que diminuam o stress oxidativo utilizando substâncias antioxidantes e que, consequentemente, revertam o quadro toxicológico instalado, especialmente o pulmonar. Como métodos de diagnóstico, entre as metodologias quantitativas disponíveis, os métodos cromatográficos são os mais relatados para materiais biológicos. Porém, a electroforese capilar e os imunoensaios podem ser utilizados. Por outro lado, uma reação simples e rápida de caracterização urinária com ditionito de sódio é muito utilizada, pois é preditiva na suspeita de intoxicações agudas. Em conclusão, perante o alto potencial de morbimortalidade nas intoxicações por paraquat, a reversão dos danos pulmonares com uso de antioxidantes tem sido muito estudada, porém não há o estabelecimento de um antídoto específico. No diagnóstico laboratorial, métodos cromatográficos, eletroforéticos e imunológicos são usados para quantificá-lo, contudo a reação urinária com ditionito ainda é valiosa na rotina da toxicologia clínica.

A presente dissertação tem como objectivo reflectir sobre as condições funcionais da Praça da República, nas Caldas da Rainha, no domínio da sua regeneração e das potencialidades económicas e sociais, no sentido de apresentar uma solução, entre outras possíveis, como contributo para futuras intervenções. Ao estudo da Praça da República das Caldas da Rainha associou-se o caso da Praça do Mercado de Cambridge, na Inglaterra, fundamental como termo de comparação no sentido de tentar regenerar a Praça da República. A dissertação desenvolve-se em três partes. A primeira parte centra-se numa apresentação generalista e exemplificativa das praças, com destaque para as suas formas e funções. Na segunda parte, procedemos a uma observação pormenorizada do caso de Cambridge, em relação ao seu funcionamento, tipo de vendas e intervenções. Por último, na terceira parte, para além de procedermos a um exame pormenorizado da Praça da República, em relação aos seus problemas, à sua importância e ao estado de conservação dos edifícios envolventes, finalizamos com uma proposta de intervenção. O estudo desenvolvido neste trabalho permitiu-nos concluir que esta praça necessita, urgentemente, de uma intervenção que corrija a situação de decadência em que se encontra e dê solução a problemas de acessibilidade, limpeza, estacionamento, para além de necessitar da construção de um edifício de apoio que torne este espaço acolhedor e agradável e que não seja apenas local fortuito de passagem.

Quando um microrganismo patogénico invade a glândula mamária, através do canal do teto, pode ocorrer infecção intramamária, desencadeando uma resposta inflamatória – a mastite. Esta possui diversas etiologias, sendo normalmente de origem bacteriana. Na resposta inflamatória, actuam as células somáticas presentes no leite. O presente estudo teve como principal objectivo avaliar a relação entre as contagens de células somáticas e os agentes causadores de mastite em amostras de leite. Foram utilizados dados referentes aos resultados de análises de leite para diagnóstico de mastite realizadas em amostras de explorações de Entre Douro e Vouga. Os resultados consistem em identificação dos agentes e respectivo antibiograma, CCS, sendo ainda registado o objectivo da análise (Secagem ou Tratamento). Determinaram-se as prevalências dos agentes, avaliando de seguida a relação entre as seguintes variáveis: agentes e CCS; CCS e objectivo da análise; e entre agentes e objectivo da análise. Concluiu-se que os Staphylococcus coagulase-negativa são os mais prevalentes, 14,5%, seguindo, Streptococcus uberis, 7,6%; Escherichia coli, 6,3%; Lactococcus garviae, 5,8% e Staphylococcus aureus, 4,1%. Concluiu-se ainda, a existência de diferenças estatisticamente significativas nas análises realizadas, confirmando a relação entre as demais variáveis.

The ECMWF full-physics and dry singular vector (SV) packages, using a dry energy norm and a 1-day optimization time, are applied to four high impact European cyclones of recent years that were almost universally badly forecast in the short range. It is shown that these full-physics SVs are much more relevant to severe cyclonic development than those based on dry dynamics plus boundary layer alone. The crucial extra ingredient is the representation of large-scale latent heat release. The severe winter storms all have a long, nearly straight region of high baroclinicity stretching across the Atlantic towards Europe, with a tongue of very high moisture content on its equatorward flank. In each case some of the final-time top SV structures pick out the region of the actual storm. The initial structures were generally located in the mid- to low troposphere. Forecasts based on initial conditions perturbed by moist SVs with opposite signs and various amplitudes show the range of possible 1-day outcomes for reasonable magnitudes of forecast error. In each case one of the perturbation structures gave a forecast very much closer to the actual storm than the control forecast. Deductions are made about the predictability of high-impact extratropical cyclone events. Implications are drawn for the short-range forecast problem and suggestions made for one practicable way to approach short-range ensemble forecasting. Copyright © 2005 Royal Meteorological Society.

An investigation is made of the impact of a full linearized physical (moist) parameterization package on extratropical singular vectors (SVs) using the ECMWF integrated forecasting system (IFS). Comparison is made for one particular period with a dry physical package including only vertical diffusion and surface drag. The crucial extra ingredient in the full package is found to be the large-scale latent heat release. Consistent with basic theory, its inclusion results in a shift to smaller horizontal scales and enhanced growth for the SVs. Whereas, for the dry SVs, T42 resolution is sufficient, the moist SVs require T63 to resolve their structure and growth. A 24-h optimization time appears to be appropriate for the moist SVs because of the larger growth of moist SVs compared with dry SVs. Like dry SVs, moist SVs tend to occur in regions of high baroclinicity, but their location is also influenced by the availability of moisture. The most rapidly growing SVs appear to enhance or reduce large-scale rain in regions ahead of major cold fronts. The enhancement occurs in and ahead of a cyclonic perturbation and the reduction in and ahead of an anticyclonic perturbation. Most of the moist SVs for this situation are slightly modified versions of the dry SVs. However, some occur in new locations and have particularly confined structures. The most rapidly growing SV is shown to exhibit quite linear behavior in the nonlinear model as it grows from 0.5 to 12 hPa in 1 day. For 5 times this amplitude the structure is similar but the growth is about half as the perturbation damps a potential vorticity (PV) trough or produces a cutoff, depending on its sign.

Partindo da premissa de que o jornal é um espaço privilegiado para a oferta de sentidos sobre o mundo, esta pesquisa tem como proposta investigar o modo como o jornal O Globo, construiu e fez circular os sentidos sobre as drogas, principalmente o crack, em 2013. Do ponto de vista da saúde pública, neste ano, travou-se o debate em torno da votação do PL 7663/2010 que, entre outros aspectos, propõe mudanças na legislação referente às alternativas de tratamento da dependência química. Nosso corpus de análise é formado pelos textos publicados n\2019O Globo, no período de janeiro a dezembro de 2013, que abordaram o eixo crack, internação compulsória e Projeto de Lei 7663/2010. Como referências teórico-metodológicas utilizamos os postulados da Semiologia dos Discursos Sociais, trabalhando ainda conceitos propostos por teóricos como Pierre Bourdieu, Mikhail Bakhtin, Eliseo Verón, Muniz Sodré e Michel Foucault a partir de reflexões sobre campo social, discurso, poder e midiatização. Tomando por base as construções enunciativas analisadas, destacamos que apesar de mostrar-se contrário a alguns aspectos da internação, o jornal O Globo defendia a mesma como alternativa para conter a epidemia de crack

In the present work, a group contribution method is proposed for the estimation of viscosity of fatty compounds and biodiesel esters as a function of the temperature. The databank used for regression of the group contribution parameters (1070 values for 65 types of substances) included fatty compounds, such as fatty acids, methyl and ethyl esters and alcohols, tri- and diacylglycerols, and glycerol. The inclusion of new experimental data for fatty esters, a partial acylglycerol, and glycerol allowed for a further refinement in the performance of this methodology in comparison to a prior group contribution equation (Ceriani, R.; Goncalves, C. B.; Rabelo, J.; Caruso, M.; Cunha, A. C. C.; Cavaleri, F. W.; Batista, E. A. C.; Meirelles, A. J. A. Group contribution model for predicting viscosity of fatty compounds. J. Chem. Eng. Data 2007, 52, 965-972) for all classes of fatty compounds. Besides, the influence of small concentrations of partial acylglycerols, intermediate compounds in the transesterification reaction, in the viscosity of biodiesels was also investigated.

The current trend toward minimal-invasive dentistry has introduced innovative techniques for cavity preparation. Chemical vapor deposition (CVD) and laser-irradiation technology have been employed as an alternative to the common use of regular burs in high-speed turbines. Objectives. The purpose of this study was to assess the influence of alternative techniques for cavity preparation on the bonding effectiveness of different adhesives to dentin, and to evaluate the morphological characteristics of dentin prepared with those techniques. Methods. One etch&rinse adhesive (OptiBond FL, Kerr) and three self-etch systems (Adper Prompt L-Pop, 3M ESPE; Clearfil SE Bond, Kuraray; Clearfil S3 Bond, Kuraray) were applied on dentin prepared with a regular bur in a turbine, with a CVD bur in a turbine, with a CVD tip in ultrasound and with an ErCr:YSGG laser. The micro-tensile bond strength (mu TBS) was determined after storage in water for 24 h at 37 degrees C, and morphological evaluation was performed by means of field -emission -gun scanning electron microscopy (Feg-SEM). Results. Feg-SEM evaluation revealed different morphological features on the dentin surface after the usage of both the conventional and alternative techniques for cavity preparation, more specifically regarding smear-layer thickness and surface roughness. CVD bur-cut, CVD ultra-sonoabraded and laser-irradiated dentin resulted in lower mu TBSs than conventionally bur-cut dentin, irrespective of the adhesive employed. Significance. The techniques, such as CVD diamond-bur cutting, CVD diamond ultra-sonoabrasion and laser-irradiation, used for cavity preparation may affect the bonding effectiveness of adhesives to dentin, irrespective of their acidity or approach. Purpose: In light of the concept of minimally invasive dentistry, erbium lasers have been considered as an alternative technique to the use of diamond burs for cavity preparation. The purpose of this study was to assess the bonding effectiveness of adhesives to Er,Cr:YSGG laser-irradiated dentin using irradiation settings specific for cavity preparation. Materials and Methods: Fifty-four midcoronal dentin surfaces, obtained from sound human molars, were irradiated with an Er,Cr:YSGG laser or prepared with a diamond bur using a high-speed turbine. One etch-and-rinse (Optibond FL/Kerr) and three self-etching adhesives (Adper Prompt L-Pop/3M ESPE, Clearfil SE Bond/Kuraray, and Clearfil S-3 Bond/Kuraray) were used to bond the composite to dentin. The microtensile bond strength (mu TBS) was determined after 24 h of storage in water at 37 degrees C. The Kruskal-Wallis test was used to determine pairwise statistical differences (p < 0.05). Prepared dentin surfaces, adhesive interfaces, and failure patterns were analyzed using a stereo microscope and Field-emission gun Scanning Electron Microscopy (Feg-SEM). Results: Significantly lower mu TBS was observed to laser-irradiated than to bur-cut dentin (p < 0.05), irrespective of the adhesive employed. Feg-SEM photomicrographs of lased dentin revealed an imbricate patterned substrate and the presence of microcracks at the dentin surface. Conclusion: Morphological alterations produced by Er,Cr:YSGG laser-irradiation adversely influence the bonding effectiveness of adhesives to dentin. We present a catalogue of galaxy photometric redshifts and k-corrections for the Sloan Digital Sky Survey Data Release 7 (SDSS-DR7), available on the World Wide Web. The photometric redshifts were estimated with an artificial neural network using five ugriz bands, concentration indices and Petrosian radii in the g and r bands. We have explored our redshift estimates with different training sets, thus concluding that the best choice for improving redshift accuracy comprises the main galaxy sample (MGS), the luminous red galaxies and the galaxies of active galactic nuclei covering the redshift range 0 < z < 0.3. For the MGS, the photometric redshift estimates agree with the spectroscopic values within rms = 0.0227. The distribution of photometric redshifts derived in the range 0 < z(phot) < 0.6 agrees well with the model predictions. k-corrections were derived by calibration of the k-correct_v4.2 code results for the MGS with the reference-frame (z = 0.1) (g - r) colours. We adopt a linear dependence of k-corrections on redshift and (g - r) colours that provide suitable distributions of luminosity and colours for galaxies up to redshift z(phot) = 0.6 comparable to the results in the literature. Thus, our k-correction estimate procedure is a powerful, low computational time algorithm capable of reproducing suitable results that can be used for testing galaxy properties at intermediate redshifts using the large SDSS data base.

Data from 58 strong-lensing events surveyed by the Sloan Lens ACS Survey are used to estimate the projected galaxy mass inside their Einstein radii by two independent methods: stellar dynamics and strong gravitational lensing. We perform a joint analysis of these two estimates inside models with up to three degrees of freedom with respect to the lens density profile, stellar velocity anisotropy, and line-of-sight (LOS) external convergence, which incorporates the effect of the large-scale structure on strong lensing. A Bayesian analysis is employed to estimate the model parameters, evaluate their significance, and compare models. We find that the data favor Jaffe`s light profile over Hernquist`s, but that any particular choice between these two does not change the qualitative conclusions with respect to the features of the system that we investigate. The density profile is compatible with an isothermal, being sightly steeper and having an uncertainty in the logarithmic slope of the order of 5% in models that take into account a prior ignorance on anisotropy and external convergence. We identify a considerable degeneracy between the density profile slope and the anisotropy parameter, which largely increases the uncertainties in the estimates of these parameters, but we encounter no evidence in favor of an anisotropic velocity distribution on average for the whole sample. An LOS external convergence following a prior probability distribution given by cosmology has a small effect on the estimation of the lens density profile, but can increase the dispersion of its value by nearly 40%.

Hepatitis C virus (HCV) infection frequently persists despite substantial virus-specific immune responses and the combination of pegylated interferon (INF)-alpha and ribavirin therapy. Major histocompatibility complex class I restricted CD8+ T cells are responsible for the control of viraemia in HCV infection, and several studies suggest protection against viral infection associated with specific HLAs. The reason for low rates of sustained viral response (SVR) in HCV patients remains unknown. Escape mutations in response to cytotoxic T lymphocyte are widely described; however, its influence in the treatment outcome is ill understood. Here, we investigate the differences in CD8 epitopes frequencies from the Los Alamos database between groups of patients that showed distinct response to pegylated alpha-INF with ribavirin therapy and test evidence of natural selection on the virus in those who failed treatment, using five maximum likelihood evolutionary models from PAML package. The group of sustained virological responders showed three epitopes with frequencies higher than Non-responders group, all had statistical support, and we observed evidence of selection pressure in the last group. No escape mutation was observed. Interestingly, the epitope VLSDFKTWL was 100% conserved in SVR group. These results suggest that the response to treatment can be explained by the increase in immune pressure, induced by interferon therapy, and the presence of those epitopes may represent an important factor in determining the outcome of therapy.

Apocynaceae sensun strictum from the Mucuge Municipal Park, Bahia, Brazil, including the valid publication of two names in Mandevilla Lindl. The flora of Rauvolfioideae and Apocynoideae, which together form Apocynaceae s.s., from the Mucuge Municipal Park, Chapada Diamantina, State of Bahia, Brazil, is presented. Eight species and five genera were recognized. Mandevilla is the most diverse genus, With four species: M. scabra (Hoffmans. ex Roem. & Schult.) K. Schum., M. tenuifolia (J.C. Mikan) Woodson, M. bahiensis (Woodson) M.F. Sales & Kinoshita-Gouvea, and M.microphylla (Stadelm.) M.F. Sales & Kinoshita-Gouvea, the last two are a new status and a new combination, respectively, whose names are being validly published here. The four remaining genera are each represented by one species: Couma rigida Mull. Arg., Stipecoma peltigera (Stadelm.) Mull Arg,, Temnadenia violacea (Veil.) Miers, and Himatanthus bracteatus (A. DC.) Woodson. Identification key, descriptions, comments and illustrations are presented for every species.
