Recently many exciting advances have been achieved in our understanding of Drosophila meiosis due to combined cytological and genetic approaches. New techniques have permitted the characterization of chromosome position and spindle formation in female meiosis I. The proteins encoded by the nod and ncd genes, two genes known to be needed for the proper partitioning of chromosomes lacking exchange events, have been identified and found to be kinesin-like motors. The effects of mutations in these genes on the spindle and chromosomes, together with the localization of the proteins, have yielded a model for the mechanism of female meiosis I. In male meiosis I, the chromosomal regions responsible for homolog pairing have been resolved to the level of specific DNA sequences. This provides a foundation for elucidating the molecular basis of meiotic pairing. The cytological techniques available in Drosophila also have permitted inroads into the regulation of sister-chromatid segregation. The products of two genes (mei-S332 and ord) essential for sister-chromatid cohesion have been identified recently. Additional advances in understanding Drosophila meiosis are the delineation of a functional centromere by using minichromosome derivatives and the identification of several regulatory genes for the meiotic cell cycle.

It is known that the phospholipids of the brain cells of fish are altered during cold adaptation. In particular, the 1-monounsaturated 2-polyunsaturated phosphatidylethanolamines (PEs) increase 2- to 3-fold upon adaptation to cold. One of the most striking changes is in the 18:1/22:6 species of PE. We determined how this lipid affected the bilayer-to-hexagonal-phase transition temperature of 16:1/16:1 PE. We found that it was more effective in lowering this transition temperature than were other, less unsaturated, PE species. In addition, it was not simply the presence of the 18:1/22:6 acyl chains which caused this effect, since the 18:1/22:6 species of phosphatidylcholine had the opposite effect on this transition temperature. Zwitterionic substances that lower the bilayer-to-hexagonal-phase transition temperature often cause an increase in the activity of protein kinase C (PKC). Indeed, the 18:1/22:6 PE caused an increase in the rate of histone phosphorylation by PKC which was greater than that caused by other, less unsaturated, PEs. The 18:1/22:6 phosphatidylcholine had no effect on this enzyme. The stimulation of the activity of PKC by the 18:1/22:6 PE is a consequence of this lipid's increasing the partitioning of PKC to the membrane.

Arabidopsis COP1 acts inside the nucleus to suppress photomorphogenic cellular development, and light inactivation of COP1 may involve a specific control of its nuclear activity in hypocotyls and cotyledons, but not in roots, of developing seedlings. To understand the molecular mechanisms of COP1 action during light-mediated development, we initiated a screen for Arabidopsis cDNAs encoding proteins which interact directly with COP1 in vitro as a step to identify the cellular components involved. We report here the isolation and characterization of a cDNA clone encoding a protein designated CIP1 (COP1-interactive protein 1). CIP1 is predominantly alpha-helical and most likely involved in coiled-coil formation. It interacts specifically with the putative coiled-coil region of COP1 in vitro. Further, CIP1 is encoded by a single gene in Arabidopsis, and its mRNA and protein levels are not regulated by light. Immunofluorescent labeling of CIP1 in Arabidopsis seedling protoplasts demonstrated that CIP1 is part of, or associated with, a cytoskeletal structure in hypocotyl and cotyledon cells, but not in roots. Our results are consistent with a possible role of CIP1 in mediating light control of COP1 nuclear activity by regulating its nucleocytoplasmic partitioning.

In aerobic organisms, protection against oxidative damage involves the combined action of highly specialized antioxidant enzymes, such as superoxide dismutase (SOD) and catalase. Here we describe the isolation and characterization of another gene in the yeast Saccharomyces cerevisiae that plays a critical role in detoxification of reactive oxygen species. This gene, named ATX1, was originally isolated by its ability to suppress oxygen toxicity in yeast lacking SOD. ATX1 encodes a 8.2-kDa polypeptide exhibiting significant similarity and identity to various bacterial metal transporters. Potential ATX1 homologues were also identified in multicellular eukaryotes, including the plants Arabidopsis thaliana and Oryza sativa and the nematode Caenorhabditis elegans. In yeast cells, ATX1 evidently acts in the transport and/or partitioning of copper, and this role in copper homeostasis appears to be directly relevant to the ATX1 suppression of oxygen toxicity: ATX1 was incapable of compensating for SOD when cells were depleted of exogenous copper. Strains containing a deletion in the chromosomal ATX1 locus were generated. Loss of ATX1 function rendered both mutant and wild-type SOD strains hypersensitive toward paraquat (a generator of superoxide anion) and was also associated with an increased sensitivity toward hydrogen peroxide. Hence, ATX1 protects cells against the toxicity of both superoxide anion and hydrogen peroxide.

A irrigação quando bem manejada, pode minimizar os riscos econômicos da atividade sucroalcooleira, particularmente em safras com presença de instabilidade climática onde a restrição hídrica, promovida pela diminuição no volume de chuvas, pode reduzir a produtividade dos canaviais. Dentre as ferramentas disponíveis para a gestão eficiente da água na agricultura irrigada, a técnica de irrigação sob déficit pode se tornar uma escolha acertada para a cana-de-açúcar, desde que sejam identificadas as fases fenológicas e épocas de cultivo onde a limitação da oferta de água não implique em reduções antieconômicas no rendimento da cultura. Diante disso, a hipótese que norteia essa pesquisa, é a de que existe uma estratégia de irrigação sob déficit, que associada a uma variedade com características específicas, possibilite a expressão de indicadores de produtividade tão satisfatórios quanto os obtidos em condições de irrigação plena. Nessa linha, os objetivos da pesquisa envolveram o estudo da dinâmica foliar, acúmulo e particionamento de biomassa e ainda, índices de produtividade da água para biomassa, açúcar e etanol de 1ª e 2ª geração de oito variedade de cana-de-açúcar, submetidas a diferentes condições de disponibilidade hídrica no solo em dois ciclos de cultivo (cana-planta e cana-soca). A pesquisa foi realizada na Escola Superior de Agricultura \"Luiz de Queiroz\", em Piracicaba/SP, onde foram estudados os dois primeiros ciclos de cultivo da cana-de-açúcar, sendo estes abordados nesta tese como Experimento 1 (cana-planta) e Experimento 2 (cana-soca). O delineamento experimental adotado para ambos os ciclos foi o de blocos casualizados, com três blocos completos. Os tratamentos foram compostos por três fatores em esquema de parcelas sub-subdivididas. Estas parcelas foram formadas por duas plantas (touceiras) alocadas em um vaso com aproximadamente 330 litros de solo. No Experimento 1, foram estudados três fatores, sendo o primeiro e segundo com quatro níveis e o terceiro com oito (4x4x8), totalizando assim 128 tratamentos, sendo eles: quatro níveis de irrigação ao longo do ciclo (125, 100, 75 e 50% da ETc); oito variedades comerciais de cana-de-açúcar e quatro procedimentos de maturação, impostos por meio de variações na intensidade do déficit hídrico aplicado. Para o Experimento 2, substitui-se o fator Maturação por Épocas de Corte, o qual consistiu em colheitas de um quarto do experimento a cada 90 dias. Os resultados encontrados apontaram que a área foliar responde positivamente a maior disponibilidade hídrica no solo, tendo sido verificado uma relação proporcional entre estes. Quanto ao acúmulo de biomassa, verificou-se que para as oito variedades estudadas houve incremento de biomassa a medida em que se aumentou o volume de água disponibilizado às variedades. No tocante ao particionamento, as folhas foram os drenos principais de fotoassimilados da planta até os 100 dias de cultivo, sendo que após este período, os colmos ocuparam o lugar de dreno preferencial. Os indicadores de produtividade da água apresentaram diferenças significativas para o fator lâmina, o que indica a existência de cultivares de cana-de-açúcar mais eficientes no uso da água. Por fim, observou-se que a produtividade da água para etanol total apresentou valores expressivos, com média para essa variável igual a 1,81 L m-3, o que denota o potencial de rendimento de etanol (1G + 2G) a partir da cana-de-açúcar quando é adotado o aproveitamento integral das plantas.

El particionado hardware/software es una tarea fundamental en el co-diseño de sistemas embebidos. En ella se decide, teniendo en cuenta las métricas de diseño, qué componentes se ejecutarán en un procesador de propósito general (software) y cuáles en un hardware específico. En los últimos años se han propuesto diversas soluciones al problema del particionado dirigidas por algoritmos metaheurísticos. Sin embargo, debido a la diversidad de modelos y métricas utilizadas, la elección del algoritmo más apropiado sigue siendo un problema abierto. En este trabajo se presenta una comparación de seis algoritmos metaheurísticos: Búsqueda aleatoria (Random search), Búsqueda tabú (Tabu search), Recocido simulado (Simulated annealing), Escalador de colinas estocástico (Stochastic hill climbing), Algoritmo genético (Genetic algorithm) y Estrategia evolutiva (Evolution strategy). El modelo utilizado en la comparación está dirigido a minimizar el área ocupada y el tiempo de ejecución, las restricciones del modelo son consideradas como penalizaciones para incluir en el espacio de búsqueda otras soluciones. Los resultados muestran que los algoritmos Escalador de colinas estocástico y Estrategia evolutiva son los que mejores resultados obtienen en general, seguidos por el Algoritmo genético.

Saproxylic beetle diversity is high at the Cabañeros National Park (central Spain), where woodland habitats exhibit remarkable heterogeneity. Our aim was to explain the diversity of saproxylic beetles, focusing on species turnover among mature woodland types. We surveyed five woodland types that represented the heterogeneity of the park’s woodland habitats. Beetles were collected using window traps over a period of 20 months. The Jaccard Similarity Index was used as indirect value of beta diversity among woodlands and to test the relation between species turnover and geographical distance. We also identified the contribution of species turnover to landscape diversity by using a partitioning model. Moreover, the presence of mixed woodlands (more than one tree species) allowed us to attempt to valorise the effect of tree species (coupled with their historical management) on species turnover among woodlands. Finally, we looked for different saproxylic beetle preferences for habitat and tree species using an indicator value method. We found that saproxylic beetle species composition varied significantly among the studied woodlands. The variation in species turnover was independent from the distance among woodlands, which suggested that beetle dispersal abilities could not explain this high turnover. Tree species within woodlands were a key factor that increased diversity turnover in woodlands and, consequently, the diversity of the park. Moreover, we found saproxylic beetle species that had different habitat and tree species preferences. We conclude that woodland heterogeneity (highly affected by woodland composition) seems to be the driving force for saproxylic beetle diversity in this protected area.

La partición hardware/software es una etapa clave dentro del proceso de co-diseño de los sistemas embebidos. En esta etapa se decide qué componentes serán implementados como co-procesadores de hardware y qué componentes serán implementados en un procesador de propósito general. La decisión es tomada a partir de la exploración del espacio de diseño, evaluando un conjunto de posibles soluciones para establecer cuál de estas es la que mejor balance logra entre todas las métricas de diseño. Para explorar el espacio de soluciones, la mayoría de las propuestas, utilizan algoritmos metaheurísticos; destacándose los Algoritmos Genéticos, Recocido Simulado. Esta decisión, en muchos casos, no es tomada a partir de análisis comparativos que involucren a varios algoritmos sobre un mismo problema. En este trabajo se presenta la aplicación de los algoritmos: Escalador de Colinas Estocástico y Escalador de Colinas Estocástico con Reinicio, para resolver el problema de la partición hardware/software. Para validar el empleo de estos algoritmos se presenta la aplicación de este algoritmo sobre un caso de estudio, en particular la partición hardware/software de un codificador JPEG. En todos los experimentos es posible apreciar que ambos algoritmos alcanzan soluciones comparables con las obtenidas por los algoritmos utilizados con más frecuencia.

One of the main challenges in biological conservation has been to understand species distribution across space and time. Over the last decades, many diversity and conservation surveys have been conducted that have revealed that habitat heterogeneity acts as a major factor that determines saproxylic assemblages. However, temporal dynamics have been poorly studied, especially in Mediterranean forests. We analyzed saproxylic beetle distribution at inter and intra-annual scales in a “dehesa” ecosystem, which is a traditional Iberian agrosilvopastoral ecosystem that is characterized by the presence of old and scattered trees that dominate the landscape. Significant differences in effective numbers of families/species and species richness were found at the inter-annual scale, but this was not the case for composition. Temperature and relative humidity did not explain these changes which were mainly due to the presence of rare species. At the intra-annual scale, significant differences in the effective numbers of families/species, species richness and composition between seasons were found, and diversity partitioning revealed that season contributed significantly to gamma-diversity. Saproxylic beetle assemblages exhibited a marked seasonality in richness but not in abundance, with two peaks of activity, the highest between May and June, and the second between September and October. This pattern is mainly driven by the seasonality of the climate in the Mediterranean region, which influences ecosystem dynamics and imposes a marked seasonality on insect assemblages. An extended sampling period over different seasons allowed an overview of saproxylic dynamics, and revealed which families/species were restricted to particular seasons. Recognizing that seasons act as a driver in modelling saproxylic beetle assemblages might be a valuable tool in monitoring and for conservation strategies in Mediterranean forests.
